전산화단층사진을 이용한 하악골 3차원 영상에서 비대칭진단 계측항목의 재현도에 관한 연구 (Reproducibility of asymmetry measurements of the mandible in three-dimensional CT imaging)

  • 본 연구는 하악골 3차원 입체영상에서 비대칭계측항목들의 재현도 비교를 통하여 안면비대칭 진단 시 유용한 계측점 설정에 도움이 되고자 시행되었다. 40명의 두경부 전산화단층사진을 이용하여 생성한 3차원 영상에서 하악골 형태를 결정짓는 부위로 하악과두, 하악우각부, 하악정중부를 선택하고 각 부위의 세부위치에 따라, 그리고 측정 시 보는 각도에 따라 구분하여 하악과두의 경우 $Cd_{sup}_{-}_{sup}$ 등 6개, 하악우각부의 경우 $Go_{inf}_{-}_{lat}$ 등 10개, 하악정중부의 경우 $Me_{-}_{ant},\;Me_{-}_{inf}$ 등 총 18개의 계측점을 설정한 후 이를 이용하여 하악골 비대칭 시 좌우 차이를 보일 수 있는 25개의 계측항목을 설정 및 계측한 후 조사자간 및 조사자내의 재현도를 비교 평가하였다. 조사자간 재현도의 경우 25개의 계측항목 중 3개를 제외한 모든 계측항목에서, 조사자내 재현도의 경우 2개 항목을 제외한 모든 계측항목에서 높은 재현도를 보였다. 아울러 본 연구에서 설정한 18개의 계측점 중 $Go_{mid}_{-}_{lat},\;Go_{mid}_{-}_{obl}$가 포함된 계측항목이 조사자간 및 조사자내 재현도가 떨어지는 경향을 보였다. 이상의 면구 결과 본 연구에서 사용된 18개의 계측점 중 16개의 계측점 (과두를 나타내는 $Cd_{sup}_{-}_{sup},\;Cd_{lat}_{-}_{ant},\;Cd_{lat}_{-}_{lat},\;Cd_{post}_{-}_{lat},\;Cd_{post}_{-}_{post},\;S$, 하악우각부를 나타내는 $Go_{int}_{-}_{lat},\;Go_{inf}_{-}_{inf},\;Go_{lat}_{-}_{ant},\;Go_{lat}_{-}_{lat},\;Go_{post}_{-}_{lat},\;Go_{post}_{-}_{post},\;Ag_{-}_{lat},\;Ag_{-}_{inf}$, 하악정중부를 나타내는 $Me_{-}_{ant},\;M_{-}_{inf}$)은 높은 재현도를 보이므로 전산화단층사진을 이용한 하악골 3차원 영상에서 안면비대칭 환자의 진단에 유용하게 사용될 수 있음을 시사하였다.

Prognostic significance of minimal residual disease detected by a simplified flow cytometric assay during remission induction chemotherapy in children with acute lymphoblastic leukemia

  • Purpose: Our study attempted to determine the prognostic significance of minimal residual disease (MRD) detected by a simplified flow cytometric assay during induction chemotherapy in children with B-cell acute lymphoblastic leukemia (B-ALL). Methods: A total of 98 patients were newly diagnosed with precursor B-ALL from June 2004 to December 2008 at the Asan Medical Center (Seoul, Korea). Of those, 37 were eligible for flow cytometric MRD study analysis on day 14 of their induction treatment. The flow cytometric MRD assay was based on the expression intensity of CD19/CD10/CD34 or aberrant expression of myeloid antigens by bone marrow nucleated cells. Results: Thirty-five patients (94.6%) had CD19-positive leukemic cells that also expressed CD10 and/or CD34, and 18 (48.6%) had leukemic cells with aberrant expression of myeloid antigens. Seven patients with ${\geq}1%$ leukemic cells on day 14 had a significantly lower relapse-free survival (RFS) compared to the 30 patients with lower levels (42.9 % [18.7%] vs. 92.0% [5.4%], $P$=0.004). Stratification into 3 MRD groups (${\geq}1%$, 0.1-1%, and <0.1%) also showed a statistically significant difference in RFS (42.9% [18.7%] vs. 86.9% [8.7%] vs. 100%, $P$=0.013). However, the MRD status had no significant influence on overall survival. Multivariate analysis demonstrated that the MRD level on day 14 was an independent prognostic factor with borderline significance. Conclusion: An MRD assay using simplified flow cytometry during induction chemotherapy may help to identify patients with B-ALL who have an excellent outcome and patients who are at higher risk for relapse.

Factors to Predict Successful Harvest during Autologous Peripheral Hematopoietic Stem Cell Collection

  • Autologous peripheral blood stem cell transplantation (PBSCT) has been used as a major treatment strategy for hematological malignancies. The number of CD34 positive cells in the harvested product is a very important factor for achieving successful transplantation. We studied the factors that can predict the number of CD34 positive cells in the harvested product of acute myelocytic leukemia (AML), multiple myeloma (MM) and Non-Hodgkin's lymphoma (NHL) patients after mobilizing them with chemotherapy plus G-CSF. A total of 73 patients (AML 19 patients, MM 28 patients, NHL 26 patients) with hematological malignancies had been mobilized with chemotherapy and granulocyte colony-stimulating growth factor from April, 2000 to February, 2012. Group's characteristics, checkup opinion of pre-peripheral blood on the day of harvest & outcome of PBSC were analyzed and evaluated using SPSS statistics program after grouping patients as below; group 1: CD34 cell counts < $2{\times}10^6/kg$ (n=16); group 2: $2{\times}10^6/kg{\leq}CD34$ cell counts < $6{\times}10^6/kg$ (n=32); group 3: CD34 cell counts ${\geq}6{\times}10^6/kg$ (n=25). We analyzed the clinical characteristics, the peripheral blood (PB) parameters and the number of CD34 positive cells in the PB and their correlation with the yield of CD34 positive cells collected from the mobilized patients. The total number of leukapheresis sessions was 263 (mean: 3.55 session per patient), and the mean number of harvested CD34 positive cells per patient was $7.37{\times}10^6/kg$. The number of CD34 positive cells in product was significantly correlated with the number of platelet and CD34 positive cells in peripheral blood (P<0.05). The number of PB CD34 positive cells was the best significant factor for the quantity of harvested CD34 positive cells on the linear regression analysis (P<0.05). Many factors could influence the mobilization of peripheral blood stem cells. Platelet count and PB CD34 positive cells count were the two variables which remained to be significant in multivariate analysis. Therefore, the number of platelet and CD34 positive cells in peripheral blood on the day of harvest can be used as an accurate predictor for successful peripheral blood stem cell collection.

CdS/Titania-나노튜브 복합 막을 이용한 광촉매적 수소제조 (CdS-Titania-Nanotube Composite Films for Photocatalytic Hydrogen Production)

  • 알칼리 수열합성법에 의해 높은 비표면적을 갖는 티타니아 나노튜브(TiNT)를 합성하였다. 가시광용 광촉매로서의 응용성을 조사하기 위해 CdS 나노입자와 조성(r=TiNT/(CdS+TiNT))을 바꿔가며 일련의 무기 복합필름을 제조하였다. 또한 비교를 위해 티타니아 나노튜브 대신 티타니아 나노입자와 CdS로 구성된 복합체를 역시 제조하였다. 합성된 티타니아 나노튜브는 $200^{\circ}C$ 이상의 소결온도에서 부분적으로 튜브 구조의 붕괴가 시작되었지만, CdS와의 복합체는 $450^{\circ}C$ 까지도 비교적 안정한 구조를 유지하였다. (CdS+TiNT)복합필름은 티타니아 나노입자 복합계와 비교할 때 가시광 흡수 측면에서는 유사한 정도를 나타내었지만, 수소생산 활성과 광전류 발생은 오히려 훨씬 낮은 값을 나타내었다. 결과적으로, 티타니아 나노튜브는 그의 높은 비표면적에도 불구하고 자기들끼리의 응집성이 강하여 CdS와의 전기적 상호작용이 약하며, 특히 얇은 튜브벽 두께(${\sim}3nm$)와 낮은 결정성에 기인하는 약한 광전류 특성은 이의 광촉매로서의 응용성을 상당히 제한하는 요소로 나타났다.

  • IL-18 has been found to have multiple effects upon various cells involved in tumor immunology. Here, we discuss opposite effects of IL-18 in tumor immunology. IL-18 has been shown that it has significant anti-tumor effects, which are mediated by T cells and NK cells, in a manner similar to IL-12. First, we investigated the evaluation of the effects of the systemic administration of IL-18 in combination with B7-1 (CD80) against murine B16 melanoma in vivo. (omitted)

경남지역 논토양의 중금속함량 조사 (Investigation of Heavy Metal Concentrations in Paddy Soils of Gyeongnam Province)

  • 경남지역 논 토양의 중금속 관리를 위한 기초 자료를 제공하고자 260 지점을 대상으로 2011년에 Cd, Cu, Ni, Pb, Zn 및 As 전함량과 0.1N HCl 가용성 Cr 함량을 분석하였다. 논토양의 중금속 평균 함량은 Cd 0.143 (범위 0.003-0.537) mg $kg^{-1}$, Cr 0.322 (0.002-0.986) mg $kg^{-1}$, Cu 25.41 (6.03-76.19) mg $kg^{-1}$, Ni 16.36 (2.63-32.94) mg $kg^{-1}$, Pb 18.67 (4.16-87.02) mg $kg^{-1}$, Zn 71.76 (22.99-153.58) mg $kg^{-1}$, As 3.516 (0.002-19.481) mg $kg^{-1}$ 였다. 논토양에서 Cd 함량은 산록경사지에서 가장 높은 반면 Ni과 As 함량은 홍적대지, Zn 함량은 해안평탄지에서 가장 높았다. 또한, Cd, Cu 및 Zn 함량은 미사질식양토에서 미사질양토, 사양토 및 양토 보다 높은 경향이었다.

둥굴레 근경의 증자 및 볶음조건에 따른 중금속(Pb, Cd) 제거 특성 (Elimination of Heavy Metals(Pb, Cd) by Steaming and Roasting Conditions of Polygonatum odoratum Roots)

  • 둥굴레 근경의 증자 및 열처리 중 수용성 고형분 및 Pb, Cd 제거율의 변화를 확인하기 위하여 반응표면분석(RSM)을 활용하였다. 중심합성계획법에 따라 증자시간($60{\sim}180$ min), 볶음온도($110{\sim}150^{\circ}C$) 및 볶음시간($10{\sim}50$ min)을 달리하여 16구간으로 제조된 시료의 수용성 고형분 함량, Pb 및 Cd 제거율을 측정하고 회귀분석을 실시하였다. 예측된 가용성 고형분의 최대값은 $71.47\%$ 이었으며, 이때의 증자 및 볶음조건은 증자시간 65.24분, 볶음온도 $126.93^{\circ}C$ 및 볶음시간 37.58분 이었다. 둥굴레 시료의 Pb 제거율은 증자시간 71.23분, 볶음온도 $119.81^{\circ}C$ 및 볶음시간 24.35분일 때 최대값 $18.87\%$이었으며, Cd 제거율은 증자시간 160.89분, 볶음온도 $126.43^{\circ}C$ 및 볶음시간 15.81분일 때 최대값 $50.85\%$로 예측되었다. 중금속 제거율을 고려한 둥굴레차의 품질특성을 극대화시킬 수 있는 증자 및 볶음조건은 증자시간 $165{\sim}180$분, 볶음온도 $120{\sim}135^{\circ}C$ 및 볶음시간 $30{\sim}45$분 범위로 예측되었다. 이상의 예측범위 내의 임의의 점에서 실제 실험한 실험치는 반응표면분석법에 의해 예측된 값과 유사한 경향을 보여 도출된 회귀식의 신뢰성을 검증할 수 있었다.

울산-온산연안 표층퇴적물 내 미량금속 오염도 및 생태위해성 평가 (Pollution and Ecological Risk Assessment of Trace Metals in Surface Sediments of the Ulsan-Onsan Coast)

  • 완전산분해법과 단일시약(1 M HCl) 추출법을 이용하여 울산-온산연안 표층퇴적물 내 미량금속의 오염도 및 생태위해성을 평가하였다. 표층퇴적물 내 미량금속 중 Cu, Cd, Pb, Zn, Hg의 농도는 울산연안에 비해 온산연안에서 2배 이상 높게 나타났다. 단일시약 추출 분석 결과, Cd과 Pb은 총함량 중 가용성부분의 평균 함량이 각각 72%, 78%로 인위적 기원에 의해서 주로 공급되는 것으로 나타난 반면, Cr, Li, Ni, As는 잔류성부분에서 평균 80% 이상 존재하여 주로 자연적 기원의 영향을 받는 것으로 나타났다. 국내 해저퇴적물 해양환경기준과 비교한 결과, Cu, Pb, Zn, Hg의 총농도는 온산연안 일부 정점에서 관리기준을 초과하였으며, Cr, Ni은 모든 정점에서 주의기준 이하였다. 미량금속의 오염도 및 생태위해성을 다양한 지수(EF, $I_{geo}$, m-PEL-Q, ERI)로 평가한 결과, 울산-온산연안에서 미량금속 중 Cu, Cd, Pb, Zn, Hg의 오염도가 높았으며, 생태계에 잠재적 위해성은 Cd과 Hg이 가장 큰 것으로 나타났다.

단기배양한 중피세포의 면역세포화학적 연구 (Immunocytochemical Characteristics of the Short-term Cultured Mesothelial Cells)

  • Reactive humsn mesothelial cells were examined by immunocytochemical stain with intermediate filaments (cytokeratin [CK1, CK7, CK8, CK18, CD19), vimentin, desmin, actin), epithelial membrane antigen, carcinoembryonic antigen (CEA), MHC class II antigen (HLA-DR), LeuM-1 (CD15), $\alpha1-antitrypsin$(ACT), $\alpha1-antichymotrypsin$ (ACHT), CD68(KP-1) and FcyRIII(CD16). The mesothelial cells were isolated from patients with liver cirrhosis and pleural effusion, and short-term cultured in RPMI 1640 media containing 10% heat inactivated fetal calf serum and 1% identical supernatant fluid of the patients' transudates. The results obtained are as follows 1. The cultured-reactive mesothelial cells were positive for the protein of cytoskeleton such as cytokeratin and vimentin, but negative for desmin and actin. The resting mesothelial cells showed positive reactions for cylokeratin, but negative for vimentin, desmin and actin. 2. The primary antibodies to the cytokeratin were strongly reactive for CK1, CK8 and CK18 but negative for CK7 and CK19 in both reactive and resting mesothelial cells. 3. Resting mesothelial cells showed negative reactions for CEA, but strong positive reactions in cultured-reactive mesothelial cells. 4. The markers for the monocytes/histiocytes(CD11b, CD14, CD16, CD68, Iysozyme and $\alpha1-antitrypsin$ and $\alpha1-antichymotrypsin$) were nonreactive in resting mesothelial cells, but lysozyme and $\alpha1-antitrypsin$ were weakly reactive in reactive and proliferative mesothelial cells. 5. MHC Class II molecule(HLA-DR antigen) was negative in both resting and reactive mesothelial cells. These results suggest that the short-term cultured, reactive mesothelial cells show a newly aberrant expression of the vimentin and calcine-embryonic antigen. The reason of the aberrant expression of the intermediate filament and oncofetal antigen in reactive and proliferative mesothelial cells should be further evaluated.

Synergistic Effect of Interleukin-18 on the Expression of Lipopolysaccharide-Induced IP-10 (CXCL-10) mRNA in Mouse Peritoneal Macrophages

  • Interleukin (IL)-18, a member of the family of IL-l cytokine, is one of the principal inducers of $interferon-{\gamma}(IFN-{\gamma})$ in T lymphocytes and natural killer cells. The objective of the present study was to evaluate the effect of IL-18 on the expression of chemokine IP-10 (CXCL-10) mRNA in mouse peritoneal macrophages. IL-18 had very weak direct effect or synergistic effect with IL-12 on the expression of IP-10 mRNA in C57BL/6 mouse peritoneal macrophages. However, IL-18 pretreatment was found to playa cooperative role in the expression of lipopolysaccharide (LPS)-induced IP-10 mRNA. For the expression of LPS-induced IP-10 mRNA, the synergistic effect was detected after 16 h of IL-18 pretreatment prior to LPS stimulation. The expression level of CD14 in cells stimulated with LPS was not changed by IL-18 pretreatment, and the level of $IFN-{\gamma}$ production during IL-18 pretreatment plus LPS stimulation was barely discernible ($0.36{\pm}0.31pg/ml$). Namely, the synergistic effect of IL-18 pretreatment was not related to a change of LPS receptor, CD14 expression, and the production of $IFN-{\gamma}$ by the interaction between IL-18 and LPS. The synergistic effect of IL-18 pretreatment on the expression of LPS-induced IP-10 was related to not NF-kB but AP-1 activation, and associated with the extracellular signal-regulated kinase (ERK) pathway, one of the mitogen-activated protein kinase signaling pathways. These results provide useful information that may elucidate the mechanisms underlying the effect of IL-18 on the expression of IP-10 mRNA.
