In order to investigate the effects of Bogigambi-tang(here in after referred to BGGBT) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BGGBT extract) and fed for 15 weeks. Items of this experimental study are as follows. Body weight change, final inclose of body weight, the weight change of the adipocytes in body, the level change of ALT, AST, total cholesterol, LDL-Cholesterol, triglyceride, glucose, free fatty acid and creatinine, the expression of ${\beta}$3AR and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}$3AR and leptin in adipocytes tissue. The following results have been obtained All experimental group have shown that the weight and the final increase of weight have decreased considerably. All experimental group have shown that the amount of the adipocyte in weight has decreased considerably. All experimental group have shown that the amount of leptin has decreased considerably. All experimental group have shown that the revelation of ${\beta}$3AR in primary adipose cell and 3T3-L1 cell has increased considerably, and that the revelation of leptin in primary adipose cell and 3T3-L1 cell has decreased considerably, All experimental group have shown that the size of adipocyte in adipocytes tissue has decreased. The high density group have shown that the adipose vacuoles in liver tissue has decreased considerably, and that the cell nucleuses has similar with normal group.
Sjögren syndrome (SS) is a systemic inflammatory autoimmune disease that involves exocrine glands. Shikonin is extracted from comfrey, which is conventionally used as an anti-tumor, antibacterial, and antiviral drug in China. However, the application of Shikonin in SS remains unreported. This study aimed to verify the potential functions of Shikonin in SS progression. Firstly, non-obese diabetic mice were used as the SS mouse model, with C57BL/6 mice serving as the healthy control. It was demonstrated that the salivary gland damage and inflammation were aggravated in the SS mouse model. Shikonin improved salivary gland function decline and injury in the SS mouse model. Moreover, Shikonin reduced inflammatory cytokines and immune infiltration in the SS mouse model. Further experiments discovered that Shikonin attenuated the MAPK signaling pathway in the SS mouse model. Lastly, inhibition of the MAPK signaling pathway combined with Shikonin treatment further alleviated the symptoms of SS. In conclusion, Shikonin ameliorated salivary gland damage and inflammation in a mouse model of SS by modulating the MAPK signaling pathway. Our findings indicate that Shikonin may be a useful drug for SS treatment.
Chrysin 7-Ο-crotonate was synthesized by condensing crotonic acid with chrysin in organic solvent, tetrahydrofuran (THF) using dicyclohexylcarbodiimide (DCC) and 4-dimethylaminopyridine (DMAP). Its structure was indentified by NMR, MS, UV, IR etc. We also investigated the physico-chemical properties and set up the quantitative anytical method of this compound. The correlation coefficient of calibration curve measured at the isobestic point (340 nm) on this compound was approximately 0.9994 by absorption spetrophtometry. Detection limit was 1.6ng at S/N=3 by using a RP column by HPLC. The hair growth effect fo chrysin 7-Ο-crotonate on the hair of black mouse (C57BL/6), was carried out using paste method. When its ethanol solution was administered to this black mouse by route of skin, this compound promoted the growth of hair.
본 실험은 phospholipase C (PLC)-$\beta$-3 및 peroxiredoxin (Prx) II 유전자가 적중 ($\Delta$)된 J1 마우스 배아주 (embryonic stem) 세포로부터 생식선이행 카이미라 마우스생산을 위한 제반조건 및 배아주세포의 배양조건을 확립하기 위하여 수행되었다. 80% 이상의 정상핵형을 보이는 유전자 적중된 4개의 클론 ($\Delta$Pu II C3, $\Delta$Pu II C3, C10 및 15)으로부터 카이미라를 생산하였을 때 형태적으로 분화정도가 높은 클론 ($\Delta$PLC$\beta$-3 C3)의 이용은 카이미라의 생산율 (21.1%)과는 무관하게 카이미리즘 (< 20%)이 낮았고, 수컷 카이미라의 생산 빈도 (7/15, 46%)도 낮아지는 것으로 나타났다. 그러나 형태적으로 안정된 3개의 클론 ($\Delta$Prx II C3, C10 및 15)은 80% 이상의 높은 카이미리즘을 지닌 마우스 (9/13, 69.2%)를 생산하였고, 수컷 카이미라의 생산율 (l1/13, 84.6%)도 증대된 것으로 나타났다. 따라서 80% 이상의 정상핵형을 지닌 배아주 세포를 형태적으로 안정하게 유지하는 것이 카이미리즘이 높은 마우스를 생산하는데 결정적 요인으로 작용할 수 있는 것으로 확인되었다. 미세주입용 배반포를 효율적으로 생산하기 위해 5~10주령 사이의 C57BL/6J 암컷마우스를 교배 한 결과, 10주령 마우스가 미세주입가능한 3.5일령 배반포를 가장 많이 생산 (2.94개/마리)하였다. 또한 미세주입된 배반포를 이식하기 위해 ICR 및 ICR$\times$C57BL/6J F1 (IBF1)위임신 대리모를 사용하였을 때, IBF1이 복당 산자수 (2.8 vs. 5.6)가 많았고 카이미라 생산율 (0 vs. 35.3%)도 매우 높았다. 따라서 공여마우스의 주령 및 대리모의 선택이 카이미라 생산효율 향상에 중요한 요인으로 부각되었다. 결과적으로 핵형이 안정된 ES 세포를 동정하는 것은 물론 클로닝 과정중에 형태적으로 분화가 없도록 ES세포를 배양하는 것이 카이미리즘이 높은 마우스를 생산하고 아울러 생식선 이행 빈도를 증가시키는데 결정적인 역할을 하는 것으로 확인되었다.
Indole-3-carbinol (I3C), one of naturally occurring main components in cauliflower vegetables, is supposed to have a chemopreventive potential in experimental animals and humans. This study was investigated to examine chemopreventive effect of I3C on colon carcinogenesis induced by azoxymethane (AOM) using C57BL/6J mice. Mice were divided into three groups (10 or 9 mice/group). All mice were subcutaneously injected with AOM (5 mg/kg body weight, four times at weekly interval). After AOM treatment, animals of group 1 were fed by AIN-76A pellets as a basal diet. Animals of groups 2 and 3 were given I3C containing diets (100 and 300 ppm in diets, respectively) for 6 weeks until sacrifice. All mice were sacrificed at week 10 and the aberrant crypt foci (ACF) of the colonic mucosa were assessed after staining with methylene blue. Total numbers of ACF/colon in group 2 ($10.1{\pm}5.1$) or group 3 ($10.6{\pm}5.3$) were decreased compared to the values of group 1 ($14.4{\pm}10.2$). Among numbers of ACF formation, 5, 7, 8 and 10 ACF in group 2 and 3 were greatly different those of group 1. Total numbers of aberrant crypts (AC)/colon of group 2 ($20.1{\pm}10.1$) or group 3 ($22.0{\pm}10.9$) were decreased compared to the value of group 1 ($33.7{\pm}24.7$). Taken together, it suggests that I3C treatment may retard mouse colon carcinogenesis even after administration of AOM.
목적: 가와사키병의 환아뿐만 아니라, 다른 전신 염증 질병의 환아들에 관상동맥 병변(coronary arterial lesion [CAL])의 발생이 보고되었다. 본 연구에서는 전형적인 전신염증반응증후군(systemic inflammatory response syndrome [SIRS])의 소견을 보이는 패혈증 마우스 모델에서 가와사키병 환아에게 관찰되는 CAL이 발생하는지 확인하고자 하였다. 방법: 생후 6주 C57BL/6 마우스에 내독소를 복강내주사하여 SIRS를 보이는 패혈증 모델을 유도하였다. 대조군과 패혈증군의 주요 장기의 조직학적 소견을 비교하였고 패혈증 마우스에서 CAL을 찾기 위한 시도를 하였다. 결과: 대조군과 비교하여, 염증세포의 침윤이 패혈증 마우스의 심장, 간, 신장에 상대적으로 증가하였다. 패혈증 마우스의 심장에서 심근(심근염)과 심장 주위 연조직에 림프구 침윤을 확인하였다. 또한, 패혈증 마우스의 관상동맥을 관찰하였지만, CAL을 확인할 수는 없었다. 결론: 본 연구에서 패혈증 마우스 모델에서 CAL의 존재를 확인하는 것은 실패하였다. 하지만, SIRS를 유발하는 많은 종류의 원인 질병에서 CAL이 발생한다는 것은 잘 알려져 있다. 가와사키병을 포함한 다양한 전신 염증 질병에 나타나는 CAL의 임상적 의미에 대한 연구가 필요할 것이다.
Amyloid ${\beta}$ ($A{\beta}$)의 신경독성은 알츠하이머병의 주된 원인이 되고 이러한 신경독성은 일련의 신경세포 사멸반응에 의해 일어난다고 알려져 있다. 본 연구에서는 알츠하이머병의 실험모델로 mouse primary neuronal cell에 $A{\beta}_{25-35}$를 처리하여 세포독성을 유도하는 세포실험모델과 C57BL/6J mouse 뇌실에 $A{\beta}_{25-35}$를 주입하여 인지장애를 일으키는 동물실험모델을 이용하여 phosphodiesterase III 억제제인 cilostazol의 신경보호 효과에 대해 조사하였다. $A{\beta}_{25-35}$를 신경세포에 처리하면 세포생존율이 감소되었고, 세포사멸이 일어난 세포의 수도 증가되었다. 이러한 $A{\beta}_{25-35}$에 의한 세포독성이 cilostazol처리에 의해 회복되었으며, peroxisome proliferator-activated receptor(PPAR)-${\gamma}$ 항진제인 rosiglitazone 또한 동일한 회복효과를 나타내었다. Cilostazol과 rosiglitazone에 의한 이러한 회복효과가 PPAR-${\gamma}$ 길항제인 GW9662에 의해 다시 억제되는 결과를 통해 cilostazol의 효과는 PPAR-${\gamma}$가 매개하는 신호전달이 관여함을 알 수 있었다. 직접 PPAR-${\gamma}$ 활성화 정도를 측정한 결과, $A{\beta}_{25-35}$ 처리에 의해 감소된 PPAR-${\gamma}$ 활성화 정도가 cilostazol과 rosiglitazone에 의해 증가함을 관찰할 수 있었고, 이는 GW9662에 의해 다시 억제됨을 확인하였다. 게다가, cilostazol은 세포사멸이 일어난 세포의 수와 세포사멸 조절단백질인 Bax/Bcl-2의 비율도 감소시켰다. Cilostazol (20 mg/kg, 구강투여)을 C57BL/6J mice 뇌실에 $A{\beta}_{25-35}$를 주입하기 2주 동안 전처리하고, $A{\beta}_{25-35}$ 주입 후 4주 동안 처리하면, 기억력과 학습능력을 증진시킨다는 결과를 water maze 실험을 통해 알 수 있었으며, rosiglitazone (10 mg/kg)을 먹인 동물에서도 동일한 결과를 얻을 수 있었다. 본 연구를 통해서 cilostazol이 PPAR-${\gamma}$ 활성화를 통해 $A{\beta}_{25-35}$로 인한 신경세포 손상과 세포사멸을 약화시켜, 신경세포의 생존을 증진시키고, 알츠하이머에서 인지장애를 개선할 것으로 생각된다. 따라서, phosphodiesterase III 억제제인 cilostazol은 알츠하이머 질병 치료에 새로운 전략이 될 수 있을 것이다.
Background: This study investigated the anti-obesity effect of the flavonoid rich fraction (FRF) and its constituent, rutin obtained from the leaf of Morus alba L., on the lipid accumulation mechanism in 3T3-L1 adipocyte and C57BL/6 mouse models. Methods and Results: In Oil Red O staining, FRF (1,000 ㎍/㎖) treatments showed inhibition rate of 35.39% in lipid accumulation compared to that in the control. AdipoRedTM assay indicated that the triglyceride content in 3T3-L1 adipocytes treated with FRF (1,000 ㎍/㎖) was reduced to 23.22%, and free glycerol content was increased to 106.04% that of the control. FRF and its major constituent, rutin affected mRNA gene expression. Rutin contributed to the inhibition of Sterol regulatory element binding protein-1c (SREBP-1c) gene expression, and inhibited the transcription factors SREBP-1c, peroxisome proliferator-activated receptor gamma (PPAR-γ), CCAAT/enhancer binding protein α (C/EBPα), fatty acid synthase (FAS) and acetyl-CoA carboxylase (ACC). In addition, the effect of FRF administration on obesity development in C57BL/6 mice fed high-fat diet (HFD) was investigated. FRF suppressed weight gain, and reduced liver triglyceride and leptin secretion. FRF exerted potential anti-inflammatory effects by improving insulin resistance and adiponectin levels, and could thus be used to help counteract obesity. The mRNA expressions of PPAR-γ, FAS, ACC, and CPT-1 were determined in liver tissue. Quantitative real-time PCR analysis was also performed to evaluate the expression of IL-1β, IL-6, and TNF-α in epididymal adipose tissue. Compared to the control group, mice fed the HFD showed the up-regulation in PPAR-γ, FAS, IL-6, and TNF-α genes, and down-regulation in CPT1 gene expression. FRF treatement markedly reduced the expression of PPAR-γ, FAS, IL-6, and TNF-α compared to those in HFD control, whereas increased the expression level of CPT1. Conclusions: These results suggest that the FRF and its major active constituent, rutin, can be used as effective anti-obesity agents.
Polygoni multiflori Ramulus (PM), the stem of Polygonum multiflorum Thunb. has been widely used as a traditional medicine for the treatment of lots of diseases. In macrophages, nitric oxide is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions in inflammation. In the present study, it was investigated that the inhibitory effects on NO and proinflammatory cytokines such as tumor necrosis factor alpha (TNF-${\alpha}$), interleukin-6 (IL-6) and the mechanism of down-regulation of immune response by 85% methanol extracts of PM in mouse (C57BL/6) peritoneal macrophages. Extracts of PM (0.1, 1 mg/ml) suppressed NO production and showed inhibition of proinflammatory cytokines like TNF-${\alpha}$, IL-6 and it attenuated iNOS and COX-2 expression via down-regulation of NF-${\kappa}$B activation. The present results indicate that the 85% methanol extracts of PM has an inhibitory effect on the production of NO through down-regulation of iNOS expression in LPS stimulated mouse peritoneal macrophages and therefore may be beneficial in diseases which related to macrophage-mediated inflammatory disorders.
Susceptibilities of 5 different mice strains, including C3H/HeN, BALB/c, C57BL6, FvB and ICR, to Echinostoma hortense infection, was evaluated. The worm expulsion rate, worm size and egg production were observed from 1 to 8 weeks after infection with 30 metacercariae. C3H/HeN and ICR mice showed the highest worm maturation rates. The worm recovery rate and the number of eggs per gram (EPG) of feces was also higher in C3H/HeN and ICR mice than in BALB/c, C57BL6, and FvB mice. It is suggested that E. hortense is highly infectious to ICR and C3H/HeN mice, but not to the other strains of mice. Based on the results obtained, we believe that the susceptibility of different mouse strains to E. hortense infection is dependent on the genetic and immunologic back-ground of mice.
