Science Teachers' Perceptions to the Utilization of Calculator Based Laboratory System with Experimental Kit in Science Experiments (Calculator-Based Laboratory system과 실험 Kit를 이용한 과학실험에 대한 교사들의 인식)

  • A Calculator-Based Laboratory (CBL) system was introduced to science teachers and their perceptions to its classroom application was assessed. A CBL survey instrument was responded by 54 middle and high school science teachers who undertook a three-hour workshop of science experiments with CBL system. There were significant differences in teachers perceptions to CBL system among gender, school level, school location, teacher's degree, and years of teaching in terms of learning CBL system, applicability of CBL system for science classrooms, and effects on science achievement. Male teachers showed significantly (p<.05) higher agreement to learning of CBL system and applicability for science classrooms than female teachers. Compared to middle school teachers, high school teachers showed significantly (p<.05) high interests in CBL applicability for science classrooms and perceptions that there will be an increase of science achievement. Teachers with 4-8 years of teaching experience also showed significantly (p<.05) higher interest toward learning CBL system and its applicability. It was concluded that science teachers perceived CBL system as a promising science teaching method in Korean middle and high schools. However, a science teacher inservice training program for CBL system should be developed in consideration of gender, school level, school location, and years of teaching.

  • The purpose of this study is to verify the effectiveness of community-based learning(CBL) on career decision-making self-efficiency of junior college students and explore the meaning. This study was conducted on 68 students and 10 departments participating in the CBL, which was supported by the D University Faculty Learning Development Center in Busan. First of all, does CBL affect the career decision-making self-efficiency for junior college students? Second, what is the meaning of CBL for career decisions for junior college students? The effectiveness of the CBL's before and after application surveys has shown statistically significant changes in the career decision-making self-efficiency. The meaning of CBL for learners' career decisions was derived from "improving understanding through on-site application of theory and creating confidence and commitment in their career paths by providing an opportunity to study." Through this, it can be seen that CBL is worth applying as a teaching method suitable for career guidance of junior college students.

  • Web 2.0 환경에서 CBL 교수 모형은 유의미한 교수모형으로 등장하고 있다. CBL 교수 모형은 Google, Wikipedia 등 대용량 데이터베이스와 Wikispaces, Voice Thread, Twitter 등 다양한 Web 2.0 도구를 활용하여 학생들이 주도적으로 도전과제를 탐구하고 해결하는 교수 모형이다. 본 논문에서는 Web 2.0 환경에 적합한 교수 모형인 CBL의 개념 및 중요 구성요소 그리고 CBL 수업을 위한 환경구축에 대해 살펴보고 CBL Lesson plan 및 초등학교 교과교육에 CBL 교수모형을 적용한 예시를 소개하고 CBL를 초등학교에 적용하기 위한 고찰을 하였다.

C-terminal truncation of a bovine B12 trafficking chaperone enhances the sensitivity of the glutathione-regulated thermostability

  • The human $B_{12}$ trafficking chaperone hCblC is well conserved in mammals and non-mammalian eukaryotes. However, the C-terminal ~40 amino acids of hCblC vary significantly and are predicted to be deleted by alternative splicing of the encoding gene. In this study, we examined the thermostability of the bovine CblC truncated at the C-terminal variable region (t-bCblC) and its regulation by glutathione. t-bCblC is highly thermolabile ($T_m={\sim}42^{\circ}C$) similar to the full-length protein (f-bCblC). However, t-bCblC is stabilized to a greater extent than f-bCblC by binding of reduced glutathione (GSH) with increased sensitivity to GSH. In addition, binding of oxidized glutathione (GSSG) destabilizes t-bCblC to a greater extent and with increased sensitivity as compared to f-bCblC. These results indicate that t-bCblC is a more sensitive form to be regulated by glutathione than the full-length form of the protein.

  • In this paper we propose a new CBL-based floorplan method that accommodates pre-placed blocks. We identify the problem of the previous CBL-based pre-placed block floorplan method, and suggest the solution method of this problem. In our method, CBLs consisting of only free blocks are perturbed and maintained during the simulated annealing. Pre-placed blocks are inserted during packing in such a way that the topology of the CBL after insertion of a pre-placed block resembles the topology before insertion. Thus, even with the inclusion of pre-placed blocks, the searching effort via simulated annealing yields acceptable results. Experimental results show that our floorplan method places pre-placed blocks effectively and efficiently.

Lack of RING Finger Domain (RFD) Mutations of the c-Cbl Gene in Oral Squamous Cell Carcinomas in Chennai, India

  • Background: In normal cells, activated epidermal growth factor receptor (EGFR) molecules are subjected to ubiquitination-mediated proteasome degradation pathway by c-Cbl, an ubiquitin ligase that checks uncontrolled proliferation. Hence expression of wild type c-Cbl molecule is essential to keep this degradation machinery in a functional state. Loss of expression or function of c-Cbl may consequently lead to sustained activation of EGFR and promote carcinogenesis, loss of function mutations in the c-Cbl gene already being reported in lung and hematopoietic cancers. However, the genetic status of c-Cbl in oral squamous cell carcinoma (OSCC) is not known. Hence in the present study we investigated the genomic DNA isolated from OSCC tissue biopsy samples for mutations in the RING finger domain coding region of c-Cbl gene, which has also been reported to be most frequently mutated in other cancers. Materials and Methods: Total genomic DNA isolated from thirty two post surgical OSCC tissue samples were amplified using primers flanking the exon 8 of c-Cbl gene that codes for the RING finger domain. The PCR amplicons were then resolved in a 1.2% agarose gel, purified and subjected to direct sequencing to screen for mutations. Results: The sequencing data of the thirty two OSCC samples did not identify mutations in the RING finger domain coding region of c-Cbl gene. Conclusions: To the best of our knowledge, this is the first time that the genetic status of c-Cbl gene in OSCC samples has been investigated. The present data indicates that genetic alteration of RING finger domain coding region of c-Cbl gene is relatively infrequent in OSCC samples.

Protection of aquo/hydroxocobalamin from reduced glutathione by a B12 trafficking chaperone

  • We identified a bovine $B_{12}$ trafficking chaperone bCblC in Bos taurus that showed 88% amino acid sequence identity with a human homologue. The protein bCblC was purified from E. coli by over-expression of the encoding gene. bCblC bound cyanocobalamin (CNCbl), methylcobalamin (MeCbl) and adenosylcobalamin (AdoCbl) in the base-off states and eliminated the upper axial ligands forming aquo/hydroxocobalamin ($OH_2$/OHCbl) under aerobic conditions. A transition of $OH_2$/OHCbl was induced upon binding to bCblC. Interestingly, bCblC-bound $OH_2$/OHCbl did not react with reduced glutathione (GSH), while the reaction of free$OH_2$/OHCbl with GSH resulted in the formation of glutathionylcobalamin (GSCbl) and glutathione disulfide (GSSG). Furthermore we found that bCblC eliminates the GSH ligand of GSCbl forming $OH_2$/OHCbl. The results demonstrated that bCblC is a $B_{12}$ trafficking chaperone that binds cobalamins and protects $OH_2$/OHCbl from GSH, which could be oxidized to GSSG by free $OH_2$/OHCbl.

  • In this study firstly we survey the calculation method and the characteristics of the way of estimating CBL(Customer BaseLine Load) that is important calculation tool for DRP internationally. Also we analyze the power consumption pattern using the 15 minutes load profiles of about 120,000 customers in domestic. Based on this pattern, we provide the CBL calculation method that can be utilized in DRP to save the cost, and analyze the accuracy of the CBL calculation proposed in this paper through the simulation.

  • CBL[1](Corner Block List)에 기반한 Non-Slicing 플로 플랜 알고리즘은 빈 공간이 없는 Non-Slicing 플로플랜만 나타낼 수 있다. 본 논문에서는 CBL 단점을 보완하고 실제 블록의 크기를 이용하여 최적의 위치에 블록을 배치 하기 위해 contour map을 이용할 것을 제시한다. 본 알고리즘은 배치시 면적을 최소화 하는 방법을 제시하므로 CBL의 단점을 해결하고 더불어 최적해를 찾기 위한 실행 시간을 단축 시키는 효과를 기대할 수 있다.

The Effects of Case-Based Learning (CBL) on Learning Motivation and Learning Satisfaction of Nursing Students in a Human Physiology Course (사례기반학습법을 적용한 수업이 간호대학생의 학습동기와 학습만족도에 미치는 효과 - 인체생리학 수업을 중심으로)

  • Purpose: The purpose of this study was to investigate the effects of case-based learning (CBL) on learning motivation and learning satisfaction of nursing students in a human physiology course. Methods: The development and application of CBL scenarios was conducted from February to June, 2013. Nursing students (n=142) who registered for a human physiology course were assigned into either a control or CLB group. The control group received traditional lectures for 14 weeks. The CBL group received the same 14-week lectures and an additional 5 CBL sessions. The learning motivation and satisfaction were measured by questionnaires at the beginning and the end of the semester. Seven students in the CBL group were randomly selected for a focus-group interview. Quantitative data were analyzed by ${\chi}^2$-test and t-test, and qualitative data were analyzed by content analysis. Results: The learning motivation and learning satisfaction were not significantly different between the two groups. However, 59% of the CBL group answered with a positive impression on the CBL approach as it helped them to feel a sense of achievement, excitement, to form their identity as nursing students, and so on. Conclusion: These findings suggest that the CBL could be a challenging but useful learning method in a physiology course for nursing students. Further studies with guidance, such as instructors' questions and feedback design are needed to utilize CBL more effectively.
