A comparative study between sterile freeze-dried and sterile pre-hydrated acellular dermal matrix in tissue expander/implant breast reconstruction

  • Background In implant-based breast reconstruction, acellular dermal matrix (ADM) is essential for supporting the inferolateral pole. Recent studies have compared non-sterilized freeze-dried ADM and sterilized pre-hydrated ADM, but have not assessed whether differences were attributable to factors related to sterile processing or packaging. This study was conducted to compare the clinical outcomes of breast reconstruction using two types of sterile-processed ADMs. Methods Through a retrospective chart review, we analyzed 77 consecutive patients (85 breasts) who underwent tissue expander/implant breast reconstruction with either freeze-dried ADM (35 breasts) or pre-hydrated ADM (50 breasts) from March 2016 to February 2018. Demographic variables, postoperative outcomes, and operative parameters were compared between freeze-dried and pre-hydrated ADM. Biopsy specimens were obtained for histologic analysis. Results We obtained results after adjusting for variables found to be significant in univariate analyses. The total complication rate for freeze-dried and pre-hydrated ADMs was 25.7% and 22.0%, respectively. Skin necrosis was significantly more frequent in the freeze-dried group than in the pre-hydrated group (8.6% vs. 4.0%, P=0.038). All other complications and operative parameters showed no significant differences. In the histologic analysis, collagen density, inflammation, and vascularity were higher in the pre-hydrated ADM group (P=0.042, P=0.006, P=0.005, respectively). Conclusions There are limited data comparing the outcomes of tissue expander/implant breast reconstruction using two types of sterile-processed ADMs. In this study, we found that using pre-hydrated ADM resulted in less skin necrosis and better integration into host tissue. Pre-hydrated ADM may therefore be preferable to freeze-dried ADM in terms of convenience and safety.

  • 눈개승마 (Aruncus dioicus)는 장미과(Rosaceae) 눈개승마속(Aruncus)에 속하는 다년생 초본식물로 울릉도에서 자생하는 잎줄기 채소이다. 독특한 향기가 특징이며, 인삼맛, 두릅맛, 고기맛이 나는 산채로 세가지 맛이 난다고 하여 예로부터 삼나물로도 불러왔다. 눈개승마에는 caffeic acid, 1-O-Caffeoyl-β-D-glucopyranose 등의 항산화 성분이 다량 함유되어 있으며, 전초는 해독, 편도선염에 효과가 있어 약용으로 쓰이고 어린순은 탄수화물과 무기물 함량이 풍부하다. 비지는 대두를 이용하여 두부를 제조하는 과정에서 발생되는 것으로서 대두로부터 수용성 단백질이 추출되었지만 나머지 약 23%의 단백질, 57%의 식이섬유와 이소플라본 등의 유용한 성분을 다량 포함하고 있다. 또한 비지의 단백질은 다른 식품에는 부족한 황 함유 아미노산과 리신의 함량이 비교적 많고, 두부에 거의 없는 식이섬유가 다량 함유된 고 식이섬유 소재이다. 그리하여, 이러한 콩비지를 고부가가치 식품 소재로 활용하기 위해 건조 미세분말 비지를 제조하고, 이를 산채와의 혼합 가공을 통한 기능증진 효과를 연구하였다. 일반적으로 산채는 쓴맛 제거와 보관성 증진을 위해 데침공정을 주로 하나 산채를 물에 데칠 경우 산채의 유용성분이 대부분 손실된다. 눈개승마의 경우 유용성분이 17.25%, 그 중 폴리페놀 성분이 약 90% 손실된다. 본 연구에서는 기존의 데침공정 대신 찜공정을 이용하여 산채의 유용성분을 보존하고 콩비지를 첨가하여 산채 유용성분의 생이용성 증진 효과를 연구하였다. 눈개승마를 콩비지와 혼합하여 고온고압 가공함으로써 산채의 쓴맛을 줄이고 보관성을 높이는 동시에 눈개승마 폴리페놀의 단량체화를 통해 눈개승마 단독가공에 비하여, 단량체 비율이 약 100~200배 증가하는 것을 확인하였다. 본 연구를 통하여 콩비지의 고부가가치 소재로의 가능성을 확인하였을 뿐만 아니라 산채의 다이어트·건강기능식품 소재로의 산업화에도 기여할 수 있을 것으로 기대된다.

Effects of IgY Supplementation on Hanwoo Calves Fed withHigh Quality Roughage (양질 조사료 급여에 따른 한우 송아지의 IgY 처리효과)

  • This study was conducted to examine the effect of IgY supplementation on growth performance, blood metabolism, and disease occurrence in the weaned calves at 3 or 4 month of age. Average daily gain (ADG) and feed intakes were not affected by IgY supplementation, regardless of weaning months. White blood cell (WBC) count of the control group was higher in the final stage than in the initial stage (p<0.05), while that of the IgY supplemented group was lower in the final stage compared to the initial stage (p<0.05). regardless of treatments. Hematocrit (HCT) and red blood cell (RBC) counts were lower in the final stage than in the initial stage (p<0.05), but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) counts were higher in the final stage than in the initial stage, regardless of treatments. In the control group, concentrations of albumin and globulin were higher in the final stage compared to the initial stage in the weaned calves at 3 month of age (p<0.05), and concentration of blood urea nitrogen (BUN) was increased in the IgY supplemented group. In the control group, concentrations of aspartate aminotransferase (AST) and gamma (${\gamma}$)-glutamyl transferase (GGT) were higher in the final stage compared to the initial stage (p<0.05). In contrast, concentrations of aspartate aminotransferase (AST) and gamma(${\gamma}$)-glutamyl transferase (GGT) were lower in the final stage compared to the initial stage for the IgY supplemented group. A diarrhea calf was only found in the control group. Manure score was higher in the control group compared to IgY supplemented group (p<0.05). Thus, these results indicates that IgY supplementation had positive effects on some blood metabolites, fecal condition score and diarrhea without negative effect on growth performance of the weaned Hanwoo calves.

Estimation of the Characteristics of Delayed Failure and Long-term Strength of Granite by Brazilian Disc Test (압열인장시험을 이용한 화강암의 지연파괴특성 및 장기안정성 평가)

  • Long-term stability and delayed failure of granite were evaluated through the laboratory test based on Wilkins method and Brazilian disc test (BDT) which yields tensile strength, mode I fracture toughness and subcritical crack growth parameters. Then, the long-term strength of granite was estimated by using analytical models and long-term stability of compressed air-energy storage (CAES) pilot cavern pressurized up to 5 ~ 6 MPa was evaluated using numerical code, FRACOD with the determined subcritical crack growth parameters. The results of test and analyses showed that the subcritical crack growth index, n was determined as 29.39 and the inner pressure of 5 ~ 6 MPa had an insignificant effect on the long-term stability of pilot cavern. It was also found that the measurement and analysis of acoustic emission events can describe the accumulation of damage due to subcritical crack growth quantitatively. That is, AE monitoring can provide the current status of rock under loading if we make an identical installation condition in the field with that of the laboratory test.

  • Microalgae can offer an attractive way of generating renewable and sustainable biodiesel. Biodiesel derived from microagae can have lower impact on the environment and food supply than biodiesel produced from crops. But biodiesel derived from microagae have poor fuel properties at low temperature depending on their species. In this study, it was investigated that fuel characteristics of biodiesel derived from Tetraselmis sp. and cold weather performance of biodiesel blend (BD3, 3 vol.% biodiesel - 97 vol.% diesel). The startability and operability of the passenger car in BD3 was good at $-20^{\circ}C$.

PLZF+ Innate T Cells Support the TGF-β-Dependent Generation of Activated/Memory-Like Regulatory T Cells

  • PLZF-expressing invariant natural killer T cells and CD4 T cells are unique subsets of innate T cells. Both are selected via thymocyte-thymocyte interaction, and they contribute to the generation of activated/memory-like CD4 and CD8 T cells in the thymus via the production of IL-4. Here, we investigated whether $PLZF^+$ innate T cells also affect the development and function of $Foxp3^+$ regulatory CD4 T cells. Flow cytometry analysis of the thymus and spleen from both CIITA transgenic C57BL/6 and wild-type BALB/c mice, which have abundant $PLZF^+$ CD4 T cells and invariant natural killer T cells, respectively, revealed that $Foxp3^+$ T cells in these mice exhibited a $CD103^+$ activated/memorylike phenotype. The frequency of $CD103^+$ regulatory T cells was considerably decreased in $PLZF^+$ cell-deficient $CIITA^{Tg}Plzf^{lu/lu}$ and $BALB/c.CD1d^{-/-}$ mice as well as in an IL-4-deficient background, such as in $CIITA^{Tg}IL-4^{-/-}$ and $BALB/c.IL-4^{-/-}$ mice, indicating that the acquisition of an activated/ memory-like phenotype was dependent on $PLZF^+$ innate T cells and IL-4. Using fetal thymic organ culture, we further demonstrated that IL-4 in concert with TGF-${\beta}$ enhanced the acquisition of the activated/memory-like phenotype of regulatory T cells. In functional aspects, the activated/ memory-like phenotype of Treg cells was directly related to their suppressive function; regulatory T cells of $CIITA^{Tg}PIV^{-/-}$ mice more efficiently suppressed ovalbumin-induced allergic airway inflammation compared with their counterparts from wild-type mice. All of these findings suggest that $PLZF^+$ innate T cells also augmented the generation of activated/memory-like regulation via IL-4 production.

Isolation and Identification of an Antioxidant Enzyme Catalase Stimulatory Compound from Garnoderma lucidum

  • Antioxidant enzymes are scavenger reactive-oxygen intermediates and are involved in many cellular defense systems. We previously reported that a crude extract of Garnoderma lucidum, a medicinally potent mushroom, profoundly increased the catalase gene expression and enzyme activities in mouse livers (Park et al., J. Biochem. Mol. Biol. 34. 144-149, 2001). In this study, we elucidated the detailed mechanism whereby G. lucidum stimulates the catalase activity and expression. The major active fraction was isolated from G. lucidum and methyl linoleate was considered the most major component of the fraction. In order to determine whether methyl linoleate increases mRNA and protein synthesis of catalase, Northern and Western blot analyses were performed in vivo with methyl linoleate-treated mouse liver homogenate after feeding methyl linoleate to the mice. Northern and Western blot analyses of the crude liver homogenates in the mice that were administered methyl linoleate revealed that the expression catalase was significantly increased when compared to the untreated controls. In addition, the catalase protein levels and enzymatic activities increased in the mouse liver homogenates. These results suggest that methyl linoleate that is produced by G. lucidum stimulates the catalase expression at the transcription level.

  • Objectives : The objective of this study was to assess whether there were sex differences in stroke types, risk factors, warning signs, and stroke complications among patients with first-ever stroke. Methods : Six-hundred seventy six patients with first-ever stroke were recruited at the Department Cardiovascular and Neurologic Diseases (Stroke center) of KyungHee University Oriental Hospital, DongGuk University International Hospital, and Kyungwon University In-cheon Oriental Medical Hospital from September 2005 to June 2007. Patients were hospitalized within 28 days after the onset of stroke. We investigated their stroke types, ischemic stroke subtypes by TOAST classification, risk factors, warning signs, stroke complications, general characteristics such as age, sex, etc. Results : Overall, 347 patients were male and 279 female. Compared with males, female patients were significantly older (mean age 67.3${\pm}$1.1 versus 62.4${\pm}$1.6 years) (P=0.000). We did not find significant sex differences in stroke types or ischemic stroke subtypes by TOAST classification. History of hypertension was significantly more frequent in female than male patients (P=0.000). Among stroke complications, urinary tract infection was significantly more frequent in female than male patients (P=0.003). Among warning signs, blepharospasm was significantly more frequent in female than male patients (P=0.006). Conclusions : Knowledge of sex differences of stroke patients can help us gain better insights on the characteristics of stroke patients. We need further and larger scale research to acquire more concrete conclusions on this theme.

  • Translational research (TR) as high quality research can accelerate collaboration strongly between biotechnology-based researchers and clinical-research experts for overcoming diseases. TR facilitates basic science translated to clinical efficacy and effectiveness from bench (basic science) to bedside (clinical practice) for the enhancement of human health. Disease-oriented TR programs were defined as unilateral, bilateral and multilateral TR in this patent performance analysis. Patent performance was measured in a R&D project on Health and Medical Technology to enhance the productivity of R&D investment on disease-oriented TR in Health Technology (HT). Patent Map (PM) analysis and Bibliometrics were conducted to collect information for the assessment of research patents of TR programs. Futhermore, PIAS (Patent Information Analysis System) and Thinklear programs were applied for quantitative and qualitative analysis successfully. These indicate that multi-dimensional analysis of patent performance for disease-oriented TR could promote the connection of R&D-IP (Research and Development-Intellectural Property) and R&BD (Research and Business Development) supporting system significantly.

Effect of Serum and Serum Free Media on the Developmental Competence of OPU Derived Bovine IVP Embryo

  • Embryos produced with serum show the alterations in their ultrastructure, impaired compaction, abnormal blastulation, aberrant mRNA expression profiles and large calf syndrome with greater incidences of stillbirths and deaths after birth. The aim of the present study was to describe in vitro embryo production by analyzing embryo production, fetal production and pregnancy rate in free-serum medium. The OPU-IVP data used in this study from 2016. Approximately, sixteen cows (Hanwoo), which belonged to the Institute of Gyeongsang National University, were used. Two experimental group is used in this study. Serum groups were conducted in March to July and free-serum group was conducted in September to December. The recovered cumulus-oocyte complexes were morphologically classified to four grades based on the compaction of cumulus cells layers and homogeneity of the cytoplasm. The number of oocyte was significantly greater in serum groups than that in free-serum groups (29.61 ± 0.63 vs. 15.6 ± 0.62; p < 0.05). Between serum and free-serum groups indicate that average of 1st and 2nd grade oocytes were no difference (2.38 ± 1.67 vs. 2.38 ± 1.48; p > 0.05), but number of 3rd and 4th grade oocytes were greater in serum groups than that in free-serum groups (7.31 ± 7.64 vs. 5.60 ± 6.29; p < 0.05). Embryo cleaved competence was higher in rate in free-serum groups than that in serum groups (62.1% vs. 58.3; p < 0.05). However, blastocyst developmental rate was no difference between serum and free-serum groups (33.1% vs. 43.5%; p < 0.05). 986 recipients were used for embryo transfer. Pregnancy rate was indicated that between serum and free-serum group was no difference (54.6% vs. 56.3%; p < 0.05). In conclusion, we developed the free-serum system for production of in vitro bovine embryos in order to meet the developmental and qualitative requirements for large scale commercial use.
