Evaluation of coat color inheritance and production performance for crossbreed from Chinese indigenous Chenghua pig crossbred with Berkshire

  • Objective: This work was to determine coat inheritance and evaluate production performance for crossbred pigs from Berkshire×Chenghua (BC) compared with Chinese indigenous Chenghua (CH) pigs. Methods: The coat color phenotypes were recorded for more than 16,000 pigs, and the genotypes of melanocortin 1 receptor (MCIR) gene were identified by sequencing. The reproductive performance of 927 crossbred BC F4 gilts and 320 purebred CH gilts was recorded. Sixty pigs of each breed were randomly selected at approximately 60 days of age to determine growth performance during fattening period, which lasted for 150 days for BC pigs and 240 days for CH pigs. At the end of the fattening period, 30 pigs of each breed were slaughtered to determine carcass composition and meat quality. Results: The coat color of BC pigs exhibits a "dominant black" hereditary pattern, and all piglets derived from boars or sows genotyped ED1 ED1 homozygous for MC1R gene showed a uniform black coat phenotype. The BC F4 gilts displayed a good reproductive performance, showing a higher litter and tear size and were heavier at farrowing litter and at weaning litter than the CH gilts, but they reached puberty later than the CH gilts. BC F4 pigs exhibited improved growth and carcass characteristics with a higher average daily live weight gain, lower feed-to-gain ratio, and higher carcass lean meat rate than CH pigs. Like CH pigs, BC F4 pigs produced superior meat-quality characteristics, showing ideal pH and meat-color values, high intramuscular fat content and water-holding capacity, and acceptable muscle-fiber parameters. C18:1, C16:0, C18:0, and C18:2 were the main fatty acids in M. longissimus lumborum in the two breeds, and a remarkably high polyunsaturated/saturated fatty acid ratio of ~0.39 was observed in the BC F4 pigs. Conclusion: The BC F4 pigs exhibit a uniform black coat pattern and acceptable total production performance.

Statistical Genetic Studies on Cattle Breeding for Dairy Productivity in Bangladesh: II. Estimation of Reciprocal and Heterosis Effects and Optimum Crossbreeding System between the Local Breeds and Exotic Breeds for Milk Performance

  • Reciprocal effects among local breeds and additive and heterosis effects were estimated for total milk production per lactation (TLP), lactation length (LL) and daily milk yield (DMY). Procedures for an optimum crossbreeding system were examined by comparing various crossbred combinations. Three data sets were chosen from the total records collected from 1962 to 1997. Data set I with a total of 5,938 records composed of 9 breed groups for reciprocal crosses, data set II with a total of 6,064 records composed of 5 breed groups for additive and heterosis effects and data set III with a total of 10,319 records composed of 42 breed groups for optimum crossbreeding system were analyzed. The least squares ANOVA was used. The reciprocal crosses between Red Sindhi and the Bangladeshi Local showed significant difference. When the Red Sindhi was used as cow, the cross was superior to the opposite in terms of the three performance traits. The results showed that additive breed effects on both TLP and DMY were positive and highly significant between Bos indicus and Bos taurus, whereas the individual heterosis effects were not significant. In general, any local breeds were improved two to three times by crossing with the Friesian once or twice. However, the effect was reduced when the grading up with the Friesian was repeated more than twice. On the other hand, when the local breed was crossed with the other breeds such as Holstein and Jersey, the improvement of TLP was less. Crossing the F1 cows with the Friesian bull gave further improvement. Almost the same trends were found also for LL and DMY. The local breeds and their crosses are raised mainly for draft animals, then it is recommended that the crossbreds between the local cows and Friesian are used for dairy purposes and more than 75% Friesian blood is used for beef.

FREQUENCY OF COTTONSEED CAKE SUPPLEMENTATION FOR YEARLING HEIFERS ON BLUE GRAMA RANGE

  • Yearling replacement heifers were used in a two-year study at the Fort Stanton Experimental Ranch near Capitan, New Mexico, USA to compare frequency (once versus three times weekly) of feeding a cottonseed cake supplement. The study involved periods of 130 days (January 4 through May 14) in 1985, and 146 days (December 4 through April 29) in 1986. In each year, supplemental periods were just before the breeding season (natural breeding in multiple-sire groups) that lasted for 75 days during first year and 60 days during second year. During the first year, all (83) heifers (Angus $\times$ Hereford) were supplemented at a level equivalent to 45 kg/head daily and during second year, all (69) heifers (Angus $\times$ Hereford, Hereford) were fed at a level equivalent to .68 kg/head daily. In the first year, heifers were sorted by initial weight into two replications, i.e., those weighing under 223 kg were placed in replicate 1 and those weighing over 223 kg were placed in replicate 2. During the second year, heifers were sorted by breed into two replications, i.e., Angus $\times$ Hereford heifers were placed in replicate 1 and Hereford heifers in replicate 2. Each year, half of the heifers in each replication were supplemented once weekly and the other half three time weekly. Weight gains of heifers were similar for the two treatments over both years of study. During the first year, weight gains for heifers supplemented once or thrice weekly (.23 vs .21 kg daily, respectively) did not differ (p>.05) and the same trend for the second year (.15 vs .16 kg daily, respectively) was observed. Similarly, breeding performance was consistent with weight gains and no difference (p>.05) between treatments was observed either year. Lower conception rates during the second tear of study (92 vs 72% for years 1 and 2, respectively) were due mainly to involvement of Hereford heifers that had an average weight of 232 kg at the start of breeding season and conception rate of 54% (averaged across both supplemental treatments). Based on the combined results of both trials, frequency of supplementation did not affect growth rate of yearling heifers nor did it influence their subsequent conception rate.

Effects of Concentrate Feeding Level after Sexual Maturity on the Growth and Breeding Performances, Blood Composition, and Ultrasonic Carcass Traits in Hanwoo Cows (성 성숙기 이후 농후사료 급여수준이 한우 암소의 성장과 번식능력, 혈액성상 및 초음파 도체성적에 미치는 영향)

  • This study was conducted to investigate the effect of low feeding level of concentrate after sexual maturity(14 month old) on the growing and breeding performances in Hanwoo cows. Eighteen cows(average body wt. 251kg) were alloted to two groups by completely randomized design, and fed a commercial formula feed by 0.8%(T1) and 1.2%(T2) of body weight for 30 months of total experimental period(feeding trial of 20 months, breeding trial of 10 months). When the results of this experiment compared to the Korean feeding standard for Hanwoo(2012), the reason of low growing performance in T1 was induced from low protein intake rather than energy. The change of body size by feeding levels was significantly appeared at chest width and body length, particularly chest width of cows in T1 was rather reduced by 3.7% in the final than the initial measurement. Ovarian size of T1 was only 63% of T2, and pregnancy rate(33.3% : 77.8%) and birth wight of calf(22.5kg : 29.5kg) were also seriously repressed by low intake of dietary protein after sexual maturity in Hanwoo cow. It was appeared that the growing and breeding performances of Hanwoo cows were more affected by the level of dietary protein rather than energy in the feeding after sexual maturity.

Effect of Oxygen and Temperature Levels on the Seedling Characteristics of Korean and Anaerobic Germination-tolerant Rice under Flooding Conditions (담수 직파에서 산소 및 온도조건에 따른 혐기발아 내성 자원과 국내 직파 적응성 벼 품종의 입모특성 비교)

  • The aim of the present study was to compare the germination and seedling characteristics of rice varieties grown under various flooding conditions and different temperature and oxygen levels and to identify germplasm suitable for wet direct seeding. Three anaerobic germination tolerant (AGT) genotypes (PBR, WD3, KHO) and eleven Korean rice varieties (KVs) adapted for direct seeding were evaluated for seedling performance under different temperatures (15, 18, 21, and 24℃ ) and oxygen levels (Low, Normal, High). Compared with the KVs, the AGT genotypes (especially KHO and PBR) exhibited relatively high germination and survival rates and coleoptile and radical growth rates under low temperature and low oxygen conditions, thereby indicating their suitability for wet direct seeding. Among the KVs, 'Dongan,' 'Jungan,' and 'Cheongdam' rice exhibited the highest survival rates under low temperature and low oxygen conditions. Three-way ANOVA indicated that temperature had the greater effects on seedling characteristics (43.2-78.0%) than either oxygen level (15.4-37.5%) or genotype (2.0-29.8%) did. Therefore, in direct seeding cultivation, temperature was the most important environmental factor for seedling establishment.

MicroRNA analysis reveals the role of miR-214 in duck adipocyte differentiation

  • Objective: Fat deposition in poultry is an important factor in production performance and meat quality research. miRNAs also play important roles in regulating adipocyte differentiation process. This study was to investigate the expression patterns of miRNAs in duck adipocytes after differentiation and explore the role of miR-214 in regulating carnitine palmitoyltransferases 2 (CPT2) gene expression during duck adipocyte differentiation. Methods: Successful systems for the isolation, culture, and induction of duck primary fat cells was developed in the experiment. Using Illumina next-generation sequencing, the miRNAs libraries of duck adipocytes were established. miRanda was used to predict differentially expressed (DE) miRNAs and their target genes. The expression patterns of miR-214 and CPT2 during the differentiation were verified by quantitative real-time polymerase chain reaction and western blot. Luciferase reporter assays were used to explore the specific regions of CPT2 targeted by miR-214. We used a miR-214 over-expression strategy in vitro to further investigate its effect on differentiation process and CPT2 gene transcription. Results: There were 481 miRNAs identified in duck adipocytes, included 57 DE miRNA candidates. And the 1,046 targets genes of DE miRNAs were mainly involved in p53 signaling, FoxO signaling, and fatty acid metabolism pathways. miR-214 and CPT2 showed contrasting expression patterns before and after differentiation, and they were selected for further research. The expression of miR-214 was decreased during the first 3 days of duck adipocytes differentiation, and then increased, while the expression of CPT2 increased both in the transcriptional and protein level. The luciferase assay suggested that miR-214 targets the 3'untranslated region of CPT2. Overexpression of miR-214 not only promoted the formation of lipid droplets but also decreased the protein abundance of CPT2. Conclusion: Current study reports the expression profile of miRNAs in duck adipocytes differentiated for 4 days. And miR-214 has been proved to have the regulator potential for fat deposition in duck.

A Simple Method for Evaluation of Pepper Powder Color Using Vis/NIR Hyperspectral System (Vis/NIR 초분광 분석을 이용한 고춧가루 색도 간이 측정법 개발)

  • Color is one of the quality determining factors for pepper powder. To measure the color of pepper powder, several methods including high-performance liquid chromatography (HPLC), thin layer chromatography (TLC), and ASTA-20 have been used. Among the methods, the ASTA-20 method is most widely used for color measurement of a large number of samples because of its simplicity and accuracy. However it requires time consuming preprocessing steps and generates chemical waste containing acetone. As an alternative, we developed a fast and simple method based on a visible/near infrared (Vis/NIR) hyperspectral method to measure the color of pepper powder. To evaluate correlation between the ASTA-20 and the visible/near infrared (Vis/NIR) hyperspectral methods, we first measured the color of a total of 488 pepper powder samples using the two methods. Then, a partial least squares (PLS) model was postulated using the color values of randomly selected 3 66 samples to predict ASTA values of unknown samples. When the ASTA values predicted by the PLS model were compared with those of the ASTA-20 method for 122 samples not used for model development, there was very high correlation between two methods ($R^2=0.88$) demonstrating reliability of Vis/NIR hyperspectral method. We believe that this simple and fast method is suitable for highthroughput screening of a large number of samples because this method does not require preprocessing steps required for the ASTA-20 method, and takes less than 30 min to measure the color of pepper powder.

Difference in Black-tailed Gull (Larus crassirostris) diet during the breeding season for the last 10 years in the South Sea of Korea

  • Sea temperature in the South Sea of Korea has been increased over the last decades. Seabirds are sensitive to changes in food availability in marine environment. In this study, we investigate the diet of Black-tailed gulls (Larus crassirostris) during the breeding season to identify changes of marine environment and biological response such as breeding performance in the South Sea of Korea. A total of 22 fish species or family (n = 128) from regurgitates by chicks were collected on Hongdo Island in 2002 and 2012. The most important prey item was Japanese anchovies (Engraulis japonicas). Proportion of Anchovy in diet increased in 2012 (70.5%) compared to 10 years ago (27.5%). Some species were newly found in 2012: Spotted chub mackerel (Scomber australasicus), Pacific sand lace (Ammodytes personatus), White ventral goby (Acanthogobius lactipes), Silver-strip round herring, Multicolorfin rainbowfish (Halichoeres poecilopterus), Silverside (Hypoatherina tsurugae), Surfperch (Neoditrema ransonneti) and Spotnape ponyfish (Leiognathus muchalis), but not in 2002. Especially, sub-tropic fish such as Kammal thryssa (Thryssa kammalensis), and Rosefish (Helicolenus hilgendorfi) were frequently observed in the diet of 2012. These results might reflect the increase of sea temperature in the South Sea of Korea.

Shield Material Consideration in the LAR Tokamak Reactor

  • For the optimal design of a tokamak-type reactor, self-consistent determination of a radial build of reactor systems is important and the radial build has to be determined by considering the plasma physics and engineering constraints which inter-relate various reactor systems. In a low aspect ratio (LAR) tokamak reactor with a superconducting toroidal field (TF) coil, the shield should provide sufficient protection for the superconducting TF coil and the shield plays a key role in determining the size of a reactor. To determine the radial build of a reactor, neutronic effects such as tritium breeding in the blanket, nuclear heating, and radiation damage to toroidal field (TF) coil has to be included in the systems analysis. In this work, the outboard blanket only is considered where tritium self-sufficiency is possible by using an inboard neutron reflector instead of breeding blanket. The reflecting shield should provide not only protection for the superconducting TF coil but also improved neutron economy for the tritium breeding in outboard blanket. Tungsten carbide, metal hydride such as titanium hydride and zirconium hydride can be used for improved shielding performance and thus smaller shield thickness. With the use of advanced technology in the shield, conceptual design of a compact superconducting LAR reactor with aspect ratio of less than 2 will be presented as a viable power plant.

Effects of Stubble Height, Irrigation and Nitrogen Fertilization on Rice Ratooning in Korea

  • Rice (Oryza sativa L.) ratooning is the production of a second rice crop from the stubble left behind after the main crop harvest. The objectives of this study were to evaluate the effects of main-crop stubble height, irrigation and fertilization on ratoon grain yield. Main crop 'Jinbuol' rice cultivar was harvested to leave with 10, 20, 30, or 40 cm stubble height. When the main crop stubble was harvested with 10 cm height, ratoon rice grain yield was increased by 2,810 kg/ha. Irrigation on stubbles after main crop harvest did not affect the ratoon crop yield and rice quality. The results showed a large variation in the ratoon performance by fertilizer application methods. Top-dressed nitrogen fertilizer on the stubble after harvest caused increase in panicle production and higher maturity rate. However, there was no significant difference in protein content, amylose content of milled rice and cooked rice characters between plots managed with and without nitrogen fertilizer.
