Adhesion of CD40-stimulated Germinal Center B Cells to HK Cells Employs the CD11a/CD18-CD54 Interactions

  • Background: The molecular basis of follicular dendritic cells (FDC)-germinal center (GC) B cell interaction is largely unknown, although this cellular interaction is thought to be important for the whole process of GC B cell differentiation. Methods: Using FDC-like cells, HK, and highly purified GC B cells, we attempted to identify the molecules that play critical roles in the interactions between FDC and B cells. GC B cells were co-cultured with HK cells and soluble CD154 in the presence or absence of various function-blocking monoclonal antibodies to examine their effect on GC B cell binding to HK cells and B cell proliferation. Results: Anti-CD11a and anti-CD54 antibodies inhibited GC B cell binding to HK cells while anti-CD49d and anti-CD106 antibodies did not. GC B cell proliferation was not impaired by the disruption of GC B cell-HK cell adherence. Conclusion: Our results suggest that CD11a/CD18-CD54 interactions play an important roles in the initial binding of GC B cells to FDC and diffusible growth factors from FDC may be responsible the massive proliferation of GC B cells.

Heavy Metal Contents in Ginseng and Ginseng Products (인삼 및 인삼제품류의 중금속 함량)

  • Contents of heavy metals, mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), manganese (Mn), zinc (Zn), and copper (Cu), In ginsengs and ginsengs products their safety were evaluated using a mercury analyzer, atomic absorption spectrophotometer and Inductively coupled plasma spectrometer. Contents of heavy metals in ginsengs were [min-max(mean), mg/kg]; Hg 0.002-0.02(0.007), Pb 0.018-0.39(0.106), Cd 0.004-0.413(0.106), As ND-0.094(0.013), Cu 0.24-5.5(3.2), Mn 3.18-50.4(20.09), Zn 0.76-44.27(13.42). Ginseng products values were; Hg 0.0001-0.002(0.001), Pb 0.001-0.133(0.017), Cd ND-0.07(0.004) As ND-0.181(0.008), Cu ND-1.1(0.13), Mn 0.73-30.15(1.61). Zn 0.02-13.42(1.02), similar to those reported by other countries. Average weekly Intakes of Hg, Pb and Cd from ginseng and ginseng products were 0,003, 0.01 and 0,02% of provisional tolerable weekly intake established by FAO/WHO, respectively. Our result could be utilized as important references to establish the standard of lead in ginseng and ginseng products.

Photocurrent study on the splitting of the valence band and growth of $CdGa_2Se_4$ single crystal thin film by hot wall epitaxy (Hot Wall epitaxy(HWE)법에 의한 $CdGa_2Se_4$ 단결정 박막의 성장과 가전자대 갈라짐에 대한 광전류 연구)

  • Single crystal $CdGa_2Se_4$ layers were grown on a thoroughly etched semi-insulating GaAs(100) substrate at $420^{\circ}C$ with the hot wall epitaxy(HWE) system by evaporating the polycrystal source of $CdGa_2Se_4$ at $630^{\circ}C$. The crystalline structure of the single crystal thin films was investigated by the photoluminescence and double crystal X-ray diffraction(DCXD). The carrier density and mobility of single crystal $CdGa_2Se_4$ thin films measured with Hall effect by van der Pauw method are $8.27{\times}10^{17}cm^{-3},\;345cm^2/V{\cdot}s$ at 293 K, respectively. The photocurrent and the absorption spectra of $CdGa_2Se_4/SI$(Semi-Insulated) GaAs(100) are measured ranging from 293 K to 10 K. The temperature dependence of the energy band gap of the $CdGa_2Se_4$ obtained from the absorption spectra was well described by the Varshni's relation $E_g(T)=2.6400eV-(7.721{\times}10^{-4}eV/K)T^2/(T+399K)$. Using the photocurrent spectra and the Hopfield quasicubic model, the crystal field energy(${\Delta}cr$) and the spin-orbit splitting energy(${\Delta}so$) far the valence band of the $CdGa_2Se_4$ have been estimated to be 106.5 meV and 418.9 meV at 10 K, respectively. The three photocurrent peaks observed at 10 K are ascribed to the $A_{1^-},\;B_{1^-},\;and\;C_{11}-exciton$ peaks.

Heavy Metals in Sediments and Organisms from Tidal Flats along the Mokpo Coastal Area (목포연안 갯벌 및 서식생물에서의 중금속 함량)

  • Total and extractable contents of heavy metals were measured in sediment and seafood from Bukhang and Boggil-ri tidal flats along the Mokpo coastal area, south-western part of Korean peninsular in order to assess the degree of metal pollution, metal bioavailability and metal hioaccumulation. The metal concentrations, except Pb were found to be greater than the background concentrations of sediments indicating the progress of cumulative contamination by anthropogenic origin of metals. The order of extractable metal concentrations in sediments were Mn(32-53 mg/kg)>Zn(14-42 mg/tg)>Cu(2.5-17.0 mg/kg)>Pb(2.4-6.8 mg/kg)>Cd(0.5-0.7 mg/kg). However, the amount of metals associated with extractable fraction of sediments were significantly high in Bukhang relative to Boggil-ri. Significant bioaccumulation of all metals, except Pb were observed in seaweed and benthos. The order of bioaccumulation of metals were: in concentration; all biota commonly, Mn(129-374 mg/kg)>Zn(19-106 mg/kg)>Cu(6-87 mg/kg)>Cd(4.6-7.6 mg/kg)>Pb(0.2-3.7 mg/kg), in BCF; Enteromorpha, Cd>Mn>Cu>Zn>Pb, Ilyoplax deschampsi, Cu>Cd>Mn>Zn>Pb, Urechis unicinctus, Cd>Zn>Mn>Cu>Pb. Some metal concentrations of Enteromorpha, especially Cu, Zn, weakly Pb in bukhang, Mn, Cd in boggili-ri, were correlate well with concentrations in sediment, indicating relatively more contaminated by the anthropogenic origin of metals in each tidal flat. The results clearly indicate that the seafood of bukhang, even of Boggili-ri known as clean area, are contaminated with metals.

  • Cadmium telluride (CdTe) films have been prepared on Corning 7059 glass, molybdenum (Mo), and polyimide (PI) substrates by r.f. magnetron sputtering technique. The influence of the sputter pressure on the structural and optical properties of these films was evaluated. In addition, a comparison of the properties of the films deposited on fferent substrates was performed.

Growth and Characterization of $CdGa_2Se_4$ Single Crystal Thin Films by Hot Wall Epitaxy (Hot Wall Epitaxy (HWE)에 의한 $CdGa_2Se_4$ 단결정 박막 성장과 특성)

  • The stochiometric mix of evaporating materials for the $CdGa_2Se_4$ single crystal thin films was prepared from horizontal furnace. To obtain the single crystal thin films, $CdGa_2Se_4$ mixed crystal was deposited on thoroughly etched semi-insulating GaAs(100) substrate by the Hot Wall Epitaxy (HWE) system. The source and substrate temperature were $630^{\circ}C$ and $420^{\circ}C$, respectively. The crystalline structure of single crystal thin films was investigated by the photoluminescence and double crystal X-ray diffraction (DCXD). The carrier density and mobility of $CdGa_2Se_4$ single crystal thin films measured from Hall effect by van der Pauw method are $8.27{\times}10^{17}cm^{-3}$, $345\;cm^2/V{\cdot}s$ at 293 K, respectively. From the photocurrent spectrum by illumination of perpendicular light on the c-axis of the $CuInSe_2$ single crystal thin film, we have found that the values of spin orbit splitting ${\Delta}So$ and the crystal field splitting ${\Delta}Cr$ were 106.5 meV and 418.9 meV at 10 K, respectively. From the photoluminescence measurement on $CdGa_2Se_4$ single crystal thin film, we observed free excition ($E_x$) existing only high quality crystal and neutral bound exiciton ($D^{\circ}$, X) having very strong peak intensity. Then, the full-width-at -half-maximum(FWHM) and binding energy of neutral donor bound excition were 8 meV and 13.7 meV, respectivity. By Haynes rule, an activation energy of impurity was 137 meV.

Isolation and Characterization of Cells from Human Adipose Tissue Developing into Osteoblast and Adipocyte (인간 지방조직에서 분리된 줄기세포의 표면항원 및 다분화능 확인)

  • Bone marrow derived mesenchymal stem cells (BMSCs) are largely studied for their potential clinical use. But it is hard to get enough number of those cells for clinical trials and give serious pain to the patients. Adipose tissue is derived from the embryonic mesenchyme and contains a stroma that is easily isolated with large amount. This cell population (adipose derived stem cells: ADSCs) can be isolated from human lipoaspirates and like MSCs, differentiate toward the osteogenic, adipogenic, myogenic and chondrogenic lineages. To confirm whether adipose tissue contains stem cells, the ADSCs extracted from omental or subcutaneous fat tissue were expanded during third to fifth passages. The phenotype of the ADSCs was identified by the conventional cell surface markers using flow cytometry: positive for CD29 and CD44, but negative for CD34, CD45, CD117 and HLA-DR that similar to those observed on BMSCs. The ADSCs were able to differentiate into the osteoblast or adipocytes with induction media. Finally, ADACs expressed multiple CD marker antigens similar to those observed on BMSCs and differentiated into osteoblast, adipocyte. With this, human adipotissue contains multipotent cells and may represent an alternative stem cell source to bone marrow-derived MSCs.

Characterization of Human Thigh Adipose-derived Stem Cells (사람의 허벅지지방유래 줄기세포의 특성 분석)

  • Human adipose stem cells are an abundant, readily available population of multipotent progenitor cells that reside in adipose tissue and these cells have characteristics very similar to bone marrow mesenchymal stromal cells (BMMSCs). However, liposuction procedure, donor age, body mass index, and harvesting sites might generate differences in the initial cell population and the preparations are a heterogeneous mixture of precursors with different subsets. Therefore, in this study, we investigated the characteristics of human thigh adipose stem cells and the differentiation potential into mesodermal and endodermal lineage. Thigh adipose stem cells maintained fibroblast-like morphology similar to BM-MSCs and they underwent average 56.5 doublings and produced $5{\times}10^{22}$ cells. These cells expressed SCF, Oct4, nanog, vimentin, CK18, FGF5, NCAM, Pax6, BMP4, HNF4a, nestin, GATA4, HLA-ABC, and HLA-DR genes at p3 and they also expressed Oct4, Thy-1, FSP, vWF, vimentin, desmin, CK18, CD54, CD4, CD106, CD31, a-SMA, HLA-ABC proteins. Moreover, they could differentiate into mesodermal lineage cells such as adipocyte, osteoblast and chondrocyte. In addition, they also differentiated into insulin secreting cells in our culture condition. In conclusion, human thigh adipose stem cells retain proliferative potential and expression patterns similar to BM-MSCs and they also differentiate into various cell types. Thus, human thigh adipose stem cells might be useful alternative cell source for clinical application.

Distribution of Heavy Metal Content in Plants and Soil from a Korean Shooting Site

  • In this research we determined the levels of heavy metals in soil and metal-accumulating plants from a D military shooting site in the Kyungkido district of Korea. The data obtained may be useful in the development of methods for the efficient phytoremediation of contaminated soil. The total Cd, Cu, Pb, and Zn concentrations in the soil were found to be 1.67-5.04 mg/kg, 52.51-106.26 mg/kg, 37.24-90.32mg/kg, and 111.45-188.19mg/kg, respectively. These results show that the soil is contaminated with Cd and Cu, and this contamination is particularly severe in the case of Cd because of its high bioavailability (25-57% of the total metal in the soil is exchangeable). The high concentrations of heavy metals in the shoots of Persicaria thunbergii and Artemisia princeps var. orientalis indicate that these plants (all perennial herbs) accumulate heavy metal efficiently. Further, these plants were found to contain more Cd in its shoots (>60% of the total metal found in the plant) than any other plant; these results indicate that these native species are particularly suited to use in Cd phytoextraction.
