Detection of BLV Proviral DNA in Korean Native Goats Experimentally Infected with Bovine Leukemia Virus by Polymerase Chain Reaction (소백혈병 바이러스 (Bovine Leukemia Virus)에 감염된 한국 재래산양에서 PCR기법을 이용한 BLV 유전자 검출)

  • PCR amplication using the primers for gag, pol and env genes in BLV (bovine leukemia virus) proviral DNA and syncytium assay were carried out for the Korean native goats experimentally infected with bovine leukemia virus to investigate pathogenesis of BLV in the goats, and to establish a model animal for BLV infection. The oligonucleotide primers used in PCR revealed very high specificity. The minimal amount of FLK-BLV cellular chromosomal DNA to detect the integrated BLV proviral DNA was 10 ng. The peripheral blood lymphocytes from the goat infected with BLV were examined at regular intervals by PCR amplification and syncytium assay. Pol or gag genes were detected in none of three infected goats at the 1st week post-infection (p.i.). At the 4th week p.i., one of three goats showed the amplified gag gene. Thereafter detection rates for the genes were increased, indicating that the BLV proviral genes were integrated in all of the lymphocytes from three goats, at the 16th weeks p.i., when it was evident in syncytium assay that the lymphocytes from all of three goats were infested with infective BLV. Investigating the tissues from the necropsied goats at the 8th month p.i., the amplified BLV proviral genes and infective BLV were detected in all of the peripheral lymphocytes from three infected-goats. Among various tissues examined, the amplified BLV proviral genes were observed in spleen and superficial cervical, mandibular and retropharyngeal lymph nodes, and the infective BLV, in superficial cervical and mandibular lymph nodes. It was assumed that the Korean native goat was quite susceptible to BLV infection, indicating that the goat could be a good model animal for BLV.

  • Nitric oxide (NO) is a simple combined molecule of oxygen and nitrogen, and has a wide variety of action on the physiological and pathophysiological function of the body. It is a key transducer of the vasodilator message from the endothelium to vascular cells. However, its different roles have been elucidated by numerous researches, which was undertaken in the 80's and 90's. Three types of NO synthase were involved in synthesizing NO and they are identified in different tissues and cells including macrophage, endothelial cells and even tumor cells. In the late 90's, we undertook a number of researches for elucidating the effect of NO on embryo development, since developmentally arrested bovine embryos contained large amount of NO metabolites in their cytoplasm. Subsequently, we found that the addition of a spontaneous NO donor to culture medium markedly inhibited embryo development and that its inhibitory role was independent of embryonic genome activation. Research was focused to find a way to prevent the inhibitory action of NO on embryo development and demonstrated that the addition of hemoglobin, a NO scavenger, to embryo culture medium greatly stimulated in vitro-development of bovine and mouse embryos. Based on these research outcomes, we developed a NO action-free culture system for embryos and other tissues. The efficacy of such system has subsequently been confirmed by achieving the high rates of preimplantation development and blastocyst formation in the NO action-free culture of mouse and bovine embryo. In this article, we briefly introduced the nature of NO and our research outcomes on the role of NO in embryo development.

Studies on the Specific Gene Expression in Lung Tissue of the Cloned Hanwoo (복제한우 폐조직에서 특이 유전자 발현에 관한 연구)

  • This study was conducted to investigate the specific expression genes in the cloned bovine tissues. Donor cells, cloned tissues were analysed by RAPD-RFLP method. The results were detected three genes (CH-U7B, CH-U7M and CH-U7P) in the cloned fetus. It was found a single copy genes by southern hybridization. Sequence analysis of CH-U7M gene was shown 99% homology to a previously reported EST from a cloned bovine fetus. The putative ORF was encode a protein of hydrophobicity index 0.03. Semi-quantitative RT-PCR by using the CH-LS001 specific primer was remarkably detected in the lung tissue of cloned fetus. Further investigation of these genes may provide one of the key information to explain the early death, abnormal fetus, large off-spring and the low pregnancy rate in the production of cloned bovine.

  • Enzootic bovine leukosis(EBL) is chronic disease caused by bovine leukemia virus(BLV), retroviridae. The characteristic feature of this disease is proliferation of lymphocytes in circulating blood or lymphoid tissues. Because EBL concern lymphocytes, immunological disorder or alteration in the lymphocyte subpopulation is suggested. In this study, we investigated the changes of the lymphocyte subpopulation in the circulating blood of Korean native cattle infected with bovine leukemia virus. (omitted)

Molecular Characterization of Porcine DNA Methyltransferase I

  • During normal early embryonic development in mammals, the global pattern of genomic DNA methylation undergoes marked. changes. The level of methylation is high in male and female gametes. Thus, we cloned the cDNA of the porcine DNA methyltransferase 1 (Dnmt1) gene to promote the efficiency of the generation of porcine clones. In this study, porcine Dnmt1 cDNA was sequenced, and Dnmt1 mRNA expression was detected by reverse transcription-polymerase reaction (RT-PCR) in porcine tissues during embryonic development. The porcine Dnmt1 cDNA sequence showed more homology with that of bovine than human, mouse, and rat. The complete sequence of porcine Dnmt1 cDNA was 4,774-bp long and consisted of an open reading frame encoding a protein of 1611 amino acids. The amino acid sequence of porcine DNMT1 showed significant homology with those of bovine (91%), human (88%), rat (76%), and mouse (75%) Dnmt1. The expression of porcine Dnmt1 mRNA was detected during porcine embryogenesis. The mRNA was detected at stages of porcine preimplantation development (1-cell, 2-cell, 4-cell, 8-cell, morula, and blastocyst stages). It was also abundantly expressed in tissues (lung, ovary, kidney and somatic cells). Further investigations are necessary to understand the complex links between methyltransferase 1 and the transcriptional activity in cloned porcine tissues.

Development of Calcification-resistant Bovine Pericardium with PEO-$SO_3(II)$ -An implantation study of bovine pericardium at artery and peritoneum- (PEO-$SO_3$를 이용한 항석회화 조직첨포의 개발(II) -동맥과 복막 이식 실험연구-)

  • Background: Calcific degeneration limits durabilities of the bioprosthetic tissues implanted in the human body. The direct coupling sulphonated polyethyleneoxide(PEO-SO3) to the bioprosthetic tissues after glutaraldehyde(GA) fixation and the removal of residual aldehyde groups from the tissues can augment the effect of calcification-resistance. Materials and methods: To study the anti-calcification effect by PEO-SO3 modification and the removal of the residual aldehyde groups of tissues, surface modified bovine pericardia(BP-PEO-SO3) were preserved in aseptic saline to wash out GA(saline group) and 0.65% GA solution(GA group). And then above two groups and PERIGUARD (Bio-vascular. Co.) (product group) were evaluated with respects to calcium contents and microscopic findings using in vivo implantation models at carotid and femoral artery and peritoneum of 8 adult dogs. Results: In the tissues retrieved from carotid artery, calcium content was significantly decreased in saline group than in other two groups(saline; 2.89±0.31 vs. GA; 6.14±1.08 vs. product; 22.82±5.00 mg/g of dried tissue; p<0.05). In the tissues retrieved from femoral artery and peritoneum, calcium amount was also decreased in saline group than in other two groups, but not reached the significant difference between groups. On the other hand, the pathologic findings of pericardial tissues showed marked destructuction in GA group compared to the other two groups. Conclusions: In this study, covalently PEO-SO3 bound to bovine pericardium decreased calcifications and the anti-calcification effect of BP-PEO-SO3 could be augmented by the washing out the residual aldehyde groups using saline after GA fixation. Conclusively, the PEO-SO3 modified bovine pericardium is highly resistant to calcification and can be useful for the development of calcification-resistant cardiovascular patches and valves.

Hemorrhagic disease caused by bovine viral diarrhea virus-2a in Korean Indigenous Cattle: case reports

  • Two 1-year-old Korean native steers in the same herd presented severe hemorrhagic diarrhea. Case 1 had severe dehydration and died after 3 days, whereas case 2 had anorexia, depression, and severe diarrhea with mucus and blood. Only case 2 was necropsied, and bovine viral diarrhea virus-2a (BVDV2a) was detected in the tissues of its alimentary tract. Gross lesions, including erosion, ulceration, and extensive hemorrhage, were observed in the digestive tract mucosa. Immunohistochemistry revealed marked positive staining for BVDV2a antigen in the large intestine. These findings are indicative of hemorrhagic disease caused by BVDV2a in a native Korean steer.

Anticalcification Treatment of Glutaraldehyde-fixed Bovine Pericardium with Amino Acids (The Effect of Ethanol, Glutamic Acid and Homocysteic Acid Treatment) (글루타르알데하이드로 고정한 소 심낭의 아미노산을 이용한 항석회화 처리(에탄올, 글루타믹 산, 호모시스테익 산 처리의 효과))

  • Background: Glutaraldehyde-fixed heterografts are prone to calcification after long-term implantation in human, and this is one of the limiting factors for the longevity of the heterografts used in cardiovascular surgery. The aim of the study was to evaluate the anticalcification effect of an ethanol and amino acids treatment on glutaraldehyde-fixed bovine pericardium. Material and Method: Bovine pericardial tissues were divided into 5 groups. Group 1 consisted of tissues fixed with glutaraldehyde, group 2 consisted of commercially available bovine pericardial valve tissues (Carpentier-Edwards PERIMOUNT), group 3 consisted of glutaraldehyde-fixed tissues treated with ethanol, group 4 consisted of glutaraldehyde-fixed tissues treated with ethanol and L-glutamic acid, and group 5 consisted of glutaraldehyde-fixed tissues treated with ethanol and homocysteic acid. The tissue microstructure was examined by light and electron microscopy. Tissue samples of each group were implanted into rat subcutaneous tissue for 3 $\sim$ 4 months and the calcium contents were measured after harvest. Result: The collagen fibers appeared to be well preserved in all the groups. The calcium contents of groups 2, 3, 4 and 5 (13.46$\pm$11.74, 0.33$\pm$0.02, 0.39$\pm$0.08 and 0.42$\pm$0.06 $\mu$g/mg, respectively) were all significantly lower than that of group 1 (149.97$\pm$28.25 $\mu$g/mg) (p<0.05). The calcium contents of groups 3, 4 and 5 were all significantly lower than that of group 2 (p<0.05). Conclusion: Treatment with ethanol alone or in combination with amino acids (L-glutamic acid or homocysteic acid) strongly prevented the calcification of glutaraldehyde-fixed bovine pericardium.

An Integrated Genomic Resource Based on Korean Cattle (Hanwoo) Transcripts

  • We have created a Bovine Genome Database, an integrated genomic resource for Bos taurus, by merging bovine data from various databases and our own data. We produced 55,213 Korean cattle (Hanwoo) ESTs from cDNA libraries from three tissues. We concentrated on genomic information based on Hanwoo transcripts and provided user-friendly search interfaces within the Bovine Genome Database. The genome browser supported alignment results for the various types of data: Hanwoo EST, consensus sequence, human gene, and predicted bovine genes. The database also provides transcript data information, gene annotation, genomic location, sequence and tissue distribution. Users can also explore bovine disease genes based on comparative mapping of homologous genes and can conduct searches centered on genes within user-selected quantitative trait loci (QTL) regions. The Bovine Genome Database can be accessed at http://bgd.nabc.go.kr.

  • The frequency distribution and morphological characteristics of tissue mast cells in the various tissues of normal domestic animals (bovine, swine, dog and chicken) and systematically infected (Hog cholera, Canine distemper and Newcastle disease) animals were studied. The results were as follows: 1 In cattle, density of mast cells was higher in foetus (bovine) and young animals than in adults Differences in frequency distribution among individual animals were also observed. 2. In chicken, the highest number of mast cells was found in age group of 15 to 40 days, the moderate number in age group of one to 10 days, and the lowest number in age group of 40 days or older. 3. The morphologically, mast cells were usually round, ovoid, spindle, oval and irregular, and particularly in ovary of bovine, it was usually round and ovoid forms. 4. The largest numbers of mast cells were shown in ovary of bovine, intestine of swine and dog, and proventriculum of chicken. 5. In the systemic infections, the number of mart cells usually tends to increase.
