Seed Characteristics and Accelerating Method of Germination in Bupleurum falcatum (시호 종자의 특성 및 발아촉진 방법 연구)

  • The experiment was conducted to determine the seed characteristics and preferable methods to enhance the seed germination rate in Bupleurum falcatum. The optimum temperature for the seed germination of Bupleurum falcatum is 2$0^{\circ}C$. Any significant promoting effects were not found in seed germination with hormone treatments and physical methods. At 15$^{\circ}C$, prechilling combined with 50~200ppm of GA$_3$ treatment raised germination rate by 2 times of control ones. The most positive effect was observed in the treatment of 10$^{-2}$ ~10$^{-3}$ M potassium nitrate only at 15$^{\circ}C$ for 12 and 24 hours. The leachate of Bupleurum falcatum didn't inhibit the germination of Lactuca sativa, showing almost 100% of germination rate, which is suggested that no inhibitors contained in the seeds of Bupleurum falcatum. Observation of embryo conditions under stereoscopic microscope showed that the ratio of seeds with or without embryo is almost 50/50. The results suggested that the lower rate of germination in Bupleurum falcatum was caused by embryolessness of seeds.

Comparison on Morphology and saikosaponin Contents of Bupleurum falcatum Produced in Korea and China (한국 및 중국산 柴胡의 형태와 saikosaponin 함량비교)

  • To determine the differences among cultivated , non-cultivated and imported bupleurum falcatum, contents and compositions of fatty acids and saikosaponins which are known as a major biologically acitive compounds in Bupleurum falcatum were meausred with GC and HPLC. Color of grinded powder of Bupleuri radix determined by colorimeter showed that the imported one has the highest white and yellow coloer Among the Bupleuri radix cultivated in China, Hong-siho(red-Bupleurum) produced in Yungil Province contained the highest red color. Cultivated Korean Bupleurum radix contained higher saikosaponi a, c, and than the imported ones which were cultivated in Yungil, Ankuk , and Gillium Province in China. Levels of saikosaponin c are 3 times higher in cultivated Korean Bupleuria radix compared the with imported ones. Furthermore, Bupleurum falcatum cultivated in Korea contained higher amount of palmitic and linoleic acids than those of improted and non-cultivated ones. Particularly they contain two to six times higher linoleic acid.

Agrobacterium tumefaciens-mediated Transformation in Colletotrichum falcatum and C. acutatum

  • Agrobacterum tumefaciens-mediated transformation (ATMT) is becoming an effective system as an insertional mutagenesis tool in filamentous fungi. We developed and optimized ATMT for two Colletotrichum species, C. falcatum and C. acutatum, which are the causal agents of sugarcane red rot and pepper anthracnose, respectively. A. tumefaciens strain SK1044, carrying a hygromycin phosphotransferase gene (hph) and a green fluorescent protein (GFP) gene, was used to transform the conidia of these two Colletotrichum species. Transformation efficiency was correlated with co-cultivation time and bacterial cell concentration and was higher in C. falcatum than in C. acutatum. Southern blot analysis indicated that about 65% of the transformants had a single copy of the T-DNA in both C. falcatum and C. acutatum and that T-DNA integrated randomly in both fungal genomes. T-DNA insertions were identified in transformants through thermal asymmetrical interlaced PCR (TAIL-PCR) followed by sequencing. Our results suggested that ATMT can be used as a molecular tool to identify and characterize pathogenicity-related genes in these two economically important Colletotrichum species.

  • Saikosaponin derivatives such as saikosaponins A, B1, B2, B3, B4, C, and D present in Bupleurum falcatum were analyzed by a high performance liquid chromatograph equipped with an evaporative light scattering detector, using different extraction solvents (water and 70% ethanol). The samples were injected into a YMC Pack Pro C18 column and separated using a gradient elution system with a mobile phase composed of acetonitrile and water at a flow rate of 1.1 mL/min. The content of saikosaponin derivatives was higher in 70% ethanol extract than in water extract. This study provides an efficient analytical method for determining the optimal conditions for extraction of saikosaponin derivatives, which can be used as a basis for development of functional foods and pharmaceutical products from B. falcatum.

  • Macromyzus woodwardiae(Takahashi, 1921) is recognized on Cyrdomium falcatum(L.f.) C. Presl (Dryopteridaceae), a htherto unknown host plant, from Jeju Island, Korea. Apterous viviparous female and alate viviparous female are described. This is the first record of genus Macromyzus in Korea.

Methanolic extract from the root of Bupleuri falcatum L. attenuates cocaine-induced c-Fos expression in rat brain. (시호 메탄올 추출물이 코카인 약물중독에 의한 흰쥐 뇌의 c-Fos 발현에 미치는 영향)

  • Objectives : The aim of present study is to examine the effect of methanolic extract from the root of Bupleurum falcatum L. (BF) on acute cocaine-induced c-Fos expression in the rat caudate putamen (CPu), a major dopaminergic terminal. Methods : Male Sprague-Dawley rats were subjected to an intraperitoneal injection with either cocaine hydrochloride (20 mg/kg) or saline 30 min after an administration of either extract of BF (100 mg/kg, i.p.) or vehicle. Animals were sacrificed 2 hr after treatment with cocaine or saline for immunohistochemistry. Quantification of brain slices was examined for c-Fos positive nuclei using light microscopy. Results : Pretreatment with BF significantly attenuated cocaine-induced c-Fos expression in the rat CPu. Conclusions : This finding suggests that BF has the inhibitory effect on cocaine-induced c-Fos expression in the rat CPu via possibly modulating the activities of central dopaminergic systems.

Toxin Produced by Colletotrichum falcatum Causing Red Rot of Sugarcane

  • Toxin produced by Colletotrichum falcatum Went, the incitant of red rot of sugarcane was isolated, purified and assayed to determine host specificity and identify its chemical nature. The toxin was found to be not host specific as it inhibited germination of various seeds(gram, greengram, blackgram, pea, cowpea, rice and sugarcane) as well as different seedlings viz. tomato, coriander, pea and rice. The toxin consists of two distinct fraction-one fraction having $R_f$, value at 0.36 producing identical red rot lesion when inoculated at leaf midrib of sugarcane, and the other having $R_f$, value at 0.72 not showing any red rot lesion. Chromatogram of high performance liquid chromatography(HPLC) of the red rot lesion causing fraction showed a sharp peak at 1.62 min of retention time(RT), and spectral analysis indicated the presence of following chemical $CH_3$ - groups-C-H, C=O, C-N, $-CH_3,\;-CH_2$ -CH and molecular mass of the compound was 203. - ($M^+,\;C_{11}H_{11}N_2O_2$).

  • The gametophytes of Cyrtomium falcatum and Cyrtomium caryoptideum var. coreanum arising from spores were mechanically homogenized and cultured In vitro, to study their gametophyte ontogeny and sporophyte development. Homogenized gametophytic tissues formed as one-dimensional filaments after 2 weeks in culture and then glowed into blanched gamatophytes after 4 weeks. After 6 weeks, which were developed to two dimensional plates with apical notch and meristem in central zone. After 8 weeks in culture, apomictic buds were formed on the midribs without archegonium formation and these buds developed to sporophytes after 10 weeks in culture. Flow cytometric analysis of gametophytes and apomictic sporophytes revealed that both forms had the same ploidy level in C. falcatum and C. caryoptideum vu. coreanum, respectively. This is to certify that C. caryoptideum var. coreanum was an apomictic fern as well as C. falcatum.

Pigment and Saikosoponin Production Through Bioreactor Culture of Carthamus tinctorius and Bupleurum falcatum

  • Traditional culture technology of medicinal plants mainly depends on the field culture, which has many problems. With progress of modern culture technology, it has become possible to produce valuable secondary metabolites from medicinal plants. In this paper, we discuss about the pigment and saikosaponin production from too medicinal plants, Carthamus tinctorius and Bupleurum falcatum, through bioreactor culture system. A two-stage bioreactor culture system was established for the production of yellow and red pigments and saikosaponins by cell suspension cultures of Carthamus tinctorius and Bupleurum falcatum. In Carthamus tinctorius, balloon type airlift bioreactors and column type airlift bioreactors were employed for the tell culture and for the pigment production, respectively. The greatest pigment production was obtained on White medium supplemented with 4 mg/L kinetin, high levels of sucrose concentration and photosynthetic photon flux. In Bupleurum falcatum, adventitious roots were cultured in balloon type airlift bioreactors and the root growth was greatest on SH medium containing 5 mg/L IBA and 0.2 mg/L kinetin. HPLC analysis showed that the contents of main active saikosaponins a, c, and d in adventitious roots were almost the same as those in field cultured root.
