Two subspecies, japonica and indica, have been reported in rice, which differ in several ecotypic traits. However, reproductive barriers in hybrid progenies between subspecies have been major obstacles in breeding programs using inter-subspecific hybridization. As the first step to elucidate the reproductive barriers, we developed subspecies-specific(SS) STS markers in this study. A total of 765 STS primers were designed through comparing DNA sequences at every $2{\sim}3$cM interval between japonica and indica rices, which are available at Web DBs such as IRGSP, NCBI, TIGR, and GRAMENE, and tested for subspecies-specificity using 15 indica and 15 japonica varieties of diverse origin. Of them, 67 STS markers were identified as SS STS markers and their subspecies-specificity scores were estimated. The SS markers were dispersed throughout the genome along chromosomes. Of them, 64 SS markers were mapped on an RIL population derived from a Dasanbyeo(indica)/TR22183(japonica) cross. Genomic inclination of RILs was evaluated based on the genotyping with different types of markers. Association test between markers and segregation distortion revealed that segregation distortion might not be the cause of generating SS markers. The SS markers will be applicable to estimate the genomic inclination of varieties or lines and to study the differentiation of indica and japonica, and ultimately to breed true hybrid rice varieties in which desirable characters from both subspecies are recombined.
Non-heading Chinese cabbage (Brassica rapa L. ssp. chinensis) is a popular vegetable in Asian countries. The diamondback moth (DBM), Plutella xylostella (L.), an insect with worldwide distribution, is a main pest of Brassicaceae crops and causes enormous crop losses. Transfer of the anti-insect gene into the plant genome by transgenic technology and subsequent breeding of insect-resistant varieties will be an effective approach to reducing the damage caused by this pest. We have produced transgenic non-heading Chinese cabbage plants expressing the potato proteinase inhibitor II gene (pinII) and tested the pest resistance of these transgenic plants. Non-heading Chinese cabbages grown for 45 days on which buds had formed were used as experimental materials for Agrobacterium-mediated vacuum infiltration transformation. Forty-one resistant plants were selected from 1166 g of seed harvested from the infiltrated plants based on the resistance of the young seedlings to the herbicide Basta. The transgenic traits were further confirmed by the Chlorophenol red test, PCR, and genomic Southern blotting. The results showed that the bar and pinII genes were co-integrated into the resistant plant genome. A bioassay of insect resistance in the second generation of individual lines of the transgenic plants showed that DBM larvae fed on transgenic leaves were severely stunted and had a higher mortality than those fed on the wild-type leaves.

This study aimed to evaluate six cauliflower genotypes regarding their resistance to black rot and their production performance. To do so, it was conducted two field experiments in Ipameri, $Goi\acute{a}s$, Brazil, in 2012 and 2013. It was used a randomized block design, with four replications (total of 24 plots). Each plot consisted of three planting lines 2.5 m long (six plants/line), spaced 1.0 m apart, for a total area of $7.5m^2$. Evaluations of black rot severity were performed at 45 days after transplanting, this is, 75 days after sowing (DAS), and yield evaluations at 90 to 105 DAS. The Verona 184 genotype was the most resistant to black rot, showing 1.87 and 2.25% of leaf area covered by black rot symptom (LACBRS) in 2012 and 2013. However, it was not among the most productive materials. The yield of the genotypes varied between 15.14 and 25.83 t/ha in both years, Lisvera F1 (21.78 and 24.60 t/ha) and Cindy (19.95 and 23.56 t/ha) being the most productive. However, Lisvera F1 showed 6.37 and 9.37% of LACBRS and Cindy showed 14.25 and 14.87% of LACBRS in 2012 and 2013, being both considered as tolerant to black rot.
Microsatellites or simple sequence repeats (SSR) are widely distributed in eukaryotic genomes and are informative genetic markers. Despite many advantages of SSR markers such as a high degree of allelic polymorphisms, co-dominant inheritance, multi-allelism, and genome-wide coverage in various plant species, they also have shortcomings such as low polymorphic rates between genetically close lines, especially in Capsicum annuum. We developed an alternative technique to SSR by normalizing and alternating anchored primers in random amplified microsatellite polymorphisms (RAMP). This technique, designated reverse random amplified microsatellite polymorphism (rRAMP), allows the detection of nucleotide variation in the 3' region flanking an SSR using normalized anchored and random primer combinations. The reproducibility and frequency of polymorphic loci in rRAMP was vigorously enhanced by translocation of the 5' anchor of repeat sequences to the 3' end position and selective use of moderate arbitrary primers. In our study, the PCR banding pattern of rRAMP was highly dependent on the frequency of repeat motifs and primer combinations with random primers. Linkage analysis showed that rRAMP markers were well scattered on an intra-specific pepper map. Based on these results, we suggest that this technique is useful for studying genetic diversity, molecular fingerprinting, and rapidly constructing molecular maps for diverse plant species.
This study was conducted to evaluate the growth characters and yield components of 18 collected sesame cultivars to get basic information on the variation for the sesame breeding using principal component analysis. All characters except days to flowering, days to maturity and 1,000 seed weight showed significantly different. Seed weight per 10 are showed higher coefficient of variance. Capsule bearing stem length and liter weight showed positive correlation with seed yield per 10 are. The principal components analysis grouped the estimated sesame cultivars into four main components which accounted for 83.7% of the total variation at the eigenvalue and its contribution to total variation obtained from principal component analysis. The first principal component ($Z_1$) was applicable to increase plant height, capsule bearing stem length and 1,000-seed weight. The second principal component ($Z_2$) negatively correlated with days to flowering and maturity by which it was applicable to shorten flowering and maturity date of sesame. At the scatter diagram, Yangbaek, Ansan, M1, M2, M4, M7 and M9 were classified as same group, but M10, Yanghuk, Kanghuk, M5, M6, M12 and M13 were classified as different group. This results would be helpful for sesame breeder to understand genetic relationship of some agronomic characters and select promising cross lines for the development of new sesame variety.

Improvement of cold-tolerance of Italian ryegrass(Lo1ium multiflorum L.) is an important breeding objectivefor enlargement of cultivation area in Korea. To develop the cold-tolerant variety of Italian ryegrass,cold-tolerant clones survived under - 13- - 14$^{\circ}$C of minimum average air temperature in January wereselected in the area of Dun Nae, Kwangwon Province. The 5 lines of Italian ryegrass clones selected werepolycrossed for seed production. Synthetic seeds were examined on growth characters and forage productionsin Suwon from 1996 to 1998, in Unbong 1997 and in Younchun 1998, respectively. The growth charactersand forage productions of Italian ryegrass variety named as "Hwasan 101" were summarized as follow ;ltalian ryegrass variety, Hwasan 101 was tetraploid and showed semi-prostrate growth habit in autumn andwas not only dark green in leaf color and broad in flag leaf width but also excellent in leafiness. Also, itshowed low plant height at first heading date of 20th May and excellent lodging tolerance. Expecially,Hwasan 101 in all trial regions was 80 -90% of winter survivals that was higher than 60 -90% of Florida80 and Marshall varieties. Therefore we estimate that Hwasan 101 can be cultivated in regions that is higherthan -9$^{\circ}$C in minimum average air temperature and lower than 400m in sea level. In forage production ofHwasan 101, fresh weight and total digestible nutrient(TDN) yield were 66,940 and 6,551kghq and drymatter yield was 10,050kgha. In conclusion, Italian ryegrass, Hwasan 101 was medium and late maturingvariety but excellent in cold-tolerance, lodging-tolerance, leafiness, and forage production.(Key words : Italian ryegrass, Cold tolerance, New variety)ian ryegrass, Cold tolerance, New variety)

In order to obtain basic information for pure line breeding of hemp which is effective on the pure line preservation and propagation, three varieties were investigative from 1995 to 1996 at Mokpo Experiment Station, Honam Agriculture Experiment Station, and cutting parts, cutting times, cutting length and storage duration after cutting of plant were observed. Higher rooting ratio were observed in cutting of shoot and later branch, in July 5, optimum harvesting time and 5 days after harvesting time, in shorter storage duration,0 and 2 days. Also there were the highest rooting ratio increased by from 12% to 16% in shorter cutting of 10cm than 15cm long cutting in shoot, lateral branch and stem cuttings. Hemp cv. Korea showed higher rooting ratio than the other varieties, Japan and Turkey. Therefore, it was concluded that hemp, a dioecious plant, could be effectively propagated for the pure lines by cutting method, especially in cv. Korea.

Genetic background and phylogenetic relationships among 20 Korean wheat cultivars were assessed using microsatellites after amplifying with 13 SSR primer pairs. Average allele number per primer pair was 3.36. Genetic similarities for every pair of cultivars ranged from 0.42 to 0.97, with 0.69 of overall average. Korean cultivars were divided into two major groups based on microsatellite DNA polymorphisms. Group I consisted of relatively old cultivars developed until 1970s, and group II contained the recent cultivars developed during 1980s and 1990s. Amongst old elite cultivars/lines, ‘Yukseung 3’, ‘Norin 12’ and ‘Norin 72’ contributed most to the genetic background of cultivars belonging to group I, and ‘Norin 4’, ‘Norin 12’, ‘Norin 43’ and ‘Norin 72’ to group II, respectively. The phylogenetic relationship of Korean wheat cultivars was in accordance with the genealogical data of each cultivar. The genetic background of each cultivar was assessed from the point of breeding and germplasm management such as variety identification and duplicated accessions for assisting in developing a system for the registration of new variety based on the molecular characterization in future.
The inheritance and allelism tests of the genes resistant to bacterial blight in some rice breeding lines and varieties were studied. Resistance to isolate JB 8206 was found to be controlled by a single dominant gene in the rice varieties such as, Cheongcheongbyo, Yeongpungbyo, Nampungbyo, Samgangbyo, Hangangchalbyo, and Milyang 42. The resistance in varieties like Pungsanbyo and Baegyangbyo, to isolates JB 8206 and KN 8298 appeared to be governed by a single dominant gene. Evidence from the allelism test indicates that Pungsanbyo, Cheongcheongbyo, Milyang 30, and Baegunchalbyo may have the same dominant gene for resistance to isolate JB 8206, and that Suweon 312, Baegyangbyo as well as Baegunchalbyo may have the other same dominant gene.
Fifiy-five wheat samples were studied to obtain the basic information on the varietal differences of physicochemical characteristics and cookie baking potentiality in wheat breeding lines. Alkaline water retention capacity (AWRC) value had a correlation coefficient of $0.51^{**}$ with sedimentation value, of $0.45^{**}$ with protein content, and of $0.47^{**}$, with Pelshenke value. The highly significant correlation coefficient was obtained for cookie diameter vs. AWRC value $(-0.98^{**})$ and cooki diameter vs. protein content $(-0.46^{**})$. Farinograph absorption is highly correlated with AWRC value $(r=0.77^{**})$ and with cookie diameter $(r=-0.73^{**})$. Farinograph band width had a correlation coefficient of $-0.49^{**}$ with AWRC value, and of $0.44^{**}$ with cookie diameter.
