Effect of Xylan on Production of Xylanolytic Activity from Penicillium verruculosum (Penicillium verruculosum의 Xylan분해활성도의 생성에 대한 Xylan의 영향)

  • During the cultivation of Penicillium verruculosum in the medium containing xylan as a sole carbon source for 26 days, xylanolytic activity and some changes were investigated. Protein content and xylanolytic activity, p-Nitrophenyl-$\beta$-D-xylopyranoside (PNPX), p-Nitrophenyl-$\beta$ -D-glucopyranoside (PNPG) hydrolytic activities were increased until 8 days but reducing sugar content was not correlated to protein content. When crude proteins from the culture broth were separated on SDS-PAGE, distribution of proteins was different from the culture broth of cellobiose octaacetate (COA) medium. The culture broth of xylan medium had high hydrolytic activity on xylan but not on cellulose. Furthermore, xylanolytic products were showed xylose, xylobiose and oligosaccharides on thin layer chromatography, and xylobiose was major product. Those result suggested that xylanolytic activity of culture broth was endo-type hydrolysis. Optimum temperatures of xylanolytic activity and PNPX hydrolytic activity of culture broth were 50~6$0^{\circ}C$ and 60~7$0^{\circ}C$, respectively and optimum pHs were 3.0~4.0 and 4.0~5.0, respectively.

Effect of Stewing Time on the Small Molecular Metabolites, Free Fatty Acids, and Volatile Flavor Compounds in Chicken Broth

  • Chicken broth has a taste of umami, and the stewing time has an important effect on the quality of chicken broth, but there are fewer studies on the control of the stewing time. Based on this, the study was conducted to analyze the effects of different stewing times on the sensory, small molecular metabolites, free fatty acids, and volatile flavor compounds contents in chicken broths by liquid chromatography-quadrupole/time-of-flight mass spectrometry, gas chromatography-mass spectrometry, headspace solid-phase microextraction, and gas chromatography-mass spectrometry. Eighty-nine small molecular metabolites, 15 free fatty acids, and 86 volatile flavor compounds were detected. Palmitic and stearic acids were the more abundant fatty acids, and aldehydes were the main volatile flavor compounds. The study found that chicken broth had the best sensory evaluation, the highest content of taste components, and the richest content of volatile flavor components when the stewing time was 2.5 h. This study investigated the effect of stewing time on the quality of chicken broth to provide scientific and theoretical guidance for developing and utilizing local chicken.

Effects of Medicinal Plants Extract on Naengmyeon Broth (약용식물 추출물이 냉면육수에 미치는 영향)

  • When the extract of the medicinal plants, Kaempferia galanga L., Zanthoxylum bungeanum, Eugenia caryophyllata, Foeniculum vulgare, was added to Naengmyeon broth with the concentration of 0.1% and 0.3% each, its effect during the preservation time of broth was investigated. pH of the extract-added broth was lower than control at the initial, but higher after 72 hours of preservation, which showed that when it added 0.1% and 0.3% of extract to the broth, pH of Kaempferia galanga L. was 4.92 and 5.08 respectively, whereas control was 4.60. Titratable acidity was lowered after 48 hours and also Kaempferia galanga L. showed the lowest acidity with 0.66 for adding 0.1% of its extract and 0.55 for 0.3% of adding, but control was 0.89 at the time of 90 hours of preservation, and then it showed to be lowered in the order of Zanthoxylum bungeanum, Eugenia caryophyllata and Foeniculum vuigare. Turbidity of each broth added the extracts of four of the medicinal plants was 7.5∼7.9 and 7.9∼8.2, respectively for 0.1% and 0.3% of concentration at the initial, but it began to lower and 90 hours later it was 8.8∼9.5 and 8.7∼9.0 respectively, whereas control was 10.8. Total viable cells(TVC) and coliform bacteria(CB) were increased with great at the 72 hours of preservation time, and Kaempferia galanga L. was the most effective, which when control was 4.8${\times}$10 CFU/ml at 72 hours, TVC was 1.7${\times}$10 CFU/ml for the addition of 0.1% of extract and 0.9${\times}$10 CFU/ml for 0.3%. CB was 3.2${\times}$10 CFU/ml for 0.1% and 1.7${\times}$10 CFU/ml for 0.3% respectively and 6.0 ${\times}$ 10 CFU/ml for control at the time of 72 hours, and it was lowered in the order of Zanthoxylum bungeanum, Eugenia caryophyllata and Foeniculum vulgare. Volatile basic nitrogen content detected that control was 2.67mg% at first, and then increased to 3.96mg% at 90 hours of preservation, but the broth added with the extract of Kaempferia galanga L. was 2.58mg% for 0.1% and 2.47mg% for 0.3% at the initial, and at 90 hours it was 3.64mg% and 3.33mg% respectively. The results of adding the extracts of four medicinal plants for the improvement of the preservation time of Naengmyeon broth, were that the most effective medicinal plant was Kaempferia galanga L. and the antimicrobial activity of the medicinal plant extracts for Naengmyeon broth was highly effective after 3 days of preservation time.

Neuraminidase Inhibitors from the Culture Broth of Phellinus linteus

  • During the search for neuraminidase inhibitors from medicinal fungi, we found that the culture broth of Phellinus linteus exhibited potent inhibitory activity. Solvent partition, Sephadex LH-20 column chromatography, and high-performance liquid chromatography (HPLC) were performed for purification of two active substances from the culture broth. According to $^1H$ NMR measurements and comparison of HPLC retention times with those of authentic compounds, their chemical structures were identified as hispidin and hypholomine B. Compounds (hispidin) 1 and 2 (hypholomine B) inhibited neuraminidase, with $IC_{50}$ values of 13.1 and 0.03 ${\mu}M$, respectively.

  • The inhibitory effects of vegetables, collected from Jinju district, on the growth of Listeria monocytogenes were investigated. Among 12 vegerables, garlic, leek, onion and cabbage were remarkably effective to inhibit the growth of the strain. Especially with the addition of extract equivalent 0.25g of raw garlic per ml broth, the population of the strain was decreased wignificantly, and completely inhibited with the addition of 0.625g or more. During the incubation with the addition of extract equivalent 0.25g of raw garlic per ml broth, the degree of inhibition was gradually increased with the lapse of time, and completely inhibited the growth of the strain after 48 hrs. While the pH value of treated broth remained unchanged almost, those of control were slightly reduced.

  • In this study, we investigated the antimicrobial activity of herbs related with treatments of intestinal diseases against intestinal pathogens under anaerobic broth system. The water extract of Saussurea lappa Clarke and Myristica fragrans Houtt. showed no growth inhibition against tested pathogens(Eubacterium limonsum ATCC 10825, Escherichia coli ATCC 25922, Bacteroides fragilis KCTC 5013, Clostridium perfringens STCC 3627, Staphylococcus aureus KFCC 11764 및 Salmonella typhimurium ATCC 14028). All tested pathogens were not inhibited in broth containing 100$\mu\textrm{g}$/$m\ell$ of Areca catachu L. Water extract but its extract strongly inhibited the growth of Eubacterium limonsum STCC 10825, Bacteroides fragilis KCTC 5013, Clostridium perfringens ATCC 3627 and Salmonella typhimurium ATCC 14028 at 1,000 to 2,000$\mu\textrm{g}$/$m\ell$ of concentration. Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew in broth containing 2,000$\mu\textrm{g}$/$m\ell$ of Terminalia chebula Retz. water extract.

Isolation and Properties of Amino Acid Antimetabolite from Streptomyces sp. YS-943 (Streptomyces sp. YS-943균주가 생산하는 아미노산 대사길항물질의 정제와 성상)

  • A Streptomyces strain YS-943, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broth of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-methionine and L-histidine on the synthetic minimal agar medium in the culture broth.The morphological and cultural characteristics serve to identify the producing organism strain YS-943 as the genus Streptomyces. Fermentation was carried out in the synthetic medium at 28$\CIRC$C for 48 hours. The fermentation yield reached about 12 mg per liter of the broth. The YS-943 substance was obtained as white powder, mp 194$\CIRC$C and has the molecular formular of C$_{4}$H$_{8}$N$_{2}$O$_{4}$. Its structure was determined to be o-carbamyl-D-serine by spectroscopic data. It is active against some Gram-positive bacteria and reversed by L-methionine and L-histidine.

  • For using additives of foods, or cosmetics, a strain A80 producing blue pigment was isolated from soil. The strain A80 was identified as a strain of Azotobacter vinelandii based on morphological and physiological characteristics. The strain A80 was extracellulaly secreted the blue pigment on PYG agar plate, but not secreted it into PYG broth. And then, the strain A80 was extracellulaly secreted the blue pigment in PYG broth containing 2.0% chitin, while the strain A80 was not secreted the blue pigment in PYG broth containing 2.0% chitin and 1% NaCl simultaniously.

  • Glycopeptide antibiotics, teicoplanin was purified from a mutant strain of Actinoplanes teichomyceticus ATCC31121, A. teichomyceticus MSL2211. We developed a simple procedure to separate and purify the teicoplanin from the fermentation broth. Teicoplanin was purified by two-step purification system, hydrophobic adsorption and sugar affinity chromatography in combination with HPLC analysis based on the properties of hydrophobic acyl chain and sugar moiety in teicoplanin. Teicoplanin was separated from the culture broth by Diaion HP-20 and further purified by concanavalin A affinity column chromatography. As an adsorbent resin, Diaion HP-20 in broth eliminated toxic effects on growth, reduced feedback repression of teicoplanin production, and assisted In rapid recovery of teicoplanin. The teicoplanin displayed the final yield of 80% and 95% of purity.

리보핵산 관련물질을 함유한 Yeast Extracts 제조에 Streptomyces faecalis MSF 배양액의 이용

  • RNA accumulating strain of Torulopsis versatilis MT-1 was cultured in molasses medium for higher contents of RNA in cell. Yeast cells were harvested at logarithmic phase on synchronous culture. Yield of cells on dry base to input sugar was 59.5%. Crude protein content was 55.1% in cell. RNA content was 13.9%. Some problems found in the process for the preparation of yeast extracts were improved by the addition of culture broth of Streptomyces faecalis MSF which secrete RNase (5' nuclease and 5' adenylic acid deaminase). When the culture broth of S. faecalis MSF was added in autolysis process 46% of RNA in cell was converted to I and G(5' inosinic acid and 5' guanylic acid) in extract. By addition of 3-7% culture broth of S.faecalis MSF in autolysis or enzymolysis process at the start or early stage, RNA in extract was converted easily to I and G and protein in cells was easily extracted and hydrolyzed to amino acid. Taste of those yeast extracts was delicious. The yeasty smell in yeast extracts was removed. And cell debris was easily removed from extract.
