Characteristics and breeding of a thermotolerant ear mushroom, Auricularia auricula-judae 'Hyeonyu' (고온적응성 목이버섯 「현유」 육성 및 특성)

  • Although ear mushroom (Auricularia auricula-judae) is cultivated worldwide, there are a limited number of commercial cultivars in Korea. Recent increase in the import of ear mushroom from China threatens the domestic farming. The present study introduces a new thermotolerant cultivar of ear mushroom, designated 'Hyeonyu', developed by mating monokaryons obtained from JBAA11 and CAA1 strains. The optimal growth temperature for Hyeonyu was 26-36?. The periods of primordia formation and fruit-body growth were 27 and 15 days, respectively. The average mushroom yield per bag (1 kg) was 350 g. Random amplification of polymorphic DNA (RAPD) analysis with OPA1 and OPA7 primers identified polymorphic DNA bands between the control, the new variety Hyeonyu, and a Chinese variety.

Genetic DNA Marker for A2 mating type in Phytophthora infestans

  • The Phytophthora infestans requires two mating types for sexual reproduction. Amplified fragment length polymorphism (AFLP) was used to specifically detect different mating types of P. infestans. The AFLP primers E+AA (5'-GACTGCGTACCAATTCAA-3') and M+CAA (5'-GATGAGTCCTGAG-TAAC AA-3') detected a fragment that is specific in the A2 mating type of P. infestans. This fragment was cloned and sequenced. Based on the sequence data, PHYB-1 and PHYB-2 primer were designed to detect the A2 mating type of P. infestans. A single 347 bp segment was observed in the A2 mating type of P. infestans, but not in the A1 mating type of P. infestans or other Phytophthora spp. Identification of mating type was performed with phenotype (sexual reproduction) and genotype (CAPs marker) methods. Two factors, the annealing temperature and template DNA quantity, were investigated to determine the optimal conditions. Using mating type-specific primers, a unique band was obtained within annealing temperatures of 57$^{\circ}C$-62$^{\circ}C$ and DNA levels of 10pg-100 ng (data not shown).

Review of PM-related Air Quality Improvement Policies of United States for PM-related Air Quality Improvement of Metropolitan Region in Korea (수도권 미세먼지 환경 개선을 위한 미국의 대기환경정책 사례 조사 연구)

  • Several fine particle-related policies in Northeast United States were investigated in support of the execution of special measures to improve air quality of Seoul Metropolitan Area (SMA). The definition of particulate matter (PM) in the Clean Air Act (CAA), components and procedures of Regional Haze Rule (RHR), and Air Quality Management (AQM) were reviewed. Several State Implementation Plans (SIPs) were also reviewed as the way to attain required air quality under the Clean Air Act. $PM_{2.5}$ attainment SIP of Maryland, 8-hr Ozone attainment SIP of New Jersey, and Regional Haze Rule attainment SIP of MANE-VU were analyzed in detail as case studies. We realized that "Special Measures for Air Quality Improvement in the Seoul Metropolitan Area" has many similarities with its US counterparts in terms of purpose, components, procedures, and implementation methods. US policies, however, have more advanced features, such as standardized procedures and methods, transparent guidelines, and stable relationship among federal/state/local governments and stakeholders, which would be helpful to improve air quality in SMA.

  • The physical phenomena of rectangular open cavity are numerically investigated in this paper Two-dimensional cavity problems with laminar boundary layers in upstream are simulated by using the compressible Wavier-Stokes equations. The high-order and high-resolution numerical schemes are used for the evaluation of spatial derivatives and the time integration. Cross-correlation is used to analyze the characteristics of wave propagation along time and spatial. Sudden phase shifting of 90 degrees is appeared near downstream edge, and this is coincident with the phase lag suggested in original Rossiter's equation. The results give a further understanding of the physical phenomenon of noise generation, and the resonance of flow and acoustic in cavity. Moreover, modified Rossiter's equation, which is more accurate and can be applied in various conditions, is suggested. The distance from the point of vortex generation to the point of vortex collapsing acts as effective distance of cavity resonance, and the phase difference between the point of vortex collapsing and the point of acoustic source acts as phase lag. Study on Pressure Variation around an Open Cavity (공동 주위에서의 압력 변화에 대한 연구)

  • Cavity tone is generated due to the feedback between flow and acoustic wave. It is recognized that the period is determined by the time required for the flow convection in one direction, the time required for the acoustic propagation in the other direction and the time for phase shift depending on the flows and mode. Most of the phenomena have been investigated by experiments and a simple but fundamental theory. But the cause of the phase shift and the correctness of the theory have not been clearly explained so far. In this paper, the phenomena are calculated numerically to obtain detail information of flow and acoustic wave to explain the mechanism including the phase. High order high resolution scheme of optimized high order compact is used to resolve the small acoustic quantities and large flow quantities at the same time. The data are reduced using cross correlation function in space and time and cross spectral density function which has phase information. Abrupt change in pressure near corner in cavity is observed and is relate to phase variation. The time required for the feedback between the flow and acoustic wave is calculated after the numerical simulation f3r various modes. The periods based on the time calculated using the above method and direct observation from the acoustic waves generated and propagated in the numerical simulation are compared. It is found that no phase shift is required if we examine the time required carefully. Rossiter's formula for the cavity tone used for quick estimation needs to be modified far some modes.

Prediction of internal broadband noise of a centrifugal fan using stochastic turbulent synthetic model (통계적난류합성모델을 이용한 원심홴 내부 광대역 소음 예측)

  • The internal broadband noise of a centrifugal fan in a household refrigerator is predicted by using hybrid CAA technique based on stochastic turbulent synthetic model. First, the unsteady flow field around the centrifugal fan is predicted using Computational Fluid Dynamics (CFD) method. Then, the turbulent flow field is synthesized by applying the stochastic turbulent synthetic technique to the predicted flow field. The aerodynamic noise sources of the centrifugal fan are modeled on a basis of the synthesized turbulent field. Finally, the broadband noise of the centrifugal fan is predicted using Boundary Element Method (BEM) and the modeled sources. The predicted result is compared with the experimental data. It is found that the predicted result closely follows the experimental data. The proposed method can be used as an effective tool for designing low-noise fans without expensive computational cost required generally for the LES and DNS simulations to resolve the turbulence flow field responsible for the broadband noise.

  • Today, high-speed trains enjoy wide acceptance as fast, convenient and environment-friendly means of transportation. However, increase in the speed of the train entails a concomitant increase in the aerodynamic noise, adversely affecting the passenger comfort. At the train speed exceeding 300 km/h, the effects of turbulent flows and vortex sheddding are greatly amplified, contributing to a significant increase in the aerodynamic noise. Drawing a biomimetic analogy from low-noise flight of owl, a method to reduce aerodynamic noise at inter-coach space of high-speed trains is investigated. The proposed method attempts to achieve the noise reduction by modifying the turbulent flow and vortex shedding characteristics at the inter-coach space. To determine the aerodynamic noise at various train speeds, wind tunnel testing and numerical CFD (Computational Fluid Dynamics) simulation for the basic inter-coach spacing model are carried out, and their results compared. The simulation and experimental results reveal that there are discrete frequency components associated with turbulent air flow at constant intervals in the frequency domain

  • The paper describes the prediction method for the unsteady flow field and the aeroacoustic noise of an small axial fan. The prediction method is comprised of various CFD conditions and acoustic analogy by using Ffowcs Williams-Hawkings equation. The diameter of tested axial fan is 170 mm and number of blade is 5. Virtual anechoic room which has same size with real one was used for CFD. URANS and LES models were used. For mesh dependence study, a different mesh type was tested and optimized mesh was selected. Calculation conditions were also studied such as time step and turbulence model for accurate noise analysis. In this paper, we got optimum analysis conditions and computational results. The unsteady pressure fluctuation at given 4 points were compared between the measured data and computational results. Also, the predicted acoustic spectrum at 3 given microphone points were compared with measured ones.

Biochemical Characterization of Glucose-Regulated Proteins, Grp94 and Grp78/BiP (Grp78/BiP과 Grp94의 생화학적 분석)

  • Glucose-regulated proteins (grp's), srp94 3nd grp78/BiP, are a group of stress proteins which are highly synthesized in cells exposed to a variety of stressful agents including tunicamycin 3nd Ca2+ ionophore. Grp78/BiP is hon to function as a molecular chaperone which regulates the folding and assembly of secretory or membrane proteins, but the biological function of grp941 remains to be elucidated. In this study, we have examined the intracellular distribution of grV's and the function of srp94. Grp's are resident in the endoplasmic reticulum (ERI 3nd a specific sequence (Lys-Asp-Glu-Leu) at their C-terminus is known to be responsible for their retention within the ER. However, it has been unclear whether upon disturbance of cellular Caa+ homeostasis by the Ca2+ ionophore, grp94 is retained within the ER or secreted into the medium. In this study, we showed that in the presence of C3a+ ionophore, grp94 and gif78/BiP are present in the cells, mainly within the ER. We have also investigated whether grp94 might function as a molecular chaperone. Here we showed that in the immunoglobulin (Ig)-secreting hvbridom3 cells, grp94 transientlY interacts with fully glycosylated Is heavy chain, suggesting that grpg94 may be involved in facilitating the folding and assembly of Ig heavy chains.

Cloning and Sequence Analysis of Ribosomal Protein S4 cDNA from Root of Panax ginseng

  • Ribosomal protein complex with ribosomal RNA to form the subunits of the ribosome serve essential functions in protein synthesis. A full-length cDNA (PRPS4) encoding ribosomal protein S4 has been isolated and its nucleotide sequence determined in ginseng plant (Panax ginseng). A PRPS4 cDNA is 1105 nucleotides long and has an open reading frame of 792 bp with a deduced amino acid sequence of 264 residues (pI 10.67). The deduced amino acid sequence of PRPS4 matched to the previously reported ribosomal protein S4 genes. Their degree of amino acid identity ranged from 68 to $92\%$. Phylogenetic analysis based on the amino acid residues showed that the PRPS4 grouped with ribosomal protein S4 of S. tuberosum (CAA54095).
