사료내 비태인과 에너지 수준이 산란계의 간지방과 콜레스테롤에 미치는 영향 (Effect of Dietary Betaine and Energy Levels on Liver Fats and Cholesterol in Laying Hens)

  • 비태인의 급여가 산란계의 지질 대사에 미치는 영향을 구명하기 위하여 혈액의 성상, 복강지방과 간 지방 함량, 난황의 콜레스테롤 함량, 조직내 중성 지방 함량 그리고 간 조직의 HMG-CoA reductase activity를 조사하기 위하여 2차례의 사양시험을 실시하였다. 시험 1에서는 18주령 ISA-Brown 산란계를 이용하여 처리구당 18수씩 총 72수를 공시하여 4주간 사양시험을 실시하였다. 처리구는 CP $16\%$, ME 2,800kcal/kg의 기초사료에 비태인 0, 300, 600, 1,200ppm 수준으로 첨가$\cdot$급여하였다. 시험 2에서는 70주령 ISA-Brown 산란계를 공시하여 처리구당 18수씩 총 72수를 공시하여 4주간 사양시험을 실시하였다 처리구는 사료내 에너지 2수준(2,800, 2,900kcal/kg)과 비태인 2수준(0, 600ppm)으로 $2\times2$ 요인분석 시험을 실시하였다. 본 시험의 결과, 난황의 콜레스테롤 함량에서는 비태인 600, 1,200ppm 첨가구가 대조구와 비태인 300ppm 첨가구에 비하여 유의하게 감소하였다(P<0.05). 혈중 total cholesterol, LDL-cholesterol 그리고 중성지방 함량은 비태인의 첨가 수준에 따라서 그 수치도 증가하는 경향을 보였으나 유의적인 차이는 없었다(시험 1). 에너지 수준(2,800kcal/kg, 2,900kcal/kg)에 따른 비태인의 첨가 효과를 조사한 결과, 혈중 total cholesterol, LDL 및 중성 지방 함량은 에너지 수준에서는 차이가 없었으나 비태인 600ppm 첨가구는 무첨가구에 비하여 현저하게 증가하였다(P<0.05). 그러나 난황 콜레스테롤 함량에서는 사료내 에너지와 비태인 수준에 영향을 받지 않았다(시험 2).총2회의 시험에서 생체중에 대한 복강지방의 비율$(\%)$은 일관적인 결과를 나타내지 못하였으며, 간의 총지질 함량은 비태인의 급여 수준이 증가할수록 감소하여 비태인 1,200ppm 급여구에서는 대조구와 비교하여 유의적인 차이를 나타내었다. 또한 간의 중성 지방 함량에 있어서도 비태인 급여구에서 유의하게 감소하였다(P<0.05). 한편,간 조직의 HMG-CoA환원 효소의 활성에 있어서는 처리구간 유의적인 차이가 없었다.

산란계 사료내 Cu-Methionine chelate 첨가가 생산성과 난황 cholesterol에 미치는 영향

  • Seven hundred twenty 56 wks old ISA brown layers were assigned to five dietary treatments for 12 weeks. Each treatment was consisted of 6 replications with 24 birds. Control diet was formulated to have 15% CP, 2800kcal/kg ME, 3.8% Ca and Cu-methionine chelate(Cu-Met) were added to control diet at the level of 25, 50, 75, 100ppm in Cu. Supplementation of 25, 75, 100ppm Cu in from of Cu-Met s increased egg productions by 2.11, 3.84, 3.66%, compare with control. Egg weight also increased by supplementation of Cu at 50, 75, 100ppm in Cu-Met. Gizzard erosion tened to increase by supplementary Cu-Met but were not significantly different. Supplementation of Cu at the level of 75, 100ppm Cu-Met treatments decreased crude fat level in liver. It concludes that supplementation of Cu at level in the form of 75 or 100ppm methionine chelate improves egg production with heavier egg weight and decreases crude fat content in the liver.

Evaluation of the inhibition of the differentiation of pre-adipocytes into matures adipocytes

  • Up until today, the key to contouring has been resumed in these two alternatives, either limiting the adipocyte storing capacity by modulating lipogenesis, or by stimulating lipolysis to eliminate adipocyte lipid content. Another interesting way could be the regulation of adipocyte differentiation. In this work, we have evaluated the effect of a brown algal extract of Sphacelaria scoparia (SSE) on the differentiation of pre-adipocytes into adipocytes. A pre-adipocyte line (3T3-L 1) was used. The differentiation was evaluated by the measure of produced lipids thanks to red oil coloration and spectrophotometry, and also by the expression of adipocyte differentiation markers: enzymes such as fatty acid synthase (FAS) and stearoyl CoA desaturase (SCD), or membrane proteins such as glucose transporters (GLUT -4) and fatty acid transporters (FAT) expressed on the surface of human adipocytes. These genes are under control of two transcription factors: CAAT-enhancer binding protein (c/EBP alpha) and sterol response element binding protein (SREBP1). All these markers were analysed at different stages of differentiation by RT -PCR. Sphacelaria extract (SSE) inhibits pre-adipocytes differentiating into adipocytes following a dose-dependant relation, using a kinetics similar to retinoic acid. It decreases the expression of mRNA specific to FAS, FAT, GLUT -4, SCD1, c/EBP alpha and SREBP1. Moreover, SSE regulated on collagen 1 and collagen 4 expression. A stimulation of collagen 1 was also measured in human skin fibroblasts. Thus, SSE performs as a genuine differentiation inhibitor and not only as a lipogenesis inhibitor, and could be used in slimming products.

Hypotriglyceridemic effects of brown seaweed consumption via regulation of bile acid excretion and hepatic lipogenesis in high fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: The present study aimed to further investigate the potential health beneficial effects of long-term seaweed supplementation on lipid metabolism and hepatic functions in DIO mice. MATERIALS/METHODS: Four brown seaweeds (Undaria pinnatifida [UP], Laminaria japonica [LJ], Sargassum fulvellum [SF], or Hizikia fusiforme [HF]) were added to a high fat diet (HFD) at a 5% ratio and supplemented to C57BL/6N mice for 16 weeks. Triglycerides (TGs) and total cholesterol (TC) in the liver, feces, and plasma were measured. Fecal bile acid (BA) levels in feces were monitored. Hepatic insulin signaling- and lipogenesis-related proteins were evaluated by Western blot analysis. RESULTS: Fasting blood glucose levels were significantly reduced in the LJ, SF, and HF groups compared to the HFD group by the end of 16-week feeding period. Plasma TG levels and hepatic lipid accumulation were significantly reduced in all 4 seaweed supplemented groups, whereas plasma TC levels were only suppressed in the UP and HF groups compared to the HFD group. Fecal BA levels were significantly elevated by UP, LJ, and SF supplementation compared to HFD feeding only. Lastly, regarding hepatic insulin signaling-related proteins, phosphorylation of 5'-AMP-activated protein kinase was significantly up-regulated by all 4 types of seaweed, whereas phosphorylation of protein kinase B was up-regulated only in the SF and HF groups. Lipogenesis-related proteins in the liver were effectively down-regulated by HF supplementation in DIO mice. CONCLUSIONS: Brown seaweed consumption showed hypotriglyceridemic effects in the prolonged DIO mouse model. Specifically, combinatory regulation of BA excretion and lipogenesis-related proteins in the liver by seaweed supplementation contributed to the reduction of plasma and hepatic TG levels, which inhibited hyperglycemia in DIO mice. Thus, the discrepant and species-specific functions of brown seaweeds provide novel insights for the selection of future targets for therapeutic agents.

품종 및 도정도별 백미와 미강의 특성 및 페놀산 함량 (Various Properties and Phenolic Acid Contents of Rices and Rice Brans with Different Milling Fractions)

  • 본 연구에서는 오대, 남평, 추청, 일미 4품종으로부터 각각 현미와 7분도, 10분도 및 12분도의 백미와 미강을 제조하고 품종과 도정도에 따른 특성을 비교하였으며 총 폴리페놀과 페놀산의 함량을 분석하였다. 현미의 성분 중 지방과 회분 함량이 도정도가 증가할수록 크게 감소하였으며 단백질 함량은 변화가 크지 않았다. 미강의 지방과 회분 함량은 품종에 따른 차이가 매우 큰 것으로 나타났으며 도정도에 따른 차이는 적었다. 도정도가 증가할수록 쌀가루의 호화개시온도가 낮아지고 피크점도(P)와 $95^{\circ}C$ 15분간 유지 후 점도(H)가 증가하였으나 반면 냉각점도, 최종점도, setback 등은 도정도에 따라 일정한 경향을 보이기보다는 품종에 따른 차이가 더 큰 것으로 나타났다. 현미에 함유된 총 폴리페놀의 함량은 93.9-88.8 mg catechin eq/100 g이고 도정한 백미 중에는 30.3-71.9 mg catechin eq/100g 범위였으며, 미강에는 310.0-541.6 mg catechin eq/100g이었다. Phenolic acid는 ferulic acid, p-coumaric acid가 주요 성분이었으며 현미의 총 phenolic acid 함량은 65.9-27.9 mg%였으나 도정도가 증가할수록 감소되고 ferulic acid 조성비가 증가하는 것으로 나타났다. 품종 중에서는 추청과 일미에서 도정으로 인한 phenolic acid 함량 감소가 컸다. 미강의 경우 ferulic acid는 157.8-240.2 mg%, p-coumaric acid는 31.8-90.4 mg% 범위로 나타났고 현미와 백미와는 달리 sinapinic acid, benzoic acid, m-hydroxybenzoic acid의 함량도 높았다.

Genetic and Phenotypic Evaluation of Milk and Fat Production Traits and Their Interrelationship in (Zebu×European) Crossbred Cattle Using Parent Group Mixed Model

  • Data pertained to 335 crossbred cows comprising of 1/2 Friesian (F) + 1/2 Hariana (H), 1/2 F + 1/4 Jersey (J) + 1/4 H, 1/2 F + 1/4 Brown Swiss (BS) + 1/4 H, 1/2 F + 1/4 Red Dane (R) + 1/4 H, FR (I) and FRH (I) genetic groups extending over a period of 21 years (1970-1990) maintained at Animal Farm of CCS HAU, Hisar. The averages for first lactation milk yield was $2,486.24{\pm}80.26kg$ and peak yield of first three lactation were $11.35{\pm}0.72kg$, $13.97{\pm}0.60kg$ and $16.02{\pm}0.42kg$, respectively. The lifetime milk production was observed as $11,305.16{\pm}1,004.52kg$ in crossbred cattle. The average first lactation fat yield was observed as $102.06{\pm}0.01kg$ and peak fat yield of first three lactation were $0.458{\pm}0.01$, $0.490{\pm}0.01$ and $0.500{\pm}0.02kg$, respectively. The lifetime fat production was estimated as $502.31{\pm}45.90kg$. LTMP and LTFP had reasonably good additive genetic variance which could be exploited either through mass selection/combined with family or pedigree selection. FLMY, peak yields and LTMP had significant positive phenotypic correlation with FLFY and LTFP and the correlation at the genetic level were also higher and positive for these traits. Finally, peak week milk yield of first lactation (PMY1) was the earliest available trait having desirable and significant correlation at phenotypic and positive at genetic level with FLFY, PFY1 and PFY2, PFY3 and LTFP and selection for this trait will help in early evaluation of sires and dams and will increase genetic advancement per unit of time.

키토산, 히비스커스 추출물 및 L-카르니틴 함유 식이가 흰쥐의 체중과 지질대사에 미치는 영향 (Effect of Feeding Chitosan, Hibiscus Extract and L-Carnitine Mixture on Body Weight and Lipid Metabolism in Rats)

  • This study was peformed to investigate effect of feeding experimental mixture containing chitosan, hibiscus extract and L-carnitine on body weight and lipid metabolism in rats. Forty-eight male rats(Charles River CD) of eight weeks old and weighing $336.5{\pm}2.3g$ were raised for five weeks with high fat diet(40% fat as calorie) to induce obesity. After induction of obesity, rats weighing $560.4{\pm}5.6g$ were blocked into four groups according to body weight and raised for eight weeks with diet containing either 0.09%(+1D group), 0.9%(+10D group) or 4.5%(+50D group) of experimental mixture. Aspartate aminotransferase(AST) and alanine aminotransferase(ALT), total protein and albumin were normal levels in plasma. Body weight gain and epididymal fat pad weight were lower in experimental mixture groups than control group However, weights of perirenal fat pad and brown adipose tissue were not significantly different among all groups. There was no significant difference in plasma and hepatic lipid levels among all groups. Liver citrate lyase and camitine acyltransferase activities were not significantly different among all groups, however, citrate lyase activity was tended to be decreased with increasing experimental mixture level in diet. Fecal total lipid and total cholesterol excretions were highest in +50D group, and triglyceride excretion was highest in +1D group. in conclusion, intake of experimental mixture containing chitosan, hibiscus extract and L-camitine was effective in reducing body weight and body fat, and its inhibitory effects might lead to obesity improvement.

개에서 겸상인대에서 유래된 분화지방육종의 영상의학증례 (Medical Imaging of Well-differentiated Liposarcoma Arising from the Falciform Fat in a Dog)

  • 7세령의 중성화수컷 코커스파니엘견이 심한 복부팽만을 주증으로 경상대학교 부속동물병원에 내원하였다. 복부방사선상에서 매우 크고 균일한 지방밀도를 보이는 종괴가 확인되었다. 복부초음파검사결과, 종괴는 불규칙한 고에코성의 피막으로 둘러싸여 있었으며, 내부는 세포성의 저에코 액체로 채워져 있었다. 컴퓨터 단층촬영결과, 내부는 지방감쇠를 보이며 주변부는 연부조직감쇠를 보이는 불균질한 종괴로 확인되었으며, 종괴의 기원은 수술적 적출시 겸상 인대에서 유래됨을 확인하였다. 절제된 종괴는 견고하고 적갈색의 표면을 나타내었다. 조직병리학적 검사결과, 분화 지방육종으로 진단되었다. 수술 1개월경과후 재검에서 환자는 특이적인 임상증상을 보이지 않았다.

고지방식이 흰쥐에서 큰실말의 지질대사에 미치는 효과 (Effects of Cladosiphon okamuranus on Lipid Metabolism in High-fat-diet Rats)

  • 본 연구에서는 큰실말의 지질대사 효과를 연구하기 위하여 체중 158±10 g의 SD계 암컷 흰쥐 25마리를 대상으로 정상군, 고지방식이 대조군, 큰실말 섭취군, 큰실말 불용성 식이섬유 섭취군, 큰실말 수용성 식이섬유 섭취군으로 분류하여 52일간 사육하였다. 체중은 큰실말 섭취군이 고지방식이군에 비하여 유의적으로 감소하여 정상군에 가까운 수치를 나타냈다. 혈액에서의 HDL 수치는 대조군보다 큰실말 및 식이섬유를 섭취한 군의 수치가 더 높게 나타났고, TC, TG, LDL, GOT, GPT의 수치는 대조군보다 큰실말 및 식이섬유를 섭취한 군의 수치가 더 낮게 나타났다. 이와 같은 연구를 통해 큰실말 및 식이섬유가 혈중 콜레스테롤 농도를 개선시켜 지질 대사에 미치는 순기능적 효과를 보여주는 것으로 사료된다.

Black soldier fly (Hermetia illucens) larvae oil as an alternative fat ingredient to soybean oil in laying hen diets

  • Objective: The objective of this study was to determine whether dietary black soldier fly (Hermetia illucens, HI) larvae oil (HILO) could serve as an alternative fat source to soybean oil (SBO) in laying hen diets. Methods: We randomly assigned 25-week-old Hy-line Brown laying hens (n = 144) to receive (n = 6 hens/group; eight replicates) a control or an experimental diet in which SBO was replaced with 50% (50HILO) or 100% HILO (100HILO). Results: Dietary HILO did not negatively affect body weight or productive performance during the study. The eggs also had similar quality parameters, proximate composition, and cholesterol levels. However, the yolk color index was significantly higher (p<0.01) in the 100HILO than in the other groups. Dietary HILO significantly altered the composition of fatty acids (FAs) in abdominal fat and eggs. Total saturated fatty acids (SFAs) and total polyunsaturated FAs (PUFAs) were significantly increased and decreased in the 50HILO and 100HILO groups, respectively, compared with those in the control group (p<0.001 and p<0.0001, respectively). Specifically, the medium-chain FAs lauric and myristic acids were remarkably increased in the abdominal fat of laying hens fed HILO (p<0.0001), whereas only myristic acid increased in eggs (p<0.0001). Undesirable heavy metal (aluminum, fluorine, arsenic, lead, mercury, and cadmium) concentrations were below permissible limits in eggs. Conclusion: We considered that HILO could be an alternative dietary fat to SBO for laying hens with maintained productive performance and good egg quality.
