Effects of 5-azacytidine, a DNA methylation inhibitor, on embryogenic callus formation and shoot regeneration from rice mature seeds (벼 성숙종자로부터 배상체 캘러스 형성 및 식물체 재분화에 DNA methylation 억제제인 5-azacytidine의 영향)

  • The modification of DNA and histone plays an important role for gene expression in plant development. The objective of this research is to observe the effects of methylation on the gene expression during dedifferentiation from rice mature seeds to callus and differentiation from callus to shoots. The embryogenic callus with ability to shoot regeneration was not induced on the N6A medium supplemented with 5-azacytidine and abnormal callus with brown color was formed. When the normal rice callus was placed on the regeneration MSRA medium supplemented with 5-azacytidine, the shoot regeneration was inhibited. The results showed that 5-azacytidine, DNA demethylating agent, had negative effects on normal embryogenic callus formation and shoot regeneration. This suggested that DNA methylation of some genes was required for normal cell dedifferentiation and differentiation in tissue culture. The microarray and $GeneFishig^{TM}$ DEG screening were used to observe the gene transcript profile in callus induction and regeneration on N6A (N6 medium + 5-azaC) and MSRA (MS regeneration medium + 5-azaC). Subsets of genes were up-regulated or down-regulated in response to 5-azaC treatments. The genes related with epigenetic regulation, electron transport, nucleic acid metabolism and response to stress were up and down regulated. The different expression of some genes (germin like protein etc.) during callus induction and shoot regeneration was confirmed using RT-PCR and northern blot analysis.

Mass Cultivation of A Hyperparasite, Ampelomyces quisqualis 94013 for Biological Control of Powdery Mildew (흰가루병 생물적 방제용 중복기생균 Ampelomyces quisqualis 94013의 대량배양)

  • An isolate of Ampelomyces quisqualis 94013(AQ94013) was selected as an effective agent for biological control against cucumber powdery mildew. In order to develop mass production technique, six cereal media made with barley, rice, mille and brown rice, sorghumand rice seed were tested. Among them, barley medium was the best for the growth and conidial production of AQ94013. Optimum temperature for the mass production of AQ94013 was $25^{\circ}C$ and followed by $20^{\circ}C$ and $30^{\circ}C$. Light radiation inhibited conidial production of AQ94013 since number of conidia formed on barely medium under continuous light or 12 hrs alternative light were much less than cultured in darkness. Tthe conidia produced on the medium at $30^{\circ}C$ maintained the parasitic ability to Sphaerotheca fusca for 30 days. A culture method of AQ94013 in barley liquid medium with adding barely power(40 g/l) in darkness for five days at $25^{\circ}C$using a 30 l-fermenter was very effective for mass production of conidia.

Successful Development of Cordyceps bassiana Stromata from Beauveria bassiana

  • A specimen of Beauveria bassiana was collected from Yang-yang of Gangwon province, Korea in October 2006. Conidial isolates were prepared from the specimen by the dilution method and inoculated in brown rice medium for fruiting body production. After nearly two months incubation for perithecial stromata developed from single isolates as well as from their combinations. They were determined as Cordyceps bassiana by observing the stromatal characters and their conidial structures. This is the first report of the development of C. bassiana from B. bassiana cultures.

  • For the purpose of application for biomass of Tricholoma matsutake, the optimum culture condition were determined. It was found that the optimum culture condition for spot culture of Tricholoma matsutake were pH 5.5 and 3% brown rice meal at $24^{\circ}C$ for 35 days with MMN medium. And the optimum culture condition of bioreactor for biomass were $18^{\circ}C$ and 60 days with PDMP broth.

  • Single ascospore isolates of Cordyceps bassiana were observed for their colony pigmentation on Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates and were inoculated in a brown rice medium for production of fruiting bodies. Colony pigmentation did not show any relationship with perithecial stromata formation. The isolates were also grown on opposite sides of SDAY agar plates and were observed for vegetative compatibility. Neither vegetative compatibility nor perithecial stromata could be found to be related to each other. It was concluded that fertile fruiting body production was independent of colony pigmentation and vegetative compatibility. Synnemata formation was found to be more common than perithecial stromata formation. This might be due to its highly conidiogenous anamorphic stage, i.e., Beauveria bassiana.

Epistatic Relationships among Genes Related to Endosperm Starch Synthesis in Rice

  • The mutants of sugary-2 (su-2), floury (flo), shrunken-1 (shr-1), and dull-1 (du-1) were crossed to waxy (wx) to produce $F_2$ seeds. Chi-square analysis on the segregating ratio of the $F_2$ seeds revealed that flo, su-2, and shr-1 were independently transmitted with wx, while wx was epistatic over du-1. The floury and sugary-2 were crossed to Hwasunchalbyeo, a waxy variety, and then the $F_4$ of floury-waxy and sugary-2-waxy seeds were developed, respectively. As the parents phenotypes of sugary-2 and floury, the grains of these two lines showed lower hardness and grain weight than normal grain of Hwacheongbyeo. For alkali digestive value (AVD), the sugary-2-waxy showed lower ADV than Hwacheongbyeo. For the gel consistency of grain flours, the floury was medium like Hwacheongbyeo, while those of the sugary-2, floury-waxy, and sugary-2-waxy were soft like Hwasunchalbyeo. The amylose contents in the grains of the sugary-2 and floury were decreased to ~15% whereas that of Hwacheongbyeo was 19.1%. All the lines showing waxy endosperm (Hwasunchalbyeo, floury-waxy, and sugary-2-waxy) showed less than 4% amylose contents. Interestingly, the free sugar content in the brown rice was increased to 9.27% in the sugary-2-waxy, showing transgressive segregation phenomenon where the free sugar contents in its parents, sugary-2 and Hwasunchalbyeo, were 5.98% and 3.98% respectively. Also, the floury-waxy showed transgressive segregation phenomenon, containing 6.15% of free sugar content in the grains.

  • Pholiota sp. were collected from the forest of Kangwon province (Mt. Samak, Mt. Odae and Kangwon National University Forest Experimentation) from the early of August to the end of October 1997. Among these fungi, Poliota adiposa was selected for culture experiment because it is suitable for edible mushroom. The Optimal temperature for the mycelial growth of Pholiota adiposa was in the range of $28^{\circ}C$ while that of fruit body formation was $15{\pm}1^{\circ}C$. Brown sugar as carbon source and soybean flour as nitrogen source were good for mycelial growth in commercial liquid culture. The fruit bodies of Pholiota adiposa were artifitially produced on plastic bottle including poplar sawdust and rice bran (4:1).

  • Hopum is a new japonica rice cultivar developed from the cross between Milyang165 and F1 crossing Milyang165 and Iksan438 at Department of Rice and Winter Cereal Crop, NICS, RDA, in 2006. This cultivar has a short grain shape and about 141 days growth duration from direct seeding to harvesting in the southern plain including Chungcheong province. This cultivar has short culm and spikelet number per panicle is similar to that of Nampyeongbyeo, while filled grain rate is lower than standard variety. This cultivar has medium size of brown rice and shows moderate resistance to leaf blast, to bacterial blight pathogens of $K_1$, $K_2$ and $K_3$ and stripe virus disease but susceptible to major virus diseases and insect pests. The milled kernel of Hopum is translucent with non-glutinous endosperm. Protein and amylose content of Hopum is about 6.5% and 18.7%, respectively. This cultivar has better palatability of cooked rice than Chucheongbyeo harvested in Gyeongki province. Its milling recovery (76.8%) and percentage of perfect-shaped milled rice (94.7%) were higher than Nampyeongbyeo. The milled rice yield of Hopum was 5.83 MT/ha (15% higher than Juan) under wet-direct seeding, 5.66 MT/ha (8% higher than Juan) under dry-direct seeding, and 6.00 MT/ha (8% higher than Nampyeong) under ordinary transplanting cultivation. "Hopum" would be adaptable for ordinary transplanting and direct seeding in the southern plain including Chungcheong province.

Proper Nitrogen Application Level for Improving the Rice Quality in Honam Plain Area (호남평야지에서 쌀 품질 향상을 위한 적정 질소시비량)

  • This study was carried out to elucidate the proper nitrogen application level considering rice quality in Honam plain area from 2002 to 2004 at the paddy field of Honam Agricultural Research Institute. The rice cultivars tested were Samcheonbyeo (Early maturing one), Hwaseongbyeo (Medium maturing one) and Nampyeongbyeo (Mid-late maturing one). The results are summarized as follows: The higher amount of nitrogen application resulted in greater number of panicle and grain in per unit area, but the rate of ripening and the 1,000-grain weight of brown rice decreased. The protein contents showed a tendency of increase with higher nitrogen level. The head rice yield increased by the higher nitrogen application up to 7 kg/10a for Samcheonbyeo and Hwaseongbyeo, and 9 kg/10a for Nampyeongbyeo respectively. The appropriate nitrogen application amounts, with respect to the yield of head rice, the rate of ripening and the quality of rice were found to be in range of 7 to 9 kg/10a.

Variation of β-carotene Concentration in Soybean Seed and Sprout

  • In this study, ${\beta}$-carotene concentrations was determined in soybean cultivar according to seed size, usage, seed coat color and cotyledon color as well as the process of seed germination. The total average concentration of ${\beta}$-carotene was $6.6{\mu}g/g$ in soybean seed, $33.3{\mu}g/g$ in soybean sprout. According to seed size, the total ${\beta}$-carotene concentration of soybean was $6.9{\mu}g/g$ in large soybean seed, $6.7{\mu}g/g$ in medium soybean seed, and $6.31{\mu}g/g$ in small soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $21.4{\mu}g/g$ in large soybean sprout, $30.5{\mu}g/g$ in medium soybean sprout, and $43.5{\mu}g/g$ in small soybean sprout. According to the utilization of seed, the total ${\beta}$-carotene concentration of soybean seed was $7.2{\mu}g/g$ in cooked with rice soybean seed, $6.1{\mu}g/g$ in paste and curd soybean seed, and $6.3{\mu}g/g$ in sprout soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $25.9{\mu}g/g$ in cooked with rice soybean sprout, $32.4{\mu}g/g$ in paste and curd soybean sprout, and $41.9{\mu}g/g$ in sprout soybean sprout. When comparison with seed coat color, the total ${\beta}$-carotene concentration of soybean with brown seed coat ($8.8{\mu}g/g$) was slightly higher than those of soybean with yellow ($6.1{\mu}g/g$). In soybean sprout, the total ${\beta}$-carotene concentration was $21.8{\mu}g/g$ in black seed coat sprout, $38.7{\mu}g/g$ in brown seed coat sprout, $34.1{\mu}g/g$ in green seed coat sprout, $39.5{\mu}g/g$ in yellow seed coat sprout, and $30.5{\mu}g/g$ in mottle seed coat sprout. The results of this study suggested the functional characteristics of soybean through quantitative analysis of ${\beta}$-carotene.
