No Association between the 5-HT6 Receptor C267T Polymorphism and Response to Citalopram Treatment in Patient with Major Depressive Disorder (주요우울증 환자에서 세로토닌 6(5-HT6) 수용체 C267T 다형성과 Citalopram 치료반응에 대한 연구)

  • The serotonin 6(5-HT6) receptor gene is a candidate gene for influencing the clinical response to treatment with antidepressants. The purpose of this study was to determine the relationship between the C267T polymorphism in the 5-HT6 receptor gene and the treatment response to citalopram in a Korean population with major depressive disorder(MDD). Methods : Citalopram was administered for 8 weeks to the 90 patients who completed study. 21-item Hamilton depression rating scale(HAMD-21) was used as a outcome measure. Results : We found that the genotype, allele, and allele-carrier distributions did not differ significantly between MDD patients and normal controls. A main effect of an interaction of genotype with time on the decrease in the HAMD-21 score during the 8 weeks study period was not found. ANOVA revealed no significant effects of the C825T polymorphism on the decrease in the HAMD-21 score at each time period. Conclusions : These results suggest that the C267T polymorphism in the 5-HT6 receptor gene is not associated with the treatment response to citalopram.

Association between Tardive Dyskinesia and 267C/T Polymorphism of $5-HT_6$ Receptor Gene in Schizophrenia (정신분열병환자에서 만발성 운동장애와 $5-HT_6$ 수용체 유전자 다형성(267C/T)과의 관계)

  • Objectives:Tardive dyskinesia(TD) is a serious side effect associated with long-term antipsychotic treatments. Some candidate genetic polymorphisms were reported to be associated with TD and possible involvement of serotonergic receptors in the pathophysiology of TD has been suggested. In the present study, we investigated the association between $5-HT_6$ receptor gene polymorphism and TD with schizophrenia. Methods:To investigate the relationship between $5-HT_6$ receptor gene polymorphism and TD, 60 patients with TD were compared with 60 patients without TD. The 267C/T allele of $5-HT_6$ receptor gene was genotyped by means of polymerase chain reaction method. TD was evaluated using the Abnormal Involuntary Movement Scale(AIMS). Results:The patients with the three 267C/T genotype showed no significant differences in age, gender, and duration of illness. No significant difference in genotype frequencies was observed between schizophrenic patients with and without TD. In addition, there was no difference in allele frequencies. Further analysis with an measure of AIMS scores showed that these scores were not significantly influenced by the $5-HT_6$ receptor gene polymorphism. Conclusion:These results suggest that 267C/T polymorphism of $5-HT_6$ receptor gene is not significantly associated with susceptibility to TD in schizophrenia.

Matrix Metalloproteinase-3 Gene Polymorphisms (A(-267)G, A658G, T813C) is Associated with Type 2 Diabetes in Koreans (제2형 당뇨병과 MMP3 (A(-267)G, A658G, T813C)의 다형성과의 연관성)

  • Type 2 diabetes is a typical polygenic disease complex, for which several common risk alleles have been identified. Proteins in the matrix metalloproteinase-3 (MMP3) family are involved in the breakdown of the extracellular matrix in normal physiological processes such as embryonic development, reproduction and tissue remodeling, as well as in disease processes. Therefore, we investigated the genotype for the A(-267)G, A658G and T813C polymorphisms in the MMP3 gene in the Korean population and compared genotypes of patients with those of the control group. 200 patients (male 108, female 92), who had previously been diagnosed with type 2 diabetes (T2DM) and 100 control subjects (male 36, female 64) participated in this study. There was a strong association between A(-267)G and A658G polymorphism in the MMP3 gene and T2DM. The present study shows that MMP3 polymorphisms (A(-267)G and A658G) may be associated with the pathogenesis of T2DM. Further studies with a larger population may be needed for the development of diagnostic methods at a genetic level, such as DNA chip.

Effects of c.494A>C and c.267T>G SNPs in OCX-32 Gene of Korean Native Chicken on Egg Production Traits (OCX-32 유전자 내 c.494A>C 및 c.267T>G SNP이 한국 재래닭 산란형질에 미치는 효과 분석)

  • The identification and utilization of potential candidate genes with significant effects on economically important traits have become increasingly important in poultry breeding programs. The ovocalyxin-32 (OCX-32) gene is located chromosome 9 in chicken, plays an important role in eggshell formation. This study was performed to assess the association between single nucleotide polymorphisms (SNPs) of OCX-32 gene and egg production traits in the Korean native chicken. Four Korean native chicken population (n = 181; including 46 females of Ogol, 46 females of white, 43 females of gray and 46 females of black) were used to analyze two SNPs (c.494A>C and c.267T>G) in the OCX-32 gene by PCR-RFLP (Polymerase Chain Reaction-Restricted Fragment Length Polymorphism). We measured egg production traits of age at first egg, first egg weight, egg production ratio and egg weight. The SNP c.494A>C was significantly associated with egg production ratio in Korean Ogol chickens (p<0.001) and egg weight in Korean white chickens (p<0.05). SNP c.267T>C was significantly associated with egg weight in Korean Ogol chickens (p<0.05). But there was no significant association in Korean gray and black chickens. Results suggest the possibility of using molecular markers in OCX-32 gene as a tool for performance and egg production traits in Korean native chicken breeding program.
