Effects of Lycium chinense Miller Fruit and its Constituent Betaine on Immunomodulation in Balb/c Mice

  • BACKGROUND: The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS: We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. CONCLUSION: These results indicate that L. chinense Miller fruit and betaine might be helpful in the immune function improvement, enhance physical stamina, and fatigue recovery.

DNA Barcoding of the Endangered Species Ellobium chinense (Mollusca, Gastropoda, Ellobiidae) from Coastal Areas of South Korea

  • The pulmonate gastropod Ellobium chinense (Pfeiffer, 1864) is an endangered marine species along the South Korean coasts due to habitat destruction and population declines. We sequenced the cytochrome c oxidase subunit 1 (COI) of 25 E. chinense specimens collected from five coastal sites in South Korea, and identified 16 unique haplotypes. The maximum intraspecific variation among individuals was 1.6%, while interspecific differences from another ellobiid species, Auriculastra duplicata (Pfeiffer, 1854), ranged from 21.9 to 23.0%. Our barcoding data will be useful to elucidate the phylogenetic relationships among pulmonate gastropods and infer the population genetic structure of E. chinense.

구기자를 첨가한 고추장의 숙성 중 이화학적 특성 (Effect of Lycium chinense Fruit on the Physicochemical Properties of Kochujang)

  • 고추장의 품질향상을 위하여 구기자의 농도를 달리하여 첨가하고 $20^{\circ}C$에서 12주간 숙성시키면서 효소활성도와 미생물상, 이화학적 특성을 비교하였다. 고추장의 효소활성도는 숙성 중기 이후에 높은 활성을 보였으며, 구기자의 첨가로 protease의 활성은 증가하였다. 고추장 숙성중의 통성 혐기성 세균은 구기자의 첨가로 감소하였나, 효모수는 구기자 1%첨가 고추장에서 많았다. 고추장의 점조성은 구기자의 첨가로 감소하였고, 색도는 구기자의 첨가량이 증가할수록 숙성 중 L값과 a값의 저하가 적어 ${\Delta}E$값의 변화가 적었다. 고추장의 수분은 숙성 중에 서서히 증가하였으나 수분활성도는 감소하였고, 구기자 첨가량이 많은 구에서 높았다. 고추장의 산도와 산화환원전위는 구기자 첨가로 증가하였고, 총당의 감소는 구기자 첨가량이 증가할수록 심하였다. 환원당은 숙성 $2{\sim}4$주에 급격히 증가하였으나, 구기자 첨가량이 증가할수록 적었다. 알코올은 숙성이 진행되면서 증가하여 3% 구기자 첨가 고추장에서 높았다. 고추장의 아미노태 질소와 암모니아태 질소는 구기자 첨가시 증가하였다. 12주 숙성시킨 고추장의 맛과 종합적인 기호도는 3% 구기자 첨가구가 우수하였고 색은 구기자 첨가량이 증가할수록 양호하여, 고추장 제조시 구기자를 $1{\sim}3%$ 첨가하면 고추장의 품질이 향상되었다.

New distribution records of two rare species of Cynanchum (Apocynaceae) in South Korea: Cynanchum thesioides (Freyn) K. Schum. and Cynanchum chinense R. Br.

  • Cynanchum L. in the subfamily Asclepiadoideae (Apocynaceae) includes four recognized species on the Korean peninsula, two of which are native to South Korea. However, the species ranges in South Korea are poorly defined. During a field survey, we discovered C. thesioides, previously unrecorded in South Korea, in Gimpo-si, Gyeonggi-do, and found an additional population of C. chinense, for which only one population has been reported in South Korea. The two taxa are considered rare species with extremely restricted distributions in South Korea, especially C. thesioides. We provide fundamental information, including descriptions, images of the habitats and morphological characters, and a taxonomic key for identification and assessments of the conservation status of Cynanchum species in Korea.

구기자 탄저병에 관여하는 2종의 Colletotrichum에 관하여 (Two Species of Colletotrichum Associated with Anthracnose of Lycium chinense)

  • 구기자(Lycium chinense Mill) 탄저병은 구기자 열매에 수침상의 흑색 병반을 형성하고 포자층을 형성한다. 일반농가 포장에서의 발병율은 $0.8\%\~23\%$(평균 $9.6\%$)의 이병과율을 보였다. 구기자 탄저병균은 Colletotrichum gloeosporioides Penz와 Colletotrichum dematium(Fr.) Grove의 2종으로 동정되었다. 구기자에 대한 병원성은 공시한 C. gloeosporioides와 C. dematium 모두 어린 잎이나 줄기에는 병원성이 없었고 열매에는 강한 병원성을 나타냈다. 성숙과에는 C. dematium Isolate I이 특히 병원성이 강하였고 미숙과에 대하여는 C. dematium Isolate II만이 약하게 나타났으나 10일정도 경과하면 병원성 정도의 구분이 어려웠다.

Resistance to Anthracnose Caused by Colletotrichum acutatum in Chili Pepper(Capsicum annuum L.)

  • Pepper fruit anthracnose, caused by Colletotrichum acutatum, results in serious yield loss and affects crop quality in many Asian countries, making it a disease of economic consequence. A source resistant to C. acutatum was identified by the AVRDC within the line Capsicum chinense Jacq. PBC932. The resistant breeding line C. annuum AR is the $BC_3F_6$ generation derived from C. chinense Jacq. PBC932. The inheritance of resistance to C. acutatum was analyzed in segregating populations derived from the two crosses HN 11$\times$AR and Daepoong-cho$\times$AR. Detached mature green fruits were inoculated using microinjection method. The disease response was evaluated as the disease incidence at 7 DAI. The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_R$ populations derived from the two crosses fit significantly to a 1:3 Mendelian model. This indicates that the resistance of AR to C. acutatum is controlled by a single recessive gene.

Isolation of an Angiotensin Converting Enzyme Inhibitory Substance from Lycium chinense Miller

  • An angiotensin converting enzyme (ACE) inhibitory substance was isolated and purified from Lycium chinense Miller. A crude water extract of Lycium chinense Miller was prepared by adding it to water shaking at $25^{\circ}C$ for 1 hr, followed by centrifugation at 8000 ${\times}$ g for 30 min. The crude extract was then filtered using YM-3 and YM-1 membranes. An ACE inhibitor was isolated using consecutive chromatographic methods including: ion exchange chromatography, gel permeation chromatography, and FPLC. The inhibitor was identified to have a molecular mass of 862 daltons by mass spectrometry.

A Convenient HPLC/ELSD Method for the Quantitative Analysis of Betaine in Lycium chinense

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

Analytical Method Validation of (-)Epicatechin gallate in Penthorum chinense Pursh Extract using HPLC

  • This study attempted to establish a High Performance Liquid Chromatography (HPLC) analysis method for the determination of (-)-epicatechin gallate as a part of the quality control for the development of functional cosmetic materials from Penthorum chinense Pursh extracts. HPLC was performed on a Unison US-C18 column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and methyl alcohol at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 280 nm. The HPLC method was performed in accordance with the International Conference on Harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation were 0.11 and 0.33 mg/mL, respectively. Calibration curves showed good linearity (r2 > 0.9999), and the precision of analysis was satisfied (less than 0.6%). Recoveries of quantified compounds ranged from 99.51 to 101.92%. This result indicates that the established HPLC method is very useful for the determination of marker compound in P. chinense Pursh extracts.
