Effects of Aqua-Acupuncture of Carthami Flos(GV15) on the Changes of Cerebral Hemodynamics in Rats (아문(瘂門)(GV15)의 홍화약침(紅花藥針)이 백서(白鼠)의 뇌혈류역학(腦血流力學) 변동(變動)에 미치는 영향(影響))

  • Objective : Carthami Flos has been used as a herb to promote blood circulation to remove blood stasis in oriental medicine for many centuries, and Amun(GV15) has been used as a meridian point to treat apoplexy etc. To investigate treatment of cerevral vascular disease(CVA) by promoting blood circulation and removing blood stasis(活血化瘀法), we observed the experimental effects and mechanism of auqa-acupunture of Carthami Flos(ACF) injected into GV15 on cerevral hemodynamics and cardiovascular system of rats. Method : Aqua-acupuncture of Carthami Flos(ACF) was injected into GV15, and then we investigated experimental effects and mechanism of ACF on the cerebral hemodynamics[regional cerebral blood flow(rCBF), pial arterial diameter(PAD), meal arterial blood pressure(MABP)] and cardiovascular system[cardiac muscle contractile force(CMF), heart rate(HR)I by pretreatment with methylene blue(MTB) and indomethacin(IDN). The changes in rCBF, MABP, CMF and HR were tested by Laser Doppler Flowmetry(LDF), and the changes in PAD was determinated by video microscopy methods and video analyzer. Results :The results were as follows in normal rats ; The changes of rCBF and PAD were significantly increased by ACF($120{\mu}{\ell}/kg$) in a injected time-dependent manner, but MABP was not changed by ACF. The changes of cardiovascular system were increased by ACF in a injected time-dependent manner. And pretreatment with MTB was significantly inhibited ACE induced increase of rCBF and PAD, and was decreased ACF induced increase of HR. And pretreatment with IDN was increased ACF induced MABP and CMF. And the results were as follows in cerebral ischemic rats ; The changes of rCBF was increased stabilizly by treatment with ACF($120{\mu}{\ell}/kg$) in during the period of cerebral reperfusion, but pretreatment with MTB was increased ACF induced increase of rCBF during the period of cerebral reperfusion. The results were as follows in normal rats ; The changes of rCBF and PAD were significantly increased by ACF($120{\mu}{\ell}/kg$) in a injected time-dependent manner, but MABP was not changed by ACF. The changes of cardiovascular system were increased by ACF in a injected time-dependent manner. And pretreatment with MTB was significantly inhibited ACF induced increase of rCBF and PAD, and was decreased ACF induced increase of HR. And pretreatment with IDN was increased ACF induced MABP and CMF. And the results were as follows in cerebral ischemic rats ; The changes of rCBF was increased stabilizly by treatment with ACF($120{\mu}{\ell}/kg$) in during the period of cerebral reperfusion, but pretreatment with MTB was increased ACF induced increase of rCBF during the period of cerebral reperfusion Conclusions : In conclusion, ACF causes a diverse response of rCBF, PAD an HR, and action of ACF is mediated by cyclic GMP. I suggested that ACF has an anti-ischemic effect through the improvement of crebral hemodynamics in a transient cerebral ischemia.

Method : When aqua-acupuncture of Carthami Flos(ACF) was injected into LI4, Liv3, B23, B62, GV16, experimental effects of ACF on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) is not known. Therefor, this study was designed to investigate the effects of ACF on the change of cerebral hemodynamics. The changes of rCBF and MABP were tested by Laser Doppler Flowmetry(LDF). Results : When ACF was injected into LI4, change of rCBF and MABP were decreased in a injected time-dependent manner. When ACF was injected into Liv3, change of rCBF and MABP were decreased in a injected time-dependent manner. When ACF was injected into B23, change of rCBF was significantly increased in a injected time-dependent manner, MABP did not change. When ACF was injected into B62, change of rCBF was increased in a injected time-dependent manner, change of MABP was significantly decreased after injection 60min. When ACF was injected into GV16, change of rCBF and MABP were similar to the change of normal. Conclusions : In conclusion, I suggested that ACF(LI4, Liv3) has an effect that depress the blood pressure & cerebral hemodynamic acceleration and ACF(B23, B62) has an anti-ischemic effect through the improvement of crebral hemodynamics.

  • A new CRT binding factor (CBF) gene designated Cbcbf25 was cloned from Capsella bursa-pastoris, a wild grass, by the rapid amplification of cDNA ends (RACE). The full-length cDNA of Cbcbf25 was 898 bp with a 669 bp open reading frame (ORF) encoding a putative DRE/CRT (LTRE)-binding protein of 223 amino acids. The predicted CbCBF25 protein contained a potential nuclear localization signal (NLS) in its N-terminal region followed by an AP2 DNA-binding motif and a possible acidic activation domain in the C-terminal region. Bioinformatic analysis revealed that Cbcbf25 has a high level of similarity with other CBF genes like cbf1, cbf2, and cbf3 from Arabidopsis thaliana, and Bncbf5, Bncbf7, Bncbf16, and Bncbf17 from Brassica napus. A cold acclimation assay showed that Cbcbf25 was expressed immediately after cold triggering, but this expression was transient, suggesting that it concerns cold acclimation. Our study implies that Cbcbf25 is an analogue of other CBF genes and may participate in cold-response, by for example, controlling the expression of cold-regulated genes or increasing the freezing tolerance of plants.

Effects of Tonggok($BL_{66}$) ${\cdot}$ Hyeopgye($GB_{43}$) Supplement and Sangyang($LI_1$) ${\cdot}$ Gyueum($GB_{44}$) Draining in Acupuncture on the Improvement of Cerebral Hemodynamics in Rats (족소양담경(足少陽膽經) 정격(正格) 척침(刺鍼)이 백서(白鼠)의 뇌혈류력학(腦血流力學) 변동(變動)에 미치는 영향)

  • Objectives: The purpose of this report is to examine the effect of $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and LIl ${\cdot}$ $GB_{44}$ draining in acupuncture for cerebral apoplexy treatment. Methods : I designed to investigate whether $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture affects cerebral hemodynamics [regional cerebral blood flow(rCBF), mean arterial blood pressure(MABP)] in normal rats, and to make manifest whether $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture is mediated by cyclooxygenase or guanylate cyclase. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF). Results : 1. $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and LI1 ${\cdot}$ $GB_{44}$ draining in acupuncture increased rCBF significantly, but decreased MABP. These results suggest that $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture increased significantly rCBF by dilating pial arterial diameter(PAD). 2. By pretreatment with indomethacin(1 mg/kg, i.p.), the effect of $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture induced increase of rCBF was significantly inhibited, the decreased MABP by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was increased. 3. By pretreatment with methylene blue($10{\mu}/kg$, i.p.), the increased rCBF by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was decreased conversely, the decreased MABP by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was decreased. and then increased. Conclusion : I suggest that $BL_{66}$ ${\cdot}$ $GB_43$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture has an anti-ischemic effect through the improvement of cerebral hemodynamics.

  • This study was designed to investigate the effects of Sagunja-tang Extract & Zingiberis rhizoma recens Juice (SEZJ) and Zingiberis rhizoma recens Juice (ZRJ) on the changes in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. The results were as follows ; SEZJ and ZRJ significantly increased rCBF in a dose-dependent manner, while it did not change MABP. This results suggest that SEZJ and ZRJ significantly increased rCBF by dilating pial arterial diameter. Increase of SEZJ-induced rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase, and indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. SEZJ-induced MABP was significantly increased by pretreatment with indomethacin but was not changed by methylene blue. These results suggested that the action of SEZJ was mediated by cyclic 3',5'-guanosine monophosphate.

Alterations in Cerebrovascular Reactivity by Trigeminovascular System Injury in Rats

  • Trigeminovascular system plays an important role for the cerebral memodynamics. The aim of this study was to investigate the alterations in cerebrovascular reactivity by trigeminovascular system injury in rats. Trigeminovascular system of male Sprague-Dawley rats was injured by either denervation of nasocilliary nerve or neonatal capsaicin treatment. Trigeminovascular system was stimulated by controlled hemorrhagic hypotension or somatosensory (whisker) stimulation. Changes in regional cerebral blood flow (rCBF) and pial arterial diameter were continuously measured by laser-Doppler flowmetry and videomicroscopy, respectively. Nitric oxide synthase (NOS) activity in cerebral cortex was determined by measuring the conversion of $L-^3H-arginine\;to\;L-^3H-citrulline$. Cyclic GMP levels in cerebral cortex and pial artery were determined using the cyclic GMP $^{125}I$ scintillation proximity assay system. rCBF autoregulation was impaired or almost abolished by trigeminovascular system injury. rCBF response to whisker stimulation was significantly attenuated by trigeminovascular system injury. NOS activity as well as cyclic GMP level in cerebral cortex and pial artery were significantly reduced in the group of trigeminovascular system injury. These results suggest that trigeminovascular system injury causes prominent alterations in cerebrovascular reactivity, and that NO, which is generated by neuronal NOS in the trigeminovascular system, is implicated in the regulation of rCBF.

  • Objectives : Folium perillae (FP) can relieve superficial pathogenic factors to dissipate cold and promote the circulation of qi and regulate the function of the stomach and is often used for interior qi-stagnation. We hypothesized that FP could rescue cerebral ischemia in rats. Methods : The present study was carried out to investigate the effects of FP on cerebral ischemia in terms of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. Finally, lactate dehydrogenase (LDH) release was investigated, too. Results : In this study, treatment with FP elevated rCBF and MABP levels in dose-dependent manner. Pre-treatment with indomethacin, an inhibitor of cyclooxygenase, inhibited rCBF increase induced by FP effectively. However, FP did not affect stability during cerebral reperfusion. Finally, FP significantly inhibited LD H activity in vitro Conclusions : These results suggest that FP is useful to treat patient with diseases related to cerebral ischemia, because FP can increase rCBF and MABP.

Effect of Cheonmabanhwa-tang on the Cerebral Hemodynamics in Rats (천마반하탕이 뇌혈류역학에 미치는 영향)

  • Cheonmabanhwa-tang(CBT) has been used in the Oriental Medicine for many centuries as a therapeutic agent for dizziness due to Poong-Dam. This study was designed to investigate whether CBT has a cytotoxicity in vitro, and affects the cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), and mean arterial blood pressure(MABP)) in normal and cerebral ischemic rats. The changes of rCBF were determinated by laser-doppler f1owmeter(LDF), and the change of PAD was determinated by video-microscopy. The results in normal rats were as follows: CBT had no cytotoxicity in neuronal cells. CBT significantly increased rCBF. MABP and PAD in a dose-dependent manner, respectively. Both rCBF and PAD were significantly and stably increased by CBT(10 mg/kg, i.p.) during the period of cerebral reperfusion. which contrasted with the findings of rapid and marked increase in control group. In conclusion, it is suggested that CBT causes a diverse effect on cerebral hemodynamics thereby has an anti-ischemic action.

Effects of Nelumbinis Rhizomatis Nodus Extract on Cerebral Hemodynamics in Rats (연근 추출물이 뇌혈류역학에 미치는 효과)

  • This Study was designed to investigate the effects of Nelumbinis Rhizomatis Nodus (NRN) on the change of cerebral hemodynamics [regional cerebral blood flow (rCBF), pial arterial diameter (PAD) and mean arterial blood pressure (MABP)] in normal and cerebral ischemic rats. And, this study was designed to investigate the inhibition of lactate dehydrogenase activity in neuronal cells The results were as follows NRN significantly increased rCBF and PAD in a dose-dependent manner, and NRN increased MABP in a dose-dependent manner. This results suggested that NRN significantly increased rCBF by dilating PAD. Both rCBF and PAD were significantly and stably increased by NRN (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. NRN significantly inhibited lactate dehydrogenase activity in neuronal cells. This results suggested that NRN prevented the neuronal death. It is suggested that NRN had an anti-ischemic effect through the improvement of cerebral hemodynamics and inhibitive effect on the brain damage.

Effects of LEONURI HERBA Extract on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal Rats (익모초(益母草) 추출액이 국소 뇌혈류량 및 평균혈압에 미치는 영향)

  • The study was designed to investigate the effects of Leonuri herba extract (LHE) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and further to determine the mechanism of action of LHE. The results in normal rats were as follows, LHE significantly increased rCBF in a dose-dependent manner, and MABP did not change in a dose-dependent manner. This results were suggested that LHE significantly increased rCBF by dilating pial arterial diameter. The LHE-induced increase in rCBF was significantly inhibited by Pretreatment with indomethacin(1 mg/kg, i.p.), an inhibitor of cyclooxygenase, and was significantly inhibited by methylene blue($10{\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase. The LHE-induced MABP did not change by pretreatment with indomethacin but was significantly inhibited by methylene blue. This results were suggested that the mechanism of LHE was mediated by cyclooxygenase.
