Comparison of Methods for Detection of Escherichia coli O157:H7 in Ground Beef and Radish Sprouts

  • Escherichia coli O157:H7 is a food-borne pathogen that causes bloody diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome (HUS). We compared three selective media and evaluated the performance of immunomagnetic separation (IMS) for the detection of low levels of E. coli O157:H7 in ground beef and radish sprouts with different levels of background flora. Bulk food samples (500 g for each trial) were artificially inoculated with nalidixic acid-resistant E. coli O157:H7 at the lowest dose that would generate 20 partial-positive samples of 25 g each. All samples were homogenized in mTSB (225 mL) and incubated overnight at $37^{\circ}C$. IMS was performed using the enriched mTSB samples (1 mL) along with conventional spreads plated onto three different selective media: Sorbitol MacConkey agar (SMAC), Sorbitol MacConkey agar with cefixime and tellulite (CT-SMAC), and Sorbitol MacConkey agar with nalidixic acid (NAL-SMAC) as the gold standard. Two suspicious colonies from each medium were selected and confirmed usinga serological test after transfer to tryptic soy broth with yeast extract (TSAYE). CT-SMAC was better than SMAC for detecting E. coli O157:H7 in all food types. Although there was no statistical difference in the number of positive samples when using IMS vs. non-IMS techniques, more positive samples were detected when IMS was used in both ground beef and radish sprouts. It appears that the improvement was more significant in radish sprouts, which had a higher level of background flora than ground beef. The results also suggest that the combination of CT-SMAC and IMS is sufficient to recover low levels of E. coli O157:H7 in high background flora food samples.

Effect of Wheat Flour Brew with Bifidobacterium bifidum on Rheological Properties of Wheat Flour Dough (Bifidobacterium bifidum을 이용한 밀가루 brew가 반죽의 이화학적 성질에 미치는 영향)

  • In order to economically utilize flour brew with Bifidobacterium bifidum as a bread improver, the effect of flour brew on the rheological properties of dough, growth curve and acid production, and symbiosis with yeast were investigated. Growth of bifidobacteria was not increased more than initial seed volume but was consistent during 24 hours of incubation. pH was decreased and T.T.A was increased up to 12 hours of incubation. Symbiosis between bifidobacteria and yeast was little. Bifidobacteria produced more lactic acid than acetic acid in flour brew and the opposite in skim milk broth. This result was inferred from Lactobacillus sp. inherent in flour. On rheological properties of dough, farinograms of flour showed progressively decreasing baking absorption, mixing time and stability as the amount of flour brew increased. The validation of extensograms showed that R/E ratio linearly increased with increment of flour brew, and nearly doubled in all treatments comparing to that of control, which suggest the reduction of actual fermentation time. On visco/amylograms, malt index increased with addition of flour brew, accordingly showing the decrease in viscosity. Break down and set back value decreased with increment of flour brew, suggesting that staling rate of bread can be delayed.

Isolation and Antimicrobial Action of Growth Inhibitory Substance on Food-borne Microorganisms from Dryopteris crassirhizoma Nakai (관중(Dryopteris crassirhizoma Nakai)의 식중독 미생물 증식 억제 물질의 분리 및 항균작용)

  • The ethanol extract of Dryopteris crassirhizoma Nakai showed strong growth inhibition against 5 strains of Listeria monocytogenes at the concentrations of $100{\sim}500$ ppm and the minimum inhibitory concentration of n-hexane fraction was under 50 ppm. The D8-2-5 fraction isolated from n-hexane fraction of Dryopteris crassirhizoma Nakai showed a strong bactericidal activity on 5 strains of L. monocytogenes at 20 ppm level in tryptic soy broth medium. At the level, the viable count was reduced $4{\sim}6$ log cycle compared to initial cell number. Observation by the measurement of adenosine triphosphate (ATP) contents and transmission electron microscope showed that disruptions of the cell wall and elution of intracellular ATP are assumed to be due to the bactericidal activity. In addition, the n-hexane fraction of Dryopteris crassirhizoma Nakai showed the strong growth inhibitions at 50 ppm on Vibrio parahaemolyticus and Bacillus cereus, and at 25 ppm on Staphylococcus aureus.

Studies on the Acid Tolerance of Acetobacter sp. Isolated from Persimmon Vinegar (감식초로부터 분리한 Acetobacter sp.의 내산성에 관한 연구)

  • The microbial properties and acid tolerance of the three kinds of Acetobacter sp. isolated from persimmon vinegar were investigated. Acid tolerance was also evaluated. Acetobacter sp. were gram negative, short rod, nonspore forming and motile. They reacted positively catalase, methyl red, oxidation fermentation, Voges-Proskauer and nitrate reduction tests and negative to hydrogen sulfide test and ONPG. Acetobacter sp. showed normal growth curve in Carr broth and there was no significant difference between isolates and (standard on) typical strains such as Acetobacter aceti (KCTC1010), Acetobacter liquefaciens (KCTC2804), Acetobacter diazotrophicus (KCTC 2859). Optimum temperature and initial ethanol concentration in incubation were $30^{\circ}C$ and 6%, respectively. Growth and acid production of Acetobacter sp. were suppressed by the concentration of above 4% acetic acid. The amount of $Mg^{++}$ release from Acetobacter sp. cells in medium was increased by acetic acid, and almost in the concentration of 6% acetic acid. Glycolysis by Acetobacter sp. had optimal pH about 6.0 to 7.0 and more stable in acidic condition than in alkalic. The $H^+-ATPase$ of Acetobacter sp. S-1 and S-3 showed a maximal activity between pH values of approximately 5.5 to 7.5 and 6.0 to 7.5, respectively.

Inhibitory Effect of Major Food Components on the Activity of Antimicrobial Active Substance from n-Hexane Fraction of Mallotus japonicus Muell on Listeria monocytogenes (식품의 주요 성분이 예덕나무 헥산추출물의 Listeria monocytogenes에 대한 항균활성에 미치는 영향)

  • Inhibitory action of major food components on antimicrobial active substance (linolenic acid) was tested against Listeria monocytogenes ATCC 19111 for 72 hr at $30^{\circ}C$. Linolenic acid (50, 1000 ppm) and n-hexane fraction of Mallotus japonicus Muell (50, 1000 ppm) were added to the broth culture medium containing casein(1%, 3%), soybean oil(1%, 3%) and soluble starch (1%, 3%), respectively. Linolenic acid 1000 ppm and n-hexane fraction 1000 ppm exhibited strongly antimicrobial actions on L. monocytogenes which were not detected viable cell after 24 hr. But casein (3%) and soybean oil (3%) strongly diminished the antimicrobial action of 1000 ppm of n-hexane fraction. Soluble starch (1%, 3%) did not affect the antimicrobial action of 1000 ppm of linolenic acid and n-hexane fraction.

  • The purpose of this study was availability for the classical test method. The test were called CLSI(Clinical and Laboratory Standards Institute) that was disk diffusion method, the newly designed E-test(made use disk diffusion method) can estimate the MIC and modified broth microdilution method that was standardized. Those tests were observed by MicroScan MicroSTREP plus panel. Target strains were 53 strains of S.pneumoniae and 51 strains of ${\alpha}$-hemolytic streptococci which were separated from the inpatient in university hospital for 6 months from February to August, 2009. The 9 antimicrobial agent of target evaluation were cefotaxime, chloramphenicol, clindamycin, erythromycin, levofloxacin, penicillin, tetracycline, trimethoprim/sulfamethoxazole, and vancomycin. researched comparative analysis both S.pneumoniae and ${\alpha}$-hemolytic streptococci. The result of the high concordance rates in ${\alpha}$-hemolytic streptococci was recognized formally in clinical microbiology laboratory.

Identification of an Actinomycetes Strain, MSA-1, Originated from Sponge, Halichondria okadai, and its Antimicrobial Component (검정해면으로부터 항균성을 가진 방선균의 분리 동정 및 항균물질의 구조)

  • An Actinomycetes strain, MSA-1, containing antimicrobial component was isolated from the black sponge, Halichondzia okadai, and was identified to a genus level by morphological and chemotaxonornic methods. The gray colored spores were oval type with smooth surface and formed flexibilis spore chains. The cell wall of this strain was type I containing D-aminopimellic acid (D-DAP) and no specific sugar was detected. Phospholipid of the cell membrane was PII type including phophoethanolamine and the major fatty acids of total lipid were branched anteiso-15 : 0, iso-16 : 0, 16 : 0 and iso-17 ; 0. From these results and other characteristics described in the Bergey's Manual, this strain was identificated as a Streptomyces sp. Meanwhile, 10mg of pale yellow colored antimicreobial component was isolated by HPLC method from the cultured Streptomyces sp. (70g of cryophillized mycellis). By crystallographyc analysis, HIRESMS and NMR assignment, the antimicrobial component produced from the strain MSA-1 was elucidated as the staurosporine (indolo[2,3-a]carbazole alkaloid).

  • This work was conducted to study the use of L. acidophilus, which exists in humun intestine for the fermentation of Chinese cabbage kimchi. The changes in vitamins, the number of microflora and sensory quality were observed during fermentation after the microflora which was not related to kimchi fermentation was eliminated by treatment with ozone water or ozone gas. The growth rate of L. acidophilus in the cabbage juice was higher than that in MRS broth. The growth of L. acidophilus was slightly promoted by adding 1~2% hot pepper powder while that was inhibited by ginger and garlic. Therefore, it was shown that the regulation of fermentation was possible by addition of spices. The result of treating spice with ozone gas and ozone water 6mg/L/sec for 1 hour was that the survival ratio of total microflora was 6~20%. When L. acidophilus was added to materials after ozone treatment, the fermentation rate was improved and the polysaccharides in the cell wall were used when the usable free sugar was all consumed. The contents of vitamin B$_1$ and C in the ozone treated kimchi was higher than in the control.

Effect of Trehalose and Sugar Alcohol on the Viability of Lactic Acid Bacteria and Quality Characteristics during Frozen Storage of Yoghurt (Trehalose와 당류가 냉동요구르트의 저장 중 유산균 생존율과 품질특성에 미치는 영향)

  • The objective of the current study was to determine the cryoprotective effects of trehaolse on lactic acid bacteria in the frozen yoghurt during long-term frozen storage conditions. The frozen yoghurts were prepared using starter culture containing Lactobacillus bulgaricus and Streptococcus thermophilus, as well as trehalose and sorbitol as a cryoprotectant. The viable cell numbers of lactic acid bacteria in frozen yoghurt did not significantly decreased during six weeks frozen storage conditions. The MRS broth, which contains either trehalose or sorbitol, cultured with L. bulgaricus and/or S. thermophilus, and then the cultured medium was kept in the frozen condition for six weeks. The results indicated that lactic acid bacteria viability significantly increased with trehalose addition (2 and 5%) in the media compared to those of control and sorbitol supplement groups. The lactic acid bacteria viability in the yoghurts was examined on the effects of repeated freeze and thaw events. The freeze-thaw resistance of lactic acid bacteria significantly increased with trehalose supplement in the yoghurt. The major volatile aroma compounds (acetaldehyde, acetone, ethanol, diacetyl, and acetoin) in yoghurt were separated and indentified by headspace GC-FID analysis. Distinct flavor components and their ratios are known as important quality factors for yoghurt notes. Trehalose addition to the yoghurt was not influenced these factors during lactic acid fermentation. The results in this study demonstrated that trehalose potentially can be applicable as an effective cryoprotectant for lactic acid bacteria in the frozen yoghurt products.

Microbiological Characteristics and Physiological Functionalities of Unrecorded Wild Yeast Strains in the Soils of Hajodae and Gyungpodae Beaches in Korea (동해안 하조대와 경포대 해수욕장 주변 토양으로부터 분리한 국내 미기록 야생효모들의 균학적 특성과 생리활성)

  • The goal of this study was to isolate and characterize the wild yeast strains in the soils of Hajodae and Gyungpodae Beaches, Gyungpo Lake in Kangwon-do, Korea and evaluate their anti-demential and tyrosinase inhibitory activities. Among the 27 yeast strains isolated from 30 soil samples from Hajodae Beach, Cyberlindnera culbertsonii HJ31-1, Dothichiza pithyophila HJ22-4, Metschnikowia bicuspidata HJ16-1, and Sakaguchia cladiensis HJ14-1 were recorded for the first time in Korea. Additionally, six of the 55 wild yeast strains isolated from 45 soil samples from Gyungpodae Beach and Gyungpo Lake had not previously been detected in Korea, such as Aureobasidium melanogenum JR3-1. The microbiological characteristics and anti-demential and tyrosinase inhibitory activities of these previously unrecorded yeasts were investigated. Almost all of them were oval-shaped and had ascospores except for Dothichiza pithyophila HJ22-4, Diutina siamensis JR37-5, and Canadida gelsemii DC 35-1. Furthermore, they were all sugar-tolerant and able to grow in 20% glucose-containing YPD broth, except for Cyberlindnera culbertsonii HJ31-1. Diutina siamensis JR37-5 had the highest tyrosinase inhibitory activity (30.9%).
