Acute and subacute toxicity of folpet to fingerings of common carp, Cyprinus carpio and goldfish, Carassius auratus (잉어치어(稚魚)와 금붕어에 대한 folpet의 급성(急性) 및 아급성독성(亞急性毒性)에 관한 연구)

  • The acute and subacute toxicity of fungicide folpet was evaluated in fingerings of common carp, Cyprinus carpio and goldfish, Carrassius auratus. Dipping of fishes for acute toxicity was performed for a period of 24h, and the TLm value(median tolerance limit) was 1.52 ppm in common carp and 1.45 ppm in goldfish. Severe damages were observed in various organs and among them, clubbing of gill lamella, lytic degeneration and vacuolation of liver cells, and epithelial edema of renal tubules were relatively prominent. The most significant changes were hyperbasophilic foci of liver cells in subacute toxicity test and these can imply the possibility of hepatocarcinogenecity of folpet.

Effects of Nitrite Exposure on Plasma Nitrite Levels and Hepatic Drug-metabolizing Enzymes in the Carp, Cyprinus carpio (아질산 노출이 이스라엘잉어 혈장내 아질산 농도 및 간장 약물대사효소에 미치는 영향)

  • Effects of ambient nitrite, NO$_2$$\^$-/, at 1, 3, 10 and 30 mg/1, on the changes of plasma nitrite/nitrate and on hepatic drug - metabolizing enzyme activity were examined in the juvenile Israeli carp, Cyprinus carpio. When the fish were exposed to 1 and 3 mg/1 NO$_2$$\^$-/, there was an exposure duration-dependent increase in plasma NO$_2$$\^$-/ over the 96-hr period reaching 6∼7 fold excess the ambient concentration. In the fish exposed to 10 mg/1, a plateau concentration of less than 2-fold of the environment was attained in 12 hr. With 30 mg/1, however, the maximal plasma NO$_2$$\^$-/ was 41.25 mg/1 at 12 hr followed by a gradual decline. There was a concentration-dependent increase in methemoglobin (metHb) level in all NO$_2$$\^$-/ -exposed groups and a significant decrease in hematocrit value in 30 mg/l group after 96-hr exposure. Apart from the blunted increase in plasma NO$_2$$\^$-/ with higher NO$_2$$\^$-/ (10 and 30 mg/1) exposure, the ratio of plasma NO$_3$$\^$-/ to NO$_2$$\^$-/ was signifirantly higher in these groups compared to 1 and 3 mg/1. The imbalance in the plasma NO$_3$$\^$-//NO$_2$$\^$-/ at higher NO$_2$$\^$-/ exposure suggests a possible accelerated conversion of NO$_2$$\^$-/ to NO$_3$$\^$-/. Nitrite exposure did not affect the hepatic drug-metabolic activities in juvenile Israeli carp. All these data indicate that disposition of NO$_2$- differ depending upon exposed concentration and that metHb production may not be the exclusive toxic mechanism in carp.

Comparison of Acute Toxicity of Pesticides between Carp(cyprinus carpio L.) and Israeli Carp(Cyprinus israeli carpio L.) (농약(農藥)에 의한 참잉어 및 이스라엘잉어의 급성독성비교(急性毒性比較))

  • This study was carried out to compare the acute toxicity(96hr) of 13 chemicals to carp (Cyprinus carpio L.) and israeli carp (Cyprinus israeli carpio L.) and the activities of acetylcholinesterase (AchE) and glutathione S-transferase (GST) in israeli carp exposed to five insecticides (diazinon, malathion, carbofuran, cartap, methomyl). $LC_{50}$ values of acute toxicity of the chemicals to israeli carp were endosulfan 0.0061ppm, captafol 0.041ppm, chlorothalonil 0.073ppm, butachlor 0.48ppm, captan 0.14ppm, carbofuran 1.13ppm, cartap 1.15ppm, diazinon 1.35ppm, nitrofen 3.72ppm, methomyl 4.39ppm, propanil 10.61ppm, malathion 11.78ppm and isoprothiolane 12.81ppm. The acute toxicity of endosulfan 0.0061ppm was 2100 times higher than that of isoprothiolane 12.81ppm. $LC_{50}$ values of acute toxicity of the chemicals to carp were endosulfan 0.0026ppm, captafol 0.062ppm, chlorothalonil 0.078ppm, captan 0.14ppm, and butachlor 0.47ppm, carbofuran 0.52ppm, nitrofen 0.58ppm, diazinon 0.81ppm, cartap 0.82ppm, methomyl 5.03ppm, propanil 10.67ppm, malathion 11.92ppm, and isoprothiolane 13.20ppm. The acute toxicity of endosulfan was 5,000 times higher than that of isoprothiolane. The toxicity of diazinon, carbofuran, cartap, endosulfan, and nitrofen to carp was approximately 2-6 times as high as that to israeli carp, but the toxicity of malathion, methomyl and captafol to israeli carp was slightly higher than that to carp. AchE activity was inhibited by 31% and 52% after 96hr’s exposure of israeli carp to diazinon and malathion respectively. GST activity in israeli carp was significantly induced by methomyl exposure for 96 hr.

Comparison of Phytoplankton Growth and Species Composition in Pangasiid Catfish Monoculture and Pangasiid Catfish/Silver Carp Polyculture Ponds

  • Excessive growth of phytoplankton is a common and severe problem in intensively farmed pangasiid catfish (Pangasius hypophthalmus) culture ponds. It can lead to cyanobacterial blooms, reduced fish growth, bad-tasting fish flesh, and lower market demand. To investigate how to manage undesirable phytoplankton growth, we evaluated three stocking strategies in nine rural fishponds (0.020-0.022 ha) owned by various farmers: a pangasiid catfish mono culture (treatment 1, $T_1$), and pangasiid catfish-silver carp (Hypophthalmichthys molitrix) polycultures at two stocking ratios of 1:1 (treatment 2, $T_2$) and 2:1 (treatment 3, $T_3$). The total density of all ponds was approximately 30,000 fishes/ha. Monoculture ($T_1$) resulted in significantly higher (p < 0.05) nutrient levels (nitrate and phosphate) in ponds than did polyculture ($T_2$ and $T_3$). Nutrient loads increased with culture time, resulting in increased growth of phytoplankton, including Cyanophyceae (9 genera), Chlorophyceae (15 genera), Bacillariophyceae (8 genera), and Euglenophyceae (3 genera). The introduction of silver carp as a co-species helped to regulate phytoplankton growth and to improve the water quality of pangasiid catfish culture ponds.

Detection of DNA Damage in Carp Using Single-Cell Gel Electrophoresis Assay for Genotoxicity Monitoring

  • To investigate the potential application of the single-cell gel electrophoresis (SCGE) assay to carp as an aquatic pollution monitoring technique, gill, liver, and blood cells were isolated from carp exposed to a direct-acting mutagen, N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), or indirect mutagen, $benzo[\alpha]pyrene$ $(B[\alpha]P)$, then the DNA strand breakage was analyzed using the assay. Based on testing 5 different cell isolation methods and 6 electrophoretic conditions, the optimized assay conditions were found to be cell isolation by filter pressing and electrophoresis at a lower voltage and longer running time (at 0.4 V/cm for 40 min). In preliminary experiments, gill and liver cells isolated from carp exposed to MNNG in vitro exhibited DNA damage signals even with 0.5 ppb exposure, which is a much higher dose than previously reported. In the gill cells isolated from carp exposed to 0.01-0.5 ppm MNNG in vivo, significant dose-and time-dependent increases were observed in the tail for 4 days. As such, the linear correlation between the relative damage index (RDI) values and time for each dose based on the initial 48-h exposure appeared to provide effective criteria for the genotoxicity monitoring of direct-acting mutagenic pollution. In contrast, the in vivo exposure of carp to 0.25-1.0 ppm of $B[\alpha]P$ for 7 days resulted in dose-and time-dependent responses in the liver cells, in which 24-h delayed responses for metabolizing activation and gradual repair after 48 h were also observed. Thus, the negative-sloped linear correlation between the RDI and time at each dose based on the initial 48 h appeared to provide more effective criteria for the genotoxicity monitoring of indirect mutagenic pollution.

Comparisons of amino acid sequences of ${\beta}$-globin gene between carp and other vertebrates (잉어와 척추동물들의 ${\beta}$-globin 아미노산배열의 비교)

  • The purpose of this study was to understand the evolutionary relationships between fish and other vertebrates which had DNA with the genetic defects in homoglobin expression, with comparison to the nucleotide homologies of the ${\beta}$-globin genes. The predicted amino acid sequence from carp ${\beta}$-globin gene was compared with those of other vertebrates from the published data. The nucleotide homologies of the predicted amino acid sequence from the carp ${\beta}$-globin gene with those of goldfish and mirror carp were high, and the rates were 97.3% and 93.9%, respectively. On the other hand, with the previously reported ${\beta}$-globins of goat, frog, human, rat, goose, chicken, and duck, it showed low homology ranging from 45.9 to 58.1%. The carp ${\beta}$-globin has one inserted amino acid residue, which was also found in other fish ${\beta}$globin, but not in other vertebrate ${\beta}$-globins.

In Vivo Metabolic studies on Carbofuran Degradation in carp(Cyprinus carpio L.) (Carbofuran 의 잉어(Cyprinus carpio)체내 대사)

  • Absorption, distribution, metabolism and excretion of $^{14}C-carbofuran$(2,3-dihydro-2,2-dimethyl-7-benzofuranyl methylcarbamte) were studied in carp(Cyprinus carpio L.) after the treatment of carbofuran at the dose level of 43 parts per billion. Maximum radioactivities in tissues(liver, kidney, gut, gall bladder) and blood of carp were shown 12hrs after the treatment of $^{14}C-carbofuran$. Carbofuran was metabolized to 3-hydroxycarbofuran and 3-ketocarbofuran in liver and kindney of carp, and the major metabolite was 3-hydroxycarbofuran. Most radioactivity absorbed into the carp tissues was eliminated 3hrs after transfer of the carp to fresh water. The excretory metabolites were 3-ketocarbofuran(32.3%), 3-hydroxycarbofuran(52.8%) and an unknown metabolite(2.6%) during the period of 3hrs of the excretory experiment.

Genetic Comparison Between Crucian Carp (Carassius auratus Linnaeus) and Crucian Carp (C. cuvieri Temminck and Schlegel) (붕어(Carassius auratus Linnaeus)와 떡붕어(C. cuvieri Temminck and Schlegel)의 유전적 비교)

  • Genomic DNA isolated from two crucian carp species obtained from Yesan (Carassius auratus) and Dangjin (Carassius cuvieri) in Korea were amplified at several times by polymerase chain reaction (PCR). The amplified products were separated by agarose gel electrophoresis (AGE) with oligonucleotides decamer primer and stained with ethidium bromide. The seven arbitrarily selected primers OPC-11, OPC-14, OPC-18, OPD- 02, OPD-11, OPD-15 and OPD-20 generated the shared loci by each species, the polymorphic and specific loci. The seven primers generated the total 458 loci that can be scored from the crucian carp obtained in C. auratus species. 358 fragments were generated from the species obtained in C. cuvieri species. The size of DNA fragments varies from 150 to 1,600bp. The complexity of the banding patterns varies dramatically between the primers and two locations. In this study, 458 loci were identified in the crucian carp species from Yesan and 358 in the crucian carp species from Dangjin: 84 polymorphic loci (18.3%) in the C. auratus species and 48 (13.4%) in the C. cuvieri species. 154 shared loci by each species, the average 22 per primer, were observed in the C. auratus species and 187 loci, the average 26.7 per primer, in the Dangjin species. Based on the average bandsharing (BS) values of all samples, the similarity matrix ranged from 0.434 to 0.868 in the C. auratus species and from 0.449 to 0.924 in the C. cuvieri species. The average BS value was 0.641±0.013 within the C. auratus species and 0.684±0.013 within the C. cuvieri species. The average BS value between two crucian carp species 0.484 ± 0.007, ranged from 0.307 to 0.682. The BS value between the individual No. 09 and No. 16 was 0.682, which was the highest between two crucian carp species. Compared separately, the BS value of individuals within the C. cuvieri species was higher than the C. auratus species. The dendrogram obtained by the seven primers, indicates three genetic clusters: cluster 1 (AURATUS No. 01, 02, 03, 04, 05, 06, 07, 08, 09, 10 and 11), cluster 2 (CUVIERI No. 12, 13, 14, 15, 16, 17, 18, 19, 20 and 21) and cluster 3 (CUVIERI no. 22). The shortest genetic distance displaying significant molecular difference was between the individual AURATUS No. 09 and AURATUS No. 08 from Yesan (genetic distance=0.064). The longest genetic distance displaying significant molecular differences was between the individual CUVIERI No. 17 and AURATUS No. 11 between two crucian carp species (0.477). Inclusion of Pangasiid Catfish in Polycultures of Major Indian Carps (Catla, Rohu and Mrigal) Increases Yield and Economic Gain

  • An experiment was conducted in twelve 0.02-ha rain-fed earthen ponds for 18 weeks to evaluate the effects of including pangasiid catfish (Pangasius hypophthalmus) in polycultures of major Indian carps, catla (Catla catla), rohu (Labeo rohita), and mrigal (Cirrhinus cirrhosus), on yield and economic gain. Treatment 1 ($T_1$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal per ha, treatment 2 ($T_2$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+2,500 pangasiid catfish per ha, treatment 3 ($T_3$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+5,000 pangasiid catfish per ha, and treatment 4 ($T_4$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+7,500 pangasiid catfish per ha. The growth performance of fishes was evaluated by calculating specific growth rate, daily weight gain, and percent weight gain. Inclusion of pangasiid catfish in the carp polyculture resulted in significant increases in the growth and yield of catla and rohu. Catla and rohu grew most in $T_4$, with 7,500/ha of pangasiid catfish. The presence of pangasiid catfish did not influence the growth of mrigal. The growth of pangasiid catfish varied with stocking density, and they grew best at 5,000/ha ($T_3$). The carp and the combined fish yields were significantly higher in the carp polycultures with pangasiid catfish than with carps alone. The total carp and the combined fish yields were highest in $T_4$, in which the density of pangasiid catfish was greatest. However, the total carp and the combined fish yields in $T_3$ and $T_4$ were not significantly different. Profit differed significantly among treatments. Economic gain was highest in $T_3$, with 5,000/ha of pangasiid catfish, and lowest in $T_1$. The inclusion of pangasiid catfish in major carp polycultures results in higher fish yields and greater economic gain.
