Functional characterization of a CCCH type zinc-finger protein gene OsZF2 by ectopic overexpression of the gene in rice (과발현 형질전환벼에서 CCCH type zinc-finger protein 유전자 OsZF2 기능 분석)

  • We have previously isolated a CCCH type zinc-finger protein gene, OsZF2 (Oryza sativa Zinc Finger 2), from the cold-treated rice cDNA library. To investigate the potential role of OsZF2, transgenic rice lines over-expressing OsZF2 under the control of CaMV 35S promoter have been developed through Agrobacterium-mediated transformation. Elevated level of OsZF2 transcripts was confirmed by RNA gel blot analysis in transgenic rice. Under the 100 mM NaCl condition, the transgenic rice showed significantly enhanced growth rate in terms of shoot length and fresh weight, implicating that OsZF2 is likely to be involved in salt response of rice. In the field condition, however, the transgenic rice showed a dwarf phenotype and flowering time was delayed. Genome expression profiling analysis of transgenic plants using the 20K NSF rice oligonucleotide array revealed many up-regulated genes related to stress responses and signaling pathways such as chaperone protein dnaJ 72, salt stress-induced protein, PR protein, disease resistance proteins RPM1 and Cf2/Cf5 disease resistance protein, carbohydrate/ sugar transporter, OsWAK kinase, brassinosteroid LRR receptor kinase, and jasmonate O-methyltransferase. These data suggest that the CCCH type zinc-finger protein OsZF2 is a upstream transcriptional factor regulating growth and stress responsiveness of rice.

Vermicomposting of Leather Waste Sludge by Earthworm, Lumbricus rubellus (I) (지렁이를 이용(利用)한 피혁(皮革) 슬러지의 퇴비화(堆肥化)에 관(關)한 연구(硏究)(I))

  • The study on the ripening of leather waste sludge was performed to vemistabilize the sludge effectively using a laboratory scale darkened wood box reactor ($10{\times}10{\times}20cm$). The acceptable feed conditions for earthworm of Eh, alkalinity were obtained in the 30th and 35th days, respectively. The value of pH was gradually decreased from 7.8 to 7.1 through the ripening time. The contents of heavy metals of the ripened sludge were estimated as lower levels compared to the other regulatory standards for compost. After the ripening time for 50days, the physico-chemical properties of the sludge were estimated as pH 7.1, water content 72%, redox potential 85mV, electrolytic conductivity $2,620{\mu}mhos/cm$ and alkalinity 450 ppm as $CaCO_3$, respectively. In the ripened sludge, survival rate of the earthworms for 50 days was about 75%, and the live weight increase rate was about 230% at the temperature range of $20{\sim}25^{\circ}C$. Moreover, hatching rate of the earthworm cocoons was about 87% and the highest value was obtained in the 20~30th days. From the above results, it was evaluated that leather waste sludge could be vermistabilized effectively by earthworms, when the sludge was ripened during 50 days.

Evaluation of Phosphorus Acid Treatment as a Growth Stimulant for Red pepper (Capsicum annuum L.), Cucumber (Cucumis sativus L.), and Kimchi cabbage (Brassica campestris L. ssp. pekinensis) in the Bed Soil Environment (상토 환경에서 고추(Capsicum annuum L.), 오이(Cucumis sativus L.) 및 배추(Brassica campestris L. ssp. pekinensis)에 대한 생장촉진제로서 아인산 처리의 평가)

  • This study was conducted to evaluate the effect of phosphorus acid (H3PO3) addition to the horticultural bed soil on the initial growth of red pepper (Capsicum annuum L. cv.), cucumber (Cucumis sativus L. cv.), and kimchi cabbage (Brassica campestris L. ssp. pekinensis (Lour.) Rupr. cv.). The stem heights of red pepper and cucumber were 46.1% and 23.0% greater in the 50 mg/L of phosphorus acid treatment than the untreated (control). Further, the stem diameter of pepper and cucumber were 48.7% and 23.0% greater in the 50 mg/L of phosphorus acid treatment than the control. In addition, the number of kimchi cabbage leaves was 47.5% greater in the 50 mg/L of phosphorus acid treatment than the control. The dry weights of red pepper, cucumber and kimchi cabbage were 72.9%, 16.5%, and 30.4% heavier in the 50 mg/L than the control, respectively. Cations (K, Ca, and Mg) and total phosphorus (T - P) were quantitatively analyzed for these three horticultural crops. The concentrations of K, Ca, and Mg, and T - P were higher in the 50 mg/L of phosphorus acid than the control, respectively. Based on the results obtained in this study, it appears that treatment of phosphorus acid in horticultural bed soil enhanced the growth of red pepper, cucumber and Kimchi cabbage.

Nutritional Components and Antioxidant Activities of Commercial Loquat (Eryobotrya japonica) Leaf Tea (시판 비파나무잎차의 영양성분 및 항산화 활성)

  • Nutritional components and antioxidant activities of commercial loquat leaf tea (CLLT) were evaluated. Proximate compositions were as follows; moisture 5.34%, crude protein 8.38%, crude fat 13.26%, nitrogen free extract 46.21%, crude fiber 19.24% and crude ash 7.57%, respectively. Ascorbic acid and total phenolics content of CLLT was 0.47 mg/100 g and 42.55 mg/GAE g, respectively. Mineral elements were Ca 1,624.01 mg/100 g, K 1,099.66 mg/100 g, and Mg 192.70 mg/100 g, respectively. Amino acid contents of CLLT were very rich in glutamic acid 565.98 mg/100 g and deficient in cystine 12.97 mg/100 g. The 2,2'-azino-bis (3-ethylbenzthiazoline-6- sulfonic acid) diammonium salt (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities of hot water extract from CLLT were 72.51% and 90.11%, respectively at a concentration of $1,000{\mu}g/mL$. Reducing power and ferric reducing antioxidant power (FRAP) of hot water extract from CLLT were increased in a dose dependent manner. Therefore, these results suggest that the hot water extract of CLLT possess antioxidant activities and thus it has great potential as a source for functional food such as natural antioxidant.

Secondary Metabolites and Morphological Diversity in the Leaves of Perilla Landrace from Korea

  • Screening and identification of genetic resources based on their phytoconstituents and morphological characters potentially provide baseline data for researchers, breeders, and nutraceutical companies who wish to formulate a nutrient-dense diet and health beneficial supplement. Thus, we evaluated the amount of total phenolic content and major phenolic compounds; examined if phenolic compounds could be used as distinguishing factors for perilla genetic resources; and investigated the relation between some quantitative and qualitative morphological characters with the contents of phenolic compounds in 360 accessions obtained from National Agrobiodiversity Center gene bank, Jeonju, Korea. Total phenolic content (TPC) was estimated using Folin-Ciocalteu colorimetric assay. Individual phenolic compounds were determined using an Ultra-High Performance Liquid Chromatography system equipped with Photodiode Array detector. Considerable variations were observed in TPC (7.99 to 117.47 mg GAE/g DE), rosmarinic acid (RA) (ND to 19.19 mg/g DE), caffeic acid (CA) (ND to 0.72 mg/g DE), apigenin-7-O-diglucuronide (ADG) (ND to 1.24 mg luteolin equivalent (LUE)/g DE), scutellarein-7-O-glucuronide (SG) (ND to 4.32 mg LUE/g DE), and apigenin-7-O-glucuronide (AG) (ND to 1.60 mg LUE/g DE). RA was the most dominant phenolic compound in most accessions (95.3%) followed by SG. The adaxial leaf color was light green, green and dark green in 13.8%, 65.0%, and 21.1 % of the accessions, respectively. 78.8% of the accessions had light green color at the abaxial side with the remaining being described as green. Most of the accessions (96.9%) were cordate shape, the remaining being eclipse. Intensities of green pigment at abaxial and adaxial leaf surfaces were correlated with contents of individual phenolic compounds and TPC whereas leaf length and width had no correlation with TPC, CA and RA, and negatively correlated with ADG, AG, and SG. Leaf shape was not related with content of phenolic compounds, color of leaves, or the length or width of leaves. Accessions IT57426, IT157434, IT267710, and IT267712 which contained relatively high contents of TPC and major phenolic compounds (RA and SG) could be used for further research in breeding and bioassay test. Our study result showed the contents of total phenolics and individual phenolic compounds along with the morphological characters could be useful distinguishing factors for perilla genetic resources.

Studies on the Compositions of Mineral Components, Total Amino Acid, Free Amino Acid and Fatty Acid of Zizyphus jujuba Seed (대추씨(Zizyphus jujuba Seed)의 무기질, 구성 총 아미노산, 유리 아미노산 및 지질 구성 지방산조성에 관한 연구)

  • Chemical components of the physiological activity substance in Zizyphus jujuba seed were analyzed for the use as an functional processed products. The proximate compositions of Zizyphus jujuba seed were carbohydrate 89.40%, moisture 8.23%, crude protein 1.22%, crude fat 0.65% and crude ash 0.50%, respectively. the richest mineral contained in Zizyphus jujuba seed was Mg(198.3mg/100g), followed by Mn(113.0mg/100g), K(100.2mg/100g), Ca(25.3mg/100g), P(25.6mg/100g_, Fe(23.5mg/100g), Zn(18.7mg/100g), Cu(7.4mg/100g) and Na(5.3mg/100g). Total amino acid contents of Zizyphys jujuba seed were 1103.12mg/100g, and the major amino acids were proline(235.17mg%), aspartic acid(175.941mg%), gltamic acid(101.46mg%), leucine(50.27mg%), serine(47.71mg%), aspartic acid(47.27mg%), glycine(41.60mg%) and lysine (33.78mg%), respectively. The amount of free amino acids in the protein of Zizyphus jujuba seed were 178.62 mg/100g, and the major free amino acids were proline(121.30mg%), alanine(6.01mg%), valine(2.63mg%) and arginine(2.04mg%), respectively. The compositions of major total fatty acid in the lipid Zizyphus jujuba seed were C18:2(n5c) 28.72%, C18:1(n9t) 23.76%, C18:1(n9c) 13.45%, C16:0 13.45% and C20:5(n3) 1.77% in order, respectively.

Physicochemical Properties and Antioxidative Activities of Omija(Schizandra chinensis Bailon) (오미자의 이화학적 특성 및 항산화 활성)

  • To develop physiological functionality of Omija extracted with water was evaluated on antioxidative activity. Omija, high acid material with pH 3.6, contain $57.5{\pm}1.03%$ of moisture, and $18.8{\pm}0.12%$ of crude fat. This material have $12.6{\pm}0.04%$ of carbohydrate and $11.1{\pm}0.07%$ of crude protein as well, but ash and crude protein contents were found less than 10%. 10 mineral contents were also found, too; K and Ca showed the highest level, then Al, Mg, Na and Mn were followed. In composition amino acid contents, glutamic acid took the largest portion, $131.7{\pm}1.3$ mg/100 g, aspartic acid $51.5{\pm}0.6%$, and other composition amino acid under 50%. In case of free sugar contents, 7 types were found. Most of them were glucose and fructose. Total phenolic compounds showed the highest level, $2,862.6{\pm}31.7$ mg/100 g. $197.8{\pm}14.6$ mg/100 g of flavonoid and $225.6{\pm}18.2$ mg/100 g were included. In terms of electron donating ability, radical scavenging ability activated as the amount of Omija extract increased. In particular, Omija extract in 1,000 ${\mu}g/m{\ell}$ demonstrated almost similar electron donating ability, $72.4{\pm}0.21%$, to BHT. It was also found that antioxidant activities of electron donating ability, SOD-like ability, hydroxyl radical scavenging ability and nitrite scavenging ability were highly promoted as Omija extract concentration increased. The nitrite scavenging ability was significant when the extract belonged to strong acid region and doping concentrations increased.

The Associations of Household food Insecurity with Socioeconomic Status, Food Behaviors, Health Status and Nutrient Intake in the Elderly in Rural Areas (일부 농촌노인의 식품안정성과 사회경제학적 특성, 식생활, 건강상태, 영양소 섭취와의 관련성)

  • The purpose of this study was to examine household food insecurity and the associations of food insecurity with socioeconomic characteristics, food behaviors, health status and nutrient intake among elderly people in a rural area. This study included 191 elderly people (mean age = $72.0{\pm}7.4$) in Jeollabuk-do, Jangsu-gun. House food insecurity was measured using an adapted version of the USDA short form household food insecurity scale, and nutrient intake measured by a 24-hour dietary recall. Only 15.2% of the subjects were in food-insecure households, 84.8% of the elderly households were food-secure. The food-insecure households were significantly lower in the number of family, frequency of shopping, and perceived health status than the food-secure households. In addition, they had serious tooth problems and NSI (Nutrition Screening Index) scores. Food-insecure households were significantly lower in most areas of nutrient intake (energy, protein, fat, Ca, P, Fe, K, Na, Zn, Vit. $B_2$, niacin, Vit. E), and also lower in nutrient intake compared to dietary reference intake (energy, protein, P, Fe, K, Na, Vit. $B_1$, Vit. $B_2$, niacin, Vit. E). These results suggest that the household food insecurity measures used in this study were an important indicator of nutritional well-being among elderly persons in the rural area. Food insecurity should be considered an important issue to public health and food service programs should be expanded for food-insecure households in rural areas.

Dietary Modification for Reducing Electrical Conductivity of Piggery Wastewater

  • A total of 108 pigs (including 36 starters, 36 growers, and 36 finishers) were randomly allocated to six treatments, which involved a 2 (Crude Protein (CP): 100 and 80% of control diet)${\times}$3 (Ca, P, Salt (CPS): 100, 80 and 60% of control diet) factorial design to evaluate the effectiveness of reducing CP and CPS in reducing wastewater EC in different stages. Another 72 starters were adopted to examine the effect of the six treatment diets (as mentioned above) on the growth performance of pigs. Activated carbon and Reverse Osmosis System (RO) were adopted to examine the reducing efficiency of wastewater EC, and ion analysis was also applied to compare with the wastewater EC in different stages of the metabolism trial. The results of wastewater EC of the six treatment diets in different stages of metabolism trial demonstrated that diminishing dietary CP or CPS decreased wastewater EC. The largest decrease of EC was approximately 30%, and was achieved with 20 and 40% reduced dietary CP and CPS, respectively. Pig growth performance deteriorated somewhat when dietary CP or CPS was diminished. Wastewater ion concentration was not always consistent with dietary CP or CPS content, except for $NO_2^{-}$, $NH_4^{+}$ and $K^{+}$, which were positively correlated with dietary CP or CPS in different stages. Activated carbon is not effective for reducing wastewater EC, while, RO system is effective (90% elimination rate) in reducing wastewater EC, but the EC of concentrated (excreted) water is around 10% higher than that of intact wastewater, representing an additional problem besides the high cost of RO system treatment.

Purification and Anticoagulant Activity of a Fucoidan from Korean Undaria pinnatifida Sporophyll

  • Crude fucoidan was extracted from the sporophyll of Korean Undaria pinnatifida collected at a coastal area ofWando, Korea, mainly by dilute acid extraction, ethanol precipitation, CaCU Precipitation, with an yield of approxi-mately 3.9% in mass. It was further purified by DEAE-cellulose column chromatography and its chemical composi-don and in vitro anticoagulant activity was determined. The average molecular mass of the purified fucoidan wasestimated about 2.1 x 103 kDa by size-fractionation HPLC and it consisted of neutral sugar (52.34% in mass), uronicacid (26.2%), and sulfate esters (7.4%). From the HPAEC-PAD analysis, the monosaccharide composition of thepurified fucoidan was shown to be fucose, galactose, xylose, and mannose, with a molar ratio of 1, 0.2, 0.02, 0.15,respectively, demonstrating that major monosacd-iande was fucose (72.3% in mol percentage) and other sugars,xylose (1.5%), galactose (14.6%), and mannose (10.9%) were present as minor component. The results suggested thatthis fucoidan is a sulfated, U-type fucoidan. The activated partial thrombloplastin time (APTT) assay of the purifiedfucoidan showed that the purified fucoidan elicited anticoagulant activity in a dose-dependent manner. Five jUg ofsporophyll fucoidan delayed the blood clotting time up to 5 times than untreated control and also up to 1.5 timesthan the same amount of the commercial fucoidan, respectively. Although it is preliminary, these results suggestthat the fucoidan of Korean Undaria vinnatifida sporophyll would be promising candidates for the development ofan anticoaeulant.
