Days to Heading and Glossiness Variation of RILs derived from Hwayeong and Wandoaengmi 6

  • Improving the taste of rice in the breeding process is one of the important goals. However, it takes a lot of time and effort to select lines with good grain quality. MAS related to rice quality can help quickly and accurately select the elite lines in breeding programs. QTL qTV9, derived from Wandoaengmi 6, has been reported as a marker associated with improved glossiness of rice (Park et al., 2019). To confirm the function of QTL qTV9, 186 RILs derived from Hwayeong/Wandoaengmi6 were cultivated on ordinary planting cultivation for five years. The average DTH of Hwayeong and Wandoaengmi 6 was not significant at 99 and 97 days, respectively, but the averages of TV (toyo value) were 72.6 and 86.0, respectively. The DTH and TV of RIL vary from year to year. In 2017-2018, the average DTH was 98 days, which was significantly higher than the other three years. In 2018 and 2021, the average TV was 79.5 and 86.5, respectively, which were significantly higher than in other years. As a result of correlation analysis, DTH in the different years showed highly significant positive correlations (r = 0.71-0.92) from 0.71 to 0.92, whereas TV showed positive but weaker correlations (r = 0.42-0.71). The correlation between DTH and TV in each year was significant but weak (r = 0.25-0.64) and there was no correlation in 2017. The TV (77.6-88.7) of RILs with QTL qTV9 was significantly higher than that of RILs without qTV9 (72.6-84.9) for all five years. As a result of analyzing TV by DTH group, the TV of the lines with qTV9 in DTH groups (93-97) and (98-103) showed a significantly higher trend for all 5 years. And TV was not significant in DTH groups A, B, E, and F. This may have been influenced by factors such as insufficient populations between groups or differences in harvest timing. This study is expected to be used as data for improving the glossiness of cooked rice in breeding programs, and further study of the QTL qTV9 marker is required.

Genetic Diversity Estimation of the Rice Mutant Lines Induced by Sodium Azide

  • To investigate dose-effect of a chemical mutagen, sodium azide on a rice elite line, Suweon472, seed aliquots were treated with five different concentrations of sodium azide. The degree of mutation levels of each aizde concentration were estimated by using DNA fingerprinting techniques such as RAPD and AFLP. Some selected mutant lines ($M_4$) were also subjected for DNA fingerprinting to estimate their mutation levels by comparing the banding patterns of the wild type, Suweon 472. RAPD and AFLP fingerprinting patterns indicated that dose-effect of different azide concentrations was not clear. With allele description of detected AFLPs among favorable mutant lines, it was possible to discriminate each mutant line from others which have similar phenotypes and reactions against pathogens. AFLP fingerprinting patterns of waxy mutant lines, otherwise, were highly homogeneous as well as their phenotypic and agronomic characters.

  • Rye breeding using F$_1$ hybrid began about 30 years ago, when cytoplasmically inherited forms of male sterility (CMS) and corresponding nuclear restorers were detected. It is very important to produce inbred lines for making hybrid lines because of strong self-incompatibility in rye. Among the 456 rye germplasms used in hybrid breeding scheme, 24 lines (5.3%) had the above 60% of self-fertility, and six lines of them were selected for their good agronomic characteristics and were used for subsequent inbreeding program. The average self-fertility of selected six lines was 78.4%, ranging from 72.2 to 99.5%. Genetic analysis for the self-fertility using $F_2$ populations showed that the segregation of self-fertile and sterile plants in F$_2$populations could be fit into 3 to 1 ratio suggesting self-fertility in rye be controlled by one major gene. The four different self-fertile lines, PI237923, 5C11, 5G5 and Florida black, had the same self-fertility gene because their F$_2$ plants showed almost the same self-fertility as their parents and showed no genetic segregation.

  • Sorghum (Sorghum bicolor L.) is a major cereal grain crop of the world and It can be cultivated under high temperature and dry condition with high adaptability to the adverse environment. It is usually eaten with rice in korea and most of domestic sorghum cultivars have been developed by pure line selection and cross breeding. In prior studies, Hybrid cultivar (F1 seed) was known to improve 30-40% in yield and resist to disease and pest than pure line varieties due to hybrid vigor. Therefore, study on the hybrid sorghum is continuously necessary. In korea, hybrid sorghum cultivar have been developed, so we need to find methods seed production technology for supply and commercialization of hybrid seed cultivar. Female inbred line(A-line) and male inbred line(R-line) are needed for hybrid seed production. This study was carried out to investigate growth characteristics and yield of hybrid sorghum according to the seedling period and planting rate between female inbred line(YSA1) and male inbred line (Sodamchal, Miryang 19). When the flowering period of two inbred groups in the process of hybrid seed production is coincides, hybrid seed production is increase. The female inbred line and male inbred lines were cultivated at Daegu in 2021. Two inbred lines were sown at intervals of 3 days and 7days and were evaluated flowering period. As a result, the flowering period of the female inbred line and male inbred lines were matched at the 7days interval. To find out optimal planting rate, two inbred lines were cultivated under different plating rate (4:2, 5:2, 6:2). Yield of YSA1/Sodamchal(F1) was the highest 130(kg/10a) in the 4:2 ratio and yield of YSA1/Miryang 19 was the highest quantity of 139(kg/10a) in the 6:2 ratio. The results of this study could be helpful for hybrid Sorghum seeds production.

Genetic Diversity and Association Analyses of Chinese Maize Inbred Lines Using SSR Markers

  • We selected 68 Chinese maize inbred lines to understand the genetic diversity, population structure, and marker-trait associations for eight agronomic traits and 50 simple sequence repeats (SSRs) markers. In this study, effective traits, such as days of anthesis (DA), days of silking (DS), ear height (EH), plant to ear height ratio (ER), plant height (PH), and leaf width (LW) were divided into PC1 and PC2 by PCA analysis for maize inbred lines. Genetic diversity analysis revealed a total of 506 alleles at 50 SSR loci. The mean number of alleles per locus was 10.12. The averages of genetic diversity (GD) and polymorphic information content (PIC) values were 0.771 and 0.743, respectively. Based on a membership probability threshold of 0.80, the population structure revealed that the total inbred lines were divided into three major groups with one admixed group. A marker-trait association using Q + K MLM showed that nine SSR markers (bnlg1017, umc2041, umc2400, bnlg105, umc1229, umc1250, umc1066, umc2092, and umc1426) were related with seven agronomic traits. Among these SSR markers, eight SSR markers were associated with only one agronomic trait (DA, DS, ER, LL, LW, PH, and ST), whereas one SSR marker (umc1229) was associated with two agronomic traits (DA and ST). These results will help in optimizing the choice of inbred lines for cross combinations, as well as in selecting markers for further maize breeding programs.

  • Selection procedures for breeding genic male sterile lines for resistance to both Phytophthora blight caused by Phytophthora capsici and bacterial spot caused by Xanthomonas euvesicatoria were executed to $F_3$ and $F_4$ generations derived from a cross between a Phytophthora resistant genic male sterile (GMS) breeding line and a bacterial spot and Phytophthora resistant breeding line. Resistance to P. capsici was originally introduced from KC294(CM334) and KC263(AC2258), the well-known sources of resistance to P. capsici. Resistance to bacterial spot was introduced from KC47(PI244670). GMS lines with high resistance to P. capsici were obtained and the selected lines are expected to be quantitatively resistant also to bacterial spot.

Effect of Hybridization on Carcass Traits and Meat Quality of Erlang Mountainous Chickens

  • Native chickens hold a significant share of the market in China. In response to the huge demand from the market, the productivity of Chinese native chickens needs to be improved. Cross breeding is an effective method to increase productivity, although it might affect meat quality. In this study, two pure lines (SD02 and SD03) of Erlang mountainous chickens were hybridized with a yellow feather and faster growing line (SD01). The effect of hybridization on carcass and meat quality (physiochemical and textural traits) was measured in the $F_1$ population at d 91 of age. The hybrids exhibited higher body weight and dressed weight, and amount of semi-eviscerated, eviscerated, breast muscle and abdominal fat (p<0.05). Abdominal fat yield also increased (p<0.05) compared to the offspring of the two pure-lines. Meanwhile, there was no significant difference in meat quality traits except for the myofiber diameter and density and the shear force of the breast muscle. Overall, the offspring of cross-lines were similar to pure lines in meat color, pH value, inosinic acid, crude protein, crude fat, dry matter, moisture content and amino acid composition in the breast muscle. These results suggest that productivity can be improved via cross-breeding while maintaining meat quality of the Erlang mountainous chicken.

Preparation Method of Indigo Standard Solution and Variation of Indigo Contents in Blue Dye Extract from Breeding Lines of Persicaria tinctoria H. Gross (인디고 분석 표준액의 제조법 및 쪽 육성계통의 인디고 함량변이)

  • This study was carried out to find out the optimum method of preparation of indigo standard solution and its stability, and to investigate the indigo contents in Niram, blue dye extract, from a total of 7 indigo plants and 34 breeding lines of Persicaria tinctoria H. Gross. Proper solvent for indigo standard was dimethyl sulfoxide (DMSO), and appropriate concentration was 1 mg of indigo in 10 mL of DMSO. Absorbance value of UV/Vis Spectrophotometer at 620 nm of standard solution was changed decreasingly 12 hours after the preparation of standard solution irrespective of the storage conditions such as temperature and light. Average value of absorbance of 8-fold diluted standard solutions prepared daily during 16 days was $0.210{\pm}0.005$, indicating the powder of indigo compound was stable chemically. Calibration curve was made for quantitative analysis of indigo of 7 Niram samples, and indigo contents ranged from 0.69% to 18.76% showing relatively larger variation. Across all 34 breeding lines, the range of indigo content was from 7.9 mg to 56.4 mg per 100 g of fresh leaves, averaging 25.2 mg of indigo content and showing a 47.7% coefficient of variation.

Agronomic Characteristics of Rice Recombinant Inbred Lines (RILs) Developed from a Cross of Nonganbyeo and BG 279

  • A set of rice recombinant inbred lines was developed from a cross between a Tongil type variety, Nonganbyeo, and an indica variety, BG276, by the single seed descent method. The number of the lines in the population was 272. All the agronomic characters studied except ADV (alkali-digestion value) showed continuous variation among the RILs, implying that their inheritance mode should be quantitative. The patterns of the variation in the RILs were either normal or skewed distribution. ADVs of RILs were segregated into two groups with 1:1 ratio, indicating that ADVs in this KIL population might be controlled by one major gene. Transgressive variations were also observed in all characters. Heritability values of the characters varied from 0.488 in brown/rough rice ratio to 0.895 in alkali-digestion value. In the analysis of genotypic and phenotypic correlations, the character of yield was positively correlated with 8 different agronomic characters. The number of panicles per hill was negatively correlated with culm length, panicle length, and number of spikelets per panicle. Grain length was positively correlated with grain width, grain thickness, grain length/width ratio, white belly, ADV, and amylose. However, grain length/width ratio was negatively correlated with grain width. White core was also negatively correlated with white belly and ADV.
