Objectives: The prevalence of obesity continues rise and obesity and metabolic syndrome is a major problem in global health care. Animal models are used in the drug discovery of novel treatment for obesity. One of common models of obesity is a high fat diet induced obesity in a C5BL/6 mouse, and the development of obesity and glucose tolerance in mouse model is different according to period of diet. Therefore, this study was performed to observe the development of obesity and glucose tolerance during a high fat diet (HFD). Methods: Male C57BL/6 mice, 5 weeks of age, were fed on a standard chow diet as a normal diet (18 kcal% fat) or a HFD (60 kcal% fat) for up to 16 weeks. The various factors related with obesity and insulin resistance were measured at 8, 12, and 16 weeks. Results: The weights of body and epididymal fat were gradually increased for 8~16 weeks, however the change of hyperglycaemia and glucose tolerance have shown different with that of body weight. Blood glucose, oral glucose tolerance and insulin tolerance were increased more clearly at week 12 and 16 than week 8. Lipid accumulation of liver and body temperature were also significantly increased at week 16, compared with normal group. Conclusions: The developments of obesity and related factors were different by a HFD period in a C57BL/6 obese mice. This result suggests that the development of obesity with glucose tolerance and liver lipid may induce clearly by a HFD for 16 weeks.
To investigate the effect of Artemisia apiacea Hance on the hair growth of black mouse (C57BL/6), this study was carried out. We obtained chloroform fraction, ethyl acetate fraction, butanol fraction from ethanol extract. When these fractions were administered to the black mouse by the method of skin paste, chloroform extract increased remarkably the hair growth of black mouse. thus Chloroform fraction was devided into 5 fractions from 20:1 to 1:1 with chloroform : methanol. the detail fractions of chloroform fraction were administered to the black mouse. We obtained the result that fraction I, II, III, IV are superior to minoxidil of positive control. Most of all fraction II have an dramatical effect on hair growth of black mouse.

Objective: This study was proceeded to determine to what extent the decoction recipes and distillation recipes extracted from the infusion of the apricot seed and other herbs respectively would produce an effect on the proliferation and the differentiation of Preadipocyte 3T3-L1 Model separated from C57BL/6 mouse. Method: Preadipocyte 3T3-L1 Model was formed from the fat tissue of the epididymis organ, which was removed from 4-week-old C57BL/6 mouse and then was treated with collagenase. Results: 1. Concerning the restraint effect of proliferation on Preadipocyte 3T3-L1 Model, the decoction recipes, in the state of low consistency, were proved to be more effective than the distillation recipes. 2. Concerning the restraint effect of differentiation on Preadipocyte 3T3-L1 Model, the decoction recipes were shown to be more effective.
Objectives : The aim of this study was to investigate the effect of Peucedani Radix herbal acupuncture(PR-HA) at St36(joksamni) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week) C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week). One of the two experimental groups was just treated with needle-prick on St36 and the other group was treated with 1% concentrations of PR-HAS at St36, for the later 8 weeks (3times /week). Results : 1. The weight and total cells of lung of the mice group treated with PR-HA decreased significantly compared with those of control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with PR-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with PR-HA in Photomicrographs decreased significantly compared with those of control group. 4. According to Histological analysis of lung sections, it decreased significantly adhension of collagen in PR-HA than those of control group 5. The concentration of IgE, IL-4, IL-5, in BALF and IL-4, IL-5, Il-13 in serum of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The number of Gr-1+/CD11b+, CD11b+, CD3-/CCR3+, CD4+, CD3e+/CD69+ , CD23+B220+ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The cytokine's manifestation of mRNA of the mice group treated with PR-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : We conclude that PR-HA is effective on OVA-induced asthma of C57BL/6 mouse.
Purpose: C57BL/6 mice, which are among the most common backgrounds for genetically engineered mice, are resistant to the induction of periodontitis by oral infection with periodontal pathogens. This study aimed to develop a periodontitis model in C57BL/6 mice using coaggregation between human pathogens and the mouse oral commensal Streptococcus danieliae (Sd). Methods: The abilities of Porphyromonas gingivalis ATCC 33277 (Pg33277), P. gingivalis ATCC 49417 (Pg49417), P. gingivalis KUMC-P4 (PgP4), Fusobacterium nucleatum subsp. nucleatum ATCC 25586 (Fnn), and F. nucleatum subsp. animalis KCOM 1280 (Fna) to coaggregate with Sd were tested by a sedimentation assay. The Sd-noncoaggregating Pg33277 and 2 Sd-coaggregating strains, PgP4 and Fna, were chosen for animal experiments. Eighty C57BL/6 mice received oral gavage with Sd once and subsequently received vehicle alone (sham), Fna, Pg33277, PgP4, or Fna+PgP4 6 times at 2-day intervals. Mice were evaluated at 5 or 8 weeks after the first gavage of human strains. Results: Fnn, Fna, and PgP4 efficiently coaggregated with Sd, but Pg33277 and Pg49417 did not. Alveolar bone loss was significantly higher in the PgP4 group at both time points (weeks 5 and 8) and in all experimental groups at week 8 compared with the sham group. The PgP4 group presented greater alveolar bone loss than the other experimental groups at both time points. A higher degree of alveolar bone loss accompanied higher bacterial loads in the oral cavity, the invasion of not only PgP4 but also Sd and Fna, and the serum antibody responses to these bacteria. Conclusions: Periodontitis was successfully induced in C57BL/6 mice by oral infection with a P. gingivalis strain that persists in the oral cavity through coaggregation with a mouse oral commensal bacterium. This new model will be useful for studying the role of human oral bacteria-host interactions in periodontitis using genetically engineered mice.
Objective & Methods : To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffusae herba infusion solution(ELP-HAS), we injected ELP-HAS into Chung-wan(CV12) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results : The results were obtained as follows : 1. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the spleen cell proliferation in BALB/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$cells in C57BL/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the pulmonary colony formation number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group. Conclusions : ELP Herbal acupuncture injection into Chung-wan(CV12) of C57BL/6 mice is considered to be effective in anti-cancer, anti-metastasis and immune response improvement.
The derivative of chrysin 7-O-cyclopropanecarboxylate was synthesized by condensing cyclopropanecarboxylic acid with chrysin in organic solvent, and its structure was identified by NMR, MS, UV IR etc. We also investigated the physico-chemical properties, anti-diabetic effect and set up the quantitative analytical method of this compound. The correlation coefficient of calibration curve on this compound was approximately 0.9985 by absorption spectrophotometry. And, this study was carried out to investigate the hair-growth effect of chrysin derivative to the black mouse (C57BL/6). When this derivative in ethanol solution was administered to the back of mouse by method of skin paste, this derivative promoted the hair growth of mouse.
The hair-growth effect of Illite was suggested by some people who were using Illite as a beautifying material. We investigated the hair-growth effect of Illite powder. The hair-growth effects were investigated by two methods; the activity of hair-growth after shaving the hairs on the black mouse (C57BL/6) and the recovery activity of hair-growth after hair-loss induced by cyclophosphamide treatment. Suspension of Illite powder was applied to the back of the black mouse by method of skin paste. Illite promoted significantly the hair growth of mouse in both conditions of shaving and hair-loss. And then we investigated the toxicity which may be induced by Illite when it was administrated orally as a single dose. We could not fond out any significant toxicity induced by single dose oral administration of Illite.
The heart is a major dose-limiting organ for radiotherapy of cancer in the thoracic region. The purpose of this study was to examine the protective effect of tranilast on the radiation-induced heart fibrosis model using the C57BL/6 murine strain. A significant reduction in the expression of TGF-${\beta}1$, collagen type I and collagen type III was observed in the radiation plus tranilast group. The authors also suggest the use of tranilast in a clinical trial for the prevention of radiation-induced heart fibrosis.
