  • Brucella (B.) abortus is a facultative intracellular pathogen that infects a wide variety of animal species and human. Brucellosis is the zoonosis and an extremely important disease around the world. Although the eight species can be differentiated by conventional phenotypic tests, these species display a high degree of DNA homology in DNA-DNA hybridization assay (>90%). Various methods have been established for genotyping of Brucella species, but most of analytical methods are lack reproducibility and limited capability to differentiate them. The attempt of this study was to evaluate multiple-locus VNTR analysis (MLVA) for use of epidemiological trace-back analysis in bovine brucellosis. Ninety-four B. abortus isolates from Gyeongbuk province during 2006~2010 were analyzed using 16 VNTR locus. High resolution automatic capillary electrophoresis system was used for more throughput, simpleer, faster, and better discriminable than conventional gel electrophoresis. As a result, 13 different genotypes were identified from 94 B. abortus isolates. MLVA could contribute to epidemiological trace-back analysis of bovine brucellosis.

Increasing of Macrophage Migration Inhibitory Factor Expression in Human Patients Infected with Virulent Brucella in Iraq

  • Brucellosis is a zoonotic disease caused by Brucella infections and humans usually contract this disease from close contact with infected animals or their products, usually via the ingestion of cheese or crude milk. Macrophage migration inhibitory factor (MIF) and Pro- and anti-inflammatory cytokines play an important role in susceptibility/resistance and the immunopathogenesis of Brucella infection. These cytokines are crucial factors in the initiation and progression of protective immunity against Brucella infection but the role of MIF has not been well studied in the human response to intracellular microbes. This study was designed to investigate the effect of MIF expression on Brucella susceptibility. A total of 85 positive rose Bengal tests and 24 samples from healthy individuals were collected for this study and subjected to polymerase chain reaction assays (PCR) of the bcsp31 diagnostic gene. MIF concentrations were evaluated using Enzyme-Linked immunosorbent assay (ELISA) and the results showed that 46 (54%) of the rose Bengal test samples were positive and 39 (46%) were negative for bcsp31 (p ≤ 0.05) and used as the gold standard for all of the comparisons in this study. The ELISA results indicate that the mean concentration of MIF was significantly higher in patients with positive rose Bengal tests when compared to the control groups and that its concentration increases with increasing age in both the patient and control groups (p ≤ 0.05).

효소면역법을 이용한 Brucella abortus 항체 검출에 관한 연구 (Studies on enzyme-linked immunosorbent assay(ELISA) for detection of antibody to Brucella abortus)

  • In order to establish a rapid, sensitive and specific diagnostic method for detection of antibody to Brucella abortus, a enzyme-linked immunosorbent assay(ELISA) was adapted. The diagnostic efficacy of the established ELISA was compared with that of the standard tube agglutination test for B abortus. 1. It was found that the optimal concentration of antigen for this ELISA was 5$\mu\textrm{g}$/ml, the optimal dilution of conjugate was 1 : 2000, and the optimal dilution of serum was 1 : 200, respectively. 2. Cut off value in this ELISA was 1,102 that was determined by mean absorbance(at 492nm) of tube agglutination test negative serum added with the triple value of the standared devation. 3. The relationship between the tube agglutination test and ELISA was showen high corresponding rate with sensitivity(96.3%) and specificity(98.1%). 4. The efficacy of the ELISA for detection of B abortus antibody was compared with tube agglutination test In brucellosis outbreak farm. The sensivity of ELSIA was higher than tube agglutination test.

Brucella canis 감염견에 대한 항균제 치료 (Two cases of antibiotic therapy in dog infected with Brucella canis)

  • For examination of antibiotic therapeutic efficacy in canine brucellosis, this examination was carried out two female bitches infected with Brucella canis in Gyeongbuk province, and used combicillin, baytril and doxycycline in susceptible antibiotics at B canis. During 18 month after the termination of antibiotic therapy, blood sample of the two bitches were examined for B canis antibody and antigen. The antibody of one bitch was disappeared at 5 month after antibiotic therapy and the other was continued at 18 month, but two bitches were not detected antigen by blood culture and PCR. Examination of blood chemical value (AST, ALT, urea, creatinine) of two bitches was increased in AST value during antibiotic therapy.

  • A bacteriological and serological investigation was conducted in a commercial breeding kennel in Taegu city in which canine abortion caused by Brucella canis occurred, and the family dogs in this area were also surveyed for Br canis infection during a period from March 1999 to May 2000. Of 195 dogs in the breeding kennel, 50(25.6%) were found to be bacteremic and 82(42.1%) were shown to be positive for canine brucellosis by both bacteriological and serological test. Of 357 family dogs examined, 17(4.8%) had an agglutinin titer of 1 : 160 or higher. Of these 17 dogs, 5(2.8%) were from indoor dogs and 12(6.7%) were from outdoor dogs. Only 2 mongrel dogs(1 female and 1 male) of 17 serologically positive dogs showed an agglutinin titer of 1 : 2560 or higher and Br canis was isolated from their blood.

국내 분리 Brucella canis의 항균제 감수성 (Antimicrobial Susceptiblity of Brucella canis Isolated from Korea)

  • Little is known to data about the in vitro activity of antimicrobial agents aganist Brucella cams (B cams) isolated from Korea. Our study aimed at determining the in vitro activities of 15 antimicrobial agents against 3 isolates and 52 isolates of B cams from dogs in 1994 and 2002, respectively. In minimal inhibitory concentration (MIC) study, minocycline and doxycycline showed the lowest MICs ( < 0.06-0.5 ug/ml). Gentamicin, streptomycin, ciprofloxacin, norfloxacin and rifampin showed MICs in the range of less than 1 ug/ml. Lincomycin and sulfisox azole showed the highest MICs ( > 32 ug/ml). Interestingly, MICs of macrolides (erythromycin, spiramycin, tylosin) against 52 isolates in 2002 were 16-64 times higher than that of 3 isolates in 1994. In minimal bactericidal concentration (MBC) study, gentamicin, streptomycin, ciprofloxacin and norfloxacin showed the lowest MBCs [0.12-1 ug/ml (1-2 times higher than MIC)], but minocycline and doxycycline showed the highest MBCs [8-32 ug/ml (128 times higher than MIC)]. Rifampin showed the MBCs in the range from 2 to 4 ug/ml (2-4 times higher than MIC).

Investigation of Brucella canis infection in public animal shelters and kennels in Incheon

  • This survey was performed to investigate the seroprevalence of antibodies to Brucella canis in dogs from public animal shelters and breeding kennels in Incheon. A total of 402 dogs selected randomly were tested serologically by using immunochromatographic antibody test kit. None of 289 dogs in public animal shelters were sero-positive and 10 (8.9%) of 113 dogs in breeding kennels were sero-positive. 5 (4.4%) strains of Brucella canis were isolated from 10 sero-positive dogs' blood. Antimicrobial susceptibility test was carried out by Disk diffusion method. They were susceptible to tetracyclines, quinolones, aminoglycosides and combination amoxicillin with clavulanic acid.

Brucellosis outbreak of Korean indigenous cattle at Yeongwol and Pyeongchang county in Korea

  • This study was attempted to investigate the properties of brucellosis in Korean indigenous cattle at the Yeongwol and Pyeongchang county. Brucella spp was differentiated and identified from cotyledons, amniotic fluids and supramammary lymph nodes which confirmed with clinical, serological, epidemiological evidences (69 cases) from January to June, 2004. Isolation frequency of this causative agent from supramammary lymph nodes, cotyledons and amniotic fluids from 38 pregnant Korean indigenous cattle were $39.1\%,\;87.5\%,\;and\;63.2\%$, respectively, and finally confirmed with Brucella abortus biotype 1 through biochemical and serological test. A Brucella specific DNA with 711bp band was detected by PCR assay using BCSP primer. The two cases were definite epidemiological evidences that infected Korean indigenous cattle acrossed the border to Yeongwol and Pyeongchang from near two provinces. Effective prevention programs are urgently needed for further spreading this epidemics.

A New ELISA Kit Based on Antigenic Epitopes for Diagnosing Brucella abortus

  • Brucellosis is one of the most important zoonotic diseases that lead to a great amount of economic losses. Prevention and diagnosis are both necessary to eradicate this disease. The identification and evaluation of different antigens of Brucella spp. play a key role in the progress of diagnostic programs. In this study, we designed, produced, and evaluated a 24-kDa polypeptide containing antigenic epitopes of VirB2, 3, and 9 of Brucella abortus for use with the ELISA kit. The produced polypeptide is appropriate for diagnosing brucellosis in bovines by a laboratory diagnostic kit, with 100% sensitivity and 97.5% specificity.

집단 개사육농장에서의 Canine Brucellosis 발생 및 PCR-RFLP를 이용한 분리주의 특성조사 (Occurrence of canine brucellosis in large kennels and characterization of Brucella canis isolates by PCR-RFLP)

  • A total of 260 dogs were randomly selected from two different treed kennels that brucellosis has occurred (group 1, 126 dogs), and random selected breed kennel (group 2, 134 dogs), and monitored for Brucella canis (B. canis) by 2-mercaptoethanol rapid slide agglutination test (2ME-RSAT) and bacterial culture method. For the differentiation, PCR-RFLP using omp-31, wbkA and per genes used for 52 of B canis strains (strain I) isolated in this study and 3 of B. canis strains (strain II) isolated in 1994 in Korea. 2ME-RSAT revealed that 63/126 dogs (50.0%) and 12/134 dogs (9.0%) were positive in group I and group II, respectively. Bacterial culture revealed that 47/126 dogs (37.3%) and 5/134 dogs (3.7%) were positive in group I and group II, respectively. As the results of PCR-RFLP, $\underline{omp}-31$ was amplified from all Brucella spp, except B. abortus. All B. canis isolates showed unique PCR-RFLP pattern following digestion with Bmel8I. However, all Brucella spp. showed the same PCR-RFLP pattern following digestion with SalI. PCR-RFLP analysis of wbkA revealed that all Brucella spp. showed the same pattern following digestion with HindIII. PCR-RFLP analysis of per revealed that B. abortus 544 and B. melitensis 63/9 showed the same pattern, but different from B. suis and B. canis following digestion with HindIII.
