Nitrogen Balance in Goats Fed Flemingia (Flemingia Macrophylla) and Jackfruit (Artocarpus Heterophyllus) Foliage Based Diets and Effect of a Daily Supplementation of Polyethylene Glycol (PEG) on Intake and Digestion

  • Diets with foliage of Flemingia (Flemingia macrophylla) or Jackfruit (Artocapus heterophyllus were fed to goats with the objective to study nitrogen (N) balance and effect of a daily supplementation of polyethylene glycol (PEG) on intake and digestion. In experiment 1, three male Alpine${\times}$Jamnapary goats with initial weights varying from 26.9 to 27.7 kg were used in a $3{\times}3$ Latin square design in the dry season. Three Alpine${\times}$Bachthao crosses, 15.3-16.7 kg, were used in the same design in the wet season. The three diets were based on chopped whole sugar cane complemented with the two green foliages, Jackfruit and Flemingia, or soybean meal (SBM). The level of dry matter (DM) offered was 4% of body weight (BW), 2.7% as foliage and 1.3% as chopped whole sugar cane. The amount of SBM offered was calculated to give the same amount of crude protein (CP) as the foliages. Each experimental period lasted 32 days (14 days for adaptation, 7 days for collection and 10 days for rest). Feed intake, apparent digestibility of DM, organic matter (OM), CP, neutral detergent fiber (NDF) and acid detergent fiber (ADF) and retained nitrogen (N) were measured by total faecal and urine collection. In experiment 2, four male goats (Alpine${\times}$Jamnapary) with initial weights from 17.1 to 23.1 kg were used in a $4{\times}4$ Latin square design. The four treatments were Jackfruit or Flemingia with or without addition of PEG, which was fed at a level of 5 g/goat and day by mixing with a small amount of rice bran. Each experimental period lasted 15 days (8 days for adaptation, 7 days for collection). Measurements were done as in experiment 1. The DM digestibility was highest (65.9-74.3%) for goats fed the SBM diet in both the dry and wet season. The DM digestibility of goats fed the Jackfruit and the Flemingia diets was similar in both the dry (58.6-59.2% respectively) and the wet season (53.9-56.1% respectively). The CP digestibility was highest (73.0-73.6%) for the SBM diet followed by the Jackfruit diet (47.0-38.5%) and was lowest (36.8-30.0%) for the Flemingia diet in both dry and wet seasons, respectively. The NDF digestibility was low for both the Jackfruit (36.4%) and Flemingia (38.0%) diets in the wet season. All diets resulted in a positive N balance. The N retention was highest (0.465-0.604 g/kg $W^{0.75}$) in the SBM diets and lowest (0.012-0.250 g/kg $W^{0.75}$) in the Flemingia diet. Addition of PEG had no effect on feed intake for any of the diets. PEG added in the Flemingia diet had a positive effect only on NDF digestibility, but the digestibility of the Jackfruit diet was significantly increased. Supplementation with PEG reduced digestibility and N retention of Flemingia, possibly because of the low tannin level, but increased digestibility and N retention for Jackfruit foliage.

Effects of body weight and fiber sources on fiber digestibility and short chain fatty acid concentration in growing pigs

  • Objective: The study was conducted to determine the effects of body weight (BW) and fiber sources on nutrient digestibility, fiber fermentation and short chain fatty acids (SCFA) concentration in different intestinal segments of growing pigs fed high-fiber diets. Methods: Nine barrows with initial BW of 25.17±0.73 kg and 9 barrows with initial BW of 63.47±2.18 kg were allotted to a duplicate 9×2 Youden Square design with 3 dietary treatments and 2 periods. The dietary treatments were formulated with 3 different high-fiber ingredients: corn bran, sugar beet pulp, and soybean hulls, respectively. Each diet was fed to 3 barrows with different stage of BW in each period. Results: There were no differences in the apparent ileal digestibility (AID) of most nutrients between pigs at different BW stages. Pigs at 60 kg had greater (p<0.05) apparent total tract digestibility (ATTD) of total dietary fiber (TDF), soluble dietary fiber (SDF) and insoluble dietary fiber (IDF), and had greater (p<0.05) hindgut disappearance of IDF and cellulose than pigs at 25 kg. The acetate, propionate and total SCFA concentrations in ileal digesta and feces of pigs at 60 kg were greater (p<0.05) than those of pigs at 25 kg. In addition, fiber sources affected (p<0.05) the AID of gross energy (GE), organic matter (OM), ether extract (EE), crude protein, SDF and hemicellulose, the hindgut disappearance and ATTD of dietary fiber components, the lactate and propionate concentrations in ileal digesta and the butyrate, valerate and total SCFA concentrations in feces. There were interactions (p<0.05) between BW and fiber sources on the AID of GE, OM, EE, SDF, hemicellulose, the ATTD of EE, TDF, and IDF, and the hindgut disappearance of SDF and hemicellulose. Conclusion: Increasing BW mainly improved the digestibility of dietary fiber fractions, and the dietary fiber sources influenced the digestibility of almost all the dietary nutrients in growing pigs.

병아리 사료에서 일반대사에너지와 순대사에너지의 이론적 정확성 비교를 위한 실험 (Feeding Trials to Compare Theoretical Accuracy between Apparent and True Metabolizable Energy Systems in Chick Diets)

  • True metabolizable energy(TME) is believed a better indicator for animal performance than apparent metabolizable energy (AME) for excluding the endogenous energy losses from excreta, However few researches have been conducted to compare superiority of any energy systems through practical animal feeding tests. Present study was to compare the energy systems in young chicks in terms of predictability of energy intake for the birds performances including body energy retention and of methodological accuracy by evaluating reproducibility and additi-vity of energy values of feed ingredients and compound diets. Five ingredients such as yellow corn wheat soybean meal fish meal and wheat bran were measured for their various biological energy values. in the first feeding trial chicks were restric-ted-fed the basal diet at 80, 60 and 40% on weight basis of the amount of feed ingested by chicks fed ad libitum the same diet. chicks in the second trial were also restricted-fed diets at levels of 80, 70, 60 and 50% on energy basis of the amount consumed by the basak duet group fed ad libitum The diets in the latter trial were however composed of differeent formulations from the basal diet. One-week-old Single Comb White Leghorn male chicks were individually alloted in a cage on 10 cages/treatment basis and fed the diets for 14 days. Individual carcass energy was measured after the feeding trials. Coefficients of variation of energy measurements were lesser for nitrogen-corrected AME and TME(AMEn & TMEn respectively) than AME and TME values suggesting taht reprodu-cibility of energy determinations by former systems could be better than the latters. The coeffi-cients for AME and TME were almkost of the same values. Additivity obtained by the rations between the calculated values and catual measurements appeared quite satisfactory for all the energy systems. Those of AME and TME however were relatively better than the other systems. Regression coefficient ${r}^2$ between energy intake by various systems and chick performances appeared higher for TME, AMEn and TMEn than AME implying that the former systems could provide better predictability for body weight gain and energy retention than the AME. The ${r}^2$ values for TME and AMEn particularly for body weight gain were on the average 0.967 and 0.960 respectively. In conclusion TME or AMEn can be recommended as choice for dietary energy system in terms of performance predictability of the birds and of procedural convenience for the measurements.

Effects of Dietary Non-phytate Phosphorus Levels on Egg Production, Shell Quality and Nutrient Retention in White Leghorn Layers

  • An experiment was conducted (28 to 44 weeks) to study the laying performance, shell quality, and nutrient retention of White Leghorn layers fed different levels of non-phytate phosphorus (NPP). Six levels of NPP (0.15, 0.18, 0.21, 0.24, 0.27 and 0.30%) at a constant calcium (Ca) level (3.5%) in maize-soya-deoiled rice bran based diets were formulated, and each experimental diet was offered ad libitum for 16 weeks to five replicates with five birds in each replicate. The body weight of WL layers fed diet containing 0.15% NPP was significantly (p<0.05) lower than those fed diet with 0.30% NPP, at 44 weeks of age. However, the hen day egg production, egg weight, daily feed intake and feed consumed per dozen eggs were not influenced by the variation in the NPP levels in the diet. The bone ash content was significantly (p<0.05) higher in the birds fed 0.30% NPP as compared with those fed diets up to 0.24% NPP. Bone ash content was intermediate in the birds fed diet containing 0.27% NPP. The tibia strength followed the same trend as that of bone ash. Dietary NPP content had no influence on serum Ca and protein concentration and activity of alkaline phosphatase. However, serum inorganic P concentration increased linearly with NPP content in the diet. The concentration of P was significantly (p<0.05) higher in the birds fed 0.27% NPP or higher as compared with those fed 0.15% NPP. Levels of dietary NPP had no influence on egg quality parameters like shell wt, shell thickness, shell strength and specific gravity. The retention of nutrients such as DM, N and Ca were comparable among the WL layers fed different levels of NPP. However, the retention of P decreased linearly with increase in the level of NPP in the diet. The retention of P in the birds fed diets up to 0.24% NPP in the diet was comparable, however further increasing the content of NPP (either 0.27% or 0.30%) reduced the retention of P. Based on the results of the present study, 0.15% NPP (180 mg/b/d) in the diets of WL layers is adequate for optimum production performance during 28 to 44 weeks of age, however, WL layers require 0.27% NPP (324 mg /b/d) in the diet for optimum production with better bone mineralization.

Comparison of Indigenous Browses and Sunflower Seed Cake Supplementation on Intake and Growth Performance of Dual-purpose Goats Fed Buffel Grass (Cenchrus ciliaris) Hay

  • A study to compare the effects of supplementing Delonix elata, Grewia similis, Tamarindus indica and sunflower seed cake on intake and growth rate of dual-purpose goats fed low quality Buffel grass (Cenchrus ciliaris) hay was carried out. Twenty-eight male goats aged five to seven months (mean weight 12.93${\pm}$3.94 kg) were randomly allocated to four dietary groups in a completely randomised design. The diets were hay plus Grewia similis, hay plus Delonix elata, hay plus Tamarindus indica and hay plus sunflower seed cake. All diets were supplemented with maize bran. The experimental period was 90 days. Voluntary dry matter intake of the supplements was higher for Tamarindus indica (275.5 g/day) and Grewia similis (201.8 g/day) and lowest for sunflower seed cake (81g/day). Goats supplemented with Grewia similis had the highest hay intake (183.8 g/day) while those supplemented with sunflower seed cake had the lowest hay intake (98.9 g/day). Animals fed browse supplements gained significantly more weight (p<0.001) than those with sunflower seed cake. There were no significant differences in live weight change between goats fed the different browses. However, those fed Tamarindus indica gained an average of 20.79 g/d which was slightly higher than the gains for those on Grewia similis and Delonix elata while those fed sunflower seed cake lost weight. Correspondingly, goats supplemented with browse leaf meals had higher feed conversion ratios than those supplemented with sunflower seed cake and required 23.91 to 35.06 g DM of feed to produce one g of weight gain per day. In a separate study, the DM disappearance pattern indicated that Grewia similis and Delonix elata were highly degradable compared to Tamarindus indica. At 24 h of incubation, DM degradability was 627, 588 and 345 g/kg DM for Grewia similis, Delonix elata and Tamarindus indica, respectively. In another study in vivo DM digestibility ranged from 46.1% (for hay alone) to 56.2% (for hay plus Grewia similis). It was concluded that the addition of Tamarindus indica, Grewia similis and Delonix elata leaf meals to Cenchrus ciliaris hay resulted in increased total DM intake, in vivo digestibility and growth rate. Therefore, leaf meals of indigenous browses particularly Tamarindus indica and Grewia similis could be used as supplementary feeds for small ruminants grazing on poor quality roughages during the dry season rather than use of expensive, less effective and intermittently available sunflower seed cake.

Ensiling Techniques for Shrimp By-Products and their Nutritive Value for Pigs

  • An experiment was performed to evaluate different methods for preserving shrimp by-products and to determine their chemical composition. In the first experiment three ratios of shrimp by-product (SBP) to molasses (6:1, 4:1 and 3:1, wet weight), and to cassava root meal (3:1, 2:1 and 1:1, wet weight of shrimp by-product and air-dry weight of cassava root meal) were investigated. The pH of the SBP ensiled with molasses at a ratio of 3:1, and with cassava root meal at a ratio of 1:1, decreased during the first week to below 4.5 and remained low up to day 56 of ensiling, whereas the pH of the mixtures with higher ratios of SBP remained above 7.0, and the material deteriorated rapidly. The dry matter decreased initially in all treatments but then increased slightly from day 28 in the treatment where shrimp by-product was ensiled with cassava root meal at a ratio of 1:1. The crude protein (CP) and ammonia-N $(NH_3-N)$ contents of the preserved shrimp by-product material ensiled with molasses at a ratio of 3:1 increased significantly one week after ensiling. The CP content then remained constant, while the $NH_3-N$ concentration continued to increase up to 56 days after ensiling. When SBP was ensiled with cassava root meal at a ratio of 1:1 the CP content of the silage increased significantly up to 21 days after ensiling and then decreased back to the original level after 56 days, whereas $NH_3-N$ increased markedly up to 14 days and then remained fairly constant up to 56 days. However, the $NH_3-N$ content was significantly higher when SBP was ensiled with cassava root meal than with molasses. A balance experiment was carried out, arranged as a double Latin-square and including 6 F1 (Large White ${\times}$ Mong Cai) castrates fed randomly one of three diets based on cassava root meal, rice bran, and fish meal (FM) or shrimp by-product ensiled with molasses (SBEMO) or with cassava root meal (SBECA) as the main protein source. Apparent organic matter and CP digestibilities were significantly (p<0.001) higher for the fish meal diet than for the two shrimp by-product diets, although CP digestibility in SBEMO and SBECA was similar (p>0.05). N-retention was significantly higher for the fish meal diet than for the SBEMO diet, which in turn was significantly higher than for the SBECA diet (p<0.01). It can be concluded that shrimp by-product can be preserved by ensiling with molasses at a ratio of 3:1 or with cassava root meal at a ratio of 1:1. Nutrient digestibility and N-retention of diets based on these shrimp by-product silages were lower than for similar diets based on fish meal, probably due to their high chitin content and inferior amino acid balance.

Effects of L-carnitine, Selenium-enriched Yeast, Jujube Fruit and Hwangto (Red Clay) Supplementation on Performance and Carcass Measurements of Finishing Pigs

  • Fifty castrated crossbred ($Landrace{\times}Yorkshire$) pigs, weighing an average of $60.6{\pm}3.1kg$ were allotted to one of five treatments in a randomized block design to examine the effects of dietary inclusion of 0.1% L-carnitine (50 ppm carnitine), 0.1% selenium-enriched yeast (0.3 ppm selenium), 0.1% Jujube fruit or 0.1% Hwangto (Red clay) on pig performance and carcass quality. All diets were based on corn, wheat, soybean meal and wheat bran and were formulated to supply 13.8 MJ DE/kg. Dietary supplementation did not influence daily gain (p = 0.57), feed intake (p = 0.52), or feed conversion (p = 0.32). Digestibility of dry matter (p = 0.60), organic matter (p = 0.74), crude protein (p = 0.76), crude fibre (p = 0.70) and energy (p = 0.75) were also unaffected by inclusion of any of the additives. Tissue samples taken from the longissimus muscle showed that the levels of carnitine (p = 0.0001) and selenium (p = 0.0001) were significantly higher with dietary inclusion of carnitine or selenium-enriched yeast. Dietary treatment did not affect dressing percentage (p = 0.33), carcass lean yield (p = 0.99) or first, $10^{th}$ and last rib midline backfat depth (p = 0.45, 0.82 and 0.47, respectively). Dietary treatment also did not affect the percentages of tenderloin (p = 0.37), bacon (p = 0.36), fat and bone (p = 0.56), picnic shoulder (p = 0.25), skirt (p = 0.80), fresh ham (p = 0.31) or ribs (p = 0.79). However, pigs fed the diet containing Jujube fruit had a higher percentage of Boston butt than pigs fed the carnitine or selenium supplemented diets (p = 0.01). Pigs fed added Hwangto had a higher (p = 0.04) percentage of loin compared with pigs fed supplementary selenium or Jujube fruit. Loin muscle from pigs fed carnitine had a significantly lower Hunter colour value for L (whiteness, p = 0.004) and a higher value for $a^*$ (redness; p = 0.069). The overall results indicate that supplementation with L-carnitine and selenium-enriched yeast can produce pork containing higher levels of carnitine and selenium, which could provide health benefits for consumers of pork without detrimental effects on pig performance.

원형질체(原形質體) 융합(融合)에 의한 느타리와 여름느타리버섯의 종간(種間) 교잡(交雜) (Interspecific Hybridization between Pleurotus ostreatus and Pleurotus sajor-caju by Protoplast Fusion)

  • 느타리 Pleurotus ostreatus와 여름느타리 Pleurotus sajor-caju의 영양요구주로부터 분리한 원형질체를 polyethylene glycol로 융합하여 종간 이질핵체 heterokaryon를 획득하였다. 불화합 균주간에는 9 융합조합에서 70개 융합주를 획득하여 7 융합주의 자실체를 유도하였으며 화합 균주간에는 1조합에서 모두 자실체를 형성하였다. 화합 균주간 융합주를 제외하고는 불화합성간 융합체는 꺽쇠연결체 clamp connections가 없었고 한천배지나 액체배지에서는 자실체를 형성하지 않았다. 그러나 활엽수톱밥과 미강이 혼합된 배지에서 균사가 완전히 성장한후 일정한 광과 온도를 처리한 결과 꺽쇠연결체 있는 균사가 다시 성장하였으며 이들 균사에서 버섯자실체가 형성되었다. 버섯자실체는 양친의 중간형태로 갓 색깔도 양친이 혼합된 orange grey-brownish grey로 나타났다. 그러나 다소의 자실체 형질인 자실층 hymenium, 버섯대, 생장습성은 느타리를 닮았는데 특히 여름느타리가 단생 closely scattered인데 비해 느타리는 속생 caespitose으로 느타리와 유사하였다. 자실체는 모두 꺽쇠연결체를 가졌으며 조직배앙한 결과 균사가 모두 꺽쇠연결체를 가져 본래 융합주와는 다른 형태로 변하였다. 6융합조합 8균주의 $F^2$에서의 유전형질 분리와 유전자 재조합을 분석한 결과 원영양형 prototroph이 양친 느타리형, 여름느타리형, 영양요구성 유전자 재조합형에 비해 많이 분리되었으며 3개조합에서 양친에 없는 유전자좌가 나타났다. 자실체가 형성되지 않는 융합주 P188과 사철느타리 ASI 2-3-rib와 균사접합 한 후 임성을 유도하여 유전분석한 결과 원영양형이 적게 분리되었고 여름느타리 양친형이 분리되지 않았으며 3친주 형질이 모두 유전자 재조합형으로 분리되었다.

담자균류(擔子菌類)에 의(依)한 섬유질류(纖維質類) 분해(分解)에 관(關)한 연구(硏究) (Decomposition of Fiber by the Basidiomycetes)

  • 담자균(擔子菌) Pleurotus ostreatus, Pholiota nameko, Flammulina velutipes가 생산(生産)하는 각종(各種) 효소(酵素)에 의(依)한 톱밥류와 농산물 폐물인 고구마줄기, 옥수수대 및 보리짚의 영양사료(榮養飼料)에 관하며 검토(檢討)한 결과(結果)는 다음과 같다. 1. 활엽수, 포플라 및 밤나무의 톱밥과 고구마줄기 옥수수대 및 보리짚의 비율을 $42{\sim}49$ 대(對) 30으로 조절(調節)하고 쌀겨 $18{\sim}25$의 비율로 배합한 기질(基質)에서 수분함량(水分含量) $67{\sim}70%$, 온도(溫度) $24{\sim}26^{\circ}C$가 발육(發育)에 적합(適合)하였다. 2. 20일간(日間) 각시험구(各試驗區)에서 생장(生長)시킨 후(後) 원료기질중(原料基質中)의 섬유를 조사(調査)한 바, $47{\sim}50%$ 분해(分解)하여 다당류화(多糖類化)시켰고 리그닌은 $20{\sim}25%$ 분해(分解)하였고 단백질함량은 처음의 약 3배량(倍量)이었다. 3. 조(粗)섬유의 농후사료화(濃厚飼料化)가 가능(可能)하였다.

beta-Galactosidase의 고정화 및 응용에 관한 연구 제1보: Aspergillus niger CAD 1의 효소생산 조건 및 효소학적 성질 (Studies on immobilization and application of beta-galactosidase I. Conditions for production and properties of the enzyme from Aspergillus niger CAD 1)

  • 토양에서 분리한 여러가지 곰팡이류 가운데 beta-galactosidase 생성력이 가장 좋은 Aspergillus niger CAD 1을 효소 생산 균주로 선정 하였다. 이 균주의 효소 생산 최적 배양조건은 밀기울에 0.5% 탈지 분유를 첨가한 배지를 $30{\circ}C$에서 72시간 정치배양하는 것이었다. 아세톤으로 침전시킨 조효소(crude enzyme)는 일차로 DEAE-cellulose, 2차로 Sephadex G-100 gel filtration에 의하여 1,387배로 정제되었고 이때의 수율은 6.2%이였다. 정제된 효소의 최적 온도는 $45{\circ}C$ 최적 pH는 4.5이었으며, 기질로서 ONPG와 유당에 대한 Km값은 각각 $3.57{\times}10^3M$$83.3{\times}10^3M$, Vmax값은 각각 33.0 unit/mg protein과 15.38unit/mg protein이었다. 활성화 에너지는 9,900cal/mol이었으며 효소활성 및 안정제로 금속이온을 필요로 하지 않았다. 50ml의 탈지유, 4.8%유당 용액, 유청용액에 효소 5ml(182 unit/ml)를 침가하여 $45^{\circ}C$에서 10시간 반응시켰을 때의 유당 분해율은 각각 65%, 70%, 78%를 나타내었다.
