Effects of Baedokhwanbalhyobang (BDHBH) on Immune Modulation in Dermatitis Model of NC/Nga Mice (배독환발효방(排毒丸醱酵方)의 면역조절작용을 통한 항아토피 효능)

  • Herbal medicine has a high body absorption rate when it ferments. Biological and clinical research on the fermented herb gradually increases because it has effective materials for the treatment of a disease and it is a little bitter. In this study, we investigated the effect of fermented Baedokhwanbalhyobang (BDHBH) on attenuation of the development of atopic dermatitis in NC/Nga mice by evaluating the cytokine level in serum, the mRNA expression of cytokine and histological alteration of the skin, and the skin severity. We have come to the following conclusion. BDHBH led to a significant decrease in the skin severity score (63.1%) as compared to the control group. CD4+/CD45+, CD4+, B220+/CD23+, and CD11b+/Gr-1+cells of peripheral mononuclear cells (PBMCs) in the BDHBH-treated group were decreased to 6.7%, 31.1%, 22.4%, 36.6%, respectively. CD3+and CD11b+/Gr-1+immune cells in dorsal skin of the BDHBH-treated group were decreased to 52.9% and 28.0%. The levels of IL-5 and IL-13 in serum of the BDHBH-treated group were inhibited to 18.8% and 5.1%. The mRNA expressions of IL-5 and IL-13 in dorsal skin were also decreased to 30.6% and 27.8% after the treatment of BDHBH. BDHBH inhibited the proliferation and differentiation of eosinophils. In histological examination, BDHBH decreased the thickness of epidermis and dermis, and infilatration of mast cells as compared to the control group. These results indicate that BDHBH inhibits the pathogenic development of atopic dermatitis in NC/Nga mice. These results may indicate that BDHBH attenuates the development of atopic dermatitis-like lesions by lowering immune cells and inflammatory cytokine levels, and that it is valuable in drug development for the treatment of atopic dermatitis. Further experiments on the components of BDHBH will be needed to better understand the effect of a fermented herb as compared to a herb.

Gastritis Associated with Initially Pediatric Crohn's Disease and Ulcerative Colitis

  • Purpose: The aim of this study is to determine the involvement of the upper gastrointestinal system (GIS) in patients diagnosed with Crohn's disease (CD), ulcerative colitis (UC), and non-inflammatory bowel disease (IBD) and to compare their differences. Methods: This study included patients aged between 2 and 18 years who underwent colonoscopy and esophagogastroduodenoscopy (EGD) for the first time due to the prediagnosis of IBD. In EGD, samples were taken from duodenum, antrum, corpus, and esophagus; and gastritis, duodenitis, and esophagitis were identified through histopathologic examination. The data gathered the ends of the research were compared between IBD with non-IBD groups and between CD-UC with non-IBD groups, and the presence of significant differences between groups were determined. Results: In our study, 16 patients were diagnosed with CD, 13 with UC, 3 with undeterminate colitis, and 13 with non-IBD. In the histopathological examination of the groups, GIS involvement was found in 94.1% of patients diagnosed with IBD and in 38.5% of non-IBD patients. Moreover, the difference was found to be statistically significant (p=0.032). No significant difference was found between the CD and UC groups. Gastritis was mostly observed in 93.8% of CD-diagnosed patients, 76.8% of UC-diagnosed patients, 81.2% of IBD-diagnosed patients, and 38.5% of non-IBD-diagnosed patients. On the other hand, significant differences were found between CD and non-IBD groups (p=0.03), UC and non-IBD groups (p=0.047), and IBD and non-IBD groups (p=0.03). Conclusion: The results of the study show that gastritis was highly observed in UC- and CD-diagnosed patients than in non-IBD-diagnosed patients.

The Effect of Ginseng-Saponin on Cd-Induced (인삼사포닌이 카드뮴의 면역독성에 미치는 영향)

  • The purpose of this study is to investigate the effects of Ginseng saponin on the cadmium which is widely distributed in the environment, results in immune system alteration. For the experiments, 125 mice of ICR strain were used. The experimental groups were divided into 5 groups a control, a cadmium alone treatment group, three Cd and saponin (10, 50, 100 mg/kg) combined treatment groups. The mice were allocated 25 to each group and observed for 8 weeks. The results of experiment are as follows: 1. Body weight growth rates during 8 weeks were as this control group 36.47%, Cd alone group 32.48%, saponin combined treatment group (10, 50, 100 mg/kg) 32.49%, 39.17%, 24.27% respectively. 2. In all groups, the relative weights of liver and kidney were increased, compared with control group. In the case of spleen, saponin combined treatment group (50, 100 mg/kg) was high to the significant level compared with a control group (p<0.05). Thymus was not. 3. On blood lymphocyte count observation, Cd alone treament group has 25.6% less than control group, and saponin combined treatment group have increasing trends. but in thymus and spleen, there was no trends like blood. 4. On antibody titer, there was no difference among groups. 5. On total serum protein, saponin (100 mg/kg) combined treatment group was high to significant level compared with control group (p<0.05), and other treatment groups have increasing trends. 6. Cd accumulation in kidney was higher than in liver, and all treatment groups were high to the very significant level compared with the control group (p<0.05), but there was no difference among groups. From the results of this study, it can be concluded that the oral administration of Cd results in alteration of immune system and Ginseng saponin prevents this effect. But, Cd accumulation was not affected by saponin.

Effects of Chitosan on Cadmium Contents of Blood and Tissues in Rats Exposed to Various Levels of Cadmium (카드뮴 수준별 투여 흰쥐의 혈액과 조직의 카드뮴 함량과 키토산 섭취 효과)

  • This study investigated the effects of chitosan on cadmium(Cd) toxicity and mineral metabolism in rats exposed to cadmium by oral administration. Six week-old Sprague-Dawley rats were divided into eight groups. Four groups were fed AIN-93G based 3% ${\alpha}$-cellulose diets while the others were fed 3% chitosan diets for four weeks with oral administration of 0, 0.5, 1.0, 2.0 mg Cd/2ml distilled water three times a week, respectively. Cd contents in the serum, liver, kidney, testis and bone, and the excretion of cadmium in feces were determined. There was no significant difference in weight gain and food intake among groups. Cadmium contents in the serum, liver, kidney, testis, femur and lumbar were significantly increased in proportion to the administration level of Cd (p<0.05). A protective effect of chitosan on cadmium toxicity in tissue was shown only in the high level cadmium-intake group. The fecal excretion, absorption of Cd were increased by the administration levels of cadmium. These results suggest that Cd administration may facilitate the accumulation of Cd in the blood and tissue in proportion to the amount of administration, and also, that chitosan may be effective in lowering the accumulation of cadmium.

Studies on the Immunomodulatory Effect Using Combinations of Several Kinds of Anti-cancer Prescriptions (수종 항암 처방의 조합에 따른 면역조절 작용에 관한 연구)

  • In order to understand the anti-carcinogenic effects of Boo-jung-bae-bon-bang(扶正培本方)-B1), Hwal-hyul-hwa-eo-bang(活血化瘀方-B2), Cheong-youl-hae-dok-bang(淸熱解毒方-B3), prescriptions of individual B1, B2, and B3 treatments or combined treatments (B4; B1+B2, B5; B1+B3, B6; B1+B2+83, B7; B2+83) were applied to cultured cancer cell lines. The major findings on their anti-cardnogenic effects in terms of mechanism and synergistic interactions are summarized below. Results of cytokine gene expression analyses are summarized as follows; IL-2 gene expression was increased by B1 and B6 treatments, IFN-v gene expression increased by B3, B1, B6, and 85, and CD28 gene expression increased by B1 and B5. IL-4 expression was not affected by any treatments. In the FACS analysis, increases in numbers of immunoreactive CD3/sup +//CD25/sup +/ T cells by B1 and B5 treatment, CD3/sup +//CD69/sup +/ T cells by B1, B3, B5, and B6 treatments, CD19/sup +//CD44/sup +/ B cells by B1 and B6 treatments were observed when compared to the corresponding non-treated control groups.

The Experimental Effect of Haedongpibokhap-bang (Hǎitóngpífùhé-fāng) on Collagen-induced Arthritis (해동피복합방(海桐皮複合方)이 Collagen II 유발 관절염에 미치는 실험적 연구)

  • Objectives : The aim of this study was to know the immunity response of Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) to rheumatoid arthritis in collagen-induced arthritis(CIA) mice. Methods : For this purpose, Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) was orally administerd to mice with arthritis induced by collagen II and then value of immunocyte in spleen, draining lymph node and paw joint and cytokine(IL-6, $TNF-{\alpha}$), rheumatoid factor (IgG and IgM) in serum were measured. Results : 1. The arthritis index was significantly decreased. 2. In total cell counts of spleen, DLN and paw joint, the cells in spleen decreased while there was a significant increase in DLN and significant decrease in paw joint. 3. In lymph nodes, CD3+, CD3+/CD69+, CD4+, CD8+ cells increased significantly. 4. In joints, CD3+ and CD11+b/Gr-1+ cells decreased significantly. 5. Serum IL-6 and $TNF-{\alpha}$ were decreased significantly. 6. Production of serum IgG and IgM decreased significantly. Conclusions : The results present that Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) controls abnormal activity of immune system, inhibitig collagen-induced arthritis(CIA).

Biogeochemistry of Metal and Nonmetal Elements in the Surface Sediment of the Gamak Bay (가막간 표층퇴적물 중의 금속 및 비금속 원소의 생지화확적 분포특성)

  • In order to evaluate die geochemical characteristics of sediment in a semi-enclosed bay used as shellfish and fish farming area, the concentrations of metallic(V, Cr, Mn, Fe, Co. Ni, Cu, Zn, Ag, Cd, Hg, Ph, As) and non-metallic(P, Se) elements and uranium were measured in the surface sediment samples collected from 19 stations of Gamak Bay in April 2010. Metal contamination status in the sediments were also evaluated using the sediment quality guidelines(SQGs) proposed by the National Oceanic and Atmospheric Administration(NOAA) and the enrichment factor(EF). The concentrations of elements in sediment were mainly controlled by quartz-dilution effect(V, Cr, Fe, Co and Ni), the dilution effect of organic matter(Cd and U), and metal redistribution by the decomposition of organic matter(Mn, Ag, As, and Se). The concentrations of metals, except As and Ni, in sediments from all sampling stations were lower than ERL values of NOAA. Conclusively, the surface sediment of Gamak Bay was slightly polluted with Ni, Ag, Cd, and Cd but was not polluted with other elements on the basis of EF results. Our results suggest that the surface sediment in Gamak Bay is not polluted by metallic elements.

Cadmium and Lead Levels of Loons Wintering in Korea (한국에 도래하는 아비류의 카드뮴과 납 축적 레벨)

  • We investigated Cd and Pb accumulation levels in tissues of 25 individuals of three species of loons wintering in coastal areas of Busan, Korea. Their liver, muscle, kidney and bone tissues were analyzed for Cd and Pb concentrations. Significant difference of Cd and Pb concentrations was not found among the three species (p>0.05). Individual Cd values ranged from 0.02 to $18.3{\mu}g/g$. Cd concentrations were highest in kidney and lowest in bone. Levels (3.45$\sim$6.34 ${\mu}g/g$) of all the analyzed liver Cd were similar in comparison with those (ND$\sim$7.11${\mu}g/g$) reported for the loons in different areas. All but one (Gavia arctica) of kidney Cd levels were lower than the exposed level (>30 ${\mu}g/g$). Individual Pb values ranged from 0.03 to 1.49 ${\mu}g/g$. Pb concentrations were highest in bone and lowest in muscle. Liver Pb levels in this study were similar to those of normal loons reported from different countries. Based on hepatic Pb levels, almost all of the samples were classified as background level (0.15$\sim$3.0 ${\mu}g/g$).

Conformation of Single Stranded Poly(dA) and Its Interaction with 4',6-Diamidino-2-phenylindole

  • We studied the interaction of 4',6-diamidino-2-phenylindole (DAPI) with single stranded poly(dA) using optical spectroscopic methods, including absorption, circular dichroism (CD), and fluorescence spectroscopy. The temperature-dependent conformation of poly(dA) was also investigated. The conformation of poly(dA) varied with temperature, which is explained by the stacking-destacking process of the adenine bases, resulting from the sugar conformation. The hypochromicity and red-shift in the absorption spectroscopy, the lack of CD change in the drag absorption region, and the fluorescence behavior, especially a great accessibility of the I2 quencher to the poly(dA)-bound DAPI, suggest that DAPI binds to the outside of poly(dA). The Job plot for the DAPI-poly(dA) mixture demonstrated that a stoichiometry of one DAPI molecule binds to the one phosphate of poly(dA).

Changes in Immunogenicity of Preserved Aortic Allograft (보존된 동종동맥편 조직의 면역성 변화에 관한 연구)

  • The causes of degenerative changes in allograft cardiac valves are not well known to this day. Today's preserved allografts possess highly viable endothelial cells and degeneration of allografts can be facilitated by immune reaction which may be mediated by these viable cells. To test the antigenicity of endothelial cells, pieces from aortic wall were obtained from fresh and cryo-preserved rat allograft. Timings of sampling were prior to sterilization, after sterilization, after 1, 2, 7, 14 days of fresh preservation and cryopreservation. Endothelial cells were tested by immunohistochemical methods using monoclonal antibodies to MHC class I(MRC OX-18), class II(MRC OX-6) and ICAM-1 antigens. After transplantation of each group of aortic allograft at the subcutaneous layers of rats, population of CD4$^{+}$ T cell and CD8$^{+}$ T cell were analyzed with monoclonal antibodies after 1, 2, 3, 4, 6 and 8 weeks. MHC class I expression was 23.95% before preservation and increased to 35.53~48.08% after preservation(p=0.0183). MHC Class II expression was 9.72% before preservation and 10.13~13.39% after preservation(P=0.1599). ICAM-1 expression was 15.02% before preservation and increased to 19.85~35.33% after preservation(P=0.001). The proportion of CD4$^{+}$ T-cell was 42.13% before transplantation. And this was 49.23~36.8% after transplantation in No treat group (p=0.955), decreased to 29.56~32.80% in other group(p=0.0001~0.008). In all the groups, the proportion of CD8$^{+}$ T-cell increased from 25.57% before transplantation to 42.32~58.92% after transplantation(p=0.000l~0.0002). The CD4$^{+}$/CD8$^{+}$ ratio decreased from 1.22~2.28 at first week to 0.47~0.95 at eighth week(p=0.0001). The results revealed that the expression of MHC class I and ICAM-1 in aortic allograft endothelium were increased but that of MHC class II were not changed, despite the different method of preservation. During 8 weeks after transplantation of aortic allograft, the subpopulations of CD4$^{+}$ T cell were not changed or only slightly decreased but those of CD8$^{+}$ T cell were progressively increased.ely increased.
