Animal models have been used to elucidate the pathophysiology of varying diseases and to provide insight into potential targets for therapeutic intervention. Although alternatives to animal testing have been proposed to help overcome potential drawbacks related to animal experiments and avoid ethical issues, their use remains vital for the testing of new drug candidates and to identify the most effective strategies for therapeutic intervention. Particularly, the study of metabolic diseases requires the use of animal models to monitor whole-body physiology. In line with this, the National Institute of Food and Drug Safety Evaluation (NIFDS) in Korea has established their own animal strains to help evaluate both efficacy and safety during new drug development. The objective of this study was to characterize the response of C57BL/6NKorl mice from the NIFDS compared with that of other mice originating from the USA and Japan in a chemical-induced diabetic condition. Multiple low-dose treatments with streptozotocin were used to generate a type-1 diabetic animal model which is closely linked to the known clinical pathology of this disease. There were no significantly different responses observed between the varying streptozotocin-induced type-1 diabetic models tested in this study. When comparing control and diabetic mice, increases in liver weight and disturbances in serum amino acids levels of diabetic mice were most remarkable. Although the relationship between type-1 diabetes and BCAA has not been elucidated in this study, the results, which reveal a characteristic increase in diabetic mice of all origins are considered worthy of further study.

Di-n-butyl phthalate (DBP) is not only a plasticizer and solvent used in industry but also one of endocrine disruptor chemicals, a low level contaminant found in a wide variety of different media ranging from drinking water to infant formulae. To evaluate the cytotoxic function of NK cells in mice after contact with DBP, C57BL/6 female mice were orally dosed with di-n-butyl phthalate (250, 500, or 750 mg/kg body weight) for 14 consecutive days, and the control mice were administered vehicle (corn oil).(omitted)
This study was carried out to investigate the effect of Origanum vulgare extracts on cell proliferation of human hair dermal papilla cell (HHDPC) using sulforhodamine B (SRB) assay, antioxidant activity by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method, expression of insulin-like growth factor-1 (IGF-1) by analyzing reverse transcriptase polymerase chain reaction (RT-PCR) and hair growth in a shaving animal model of C57BL/6 mice topically applying with an amount of 0.1 mL once a day for 3 weeks. The mice were divided into 4 groups including normal group (saline, N), negative control group (dimethyl sulfoxide, NC), positive control group (5 mg/mL minoxidil, PC), and experimental group (Origanum vulgare extracts, OV). Treatment of OV didn't show cytotoxicity in HHDPC up to 10 ${\mu}g/mL$ and exhibited antioxidant activity with $IC_{50}$ of 31.0 ${\mu}g/mL$. IGF-1 expression in the skin was significantly (p<0.05) increased in the PC and OV compared to the N or NC. PC and OV also showed a prominently promoted hair regrowth compared to the N or NC in hair growth observation. The hair regrowth of OV was significantly higher than that of PC (p<0.05). Therefore, these results indicate that O. vulgare extracts effectively stimulated hair growth in an animal model.

This study was to investigate the hypolipidemic effect of powdered mulberry leaves (PML) and water extract of powdered mulberry leaves (WML) on high-fat fed mice. Male C57BL/6 mice were divided into four groups; a normal group (N), a high-fat (HF) group, a high-fat group supplemented with PML (HF-PML) and a high-fat group supplemented with WML (HF-WML). The PML or WML was added to a standard diet based on 1% dried mulberry loaves (1g PML/100g diet and 0.22g WML/100g diet) for 6 weeks. Body weight and organ weights were not different among thle groups in high-fat fed mice, whereas food intake and daily energy intake were significantly (p<0.05) lowered in the HF-PML group. In plasma and liver, the supplementation of PML and WML significantly (p<0.05) lowered cholesterol and triglyceride concentrations compared to the HF group. The HDL-cholesterol/total cholesterol ratio was significantly (p<0.05) higher in the HF-PML and HF-WML groups than in the B:.w group. The fecal triglyceride and cholesterol concentrations were significantly (p<0.05) increased in the HF-PML and HF-WML groups compared to the HF group. Hepatic lipid regulating enzyme activities, fatty acid synthase, fatty acid ${\beta}-oxidation$ and carnitine palmitoyl transferase were significantly lower in the HF group than in the N group. However, the activities of these hepatic lipid regulating enzymes activities were significantly (p<0.05) elevated in the HF-PML and HF-WML groups compared to the HF group. Accordingly, these results suggest that PML and WML improve plasma and hepatic lipid levels partly by increasing fecal lipid excretion and enhancing hepatic lipid regulating enzymes activities.
Susceptibilities of 5 different mice strains, including C3H/HeN, BALB/c, C57BL6, FvB and ICR, to Echinostoma hortense infection, was evaluated. The worm expulsion rate, worm size and egg production were observed from 1 to 8 weeks after infection with 30 metacercariae. C3H/HeN and ICR mice showed the highest worm maturation rates. The worm recovery rate and the number of eggs per gram (EPG) of feces was also higher in C3H/HeN and ICR mice than in BALB/c, C57BL6, and FvB mice. It is suggested that E. hortense is highly infectious to ICR and C3H/HeN mice, but not to the other strains of mice. Based on the results obtained, we believe that the susceptibility of different mouse strains to E. hortense infection is dependent on the genetic and immunologic back-ground of mice.
We have synthesized the cinnamyl derivative of minoxidil, N-2-cinnamylamino-4-amino-6-piperidinopyrimidine 3-oxide(MINAMYL). Physicochemical properties and structural analysis on this compound were examined. And also, we have screened its antialopecic effect.
Obesity is one of the most serious health problems in developed countries. It negatively affects diverse aspects of human wellbeing. Of these, a relationship between obesity and depression is widely recognized but biomarkers for assessment of obesity-associated mood changes in animal obesity models are rarely known. Here we explored the link between obesity and the plasma levels of monoamine neurotransmitters involved in mood control using a sensitive UPLC/MSMS technique in high fat diet (HFD)-induced obesity model in male C57BL/6 mice to explore the potential utility of plasma tests for obesity-associated mood change. HFD (60% of total calories, 8 weeks) induced significantly higher weight gains in body (+37.8%) and fat tissue (+306%) in male C57BL/6 mice. Bioanalysis of serotonin, dopamine and norepinephrine in plasma at 8 weeks of HFD revealed that serotonin decreased significantly in the obese mice when compared to normal diet-fed mice ($2.7{\pm}0.6$ vs $4.3{\pm}2.0ng/ml$, N=8). Notably, a negative correlation was found between the levels of serotonin and body weight gains. Furthermore, principal component analysis (PCA) with the individual levels of neurotransmitters revealed that plasma levels of dopamine and serotonin could apparently differentiate the obese mice from lean ones. Our study demonstrated that blood plasma levels of neurotransmitters can be employed to evaluate the mood changes associated with obesity and more importantly, provided an important clue for understanding of the relationship between obesity and mood disorders.
Purpose: The purpose of this study is to investigate the effect of Boheo-tang (B) and Boheo-tang plus cervi pantotrichum cornu (B+CP) on lactation in postpartum C57BL/6N mice. Methods: Normal saline(control), Band B+CP (8mu l/g$) were administerd p.o. twice a day for 20 days. Lactating mammary gland tissues were examined through light microscope by the way of HE staining and immunohistochemical assay. Milk producing associated gene expression were accessed by RT-PCR. Results: In mammary gland, amount of adipose tissues were decreased in both Band B+CP treated group. And the ductal branches and alveolar tissues increased in both treated group. Immunoreactivity of prolactin receptors was increased both treated group, and immunoreactivity of oxytocin receptors was increased in the B+CP treated group. In both treated group, IGF-l mRNA expression was increased and TGF-$\beta$ mRNA expression was decreased. And PRL mRNA expression was increased in the B+CP treated group. PL-l mRNA expression was decreased in the B treated group but increased in the B+CP treated group. Conclusion: This study shows that treatment of Boheo-tang and Boheo-tang plus cervi pantotrichum cornu can improve postpartum lactation in C57BL/6N mice.
Purpose: The purpose of this study is to investigate the effect of Boheo-tang (B) and Boheo-tang plus cervi pantotrichum cornu (B+CP) on immune response in postpartum C57BL/6N mice. Methods: Normal saline(control), B and B+CP (8${\mu}{\ell}$/g) were administerd p.o. twice a day for 20 days. Subpopulation of T and B lymphocyte were accessed by flow cytometric analysis. Results: Splenic T and B lymphocytes were increased by the treatment of B. Subpopulation of cytotoxic T lymphocytes in the spleen, were significantly increased by both treatment of B and B+CP. Subpopulation of cytotoxic T lymphcytes in the thymus, were significantly increased by both treatment of B and B+CP. IL-4 production was significantly increased by the treatment of B+CP. Conclusion: This study shows that treatment of Boheo-tang and Boheo-tang plus cervi pantotrichum cornu can improve postpartum immune response in C57BL/6N mice.

The antihyperlipidemic and glycemic control effects of mycelia of Inonotus obliquus including protein-bound polysaccharides extract were investigated. In high fat diet-induced obese C57BL/6J mice, the mycelia of Inonotus obliquus including protein-bound polysaccharides extract showed significant decrease in epididymal fat tissue weight, blood triglyceride and VLDL level, triglyceride and total cholesterol level in liver, serum insulin and HOMA-IR level and AUC. Oral glucose tolerance test glucose level reduction were 4.9% (M150L; ${\beta}$-glucan 1.303 mg/kg bw), 9.5% (M150; ${\beta}$-glucan 2.606 mg/kg bw) after 180 min of glucose loading compared to H-C group. Besides, the mycelia of Inonotus obliquus including protein-bound polysaccharides extract treatment significantly increased glycogen contents in liver and adiponectin level in high fat diet-induced obese mice. In conclusion, the results showed that the mycelia of Inonotus obliquus including protein-bound polysaccharides extract possesses significant antihyperlipidemic and glycemic control effects in C57BL/6J mice.
