Effects of Samjawhadam-jeon on Immune Cells in Ovalbumin-induced Asthmatic Mice (삼자화담전(三子化痰煎)이 천식모델 생쥐의 면역세포에 미치는 영향)

  • Objective: The purpose of this study was to evaluate the effect of Samjawhadam-jeon (SJHDJ; 三子化痰煎) on immune cells in OVA-induced asthmatic mice. Material and Methods : C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (four times a week) for asthma induction. Two experimental groups were treated with different concentrations of SJHDJ group (400 mg/kg) extracts and cyclosporin A (10 mg/kg) for the latter 8 weeks. At the end of the experiment, the mouse lung. peripheral lymph node (PLN) and spleen were removed and immune cells were analyzed by flow cytometer. Results : Lung weight, total cells in lung, PLN, and spleen of the SJHDJ group decreased significantly compared with that of the control group. Number of $CD3e^+/CD69^+$, $CD3e^+/DX5^+$ cells in lung, PLN and spleen, number of $CD3^+$ cells in PLN and spleen, number of $CD3e^-/CCR3^+$ cells in lung and PLN, and number of $B220^+/IgE^+$ cells in PLN of the SJHDJ group decreased compared with that of the control group. Number of $CD4^+/CD25^+$ cells in PLN and spleen of the SJHDJ group increased compared with that of the control group. Conclusion : These results demonstrate that SJHDJ will be a desirable alternative therapy for allergic asthma by inhibiting the expression of immune cells.

Effects of Gamicheungpyehwadam-tang on Immune-cell Regulation in Association with Bronchial Asthma in OVA-induced Mouse Model (가미청폐화담탕이 천식 유발 병태 모델에서 천식 관련 활성 면역세포에 미치는 영향)

  • These studies were investigated the effects of Gamicheungpyehwadam-tang (CPHDT) on immune-cell regulation in association with bronchial asthma in OVA-induced mouse model. The administration of 400 mg/kg CPHDT significantly reduced the number of total cells in lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. The administration of 400 mg/kg CPHDT significantly reduced $CD3^+,{\;}CD19^+$and $CD3^+,{\;}CD69^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. CPHDT significantly reduced $CD3^+/CCR3^+,{\;}CD4^+,{\;}B220^+/IgE^+$, and $CD3^+/DX5^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model in a dose dependent manner, However, CPHDT significantly reduced $CD8^+$ cell numbers from only lung and spleen. The administration of CPHDT significantly reduced $NK^+$ cell numbers separated from lung of OVA-induced bronchial asthma mouse model in all concentrations, but 200 mg/kg CPHDT reduced $NK^+$ cell numbers separated from peripheral lymph node. These results suggest that CPHDT has anti-asthma and anti-allergy effects. In addition to, CPHDT may be useful treatment of asthma based on the further studies about the individual efficacy search of the components of CPHDT and the adding of variety drugs to CPHDT.

Suppressive Effects of Jauijeonhotang(JHG) on Immune cells in BALF of OVA-sensitized Balb/c Mice (자의전호탕(自擬前胡湯)이 천식 모델 생쥐의 BALF 내 관련 면역세포 변화에 미치는 영향)

  • In order to investigate the effects of JHG on immune cells in BALF, clinically prescribed JHG was tested using OVA induced asthma animal model. The results are shown below: 1. JHG significantly reduced the total number of cells in BALF compared to that of the control at 400 mg/kg doses. 2. JHG significantly reduced the number of CD3+/CD69+ cells in BALF at 400 and 200 mg/kg doses. 3. JHG significantly reduced the number of CCR3+ cells in BALF at 400 and 200 mg/kg doses. 4. JHG significantly reduced the number of B220+/CD22+ cells in BALF at 400 and 200 mg/kg doses. 5. JHG significantly reduced the number of B220+/IgE+ cells in BALF at 400 and 200 mg/kg doses. 6. JHG significantly reduced the number of B220+/CD45+ cells in BALF at 400 and 200 mg/kg doses. From the results above, anti-asthmatic efficacy of JHG through anti-oxidative activity as well as immune control activity has been experimentally proven. In depth study of JHG on various pathological factors and individual drug contents of JHG should follow.

Efficiency analysis of agricultural machinery rental system using the DEA model (자료포락분석법을 이용한 농기계 임대사업의 효율성 분석)

  • This study was conducted to survey and diagnose operation status of the agricultural machinery rental service, analyse and compare operational efficiency among 82 city and county ATDEC (agricultural technology development and extension center) using the DEA (Data Envelopment Analysis) method, and recommend future direction, for improvement of the business. Input variables were invested budget and labor, and output variable was rental return. Percentages of return to investment on the rental service were calculated as 68.3% and 63.9% when analyzed with CCR (Charnes, Cooper and Rhodes) and BCC (Banker, Charnes and Cooper) models, respectively, indicating inefficiency of the service operation. Increase of rental charge would increase efficiency by 63.9~68.3% depending on models, and decrease of financial and labor investment would improve the efficiency by about 11.3%. Technical efficiency would be more important than scale efficiency, therefore adjustment of over-invested budget and labor needed to be made together with increase of rental charge to improve the operation. Among the ATDECs providing the rental service, 6 (7.3%), 43 (52.4%), and 33 (40.2%) were in state of CRS (constant return to scale), IRS (increasing return to scale), and DRS (decreasing return to scale), respectively. These indicated public aspects of the rental system, over-investment, lack of output component for input component, meaning that scale income would be increased by qualitative expand of rental charge. Efficiency analysis of the rental system by region showed that efficient ATDECs to be benchmarked by others were in the order of DMU-70, DMU-54, DMU-29, DMU-5, DMU-22, DMU-2, and DMU-61. More comprehensive and extensive survey and analyses would be necessary in the future.

Lactoferrin Combined with Retinoic Acid Stimulates B1 Cells to Express IgA Isotype and Gut-homing Molecules

  • It is well established that TGF-${\beta}1$ and retinoic acid (RA) cause IgA isotype switching in mice. We recently found that lactoferrin (LF) also has an activity of IgA isotype switching in spleen B cells. The present study explored the effect of LF on the Ig production by mouse peritoneal B cells. LF, like TGF-${\beta}1$, substantially increased IgA production in peritoneal B1 cells but little in peritoneal B2 cells. In contrast, LF increased IgG2b production in peritoneal B2 cells much more strongly than in peritoneal B1 cells. LF in combination with RA further enhanced the IgA production and, interestingly, this enhancement was restricted to IgA isotype and B1 cells. Similarly, the combination of the two molecules also led to expression of gut homing molecules ${\alpha}4{\beta}7$ and CCR9 on peritoneal B1 cells, but not on peritoneal B2 cells. Thus, these results indicate that LF and RA can contribute to gut IgA response through stimulating IgA isotype switching and expression of gut-homing molecules in peritoneal B1 cells.

Proliferation of Mouse Prostate Cancer Cells Inflamed by Trichomonas vaginalis

  • Our objective was to investigate whether inflammatory microenvironment induced by Trichomonas vaginalis infection can stimulate proliferation of prostate cancer (PCa) cells in vitro and in vivo mouse experiments. The production of CXCL1 and CCL2 increased when cells of the mouse PCa cells (TRAMP-C2 cell line) were infected with live T. vaginalis. T. vaginalis-conditioned medium (TCM) prepared from co-culture of PCa cells and T. vaginalis increased PCa cells migration, proliferation and invasion. The cytokine receptors (CXCR2, CCR2, gp130) were expressed higher on the PCa cells treated with TCM. Pretreatment of PCa cells with antibodies to these cytokine receptors significantly reduced the proliferation, mobility and invasiveness of PCa cells, indicating that TCM has its effect through cytokine-cytokine receptor signaling. In C57BL/6 mice, the prostates injected with T. vaginalis mixed PCa cells were larger than those injected with PCa cells alone after 4 weeks. Expression of epithelial-mesenchymal transition markers and cyclin D1 in the prostate tissue injected with T. vaginalis mixed PCa cells increased than those of PCa cells alone. Collectively, it was suggested that inflammatory reactions by T. vaginalis-stimulated PCa cells increase the proliferation and invasion of PCa cells through cytokine-cytokine receptor signaling pathways.

The Effects of Sinapis Semen, Raphani Semen, and mixture decoction on the Asthmatic Mouse Model (백개자, 나복자 및 두 배합 약물의 천식 동물 모델에 대한 효과)

  • Objectives : To clarify the possible effects of Sinapis Semen and Raphani Semen on the development of pulmonary eosinophilic inflammation in a asthmatic mouse model. Methods : BALBav/c mice were sensitized to OVA followed intratracheally and by aerosol allergene challenges. We investigated the effect of Sinapis Semen and Raphani Semen on airway hyperresponsiveness, eosinophiic infitratio, immune cell phenotype, The2 cytokine product, and OVA-spedific IgE production. Results : Total lung cells, eosinophils, and lung leukocytes, OVA specific IgE levels, and Th 2cytokine levels such as IL-5, IL-13, IL-17, TNF-alpha, and eotaxin in BALF were reduced compared with those of OVA sensitized asthma mice (control). The absolute numbers of $CD3^+$, $CD3^+/CD69^+$, $CD3^-/CCR3^+$, $CD4^+$, $CD8^+$, $Gr-1^+/CD11b^+$, $B220^+/CD22^+$, $B220^+/IgE^+$ cells in lung tissiues significantly reduced compared to those of control. Specially total lung cells in BALF and the absolute number of $CD3^+/CD69^+$ and, $B220^+/IgE^+$ cells in lung tissiue effectively reduced in Sinapis Semen plus Raphani Semen compared to those of Sinapis Semen and Raphani Semen. Conclusions : These results indicate that Sinapis Semen plus Raphani Semen has deep inhibitory effects on airway inflammation and hyperresponsiveness in asmatic mouse model and also has effect of suppression of IL-5, IL-13, IL-17, OVA specific IgE production in BALF. The results verified that Sinapis Semen, Raphani Semen, and Sinapis Semen plus Raphani Semen could act as a immunomodulator which possess anti-inflammatory and anti-asthmatic property by modulating the relationship of Th1/Th2 cytokine imbalance.

The Effect of Nebulized Frankincense Essential Oil in an OVA-Induced Allergic Asthma Mouse Model (프랑킨센스 에센셜 오일 흡입이 OVA로 유도된 알러지성 천식 모델 생쥐에 미치는 영향)

  • In this study, we investigated the effects of frankincense essential oil (BSEO) on the immune cell change in the lung, BALF and PBMC using a mouse model of asthma. BALB/c mice after intraperitoneal OVA sensitization (day 1) were challenged intratracheally with OVA on day 14. Then, the asthma was induced by repeated OVA inhalation challenged. The asthma induced mice group inhaled 0.3% BSEO for 30 minutes per trial, three times a week, for 8 weeks using the nebulizer. After 12 weeks from the experiment, the mice was killed and the lung, bronchoalveolar lavage fluid (BALF) and peripheral blood mononuclear cell (PBMC) were obtained. Next, the change of immune cells inside the separated tissues was observed to identity the effects of BSEO on the allergic asthma mice. In conclusion, the hypersensitive reaction of airway to the bronchoconstrictor in the allergic asthma induced mice was effectively suppressed in Frankincense group, in Bermagot, Eucalyptus, Chamomile, Marjoram and Frankincense groups, the natural aromatic essential oil groups. Furthermore, it was also confirmed that the weight of lung, total number of alveolus cells and the number of BALF, MNL and DLN increased after inducing allergic asthma were reduced. BSEO suppressed the percentage of $CD3e^+/CD19^-$, $B220^+/CD23^+$ and $CD11b^+/Gr-1^+$ cells in the lung tissue of allergic asthma mice. Moreover, BSEO also reduced the percentage of $CD4^+/CD8^-$, $B220^+/CD23^+$ and $CD3^+/CCR3^+$ cells in BALF. In addition, the percentage of $CD3e^+/CD19^-$, $CD3^+/CD69^+$ and $B220^+/CD23^+$ cells in PBMC was reduced. The results of this study indicate that BSEO would be effective to treat allergic asthma by the immune control suppressing the activity of immune cells in each tissue.

Anti-CD3 Antibody Induces IL-10-producing $CD4^+CD25^+$ Regulatory T Cells, Which Suppress T Cell Response in Rheumatoid Arthritis Patients

  • Background: Regulatory T cells (Tregs) have been investigated intensively for some decades. These cells regulate the immune system, prevent overactivated immune responses and can be used therapeutically. For rheumatoid arthritis (RA), understanding the functions and status of Tregs is an important step for understanding immune regulation in this autoimmune disease. Methods: We investigated the percentages, phenotypes and suppressive functions of $CD4^+CD25^+$ Tregs in peripheral blood (PB) of patients with RA. Results: The percentages were higher in the patients (n=12) than in healthy controls (n=10), and the cells expressed the $CD45RB^{low}$, CTLA-4 and CCR7 phenotypes. We also investigated the expression of Foxp3 and secretion of interleukin (IL)-10 induced $CD4^+CD25^+$ Tcells by anti-CD3 antibody treatment. A suppressive function of the patients' cells was shown through coculture with $CD4^+CD25^-$ T cells in vitro. Conclusion: We suggest that, despite their increased numbers and suppressive function, they manage the ongoing inflammation ineffectively. It might be possible to apply IL-10 to induce the proliferation of IL-10-producing Tregs as therapy for RA.

Henoch-$Sch{\ddot{o}}nlein$ Nephritis in Children (소아 Henoch-$Sch{\ddot{o}}nlein$ 신염의 추적 관찰)

  • Purpose : This retrospective study has been undertaken to find out the clinical outcome of children with HS nephntis and its relationship with initial clinical presentation and/or renal pathologic finding. Patients and methods : Study population consisted of 59 children with HS nephritis who have been admitted to the Pediatric department of Kyungpook University Hospital from 1987 to 1999, and biopsy was done with indications of heavy proteinuria (> 1 g/m2/day) lasting over 1 month, nephrotic syndrome, and persistent hematuria and/or proteinuria over 1 year. Patients were divided clinically into 3 groups ; isolated hematuria, hematuria with proteinuria and heavy proteinuria (including nephrotic syndrome). Biopsy findings ore graded from I-V according to International Study of Kidney Disease in Children (ISKDC). Results : Mean age of presentation was $8.1{\pm}3.0$ years and slight male preponderance m noted (33 boys md 26 girls). Histopathologic grading showed Grade I ; 2, Grade II ; 44, and Grade III ; 13 cases. Clinical outcome at the follow-up period of 1-2 year (49 cases) and 3-4 years (30 cases) shooed normal urinalysis in 75 (30.6$\%$) and 18 cases (60.0$\%$), persistent isolated hematuria in 20 (40.8$\%$) and 2 cases (6.7$\%$), hematuria with proteinuria in 11 (22.5$\%$) and 8 cases (26.6$\%$), and persistent heavy proteinuria in 3 (6.1$\%$) and 2 cases (6.7$\%$) respectively. Clinical outcome according to histopathologic grading showed the frequency of normalization of urinalysis being lower in Grade III compared to grade I or II. Clinical outcome according to initial clinical presentation showed no relationship to the normalization or urinalysis at follow-up periods. However, 15-20$\%$ of children with initial heavy proteinuria showed persistent heavy proteinuria (3 out of 20 cases at 1-2 years, and 2 out of 10 case at 3-4 years of follow-up periods). Conclusion : The majority of children with HS nephritis (histopathologic grade I, II, III) improved within 3-4 years and persistent heavy proteinuria was seen only in a kw of children with initial clinical presentation of heavy proteinuria.
