미생물 유래 Dykellic Acid가 담배 녹색배양세포의 생장 및 Superoxide Dismutase 활성에 미치는 영향 (Effects of Dykellic Acid Derived from Microorganism on the Cell Growth and Superoxide Dismutase Activity in Tobacco Photomixotrophic Cultured Cells)

  • 미생물에서 분리한 dykellic acid가 식물세포에 미치는 생리적 영향을 평가하기 위하여, 담배 광혼용영양배양세포(photomixotrophic cultured cells, PM세포)에 dykellic acid를 다양한 농도로 처리하여 세포의 생장과 단백질 함량 및 superoxide diamutase (SOD)활성에 미치는 영향을 조사하였다. 담배 PM세포는 0.7 mg/L 2,4-D, 0.3 mg/L kinetin, 30g/L sucrose, 200 mM NaCl를 함유한 MS배지에서 $25^{\circ}C$, 광조건에서 현탁배양 (100 rpm) 하였다. 계대배양시 화합물을 처리한 후 12일째의 세포생장 억제와 배지의 이온 전도도를 측정한 세포막손상의 결과는 일치하였으며, 화합물은 세포생장을 강하게 억제시켰다 ($IC_{50}$/, 약 20 $\mu$M). Dykellic acid 처리농도가 증가함에 따라 단위세포 무게당 단백질 함량과 SOD 비활성도를 현저히 증가시켰다. 이상의 결과로 dykellic acid는 식물세포 생장을 저해하는 활성을 가지고 있으며, 담배 PM세포는 적은 양으로도 천연물의 생리활성을 평가할 수 있는 유용한 생물소재임이 확인되었다.

The Effect of Bacteriocin Produced by Lactobacillus plantarum on the Growth of Listeria monocytogenes

  • The inhibitory effect of Lactobacillus plantarum (Lb. plantarum) which is bacteriocin­producing strain against the growth of Listeria monocytogenes (L. monocytogenes) was examined in trypticase soy broth (TSB). TSB was inoculated with 104 cells/me L. monocytogenes and then with different numbers $(10^6\;10^4\;and\;10^2\;cells/ml)$ of Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. The most effective inhibition of was found at $37^{\circ}C$ and a less inhibition at $25^{\circ}C$. However, there was no significant change in the cell numbers of both L. monocytogenes and Lb. plantarum at $4^{\circ}C$. At same incubation temperature, the higher initial inoculum level of Lb. plantarum, the better inhibitory effect against L. monocytogenes. In addition, TSB was inoculated with L. monocytogenes at different initial inoculum levels of $10^6,\;10^4$ and $10^2$ cells/me and then supplemented with 0, 30, 60 and 100 AU/ml of bacteriocin produced by Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. L. monocytogenes of three different initial inoculum levels began to be inhibited in the presence of more than 60 AU/ml of bacteriocin at $37^{\circ}C$. In TSB containing more than 60 AU/me of bacteriocin and incubated at $25^{\circ}C$, L. monocytogenes decreased by 2 log-units during the period of 12 hrs incubation and thereafter remained steady. At $4^{\circ}C$, L. monocytogenes decreased by 1.5 log-units in the presence of 60 AU/ml bacteriocin during the period of 4 days incubation and dropped to the non-detectable level in TSB with 100 AU/ml bacteriocin.

The Inhibitory Effect of Leek (Buchu) Kimchi Extracts on MCA-induced Cytoxicity and Transformation in C3H-10T1/2 Cells

  • The anticarcinogenic effects of the methanol extracts from leek (buchu in Korean) kimchi and Korean cabbage kimchi were evaluated using cytotoxicity and transformation tests in C3H/10T1/2 cells. Various fractions of the 6-day fermented leek kimchi at 15$^{\circ}C$, hexane, methanol soluble, dichloromethane, ethyl acetate, butanol and aqueous fraction, were also studied in the same system. The inhibitory effect of the leek kimchi(6-day fermented at 15$^{\circ}C$, pH 4.29) was higher than that of the Korean cabbage kimchi(4-day fermented at 15$^{\circ}C$, pH 4.21) on the cytotoxicity induced by 3-methylcholanthrane (MCA) in the C3H/10T1/2 cell system. While the MCA-treated culture(control) formed 21.0 foci of type II plus III in C3H/10T1/2 cells, 100$\mu\textrm{g}$/ml of the methanol extract of the leek kimchi and that of the 4-day fermented Korean cabbage kimchi treated cultures reduced the formation of type II plus III foci to 7.4 and 11.3, respectively. Among the fractions of the leek kimchi, the dichloromethane fraction showed the highest inhibitory effect on MCA-induced cytotoxicity in C3H/10T1/2 cells. Fifty $\mu\textrm{g}$/ml of dichloromethane fraction from the leek kimchi suppressed the MCA-induced cytotoxicity by 77%. On the transformation test using MCA, the dichloromethane fraction considerably reduced the formation of type II plus III foci, especially thpe III foci. When 50$\mu\textrm{g}$/ml of dichloromethane fraction from the leek kimchi was treated, the numbers of type III foci mediated by MCA were decreased to 1.7 compared to 10 for the control. These results indicate that leek kimchi has stronger anticarcinogenic effects than Korean cabbage kimchi and that the dichloromethane fraction of the leek kimchi may contain the major compound(s) that suppress the carcinogenesis in the eukaryotic cells.

Production of Acrylamide Using Immobilized Cells of Rhodococcus rhodochrous M33

  • The cells of Rhodococcus rhodochrous M33, which produce a nitrile hydratase enzyme, were immobilized in acrylamide-based polymer gels. The optimum pH and temperature for the activity of nitrile hydratase in both the free and Immobilized cells were 7.4 and 45$\^{C}$, respectively, yet the optimum temperature for acrylamide production by the immobilized cells was 20$\^{C}$. The nitrile hydratase of the immobilized cells was more stable with acrylamide than that of the free cells. Under optimal conditions, the final acrylamide concentration reached about 400 g/L with a conversion yield of almost 100% after 8 h of reaction when using 150 g/L of immobilized cells corresponding to a 1.91 g-dry cell weight/L. The enzyme activity of the immobilized cells rapidly de-creased with repeated use. However, the quality of the acrylamide produced by the immobilized cells was much better than that produced by the free cells in terms of color, salt content, turbidity, and foam formation. The quality of the aqueous acrylamide solution obtained was found to be of commercial use without further purification.

cDNA microarray에 의한 치주인대세포의 광물화 결절형성에 관여하는 유전자들의 분석 (Identification of Matrix Mineralization-Related Genes in Human Periodontal Ligament Cells Using cDNA Microarray)

  • 치주인대세포는 시험관적 실험에서 광물화 결절형성을 유도할 수 있으므로 광물화 결절형성에 관여하는 유전자들을 특이하게 발현할 것으로 여겨진다. 이에 본 실험은 cDNA microarray를 이용한 동시 유전자분석을 시행하여 치주인대세포의 분화에 의한 광물화 결절형성시 나타나는 유전자의 특징적 발현 양상을 알아보고자 하였다. 교정치료를 목적으로 경북대학교병원에 내원한 환자의 제일소구치를 발치하여 통상적 방법으로 치주인대세포를 분리, 배양하였고, 3세대의 치주인대세포를 사용하여 실험을 시행하였다. 치주인대세포를 100mm 배양접시에 넣고 배양하여 매 2일 마다 배지를 교환해 주고, 10% FBS만을 투여한 대조군으로, ascorbic acid $(50\;{\mu}g/ml)$, ${\beta}-glycerophosphate$ (10 mM) 및 100 nM dexamethasone을 투여한 군을 실험군으로 하였다. 배양된 치주인대세포에 ascorbic acid, ${\beta}-glycerophosphate$, 그리고 dexamethasone을 투여한 실험군에서 21일째 광물화된 결정을 관찰할 수 있었으나 대조군에서는 관찰할 수 없었다. 3063개의 유전자를 분석한 결과 35개 유전자가 대조군에 비해 2배이상 발현이 증가하였고, 38개 유전자는 2배이상 발현이 감소하였다. 형태학적 검사에서 보여준 바와 같이 광물화 형성과정시 관여하는 JGF-2과 IGFBP2와 같은 유전자가 실험군에서 증가하였으며, 세포골격과 세포외기질 형성에 관여하는 proteogycan 1, fibulin-5, keratin 5, ${\beta}-actin$, ${\alpha}-smooth$ muscle actin, capping protein 등도 발현이 실험군에서 증가하였다. 한편 periostin and S100 calcium-binding protein A4는 대조군에서 오히려 높게 나타나므로 이는 배양된 치주인대세포가 그 자체의 표현형을 유지하고 있음을 보여 주고 있다. 그 외 apoptosis를 유발시키는데 관여하는 Dkk-1와 Nip3는 실험군에서 높게 발현되었고, apoptosis를 억제시키는데 관여하는 Btf와 TAX1BP1는 오히려 낮게 발현됨을 알 수 있으므로 이는 실험군에서 치주인대세포가 골아세포로의 분화되었음을 나타낸다.

유해적조생물 Cochlodinum polykrikoides를 살멸하는 Brachybacterium sp. SY-97의 분리 및 특성 (Isolation and Characteristics of Brachybacterium sp. SY -97 Killing the Harmful Dinoflagellate Cochlodinium polykrikoides)

  • A bacterial strain SY-97 that showed algicidal activity against Cochlodinium polykrikoides was isolated from coastal water of Uljin (eastern coast of Korea) in August, 2005. The isolated strain was identified as Brachybacterium sp. by morphological and biological tests, and analysis of 16S rDNA sequence. The optimal culture conditions for the growth of strain SY-97 were $30^{\circ}C$, initial pH 7.0, and salinity 2.0%. From the result of cell culture insert experiment, Brachybacterium sp. SY-97 is assumed to produce secondary metabolites which have algicidal activity. When 10% culture filtrate of this strain was applied to C. polykrikoides ($1.2{\times}10^4\;cells/m{\ell}$) cultures, 100% of C. polykrikoides cells was destroyed within 15 hours. The released algicides were heat-tolerant to $100^{\circ}C$ and stable in pH $6.0{\sim}10.0$. These results suggest that Brachybacterium sp. SY-97 is potentially useful for controlling outbreaks of C. polykrikoides.

생쥐 난자-난구 복합체의 성숙과 분산에 관한 연구 : 세포내 cAMP의 조절 (Studies on the Cumulus Expansion and Oocyte Maturation of Mouse Cumulus-Oocyte Complexes: Regulation of Intracellular cAMP Level)

  • 생쥐 난자-난구 복합체를 인공배양하면서 adenylate cyclase의 촉진제인 forskilin과 phosphodiesterase의 저해제인 3-isobutyl-1-methylxanthine(IBMX)을 배양액에 첨가하여 이들이 난구세포의 분산과 난자의 성숙(핵붕괴)에 미치는 효과를 관찰한 결과 다음과 같았다. 1. Forskilin은 0.001$\mu$M에서 부터 난구세포의 분산을 유도하기 시작하여 (36%) 0.1-10$\mu$M 구간에서 최대의 분산율을 나타내었고 (80-90%) 100$\mu$M에서는 그 효과가 줄어들었다(60%). 이때 난자의 핵 붕괴는 10$\mu$M까지 정상으로 일어나다 (75-80%) 100$\mu$M에서 부분적으로 억제되기 시작하였다(40%). 2. IMBX는 0.01$\mu$M에서 부터 난구세포의 분산을 유도하기 시작하여 (30%) 1-1,000$\mu$M의 전 구간에서 최대의 분산율(81-89%)을 나타내었다. 난자들은 10$\mu$M의 농도까지 정상적으로 핵 붕괴를 일으키었으나(90% 이상) 100$\mu$M 이상에서 급격히 억제되었다(14%). 3. 난구세포의 분산을 유도하는데 필요한 최소의 자극기간을 조사해 본 결과 HCG는 2분, FSH와 forskilin은 15-30분, IBMX는 2시간이었다. 위 결과로 부터 생쥐 난구세포에서 cAMP의 농도를 높이는데 adenylate cyclase 와 phosphodiesterase의 두 효소가 모두 중요한 기여를 하며 난구세포는 단 기간에 생긴 cAMP의 peak로서 분산이 유도될 수 있으나 난자의 성숙억제 과정에서는 지속적인 cAMP의 존재가 필요하다는 것을 알았다.

  • Solar cells have been more intensely studied as part of the effort to find alternatives to fossil fuels as power sources. The progression of the first two generations of solar cells has seen a sacrifice of higher efficiency for more economic use of materials. The use of a single junction makes both these types of cells lose power in two major ways: by the non-absorption of incident light of energy below the band gap; and by the dissipation by heat loss of light energy in excess of the band gap. Therefore, multi junction solar cells have been proposed as a solution to this problem. However, the $1^{st}$ and $2^{nd}$ generation solar cells have efficiency limits because a photon makes just one electron-hole pair. Fabrication of all-silicon tandem cells using an Si quantum dot superlattice structure (QD SLS) is one possible suggestion. In this study, an $SiO_x$ matrix system was investigated and analyzed for potential use as an all-silicon multi-junction solar cell. Si quantum dots with a super lattice structure (Si QD SLS) were prepared by alternating deposition of Si rich oxide (SRO; $SiO_x$ (x = 0.8, 1.12)) and $SiO_2$ layers using RF magnetron co-sputtering and subsequent annealing at temperatures between 800 and $1,100^{\circ}C$ under nitrogen ambient. Annealing temperatures and times affected the formation of Si QDs in the SRO film. Fourier transform infrared spectroscopy (FTIR) spectra and x-ray photoelectron spectroscopy (XPS) revealed that nanocrystalline Si QDs started to precipitate after annealing at $1,100^{\circ}C$ for one hour. Transmission electron microscopy (TEM) images clearly showed SRO/$SiO_2$ SLS and Si QDs formation in each 4, 6, and 8 nm SRO layer after annealing at $1,100^{\circ}C$ for two hours. The systematic investigation of precipitation behavior of Si QDs in $SiO_2$ matrices is presented.

일산화탄소를 이용하여 성장하는 acinetobacter의 분리 및 동정 (Acinetobacter Isolates Growing with Carbon Monoxide)

  • 일산화 탄소를 이용한 enrichment 배양 방법을 통하여 흙으로부터 일산화탄소를 이용하여 성장 할 수 있는 세가지 종류 (JC1, JC2, HY1)의 호기성 Acinetobacter 들을 분리했다. 이세균들은 모두Gramdma성 세균들로 운동성이 없었으며, 지수성장기에는 간균의 형태를 나타내었으나 성장이 정지되었을 때에는 구균으로 변하였다. 이들은 페니실린에 대해 내성이 있었고 $42^{\circ}C$에서도 성장을 할 수 있었다. DNA의 G+C함량은 43%-44.5%이였고 모든 세균에서 oxidase의 활성이 나타나지 않았다. 이 세균들의 coloy들은 모두 둥글고 매끄러웠으며 연한 노란색을 띄었다. 이들은 또 여러가지 종류의 당이나 유기산, 아미노산. 알코올 등을 이용하여 생장할 수 있였다. JC1과 J JC2 벚 HY1이 30%의 일산화탄소를 이용하여 $30^{\circ}C$에서 성장할 때의 doubling time은 각각 19시간. 25시간, 그리 고 35시간 이었다. JC1의 자가영양적 성장을 위한 최적조건은 pH가 6.8이였L 온도는 $42^{\circ}C$ 그리고 일산화탄소의 농도는 30%이였다. JC1 의 자가영양적 성장에는 몰리브데늄이 필요치 않았고, 또 이 세균은 100ppm의 CO만으로도 성장할 수 있는 것으로 밝혀졌다.

Highly Sulfonated Poly(Arylene Biphenylsulfone Ketone) Block Copolymers Prepared via Post-Sulfonation for Proton Conducting Electrolyte Membranes

  • A series of the block copolymers were successfully synthesized from post-sulfonated hydrophilic and hydrophobic macromers via three-step copolymerization. The degrees of sulfonation (DS) of the copolymers (10%, 30%, or 50%) were controlled by changing the molar ratio of the hydrophilic and hydrophobic parts. The resulting block copolymers were characterized by $^1H$ NMR and other technologies. The membranes were successfully cast using dimethyl sulfoxide (DMSO) solution at $100^{\circ}C$. The copolymers were characterized to confirm chemical structure by $^1H$ NMR and FT-IR. Thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) demonstrated that all sulfonated block copolymers exhibited good thermal stability with an initial weight loss at temperatures above $240^{\circ}C$. The membranes showed acceptable ion exchange capacity (IEC) and water uptake values in accordance with DS. The maximum proton conductivity was 184 mS $cm^{-1}$ in block copolymer-50 at $60^{\circ}C$ and 100% relative humidity, while the conductivity of Nifion-115 was 160 mS $cm^{-1}$ under the same measurement conditions. AFM images of the block copolymer membranes showed well separated the hydrophilic and hydrophobic domains. From the observed results it is that the prepared block membranes can be considered as suitable polymer electrolyte membranes for the application of polymer electrolyte membrane fuel cells (PEMFC).
