To analyze the linkage relationships among molecular markers recently reported to be linked to onion (Allium cepa L.) Ms, a restorer-of-fertility locus, in onion (Allium cepa L.), three single nucleotide polymorphism markers were converted into cleaved amplified polymorphic sequence (CAPS) markers based on onion transcriptome sequences and the rice genome database. Analysis of the recombinants selected from 4,273 segregating plants using CAPS and other linked markers demonstrated the jnurf13 and jnurf610 markers to perfectly co-segregate with the Ms locus. In contrast to jnurf13, the jnurf610 marker was not in perfect linkage disequilibrium with the Ms locus in diverse breeding lines. Thus, the jnurf13 marker and the marker for identification of cytoplasm types were utilized to enhance the efficiency of onion breeding through four applications. First, 89 maintainer lines containing the normal cytoplasm and homozygous recessive Ms genotypes were successfully identified from 100 breeding lines. Second, these two molecular markers were used to analyze the main sources of male-fertile contaminants frequently found in the male-sterile parental lines during F1 hybrid seed production. The majority of the contaminants contained heterozygous Ms genotypes, indicating that pollen grains harboring the dominant Ms genotype may have been introduced during propagation of the maintainer lines. Therefore, the genetic purity of the two maintainer lines was analyzed in the third application, and the results showed that both maintainer lines contained 13-21% off-types. Finally, the two markers were used to increase the seed yield potentials of two open-pollinated varieties containing sterile cytoplasms by removing the plants harboring homozygous recessive and heterozygous Ms genotypes.
Molecular markers developed from the flanking sequences of two cytoplasmic male sterility (CMS)-associated genes, orf456 and ${\Psi}atp6-2$, have been used for marker-assisted selection of CMS in pepper. However, in practice, the presence of orf456 and ${\Psi}atp6-2$ at substoichiometric levels even in maintainer lines hampers reliable selection of plants containing the CMS gene. In this study, we developed a novel CMS-specific molecular marker, accD-U, for reliable determination of CMS lines in pepper, and used the newly and previously developed markers to determine the cytoplasm types of pepper breeding lines and germplasms. This marker was developed from a deletion in a chloroplast-derived sequence in the mitochondrial genome of a CMS pepper line. CMS pepper lines could be unambiguously determined by presence or absence of the accD-U marker band. Application of orf456, ${\Psi}atp6-2$and accD-U to various pepper breeding lines and germplasms revealed that accD-U is the most reliable CMS selection marker. A wide distribution of orf456, but not ${\Psi}atp6-2$, in germplasms suggests that the pepper cytoplasm containing both orf456 and ${\Psi}atp6-2$ has been selected as CMS cytoplasm from cytoplasm containing only orf456. Furthermore, factors other than orf456 may be required for the regulation of male sterility in pepper.

Knowledge of genetic diversity and of the genetic relationships among elite breeding materials has had a significant impact on the improvement of crops. In maize, this information is particularly useful in i) planning crosses for hybrid and line development, ii) in assigning lines to heterotic groups and iii) in plant variety protection. We have used the SSR technique to study the genetic diversity and genetic relationships among 76 Korean waxy corn accessions, representing a diverse collection from throughout Korea. Assessment of genetic diversity among members of this group was conducted using 30 microsatellite markers. Among these 30 microsatellite markers, we identified a total of 127 alleles (with an average of 4.2 and a range of between 2 and 9 alleles per locus). Gene diversity at these 30 microsatellite loci varied from 0.125 to 0.795 with an average of 0.507. The cluster tree generated with the described microsatellite markers recognized two major groups with 36.5% genetic similarity. Group I includes 63 inbred lines, with similarity coefficients of between 0.365 and 0.99. Group II includes 13 inbred lines, with similarity coefficients of between 0.45 and 0.85. The present study indicates that the 30 microsatellite loci chosen for this analysis are effective molecular markers for the assessment of genetic diversity and genetic relationships between Korean waxy corn accessions. Specifically, this study's assessment of genetic diversity and relationships between a set of 76 Korean waxy corn inbred lines will be helpful for such activities as planning crosses for hybrid and line development and association mapping analyses of maize breeding programs in Korea.

This study was conducted to evaluate tomato plant resistance against bacterial wilt by Ralstonia solanacearum using tomato cultivars or tomato breeding lines maintained in RDA-Genebank of Rural Development Administration and to select resistant tomato lines for breeding purpose. We evaluated the disease responses of a total of 13 cultivars and 39 breeding lines from RDA-Genebank using R. solanacearum SL341 strain, which is a representative strain in Korea. Tomato cultivar Hawaii 7996 and Moneymaker were used as a resistant control plant and a susceptible control plant, respectively. A total of 32 cultivars were susceptible and 10 cultivars showed various disease response suggesting resistant phenotype segregation in the lines. Five commercial cultivars and 5 breeding lines exhibited strong resistance to bacterial wilt by the SL341 strain. These 5 breeding lines might be used for further study of plant defense response against bacterial wilt and cloning of the resistance gene from tomato plants. Ultimately, the selected lines could be used for tomato breeding to generate bacterial wilt resistant tomato plants.
Cytoplasmic male sterility (CMS), one of the most important traits in crop breeding, has been used for commercial seed production by $F_1$ hybrid cultivars of pepper (Capsicum annuum L.). To develop reliable molecular markers for allelic selection of the Restorer-of-fertility (Rf) gene, which is known to be a major determinant of pollen fertility restoration in peppers, a sequence of approximately 10 kb flanking an RAPD fragment closely linked to the Rf locus was obtained by genome walking. A homology search revealed that this sequence contained an LTR retrotransposon and a non-LTR LINE-like retrotransposon. Sequencing of this Rf-linked region to search for polymorphisms between a dominant and recessive allele revealed 98% nucleotide sequence identity between them. A third polymorphic haplotype of the Rf-linked sequence, which has 94-96% nucleotide sequence identity with the two previously isolated haplotypes, was identified among a large number of breeding lines. Utilizing polymorphic sequences in the haplotypes, PCR markers were developed for selection of particular haplotypes and used to examine the distribution of the haplotypes in diverse breeding lines, cultivars, and C. annuum germplasms. Surprisingly, the third haplotype was the predominant type in C. annuum germplasms, while its frequency in $F_1$ hybrid cultivars was relatively low. Meanwhile, analysis of breeding lines whose Rf allele genotypes and male-sterility phenotypes were already known revealed that the third haplotype was mainly present in exotic breeding lines that cause unstable male-sterility when combined with sterile cytoplasms.
Lines bred for resistance to Phytophthora capsici by incorporation of resistance to P. capsici in PI201234 or CM334 into 'Subi' and 'Chilseong', land races in Youngyang, and 'Punggak', a land race in Cheongdo in Gyeungbuk province, and lines bred for fortification of one of them above with resistance to viral complex, and tolerant selections from another landrace collection from Punggak (KC268) were evaluated for resistance to P. capsici by inoculation at seedling stage. Almost all the breeding lines showed high level of resistance to P. capsid and selections from KC268 showed tolerance or moderate resistance to P. capsid. The selected plants were grown in a net cage in an outdoor nursery for seed production. Utilization of the lines in breeding was discussed.
Rye has been a major winter forage crop in Korea. Varietal improvement of rye has been practiced either by hybrid or population breeding systems. Hybrid breeding offers important advantages over population breeding since it is normally a cross-pollinated crop. The hybrid breeding in rye has been possible since cytoplasmically inherited forms of male sterility (CMS) and corresponding nuclear restorer genes were found. The objectives of this research were to develop the maintainer and restorer lines of Korean inbred lines and to estimate the effect of 'Pampa' type of CMS cytoplasm on yield and its related characteristics. For easy discrimination of male-sterile status of plants, anther scoring and the restore index system in which seed-setting and pollen quantity of viability were taken into account were established. High significant correlation between pollen quantity and pollen viability was found. For "Pampa" cytoplasm, four of 14 Korean inbred lines tested turned out to be a maintainer but no restorer was found. But for "235b" CMS cytoplasm, seven inbred lines acted as complete restorers. The Korean inbred rye lines acted mainly as maintainers in "Pampa" cytoplasm but acted mainly as restorer in "235b" cytoplasm. The 'Pampa' cytoplasm inducing male sterility reduced cohn length and plant height and increased the number of tiller, so forage yield and grain yield were enhanced. The A, C, F colonies of Apis mellifera ligustica Spin. and D, V colony of Apis mellifera caucasia Gorb. bees were collected from 2005-2007. Consequently, inbred lines were derived from the bees of original colonies by matting in the isolated island with due regard for pure breeding. This project helps in the selection of colonies with higher production capacity, aiming to improve honey and royal jelly production and breeding programs. Twenty-three standard morphological traits of honeybee were evaluated, and samples were compared with the data of the two original subspecies. The result suggested that 8 traits partly preserved in bees of inbred lines, and the bees from A. m. ligustica preserved more traits than bees from A. m. caucasia. Among the studied inbred lines, the F line is distinguished by an increase in leg parameters, considered as a favorable phenotypic trait of inbred lines. Importantly, bred of beelines in the same area can be classified as remote and isolated areas. Therefore, we observed differences of inbred lines with the origin subspecies in description acquired with morphometric characteristics as a result of adaptation, breeding, purebred individual lines used as an important resource for breeding novel cross-breeding colonies.

Manifestation of heterotic genetic interaction was studied in different hybrids made between multivoltine recurrent backcross (RBL)/congenic lines (Con. L) during unfavourable season when temperature and relative humidity are > $30^{\circ}C$ and 86%, respectively. A few number of silkworm race or strain or breed like Nistari (N + p or Np) can sustain the temperature above 3$0^{\circ}C$ and RH above 86%. The present heterosis study screened a hybrid i.e., CB$_{5}$Lm5RBL1M$_{6}$DPC-LmE$^1$RBL and its reciprocal provided heterobeltiotic effect on survival by number and pupation rate at a magnitude of 20% (p < 0.01) and yield by weight of 10% (p < 0.01). Beside all the hybrids expressed heterosis over check - Nistari (N + p) with better quality silk. Therefore, aforesaid hybrid may be useful for utilization at commercial level during adverse seasons of West Bengal.gal.
The induced genetic divergence was estimated in 44 mutant lines of finger millet variety GPU 26, developed by single and combination treatments with gamma rays, EMS and NG using three multivariate analyses. The mutant lines were grouped into eight genetically diverse clusters by multivariate D2 and canonical analyses and 11 clusters by dendrogram grouping through Gower's similarity coefficient. The clustering pattern in these three methods was almost similar. Twelve mutant lines in D2 and 13 in the dendrogram grouping method were grouped in the parental cluster(Cluster I) indicating that they did not possess enough divergence from the parent to be classified as micromutant lines. However a large proportion of mutant lines showed divergence from the parent variety and also among themselves. No definite relationship of mutagenic origin and clustering of mutant lines were observed. The mutant lines developed from the same mutagenic treatments often grouped into different clusters indicating that each mutagenic treatment was effective in inducing diverse types of changes in the nine traits studied. The hybridization program between the divergent mutant lines GE 2-2 or GE 3-4 with GG 3-1 is expected to give promising and desirable segregants in subsequent generations. Traits such as days to 50% flowering and days to maturity had major contributions to the induced genetic divergence.
