Relationship between the Methane Production and the CNCPS Carbohydrate Fractions of Rations with Various Concentrate/roughage Ratios Evaluated Using In vitro Incubation Technique

  • The objective of the trial was to study the relationship between the methane ($CH_4$) production and the Cornell Net Carbohydrate and Protein System (CNCPS) carbohydrate fractions of feeds for cattle and the suitability of CNCPS carbohydrate fractions as the dietary variables in modeling the $CH_4$ production in rumen fermentation. Forty-five rations for cattle with the concentrate/roughage ratios of 10:90, 20:80, 30:70, 40:60, and 50:50 were formulated as feed samples. The Menke and Steingass's gas test was used for the measurement of $CH_4$ production. The feed samples were incubated for 48 h and the $CH_4$ production was analyzed using gas chromatography. Statistical analysis indicated that the $CH_4$ production (mL) was closely correlated with the CNCPS carbohydrate fractions (g), i.e. CA (sugars); $CB_1$ (starch and pectin); $CB_2$ (available cell wall) in a multiple linear pattern: $CH_4=(89.16{\pm}14.93)$ $CA+(124.10{\pm}13.90)$ $CB_1+(30.58{\pm}11.72)$ $CB_2+(3.28{\pm}7.19)$, $R^2=0.81$, p<0.0001, n = 45. Validation of the model using 10 rations indicated that the $CH_4$ production of the rations for cattle could accurately be predicted based on the CNCPS carbohydrate fractions. The trial indicated that the CNCPS carbohydrate fractions CA, $CB_1$ and $CB_2$ were suitable dietary variables for predicting the $CH_4$ production in rumen fermentation in vitro.

Hepatoprotective Effects of Fermented Smilax china Leaf Extract on Carbon Tetrachloride-induced Liver Injury in Mice (발효 청미래덩굴잎 추출물이 사염화탄소에 의한 마우스의 간 손상 보호 효과)

  • This study was conducted to investigate the hepatoprotective effects of fermented Smilax china leaf ethylacetate extracts by Aspergillus oryzae on carbon tetrachloride-induced liver injury in mice. Experimental mice were divided into four groups (five mice/group) (NC; normal control group, CB; basic diet supplemented before $CCl_4$ treatment group, NS ; basic diet mixed with 0.5% Smilax china leaf ethyl acetate extract supplemented before $CCl_4$ treatment group, FS; basic diet mixed with 0.5% fermented Smilax china leaf ethyl acetate extract supplemented before $CCl_4$ treatment group) fed for 4 weeks each. In the $CCl_4$-treated groups (CB, NS and FS) compared with the NC group, liver weights, activities of alanine aminotransferase, aspartate aminotransferase, xanthine oxidase and aldehyde oxidase, contents of triglycerides, total cholesterol and LDL-cholesterol in serum, and hepatic lipid peroxide levels increased, whereas body weight gain and contents of glutathione and HDL-cholesterol decreased. Furthermore, in the FS groups compared with the NS and CB groups, increased or decreased indicators by $CCl_4$ treatment significantly decreased or increased, respectively. This study suggests that fermented Smilax china L. leaf extracts may regulate xanthine oxidase and aldehyde oxidase inhibitory activities and hepatoprotective effects due to flavonoid aglycone derived from its glycoside in leaves of Smilax china by fermentation of A. oryzae.

  • A total of 20 extracts derived from different plant family commonly used in Indonesian traditional inflammation medicine were screened for their inhibitory effect on soybean lipoxygenase (SBL) and hyaluronidase (HAse) activity. Three methanol extracts, the bark of Cinnamomum burmanni (CB), the leaves of Piper betel (PB), and fruit of Barringtonia acutangula (BA) were found to have high inhibitory effects, whereas the methanol extract of the leaves of Mimusops elengi (ME) have medium inhibitory effect. The IC50 of CB, PB, BA and ME were found to be 21.7, 16.9, 39.1 and 62.8 g/$m\ell$, respectively. Among the tested extracts, only CB inhibited HAse (IC50 = 27g/$m\ell$). CB was successively fractionated with n-hexane, ethyl acetate, butanol and water. The EtOAc fraction having the strongest activity was fractionated and some compounds were isolated and purified by a preparative HPLC(Develosil ODS-HG-5 column). Coumarin and 2-hydroxy cinnamaldehyde. were identified through the analyses of UV-Vis absorption 1H-NMR, 13C-NMR and FAB+-MS spectra.

Physicochemical characteristics of wine made from domestic grapes (국내 포도로 제조한 와인의 이화학적 특성)

  • This study was conducted to investigate the quality of wine, produced from 11 different grape varieties. Black Sun, CB, Gaeryangmeoru, MW52, MW55, and Narsha with 0.6-0.8% total acids were good for wine. The total acid content of Wangmeoru wine was too high (1.41%). Black Sun, CB, Gaeryangmeoru, MW52, MW55, MBA, WB, and Wangmeoru showed excellent chromaticity (90dE or higher). Total polyphenols, total anthocyanin and tannin, contents were the highest in Gaeryangmeoru, WB, Wangmeoru, and CB and would be advantageous for wine aging. The results of this study deemed Gaeryangmeoru and CB to be suitable for red wine preparation.

Decreases Nitrous Oxide Emission and Increase Soil Carbon via Carbonized Biomass Application of Orchard Soil (과수원 토양의 탄화물 시용에 따른 아산화질소 발생량 감소와 토양탄소 증가효과)

  • BACKGROUND: Carbonized biomass is a carbon-rich solid product obtained by the pyrolysis of biomass. It has been suggested to mitigate climate change through increased carbon storage and reduction of greenhouse gas emission. The objective of this study was to evaluate carbon dioxide ($CO_2$) and nitrous oxide ($N_2O$) emissions from soil after carbonized biomass addition. METHODS AND RESULTS: The carbonized biomass was made from a pyrolyzer, which a reactor was operated about $400{\sim}500^{\circ}C$ for 5 hours. The treatments were consisted of a control without input of carbonized biomass and two levels of carbonized biomass inputs as 6.06 Mg/ha for CB-1 and 12.12 Mg/ha for CB-2. Emissions of $CO_2$ and $N_2O$ from orchard soil were determined using closed chamber for 13 weeks at $25^{\circ}C$ of incubation temperature. It was shown that the cumulative $CO_2$ were $209.4g\;CO_2/m^2$ for CB-1, $206.4g\;CO_2/m^2$ for CB-2 and $214.5g\;CO_2/m^2$ for the control after experimental periods. The cumulative $CO_2$ emission was similar in carbonized biomass input treatment compared to the control. It was appeared that cumulative $N_2O$ emissions were $4,478mg\;N_2O/m^2$ for control, $3,227mg\;N_2O/m^2$ for CB-1 and$ 2,324mg\;N_2O/m^2$ for CB-2 at the end of experiment. Cumulative $N_2O$ emission contents significantly decreased with increasing the carbonized biomass input. CONCLUSION: Consequently the carbonized biomass from byproducts such as pear branch residue could suppress the soil $N_2O$ emission. The results fromthe study imply that carbonized biomass can be utilized to reduce greenhouse gas emission from the orchard field.

Effect of passive self-ligating bracket placement on the posterior teeth on reduction of frictional force in sliding mechanics

  • Objective: The purpose of this study was to investigate the static (SFF) and kinetic frictional forces (KFF) in sliding mechanics of hybrid bracket systems that involve placing a conventional bracket (CB) or active self-ligating bracket (ASLB) on the maxillary anterior teeth (MXAT) and a passive SLB (PSLB) on the maxillary posterior teeth (MXPT). Methods: The samples consisted of two thoroughbred types (group 1, anterior-CB + posterior-CB; group 2, anterior-ASLB + posterior-ASLB) and four hybrid types (group 3, anterior-CB + posterior-PSLB-type 1; group 4, anterior-CB + posterior-PSLB-type 2; group 5, anterior-ASLB + posterior-PSLB-type 1; group 6, anterior-ASLB + posterior-PSLB-type 2) (n = 13 per group). After maxillary dentition alignment and maxillary first premolars removal in the stereolithographically-made typodont system, a $0.019{\times}0.025$-inch stainless steel wire was drawn through the right quadrant of the maxillary arch at 0.5 mm/min for 5 min. The SFF and KFF were measured with a mechanical testing machine and statistical analyses were performed. Results: Four different categories of SFF and KFF were observed among all groups (all p < 0.001). Group 1 demonstrated the highest SFF and KFF; groups 4 and 3 were second and third highest, respectively. The fourth category included groups 2, 5, and 6. Placing PSLBs on the MXPT resulted in significant SFF and KFF reductions in cases with CBs on the MXAT, but not in cases with ASLBs on the MXAT. Conclusions: These data might aid in the development of a hybrid bracket system that enables low-friction sliding of an archwire through the MXPT.

Protection of Amyloid ${\beta}$ Protein (25-35)-induced Neuronal Cell Damage by Methanol Extract of New Stem of Phyllostachys nigra Munro var. henonis Stapf in Cultured Rat Cortical Neuron

  • Caulis Bambusae in Taenia is widely used in Korea and China due to its various pharmacological activity. The present study aims to investigate the effect of the methanol extract of Caulis Bambusae in Taenia (CB) from Phyllostachys nigra Munro var. henonis Stapf (Gramineae) on amyloid ${\beta}$ protein (25-35) $(A{\beta}\;(25-35))$, a synthetic 25-35 amyloid peptide, -induced neurotoxicity using cultured rat cortical neurons. CB, over a concentration range of $10-50{\mu}g/{\mu}l$, inhibited the $A{\beta}\;(25-35)\;(10\;{\mu}M)$-induced neuronal cell death, as assessed by a 3-[4,5-dimethyIthiazole-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and the number of apoptotic nuclei, evidenced by Hoechst 33342 staining. CB $(50\;{\mu}g/{\mu}l)$ inhibited glutamate release into medium induced by $10\;{\mu}M\;A{\beta}$, (25-35) which was measured by HPLC. Pretreatment of CB $(50\;{\mu}g/{\mu}l)$ inhibited $10{\mu}M\;A{\beta}$ (25-35)-induced elevation of cytosolic calcium concentration $([Ca^{2+}]_c)$, which was measured by a fluorescent dye, fluo-4 AM, and generation of reactive oxygen species. These results suggest that CB prevents $A{\beta}$ (25-35)-induced neuronal ell damage in vitro.

Biotype, serotype and antibiotics susceptibility of Yersinia enterocolitica isolated from zoo animals (동물원(動物園) 야생동물(野生動物)에서 분리(分離)한 Yersinia enterocolitica 의 생물형(生物型), 혈청형(血淸型) 및 항생제(抗生劑) 감수성(感受性))

  • A study on the isolation of Yersiniae from the feces of wild animals(mammals 376, birds 19 and reptiles 13) in zoo and the biotype and serotype and susceptibility of 12 antibiotics was carried. Out of 408 animals, Yersiniae were isolated from 28 animals(6.9%). Of 28 isolates, 27 isolates(96.4%) were Y. enterocolitica and 1(3.6%) was Y. kristensenii. According to the species, 25(6.6%) of Y. enterocolitica and 1(0.3%) of Y. kristensenii were isolated from 376 mammals, 2(15.4%) of Y. enterocolitica from 13 reptiles but not isolated from 19 birds. According to the eating pattern, 8(5.2%) of Y. enterocolitica were isolated from 155 carnivora, 13(10%) of Y. enterocolitica from 123 herbivora, and 6(4.9%) of Y. enterocolitica and 1(0.8%) of Y. enterocolitica from 123 omnivora. Out of 27 isolates of Y. enterocolitica, all were biotype 1. And predominant serotype was 0:21(40.7%), and 0:5(37.0%), 0:6(11.1%), 0:1(3.7%), 0:9(3.7%) and untypable(3.7%). Yersiniae isolated from zoo animals were resistant to cephalothin(100%), ampicillin(96.4%), carbenicillin(96.4%) and tetracycline(14.3%) and streptomycin(3.6%) and susceptible to chloramphenicol(100%), colistin(100%), gentamicin(100%), kanamycin(100%), nalidixic acid(100%), polymyxin B(100%) and tobramycin(100%). The predominant multiple resistance pattern was Am-Cf-Cb(82.1%), and Am-Cf-Cb-Te(10.7%) and Am-Cf-Cb-Te-Sm(3.7%).

Epidemiologic and Clinical Features of Enteroviral Infections in Children; 1996~1998 (소아에서 장바이러스 감염의 역학 및 임상적 특성에 관한 연구; 1996년~1998년)

  • Purpose : As several epidemics of aseptic meningitis had been reported in Korea since 1990, the need of epidemiologic study about enteroviral infections was recognized. We studied epidemiologic and clinical features of enteroviral infections in children. Methods : We analyzed the seasonal occurrence of enteroviral infections and age, sex and clinical diagnosis of the children admitted to five branch hospitals of Hallym University and diagnosed to have enteroviral infections by culture from January 1996 to December 1998. Results : Enterovirus was isolated in 126 out of 245(51.4%) stools, 15 out of 89(16.8%) nasopharyngeal aspirates and 195 out of 1,835(10.6%) cerebrospinal fluids. There were 273 echoviruses(Echo)(197 Echo 30, 46 Echo 9, 17 Echo 6, 13 unclassified), 24 coxsackieviruses B(CB)(11 CB 2, 2 CB 5, 11 unclassified), 7 coxsackieviruses A 24 and 32 unclassified enteroviruses. Yearly number of isolates was 65(46 Echo 9) in 1996, 239(197 Echo 30) in 1997 and 32(15 CB: 9 CB 2) in 1998. Epidemics of enteroviruses occurred mainly during the months May to October. The mean age of the infected children was $62.1{\pm}38.0$ months and 74.6% of the patients were younger than 7 years of age. There were 203 boys and 100 girls and 79.3% of the patients were diagnosed as aseptic meningitis. Conclusions : We confirmed that enteroviral infections occurred in epidemics from late spring to fall during the study period. Enteroviral infections occurred predominantly in boys and the preschool children. Aseptic meningitis was the main illness in the hospitalized children.
