Characterization of Acetobacter sp. Strain CV1 Isolated from a Fermented Vinegar (고산도 생성 초산균의 분리 및 발효특성)

  • Ten types of farm-made brewing vinegars were collected and four high acetic acid-producing strains (CV1, CV3, CV5, and CV6) were isolated. Among them strain CV1, exhibiting highly alcohol-resistant and acetic acid-producing properties, was selected and its taxonomic properties were investigated by phenotypic (particularly chemotaxonomic) characterization and phylogenetic inference based on 16S rRNA gene sequence analysis. On SM broth agar, cells of strain CV1 were gram-stainingnegative and formed pale white colonies with smooth to rough surfaces. Strain CV1 produced acetate from ethanol and was resistant to up to 8% (v/v) ethanol in LM broth. Strain CV1 had a G+C content of 61.0 mol%, contained meso-DAP as the cell wall amino acid, and possessed Q-10 as the major ubiquinone. A comparison of 16S rRNA gene sequences showed that strain CV1 was most closely related to Gluconacetobacter saccharivorans (≥99.0% identity). In liquid media, the optimum growth conditions for acetic acid production were 30℃ and pH >3.0 and strain CV1 produced 9.3% and 8.4% acetic acids from 10% and 9% alcohol concentrations, respectively.

Characterization of Brewing Yeast Expressing Glucoamylase Selected by Rare Mating. (Rare Mating에 의한 양초효모에서의 glucoamylase 발현 균주 HCS 선별 및 특성)

  • Rare mating was used to select a respiratory deficient mutant of Saccharomyces cerevisiae HDC52 strain. Glucoamylase gene of S. diastaticus K114 was developed into the RD mutant which could uptake maximum amount of non-fermentable sugars through the expression of glu- coamplyase gene and the fermentation characteristics of the developed strain HCS were investigated. The size of HCS yeast and HBD52 yeast strain were 13 $\mu\textrm{m}$ and 10$\mu\textrm{m}$ respectively. HCS strain which can uptake maximum amount of non-fermentable sugar through the expression of glucoamylase gene was developed. By karyotype anal- ysis. HCS stain but not RD mutant HBC52 showed a band of 1150 kb chromosome DNA This band should include glcoamylase gene from Saccharomyces diataticus K114 THis strain has glucoamylase which can degrade starch By transduction and contrnuance of glucoamylase gene HCS strain gegraded strach and formed halo. Also, HCS strain maintained the character after 50 generations. Glucoamylase activities of Saccharomyces diastaticus K114 and HCS yeast strains are 9.5 and 2.7~3.4(unit/ml) HCS and HBC52 strain showed similar sugar fermentation patterns and low flocculation In spore and film forming test, HCS and HBC52 strain formed neither spores nor films. In the limit fermentation test, HBC52 strain showed fermentation level of 68% and HCS strain showed 76~78% As the limit attenuation of HBC52 and HCS were ($2.00^{\circ}$P) and ($0.7~0.93^{\circ}$P) This study demon- strates and HCS strain may be used for low carbohydrate beer fermentation.

  • The fruits of Acanthopanax sessiliflorus Seeman (Araliaceae) were extracted with 70% aqueous ethanol at room temperature. The concentrated extract was partitioned with ethyl acetate (EtOAc), n-butyl alcohol, and $H_2O$, successively. From the EtOAc fraction, two compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. According to the results of physicochemical and spectroscopic data including NMR, mass spectrometry, and infrared spectroscopy, the chemical structures of the compounds were determined as 3,5-dihydroxycinnamic acid (1) and protocatechuic acid (2). Compound 1 was isolated from the fruits of A. sessiliflirus Seeman for the first time. And the compounds were evaluated for the radical scavenging the antioxidant capacity using 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)diammonium salt, 1,1-diphenyl-2-picrylhydrazy, and oxygen radical absorbance capacity assay.

Studies on the Yeasts for the Brewing of Soy Sauce(3) -Isolation and Identification of Osmophilic Yeasts for Higher Concentration of NaCl (간장발효에 관여하는 효모에 관한 연구(제3보) -고농도 식염내성 효모의 분리동정-)

  • This experiment was carried out to isolate osmophilic yeasts for higher concentration of NaCl than 20 per cent may affect greatly the fermentation of soy sauce. 6 strains of yeasts were selected from the soy sauce mashes at the fermentating periods and identified observing their morphological characteristics. Their salt-resistance and effects upon the flavor of soy sauce were examined. The results obtained were as follows: (1) The strain $T_3,T_5,$ and $T_{11},\;T_8,\;T_9,\;and\; T_{10},$ selected were identified Saccharomyces cerevisiae group II, Debaryomyces nicotianae, Saccharomyces chevalieri, Saccharomyces rouxii and Saccharomyces rosei, respectively. (2) They were more salt-resistant on the liquid media than on the solid media in the case of cultures on the media containing more than 26 per cent of NaCl. (3) They were grown on the media containing 5-23 per cent of NaCl better than containing none of NaCl. (4) The strain $T_3,\;T_8,\;T_9,\;and\; T_{10}$ were grown on the media containing up to 28 per cent of NaCl, and $T_5\;and\;T_{11}$ up to 32 per cent of NaCl is the case of liquid cultures. (5) Among them, the strain $T_9$ showed the best alcohol fermentation ability and the best fermentative flavor, and $T_5\;and\;T_{11}$ formed moldy smell and pellicle.

Yeast Selection for Quality Optimization of Distilled Spirits (증류주의 품질 최적화를 위한 효모선발 연구)

  • The aim of this study was to select yeast strain for quality optimization of distilled spirit which is prepared using rice. Five yeasts strains were compared for their brewing characteristics and showed normal fermentation pattern, but songcheon yeast(Y1) and distillery yeast(Y5) revealed higher fermentation ability than other samples tested. The analyzed results of fermented mashing showed that the distillery yeasts(Y4, Y5) had significantly much higher alcohol content, and songcheon yeast(Y1), distillery yeasts(Y4, Y5) had significantly higher ester content than other samples tested respectively, while the distillery yeasts(Y3, Y4, Y5) had significantly higher organic acid content than other samples tested. The analyzed results of distilled spirits which were distilled using copper distillery apparatus showed that the songcheon yeast(Y1) and distillery yeast(Y5) had a higher yield compared to other samples tested. In addition, the results of the aroma compounds such as ester and higher alcohol of distilled spirits among the five yeasts tested were similar to the analyzed results of fermented mashing. Siha aktivhefe 6 brennereihefe(Y5) indicated the highest overall preference including sensory evaluation and was selected as best yeast strain for quality optimization of distilled spirit which is prepared using rice.

Effect of Rice Pre-treatment on Enzymatic Saccharification in the Brewing Process (주류 제조를 위한 효소 당화에 쌀의 전처리가 미치는 영향)

  • To produce sweet liquor without artificial sweeteners, 8 traditional rice pre-treatment methods (juk, beombeok, seolgitteok, gumeongtteok, mulsongpyeon, injeolmi, gaetteok, and godubap) were analyzed in this study. The formation of sugars with the help of ${\alpha}$-amylase, ${\beta}$-amylase, and glucoamylase using nuruk as a substrate has been previously confirmed. During the early stages of the pre-treatment processes, the amount of maltose produced (in descending order of its concentration) by ${\alpha}$-amylase was observed to be as follows: gaetteok > seolgitteok > beombeok > mulsongpyeon > juk > injeolmi > gumeongtteok > godubap. However, changes in maltose concentrations with respect to the pre-treatment processes after 48 hours were observed to be as follows: injeolmi > beombeok = godubap > gumeongtteok > gaetteok = mulsongpyeon > seolgitteok > juk. Maltose produced using either ${\alpha}$-amylase or ${\beta}$-amylase showed similar results. Glucoamylase produced 10 mg/ml of glucose during the godubap process, which was the highest amount of glucose among all the methods. Moreover, when ${\alpha}$-amylase, ${\beta}$-amylase, and glucoamylase were used concurrently, glucoamylase increased glucose production in the later stages. Therefore, the possibility of producing sweet liquor without employing artificial sweeteners was confirmed, even if the amount of sugar in the liquor varied with the pre-treatment process.

  • These experiments were carried out to obtain useful yeast for Korean grape wine brewing. The 213 strains of yeasts were isolated from grapes sampled in Daejeon city and suburbs and then 3 strains among the isolated yeasts were selected as useful strains and identified. The results obtained as follows; 1. The 3 strains of W-49, W-50 and W-127 selected as useful yeasts were clarified to grape wine mash rapidly. 2. In the mashes clarified after storage of 45 days, the value of optical density of the case of W-50 strain was highest and the color was very thick, compared with the case of another strains. 3. In the pannel test of the grape wines, the case of W-49 and W-50 strains were best, the case of the W-127 strain and control yeast, Hb were similar each other as next order. 4. Among 3 strains selected as useful yeasts it were identified that the strains of W-49, W-50 selected as useful strains were Saccharomyces cerevisiae and the strain of W-127 was Saccharinyces pretoriencis by the Ladder manual. 5. All of these useful yeasts were appeared red color on TT C stain.

Fermentation Characteristics of Wine Yeast Strains (포도주 양조용 효모의 발효특성)

  • In fermentation characteristics, R2 was suitable for fast start and fermentation, while UCD530 was tool suitable far complete fermentation at 25$^{\circ}C$. T73 and AC- strains produced much more total acid compared to other strains and it was concerned with producing large amount of succinic acid and acetic acid. OC-2, UCD530, Beaujolais, and BC strains revealed low fermentation efficiency (below 62.0%), but EC1118, RC212, T73 and BM45 strains showed opposit result (above 70.0%). D254 and Wadenswil 27 seemed to have excellent cohesive ability because these two yeast strains made somewhat hard precipitation at the bottom after complete fermentation. T73 and CEG gave higher amounts of acetic acid (above 630 mg/L), while UCD530, W-3 and Beaujolais recorded low concentration (below 200 mg/L). In sulfur dioxide tolerance, preferable culturing temperature and times were 25$^{\circ}C$ and 72 hr respectively. The strains R2, BM45 and L2056 revealed high sulfur dioxide tolerance (above 30mg/L), white 71B, Wadenswil 27 and Beaujolais showed the opposite result (below 5mg/L)

Fermentation Characteristics of Wine Yeast Strains for White Wine Making (백포도주 양조에 있어서 포도주 효모의 발효 특성)

  • The characteristics of used wine yeast strains were as follows, S6U showed low fermentation speed than those of other yeast strains, but this strain fermented completely later. The wine fermented by W-3 was very low contents of total acid, 0.75% and the ones fermented by UCD530 and AC- contained much extract, 3.26 mg/L and 3.22 mg/L respectively. The wine fermented by CEG and CS2 were predominant in yellowness, and EC1118 produced large amount of acetaldehyde, 49.9 mg/L than those of other strains. EC1118 and CY3079 displayed low methylene blue dyeing ratio, below 15%, meaning high alcohol tolerance yeast. UCD530 produced extremely high contents of glycerol, succinate and lactate compared with other strains. These properties revealed that UCD530 was a typical Saccharomyces bayanus species. The main organic acids produced by wine yeasts were pyruvate, lactate, succinate and acetate. The concentration of acetate in experimental wine could be divided into two parts, one group had concentration below 170 mg/L (UCD530, EC1118, AC-, CY3079, W-3), and the other had concentration up to 350 mg/L (S6U, CEG, CS2).
