Signal Transduction Network Leading to COX-2 Induction: A Road Map in Search of Cancer Chemopreventives

  • Cancer is still a major global health concern even after an everlasting strive in conquering this dread disease. Emphasis is now given to chemoprevention to reduce the risk of cancer and also to improve the quality of life among cancer afflicted individuals. Recent progress in molecular biology of cancer has identified key components of the cellular signaling network, whose functional abnormality results in undesired alterations in cellular homeostasis, creating a cellular microenvironment that favors premalignant and malignant transformation. Multiple lines of evidence suggest an elevated expression of cyclooxygenase-2 (COX-2) is causally linked to cancer. In response to oxidative/pro-inflammatory stimuli, turning on unusual signaling arrays mediated through diverse classes of kinases and transcription factors results in aberrant expression of COX-2. Population-based as well as laboratory studies have explored a broad spectrum of chemopreventive agents including selective COX-2 inhibitors and a wide variety of anti-inflammatory phytochemicals, which have been shown to target cellular signaling molecules as underlying mechanisms of chemoprevention. Thus, unraveling signaling pathways regulating aberrant COX-2 expression and targeted blocking of one or more components of those signal cascades may be exploited in searching chemopreventive agents in the future.

Differently expressed genes of soybean by ambient heat stress

  • Plants are grown under constitutive changing of environmental conditions and response to external conditions at both protein and transcription level. The effects of heat on plant growth are broad and influence the yield directly. Heat stresses could be classified depend on intensity and duration. Fundamental changes of growth condition by climate change maybe or maybe not classified as a stress on plant growth. The effects of a short and unanticipated impact of elevated heat on plant could be different with those of under longer extension of ambient heat. To examine differently expressed gene sets by ambient heat stress of soybean, we grow the soybean in normal condition for three weeks. After that, soybean plants move to growth chamber. The temperature of growth chamber increase up to $9^{\circ}C$ for four days. We have extracted mRNA and micro RNA every 24 hours and carried RNA sequence analysis. We found major metabolic pathways affected by ambient heat stress. Mainly carbon metabolism, translation machinery and amino acid synthesis are affected. We discussed the expression patterns of genes of heat sensing and hormone responses.

Characterization and expression profiles of aquaporins (AQPs) 1a and 3a in mud loach Misgurnus mizolepis after experimental challenges

  • Two distinct cDNAs encoding aquaporins (mmAQPs 1a and 3a) were isolated and characterized from mud loach Misgurnus mizolepis. The identified mud loach AQP cDNAs encode for polypeptides of 260 and 302 amino acids. Topology predictions confirmed six putative membrane-spanning domains connected by five loops and the N- and C-terminal domains being cytoplasmic. The mud loach AQPs 1a and 3a showed broad distribution in multiple tissues including immune-responsive tissues as well as osmoregulatory tissues. Hence, the diversity of AQP distribution and expression possibly indicated its differential functions in the regulation of fluid movement in response to environmental stimuli. The transcription of mmAQP genes was differentially modulated by immune challenges. In particular, the mmAQP3a expression level in the liver was more responsive to immune challenges than that of mmAQP1a. Taken together, fish stimulation or infection resulted in significant modulation of mud loach AQP genes, suggesting potential functional roles of these proteins in piscine pathophysiological process.

Construction of a CRISPR/Cas9-Mediated Genome Editing System in Lentinula edodes

  • CRISPR/Cas9 genome editing systems have been established in a broad range of eukaryotic species. Herein, we report the first method for genetic engineering in pyogo (shiitake) mushrooms (Lentinula edodes) using CRISPR/Cas9. For in vivo expression of guide RNAs (gRNAs) targeting the mating-type gene HD1 (LeA1), we identified an endogenous LeU6 promoter in the L. edodes genome. We constructed a plasmid containing the LeU6 and glyceraldehyde-3-phosphate dehydrogenase (LeGPD) promoters to express the Cas9 protein. Among the eight gRNAs we tested, three successfully disrupted the LeA1 locus. Although the CRISPR-Cas9-induced alleles did not affect mating with compatible monokaryotic strains, disruption of the transcription levels of the downstream genes of LeHD1 and LeHD2 was detected. Based on this result, we present the first report of a simple and powerful genetic manipulation tool using the CRISPR/Cas9 toolbox for the scientifically and industrially important edible mushroom, L. edodes.

국화 꽃색 변경을 위한 플라보노이드 대사공학 (Flavonoid Metabolic Engineering for Modification of Flower Color in Chrysanthemum)

  • 관상용 화훼작물에 있어서 꽃의 색깔과 형태는 중요한 형질 중 하나이다. 일반적으로 꽃색은 카로티노이드, 플라보노이드, 베타라인에 의해 결정된다. 그 중 플라보노이드는 보다 넓은 영역의 색을 나타낸다. 국화는 세계적으로 인기가 많은 관상용 화훼작물이며 꽃색을 바꾸기 위한 많은 연구가 진행되어 왔다. 국화의 경우, 시아니딘 계열 안토시아닌의 축적으로 분홍색 혹은 빨간색의 꽃색을 나타내며, 카로티노이드 계열 색소물질의 축적으로 노란색 또는 초록색의 꽃색을 나타낸다. 그러나 자연계에는 파란 꽃색의 국화는 존재하지 않는다. 지금까지 플라보노이드계 물질 생합성을 조절함으로써 파란색 꽃을 개발하기 위한 여러 연구가 시도되었다. 반면 그 외의 플라보노이드계 물질을 기반으로 한 새로운 꽃색 국화 개발연구는 거의 수행되지 않았다. 플라보노이드 생합성 조절에는 다양한 전사인자들이 관여하고 플라보노이드계 물질 기반 꽃색 변경을 위해서는 구조 유전자 및 전사인자들을 이해하는 것이 중요하다. 따라서 본 논문에서는 화훼작물의 플라보노이드 생합성 및 조절에 대하여 전반적으로 서술하였고, 그 동안 보고된 플라보노이드계 물질의 꽃색 변경 연구들을 검토하였다. 이러한 결과들은 생명공학기술을 기반으로한 국화 꽃색 변경 달성을 위한 중요한 길잡이가 될 수 있을 것이다.

당귀에서 발생한 토마토반점위조바이러스의 감염 첫 보고 (First Report of Tomato Spotted Wilt Virus in Angelica acutiloba)

  • 2019년 6월 충남 논산의 당귀 재배 농가에서 원형반점, 괴사반점, 황화, 모자이크 등의 증상을 보이는 당귀잎을 채집하였고, 이들 시료에 대한 바이러스 감염 여부를 확인하기 위하여 전자현미경 검경과 감염우려가 있는 바이러스 3종(cucumber mosaic virus, broad bean wilt virus 2, tomato spotted wilt virus [TSWV])에 대해 reverse transcription polymerase chain reaction 진단을 수행한 결과 TSWV에 대해 양성반응을 보였다. 당귀 TSWV의 생물학적 특성을 구명하기 위해 순수분리하여 5과 28종의 지표식물과 일당귀에 즙액접종하여 기주범위와 병원성을 확인하였다. TSWV의 3 segment (L, M, S)의 전체 염기서열을 결정하고 기존에 보고된 TSWV 분리주와 계통분석을 한 결과, 당귀 TSWV 분리주(NS-AG-28)는 논산지역의 머위 분리주(TSWV-NS-BB20)와 가장 유사한 것으로 나타났다. TSWV는 넓은 기주범위를 가지고 있고 특히 고추, 토마토 등 가지과 작물에 큰 피해를 주고 있기 때문에, 당귀가 TSWV의 중간기주로서 작용하지 않도록 주의하고, 당귀의 바이러스병 피해를 예방하기 위하여 지속적인 모니터링이 요구된다.

Combining In Silico Mapping and Arraying: an Approach to Identifying Common Candidate Genes for Submergence Tolerance and Resistance to Bacterial Leaf Blight in Rice

  • Several genes/QTLs governing resistance/tolerance to abiotic and biotic stresses have been reported and mapped in rice. A QTL for submergence tolerance was found to be co-located with a major QTL for broad-spectrum bacterial leaf blight (bs-blb) resistance on the long arm of chromosome 5 in indica cultivars FR13A and IET8585. Using the Nipponbare (japonica) and 93-11 (indica) genome sequences, we identified, in silico, candidate genes in the chromosomal region [Kottapalli et al. (2006)]. Transcriptional profiling of FR13A and IET8585 using a rice 22K oligo array validated the above findings. Based on in silico analysis and arraying we observed that both cultivars respond to the above stresses through a common signaling system involving protein kinases, adenosine mono phosphate kinase, leucine rich repeat, PDZ/DHR/GLGF, and response regulator receiver protein. The combined approaches suggest that transcription factor EREBP on long arm of chromosome 5 regulates both submergence tolerance and blb resistance. Pyruvate decarboxylase and alcohol dehydrogenase, co-located in the same region, are candidate downstream genes for submergence tolerance at the seedling stage, and t-snare for bs-blb resistance. We also detected up-regulation of novel defense/stress-related genes including those encoding fumaryl aceto acetate (FAA) hydrolase, scramblase, and galactose oxidase, in response to the imposed stresses.

황백(黃柏)이 전립선비대증(前立腺肥大症) Rat에 미치는 영향 (The Effects of Phellodendri Cortex Ex on Experimental Rat Model of Benign Prostatic Hyperplasia)

  • Objective : Benign prostatic hyperplasia(BPH) is one of the most common diseased among elderly men. BPH can be treated with alpha-1 adrenergic blocker or $5{\alpha}$-reductase inhibitor(Finasteride) that reduces serum dihydrotestosterone(DHT). Phellodendri Cortex Ex has been broad studied on its chemical components, pharmacological activity, and clinical effects on anti-inflammation, anti-allergy, anti-tumor, immunity, antibacteria and other bioactivities. In this study, we investigated the therapeutic effects and action mechanism of Phellodendri Cortex Ex with a BPH induced by castration and testosterone treatment. Methods : Sprague-Dawley rats were treated with testosterone after castration for induction of experimental benign prostatic hyperplasia, which is similar to human benign prostatic hyperplasia in histopathological profiles. Phellodendri Cortex as an experimental specimen, and Finasteride as a positive control, were administered orally. The prostates were evaluated by histopathological changes, and the expression of $5{\alpha}$-reductase genes. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with Phellodendri Cortex Ex showed a diminished range of the tissue damage. In the reverse transcription-polymerase chain reaction(RT-PCR) of $5{\alpha}$-reductase genes, Phellodendri Cortex inhibited the expression of $5{\alpha}$-reductase genes. Conclusions : These findings suggest that Phellodendri Cortex Ex may protect the glandular epithelial cells and also inhibit stromal proliferation in association with the suppression of $5{\alpha}$-reductase. From these results, we suggest that Phellodendri Cortex Ex could be a useful agent for treating the benign prostatic hyperplasia.

Regulation of the Edwardsiella tarda Hemolysin Gene and luxS by EthR

  • Edwardsiella tarda is a pathogen with a broad host range that includes human and animals. The E. tarda hemolysin (Eth) system, which comprises EthA and EthB, is a noted virulence element that is widely distributed in pathogenic isolates of E. tarda. Previous study has shown that the expression of ethB is regulated by iron, which suggests the possibility that the ferric uptake regulator (Fur) is involved in the regulation of ethB. The work presented in this report supports the previous findings and demonstrates that ethB expression was decreased under conditions when the E. tarda Fur ($Fur_{Et}$) was overproduced, and enhanced when $Fur_{Et}$ was inactivated. We also identified a second ethB regulator, EthR, which is a transcription regulator of the GntR family. EthR represses ethB expression by direct interaction with the ethB promoter region. In addition to ethB, EthR also modulates, but positively, luxS expression and AI-2 production by binding to the luxS promoter region. The expression of ethR itself is subject to negative autoregulation; interference with this regulation by overexpressing ethR during the process of infection caused (i) drastic changes in ethB and luxS expressions, (ii) vitiation in the tissue dissemination and survival ability of the bacterium, and (iii) significant attenuation of the overall bacterial virulence. These results not only provide new insights into the regulation mechanisms of the Eth hemolysin and LuxS/AI-2 quorum sensing systems but also highlight the importance of these systems in bacterial virulence.

Characterization and gene expression of heat shock protein 90 in marine crab Charybdis japonica following bisphenol A and 4-nonylphenol exposures

  • Objectives Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone important in the maturation of a broad spectrum of protein. In this study, an HSP90 gene was isolated from Asian paddle crab, Charybdis japonica, as a bio-indicator to monitor the marine ecosystem. Methods This work reports the responses of C. japonica HSP90 mRNA expression to cellular stress by endocrine disrupting chemicals (EDCs), such as bisphenol A (BPA) and 4-nonylphenol (NP) using real-time. reverse transcription polymerase chain reaction. Results The deduced amino acid sequence of HSP90 from C. japonica shared a high degree of homology with their homologues in other species. In a phylogenetic analysis, C. japonica HSP90 is evolutionally related with an ortholog of the other crustacean species. The expression of HSP90 gene was almost distributed in all the examined tissues of the C. japonica crab but expression levels varied among the different body parts of the crabs. We examined HSP90 mRNA expression pattern in C. japonica crabs exposed to EDCs for various exposure times. The expression of HSP90 transcripts was significantly increased in C. japonica crabs exposed to BPA and NP at different concentrations for 12, 24, 48 and 96 hours. The mRNA expression of HSP90 gene was significantly induced in a concentration- and time-dependent manner after BPA or NP exposures for 96 hours. Conclusions Taken together, expression analysis of Asian paddle crab HSP90 gene provided useful molecular information about crab responses in stress conditions and potential ways to monitor the EDCs stressors in marine environments.
