Inflammatory bowel disease (IBD) is a chronic and recurrent illness of the gastrointestinal tract. Treatment of IBD traditionally involves the use of aminosalicylic acid and steroids, while these drugs has been associated with untoward effects and refractoriness. The absence of effective treatment regimen against IBD has led to the exploration of new targets. Parasites are promising as an alternative therapy for IBD. Recent studies have highlighted the use of parasite-derived substances, such as excretory secretory products, extracellular vesicles (EVs), and exosomes, for the treatment of IBD. In this report, we examined whether EVs secreted by Giardia lamblia could prevent colitis in a mouse model. G. lamblia EVs (GlEVs) were prepared from in vitro cultures of Giardia trophozoites. Clinical signs, microscopic colon tissue inflammation, and cytokine expression levels were detected to assess the effect of GlEV treatment on dextran sulfate sodium (DSS)-induced experimental murine colitis. The administration of GlEVs prior to DSS challenge reduced the expression levels of pro-inflammatory cytokines, including tumor necrosis factor alpha, interleukin 1 beta, and interferon gamma. Our results indicate that GlEV can exert preventive effects and possess therapeutic properties against DSS-induced colitis.

Liver fibrosis is part of the wound healing process to help the liver recover from the injuries caused by various liver-damaging insults. However, liver fibrosis often progresses to life-threatening cirrhosis and hepatocellular carcinoma. To overcome the limitations of current in vivo liver fibrosis models for studying the pathophysiology of liver fibrosis and establishing effective treatment strategies, we developed a new mouse model of liver fibrosis using polyhexamethylene guanidine phosphate (PHMG-p), a humidifier sterilizer known to induce lung fibrosis in humans. Male C57/BL6 mice were intraperitoneally injected with PHMG-p (0.03% and 0.1%) twice a week for 5 weeks. Subsequently, liver tissues were examined histologically and RNA-sequencing was performed to evaluate the expression of key genes and pathways affected by PHMG-p. PHMG-p injection resulted in body weight loss of ~15% and worsening of physical condition. Necropsy revealed diffuse fibrotic lesions in the liver with no effect on the lungs. Histology, collagen staining, immunohistochemistry for smooth muscle actin and collagen, and polymerase chain reaction analysis of fibrotic genes revealed that PHMG-p induced liver fibrosis in the peri-central, peri-portal, and capsule regions. RNA-sequencing revealed that PHMG-p affected several pathways associated with human liver fibrosis, especially with upregulation of lumican and IRAK3, and downregulation of GSTp1 and GSTp2, which are closely involved in liver fibrosis pathogenesis. Collectively we demonstrated that the PHMG-p-induced liver fibrosis model can be employed to study human liver fibrosis.

Background and Objectives Low-level light therapy (LLLT) is an application of low-power light for various purposes such as promoting tissue repair, reducing inflammation, causing analgesia, etc. A previous study suggested the effect of light emitting diode (LED) light with the wavelength of 740 nm for promoting wound healing of corneal epithelial cells. This current study aimed to confirm the effect of LLLT for managing inflammation of a dry eye disease (DED) mouse model. Materials and Methods A total of 50C57BL/6 female mice were randomly grouped into 5 groups to compare the effect of LLLT:1) Control group, 2) Only LLLT group, 3) Dry eye group, 4) LLLT in dry eye group, and 5) Early treatment group. DED was induced with 4 daily injections of scopolamine hydrobromide and desiccation stress for 17 days, and LLLT at 740 nm was conducted once every 3 days. To analyze the effect of LLLT on the DED mouse model, tear volume, corneal surface irregularities, and fluorescence in stained cores were measured, and the level of inflammation was assessed with immunohistochemistry. Results The DED mouse model showed significant deterioration in the overall eye condition. After LLLT, the amount of tear volume was increased, and corneal surface irregularities were restored. Also, the number of neutrophils and the level of inflammatory cytokines significantly decreased as well. Conclusion This study showed that LLLT at 740 nm was effective in controlling the corneal conditions and the degree of inflammation in DED. Such findings may suggest therapeutic effects of LLLT at 740 nm on DED.

We investigated the probiotic characteristics and anti-obesity effect of Lactiplantibacillus plantarum MGEL20154, a strain that possesses excellent intestinal adhesion and viability. The in vitro properties, e.g., gastrointestinal (GI) resistance, adhesion, and enzyme activity, demonstrated that MGEL20154 is a potential probiotic candidate. Oral administration of MGEL20154 to diet-induced obese C57BL/6J mice for 8 weeks resulted in a feed efficacy decrease by 44.7% compared to that of the high-fat diet (HFD) group. The reduction rate of weight gain was about 48.5% in the HFD+MGEL20154 group compared to that of the HFD group after 8 weeks, and the epididymal fat pad was also reduced in size by 25.2%. In addition, the upregulation of the zo-1, pparα, and erk2, and downregulation of the nf-κb and glut2 genes in Caco-2 cells by MGEL20154 were observed. Therefore, we propose that the anti-obesity effect of the strain is exerted by inhibiting carbohydrate absorption and regulating gene expression in the intestine.
목적: 이 연구에서는 폐 전이 종양을 영상화하기 위하여 흑색종의 폐 전이 종양 마우스 모델을 제작하고 영상 획득 전처리 조건을 개선하여 폐 전이 종양의 $[^{18}F]FDG$ 소동물 PET 영상을 획득하고자 하였으며, 임상 CT를 이용하여 전이 종양의 해부학적 위치를 확인하고자 하였다. 대상 및 방법: 정상 마우스의 $[^{18}F]$FDG 영상 획득 전 조건은 $16{\sim}22$시간 금식 하고 $30^{\circ}C$의 온도를 유지하며 $[^{18}F]FDG$ (7.4 MBq) 정맥 주사 후 서로 다른 마취제(Ketamine/Xylazine, Ke/Xy과 Isoflurane, Iso)로 $[^{18}F]FDG$ 섭취 60분 동안 유지한 후 20분간 $[^{18}F]FDG$ 소동물 PET 영상을 획득하였다. 혈중 포도당 농도를 보정한 포도당 표준 섭취 계수 영상을 이용하여 관심영역 대 배경비(lung to background ratio, L/B)를 구하여 평가하였다. C57BL/6 마우스에 B16-F10 세포를 정맥내 주사하여 제작한 폐전이 종양 마우스 모델은 정상 마우스의 영상 획득 조건과 동일한 조건에서 $[^{18}F]FDG$ 소동물 PET 영상을 획득하였으며, 임상 CT를 이용하여 획득된 해부학적 영상으로 폐 부위의 종양 위치를 확인하였다. 결과: 정상 마우스의 평균 혈중 포도당 농도는 Ke/Xy으로 마취한 군에서 $128.0{\pm}23.87\;mg/dL$이었으며 Iso으로 마취한 군에서는 $86.0{\pm}21.65\;mg/dL$로, Ke/Xy으로 마취한 군이 Iso로 마취한 군 보다 1.5 배 높은 혈중 포도당 농도를 나타내었다. 포도당 표준 섭취 계수 영상에서의 L/B는 Ke/Xy으로 마취한 군에서 $8.6{\pm}0.48$ 이었으며 Iso으로 마취한 군에서는 $12.1{\pm}0.63$로, Iso로 마취한 군이 Ke/Xy으로 마취한 군 보다 주변 정상조직과의 대조도가 높은 경향을 보였다. 폐 전이 종양 마우스에서는 Iso로 마취한 군이 Ke/Xy으로 마취한 군의 $[^{18}F]FDG$ 소동물 PET 영상보다 주변 조직의 $[^{18}F]FDG$ 섭취가 낮았다. 또한 해부학적 종양의 위치를 확인하기 위하여 임상 CT 영상과 융합한 결과 폐 전이 종양이 폐 부위에 위치함을 확인하였다. 결론: 마우스와 같은 소동물에서의 폐 부위 종양을 $[^{18}F]FDG$로 영상화하는데 있어서 금식, 온도유지, $[^{18}F]FDG$ 섭취 시간 동안의 마취제 조건 등을 고려하여야 하며, $[^{18}F]FDG$ 소동물 PET과 CT 영상의 융합은 폐 부위의 전이 종양을 확인하는데 유용할 것으로 기대된다.
Objectives : This study was performed to investigate the inhibitory effects of Cheongshimyeonja-tang water extracts(CSYJ) on high fat diet-induced obesity and hyperlipidemia in C57BL/6J mice. Methods : The experimental animals were divided into four groups; normal diet-fed control(ND), high fat diet-fed control(HFD), HFD+CSYJ 150 mg/kg(CSYJ 150), and HFD+CSYJ 300 mg/kg(CSYJ 300). Obesity with hyperlipidemia was induced by feeding high fat diet(40%), and CSYJ was administrated orally into mice every day for 5 weeks. The effect of CSYJ on the serological parameters for Obesity with hyperlipidemia was evaluated. Results : CSYJ-treated groups revealed significantly reduced body weight and feed intake, as well as feed efficiency ratio, compared to HFD-fed group in dose-dependent manner. CSYJ reduced significantly the serum levels of triglyceride, total cholesterol and LDL-cholesterol elevated by intake of high fat diet feed, while the increased serum levels of HDL-cholesterol attenuated levels of atherogenic index and cardiac risk factor. It also reduced the blood levels of insulin and leptin in HFD group, and inhibited lipid accumulation in organs such as liver and abdomal adipose tissue. Moreover oral administration of CSYJ decreased significantly the blood level of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and lactate dehydrogenase(LDH), and lipid peroxide(LPO), compared to HFD-fed group in dose-dependent manner. Conclusions : These results indicate that CSYJ could reduce high fat diet-induced obesity and hyperlipidemia, suggesting its clinical usefulness for declining body fat and hyperlipidemia.
본 연구는 고지방식이(열량의 40%를 지방으로 공급)로 유도한 비만마우스에게 아피오스 분말(10%, w/w)을 12주간 급여한 후 내장지방 감소 및 지방간 개선 효과를 살펴보았다. 아피오스 분말 급여로 인한 유의적인 체중 감소는 보이지 않았으나, 부고환지방과 후복막지방 무게가 유의적으로 감소하여 총 내장지방 무게는 대조군에 비해 14.3% 감소하였다. 부고환지방조직의 지방세포 크기 역시 아피오스 분말군에서 현저히 감소되었다. 혈청의 유리지방산, 중성지질과 총 콜레스테롤 함량은 실험군 간의 유의적인 차이가 없었으나 총 콜레스테롤에 대한 HDL-콜레스테롤 비와 동맥경화 지수는 아피오스 분말군이 대조군에 비해 각각 18.9%와 47.2% 유의적으로 개선되었다. 혈청의 렙틴 함량은 아피오스 분말군이 대조군에 비해 유의적으로 낮았다. 고지방식이를 급여한 마우스에 아피오스 분말 급여는 간 조직 중의 지질축적 및 유리지방산과 중성지질 함량을 유의적으로 낮추는 것으로 나타났다. 간 조직의 지질합성 관련 유전자인 Pparg, Fasn과 Dgat2의 발현이 아피오스 분말 급여로 억제되었다. 이상의 결과에서 아피오스 분말은 고지방식이로 인한 내장지방 축적과 지방간을 개선할 수 있을 것으로 평가되었다.
알츠하이머 질환은 인지능력과 행동능력 두 가지에 모두 영향을 미치는 진행적 노인성 치매증의 일종으로 정확한 발병기전은 아직 알려져 있지 않으나 환경적 요인 및 유전적 요인이 모두 중요한 위험인자로 알려져 있다. 본 연구에서는 8주간 에탄올에 노출된 Aldh2 knockout mouse 뇌조직을 분리하여 알츠하이머 질환의 지표로 잘 알려진 아밀로이드 베타와 NF-kB 발현을 평가하고 이러한 변화가 ALDH2 효소의 활성에 따라 어떻게 달라지는 지 비교하였다. 그 결과, 8주간 에탄올을 경구 투여한 마우스에서 ALDH2 효소의 활성에 따른 NF-kB 농도의 차이는 확인할 수 없었으나, ALDH2 효소 활성은 알코올 노출에 의한 해마조직의 아밀로이드 베타 축적에서 중요한 영향을 미치는 것으로 나타났다(p<0.05). 이러한 결과는 ALDH2 효소가 결핍된 사람이 결핍되지 않은 사람에 비해 알코올에 의한 알츠하이머 질환의 발생에 보다 민감할 수 있음을 시사한다.
본 연구는 시중에서 판매되고 있는 막걸리를 알콜과 수분을 제거한 막걸리 동결건조물로 만들어 AGS위암세포주가 이식된 종양이식 마우스 모델을 가지고 종양성장에 미치는 영향을 관찰하였다. 면역세포가 결핍된 C57/BL/6누드마우스를 이용하여 AGS위암 세포 이식 군(Induction), AGS위암 세포 이식+시판막걸리 동결건조물 $100mg/kg{\cdot}day$군(ML), AGS위암 세포 이식+시판막걸리 동결건조물 $500mg/kg{\cdot}day$군(MH), AGS위암 세포 이식+5-FU $18mg/kg{\cdot}day$군으로 나누어 비교한 결과, 시판막걸리 동결건조물에서 종양의 성장을 유의하게 감소시키는 것을 관찰하였다. 이와 더불어 각 군 간에 체중변화는 유의한 차이가 없어 막걸리의 투여에 의한 체중감소 등의 특이한 부작용이 없는 것으로 판단하였다.
Cysteine protease의 일종인 CTSS가 비만에 관여한다는 것이 알려진 후 CTSS저해를 통한 비만 억제의 가능성이 제시되고 있다. 본 연구는 눈꽃동충하초로부터 CTSS저해 활성을 갖는 분획을 분리하여 항비만 효과를 확인하는 것을 목적으로 수행되었다. 눈꽃 동충하초 열수추출물을 Diaion HP-20 수지 흡착 여부에 따라서 DHP1, DHP2 두 개의 분획으로 나누었다. CTSS에 대한 $IC_{50}$값은 열수추출물, DHP1, DHP2가 각각 108.7, 890.3, 2.3 ${\mu}g$/mL이었다. 고지방식이로 비만을 유도한 마우스에 각 분획들을 4주간 섭취시키고 체중 변화, 부고환 지방 무게, 지방세포 크기 및 혈중지질 농도 등을 측정한 결과 CTSS 저해활성이 가장 높은 분획 DHP2를 섭취시킨 실험군에서 가장 뛰어난 항비만 효능을 확인할 수 있었다. 결론적으로 본 연구는 눈꽃동충하초로부터 분리된 CTSS저해 성분들이 비만을 치료하거나 예방하는데 효과적으로 활용될 수 있는 가능성을 제시한다고 볼 수 있다. 향후 눈꽃동충하초로부터 CTSS저해 물질을 분리 및 동정한다면 보다 명확한 항비만 기작을 밝혀낼 수 있을 것이다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
