약수에서 분리한 Yersinia pseudotuberculosis의 병원성과 16S rDNA 분석에 의한 분자학적 분류 (Molecular Taxonomy based on 16S rDNA Analysis and Pathogenicity of Yersinia pseudotuberculosis Isolated from Spring Waters)

  • 서울시내 25개 자치구에 산재한 약수터에서 5주의 Yersinia pseudotuberculosis를 분리하여 생화학적 특성, 병원성의 유무 및 16S rDNA 분석을 실시하였다. 분리된 Y. pseudotuberculosis는 모두 병원성 유전자인 inv를 소유하고 있었으며, 16S rDNA를 증폭한 후 염기서열을 분석하여 NCBI Genbank에 등록된 다른 Yersinia 속 및 장내세균 등과 비교해 본 결과 Yersinia 속 등과는 97.5%에서 100%의 높은 상동성(Similarity)을 나타내었고, 다른 장내세균 등과는 93.0%에서 95.1%의 낮은 상동성을 나타내었다. 165 rDNA 염기서열을 기초로 계통수를 작성한 결과 크게 3개의 cluster를 형성하였는데 특히 Y.enterocolitica (Z49830)은 Y.pseudotuberculasis (Z21939)와의 상동성(97.7%)보다 Y.intermedia (X75279)와의 상동성(97.9%)이 더 높게 나타났으며, E. coli (Z83205)는 Proteus vulgaris (AJ233425) 와의 상동성(93.2%)보다 Salmonella enteritidis (U90318)와의 상동성(97.7%)이 더 밀접한 연관성을 나타내었다.

Corynebacterium pseudotuberculosis에 의한 유산양의 가성결핵 증례 (Caseous lymphadenitis by Corynebacterium pseudotuberculosis in a Saanen dairy goat (Capra hircus aegagrus))

  • Caseous lymphadenitis (CLA) is a chronic and contagious disease of sheep and goats caused by Corynebacterium (C.) pseudotuberculosis. A four-year-old female Saanen dairy goat was submitted to the Animal Disease Diagnostic Center at National Veterinary Research and Quarantine Service. The clinical signs of the goat were emaciation, abortion and quadriplegia. The multifocal nodules of lymph nodes were encapsulated and filled with whitish caseous contents on the cut surface. Histopathologically, lymph nodes displayed suppurative and necrotizing granulomas. Caseous necrosis was diffusely observed in the center of the lymph nodes. Gram positive bacilli were shown in the lesions. C. pseudotuberculosis was isolated and confirmed by the biochemical tests and PCR assay. Based on clinical signs, histopathological examination and bacterial isolation, we diagnosed this case as CLA. To our knowledge, this is the first report of CLA in a Saanen dairy goat in Korea.

흑염소의 건락성 림프절염 제어를 위한 소독제 효능 비교 (Comparison of the Efficacy of Disinfectants to Control Caseous Lymphadenitis in Korean Black Goat Farms)

  • 최근 국내의 흑염소 사육량과 소비량이 지속적으로 증가하고 있다. 흑염소 사육방식 또한 대량화, 밀집화 형태로 변화하는 추세이며, 이에 따라 다양한 전염성 질병이 흑염소 농장에 발생하고 있다. 그중 소형 반추류에 발생하는 건락성 림프절염은 국내 흑염소 농장에서 유병률이 50% 이상에 이를 정도로 빈발하고 있으나 현재 마땅한 제어 방법이 없는 실정이다. 따라서, 본 연구에서는 흑염소의 대표적인 세균성 질병인 건락성 림프절염의 제어를 위한 노력의 일환으로 원인균인 Corynebacterium pseudotuberculosis(C. pseudotuberculosis)에 효과적인 소독제를 확인하고자 하였다. 국내 시판되고 있는 축산용 소독제를 유효성분으로 구분하여 6종의 소독제에 대하여 소독제 효력시험을 수행하였다. 실험 대상으로 선정된 6종의 소독제는 산성제(구연산, Citric acid), 할로겐계(이염화이소시아눌산나트륨, Sodium dichloroisocyanurate), 산소계(모노과황산칼륨화합물, Potassium monopersulfate triple salt), 염기제(황산구리수화물, Copper sulfate pentahydrate), 양이온성 세정제(4급 암모늄, Dialkyl dimethyl ammonium chloride), 염소계(차아염소산나트륨, Sodium hypochlorite)이다. 농림축산검역본부 소독제 효력시험지침에 준하여 실험한 결과, 경수 조건에서는 황산구리수화물을 제외한 모든 소독제가 소독제 효력시험지침에서 제시하는 기준인 99.99% 이상의 살균력을 보였으며, 유기물 조건에서 구연산, 모노과황산칼륨 화합물 및 4급 암모늄 제재의 3종만이 99.99% 이상의 살균력을 보였다. 표준 효력시험지침(30분)보다 짧은 시간(1분, 5분, 15분) 동안 처리 후 살균력을 확인하였을 때, 경수 조건에서는 이염화이소시아눌산나트륨, 모노과황산칼륨 화합물, 차아염소산나트륨 제재가 가장 효과가 좋았으며 1분간 처리 시에도 99.99% 이상의 살균력을 보였다. 유기물 조건의 경우에는 구연산과 4급 암모늄 제재가 가장 효과가 높았으며 5분간 처리 시에도 99.99% 이상 또는 근접하는 살균력을 유지하였다. 본 실험 결과를 종합하여 볼 때 축산용 소독제의 유효성분에 따라 C. pseudotuberculosis에 대한 살균력에 차이가 있으며 경수 및 유기물 조건에 따른 차이도 확인된다. 따라서 C. pseudotuberculosis에 효과적인 소독제 선정 및 활용을 통하여 흑염소 농장 내 환경 오염을 방지한다면 건락성 림프절염 저감화에 효과가 있을 것으로 기대된다.

염소의 피하농양 발생에 따른 혈액 내 염증지표 분석 (Analysis of inflammatory markers in blood related with the occurrence of subcutaneous abscesses in goats)

  • Subcutaneous abscesses, which occur mainly in goats and sheep, are lymph node abscesses caused by Corynebacterium pseudotuberculosis infection, and are divided into internal, external, and mixed types depending on the type of occurrence. While diagnostic methods for subcutaneous abscesses have been continuously studied, research reports for effective treatment and management of subcutaneous abscesses are inadequate. Therefore, this study was conducted to determine the changes in biometric information related to the inflammatory markers of goats induced by subcutaneous abscesses by infection with C. pseudotuberculosis. For this, hematological tests, analysis of inflammatory indicators, and analysis of serum proteins through electrophoresis separation of goats with healthy goats and goats inoculated with C. pseudotuberculosis to induce subcutaneous abscesses were compared and analyzed by date, and the differences and characteristics were identified periodically. As a result, in goats induced with subcutaneous abscesses, anemia findings related to a rapid decrease in red blood cell (RBC), hematocrit (HCT), and hemoglobin (Hb) were observed, and a significant increase in inflammatory cells expressed in total white blood cell (WBC), neutrophil, and monocytes was observed. And the levels of acute phase protein (APP) such as fibrinogen, haptoglobin, and serum amyloid A (SAA) were observed to increase rapidly immediately after infection. In addition, in the results of electrophoretic analysis of serum proteins, it was observed that the levels of α-globulin and β-globulin were significantly increased in goats with subcutaneous abscesses. That is, when looking at these changes, it was found that the systemic inflammatory response of goats was rapidly induced immediately after infection with the C. pseudotuberculosis pathogen. Through this study, it was possible to identify changes in the biomarkers of goats with subcutaneous abscesses, which had not been reported. Furthermore, these analyzed data are thoughts to be of great help in identifying, treating, and managing the goats of subcutaneous abscesses.

우리나라 전염성 질환의 변화 양상 (Changing Patterns of Communicable Diseases in Korea)

  • Before twentieth centuries and during early twentieth centuries, communicable diseases were the major cause of morbidity and mortality in Korea. But reliable data are not available. After 1975, the overall morbidity and mortality from communicable diseases, rapidly declined. Recently many new pathogenic microbes were recognized: L. monocytogenes, Hantaan virus, Y. pseudotuberculosis, P. multocida, L. pneumophilia, Human immunodeficiency virus (HIV), G. seoi, H. capsulatum, C. burnetii, V. cholerae O139, C. parvum, F. tularensis, E. coli O157:H7, B. burgdorferi, S. Typhimurium DT104, Rotavirus, hepatitis C virus and so on. Since the first HIV infection recognized in 1985, the reported cases of infection and deaths from HIV/AIDS have been steady increased each year. Legionnaire's disease, E. coli O157:H7 colitis, listeriosis and crytosporidiasis have been occurring just sporadically among immunocompromized cases. Many re-emerging communicable diseases were occurred in Korea: leptospirosis, malaria, endemic typhus, cholera, tsutsugamushi disease, salmonellosis, hepatitis A, shigellosis, mumps, measles, acute hemorrhagic conjunctivitis, brucellosis and so on. Leptospirosis and tsutsugamushi diseases have been noticed as major public health problems since 1980s. The malaria that had been virtually disappeared for a decade has reappeared from 1993 with striking increase of patients in recent 3-4 years. The distributions of salmonella and shigella serotypes have been changed a lot in recent few decades. Furthermore rapid emergence of antibiotic-resistant bacterial strains induces more difficult and complex problems in control of communicable diseases. We must recognize on the importance of environment and ecosystem conservation and careful prescription of anti-microbial agent in order to prevent communicable diseases.

A Newly Identified Glutaminase-Free L-Asparaginase (L-ASPG86) from the Marine Bacterium Mesoflavibacter zeaxanthinifaciens

  • L-Asparaginase (E.C. 3.5.1.1) is an enzyme involved in asparagine hydrolysis and has the potential to effect leukemic cells and various other cancer cells. We identified the L-asparaginase gene (L-ASPG86) in the genus Mesoflavibacter, which consists of a 1,035 bp open reading frame encoding 344 amino acids. Following phylogenetic analysis, the deduced amino acid sequence of L-ASPG86 (L-ASPG86) was grouped as a type I asparaginase with respective homologs in Escherichia coli and Yersinia pseudotuberculosis. The L-ASPG86 gene was cloned into the pET-16b vector to express the respective protein in E. coli BL21 (DE3) cells. Recombinant L-asparaginase (r-L-ASPG86) showed optimum conditions at 37-40℃, pH 9. Moreover, r-L-ASPG86 did not exhibit glutaminase activity. In the metal ions test, its enzymatic activity was highly improved upon addition of 5 mM manganese (3.97-fold) and magnesium (3.35-fold) compared with the untreated control. The specific activity of r-L-ASPG86 was 687.1 units/mg under optimum conditions (37℃, pH 9, and 5 mM MnSO4).
