Histopathological Study on Teeth, Jawbone, and Oral Tissues in Poisoned Rabbit (독극물중독 가토의 치아 및 악골조직의 병리조직학적 변화에 관한 연구)

  • The authors administered KCN, NaF,AS2O3 orally to rabbits and caused acute and chronic poisoning, then studied the teeth, jaw bones, and other oral tissues histopathologically. The results were as follows : 1. There was no significant difference between acute poisoned group by NaF and control group. But, vasodilatation in the connective tissues, esepcially marginal area of jaw bone, atrophy and destrution of glandualr cells was observed. 2. Chronic poisoned group by NaF showed degeneration and thicking of subcutanece fibrosis ective tissues, atrophy and degeneration of subcutaneous connective tissues, atrophy and degeneration of muscle fibers, vasodilation of subcutaneous in bone cavities(lacunae), and degeneration of odotlblasts in pulp tissue. 3. Acute poisoned group by KCN showed almost similar appearances as control group, and chronic poisoned group showed hyperplasia of baal layer in epitheilium, degeneration of subcutaneous connective tissues, vasodilation and huperemia, severe hemorrhage of marginal area of jaw bone. hyperplasia of salivary gland ducts, but normal arrangement of muscle fibers and narrow bone carity(lacunae) due to active osteoblastic action, osteodentin were observed. 4. Acute poisoned group by AS2O3 showed degeneration of basal cell, atrophy of blood vessels in palatal muscosa. Chronic poisoned group showed irregular cell arrangement and degeneration, reduction of capillaries in palatal mucosa. Osteoclasts in jaw bone were observed. 5. In Masson's Trichrome and Van Gieson Staining, chronic poisoned group by NaF showed thicking and loosening of subcutaneous connective tissues. Hyperplasia of intermuscular connective tissue was observed in chronic poisoning by KCN and NaF. In PAS staining, negative reation in outer layer of palatalmucosa, positive reaction in keratin layer and mild reaction of basal layer in palate and tongue mucosa was observed.

A STUDY OF HOLOGRAPHIC INTERFEROMETRY ON THE INITIAL REACTION OF MAXILLOFACIAL COMPLEX TO THE INTERMAXILLARY FORCES ON THE ORTHODONTIC ARCHWIRES (교정용 호선에 악간 교정력 적용시 악안면골의 초기반응에 관한 Holographic Interferometry 연구)

  • This study was performed to evaluate the initial reaction of maxillofacial complex to the Class II intermaxillary and the anterior vertical elastic forces on the six types of archwires including multiloop edgewise arch wires(MEAW). A human dry skull was used for this purpose and this investigation was done by holographic interferometry. Based on such investigation, the fringe pattern and the number of fringes of each condition were compared and analyzed. The findings of this study were as follows: 1. As the orthodontic forces increased, the amount of displacement increased. 2. As the orthodontic forces were applied, the fringes were shown not only in the teeth and the maxilla but also in the adjacent bones, i.e., temporal bone, zygomatic bone, nasal bone, frontal bone and sphenoid bone. And the direction of fringe pattern and the number of fringes were different from each other by the sutures. 3. As the long Class II elastic forces were applied, the backward-downward displacements of the anterior teeth and the maxilla were shown, and backward displacement of the former were grater than those of the latter. And backward displacements were greater by the long Class II elastic forces than by the short Class II elastic forces. 4. As the anterior vertical elastic forces were applied, downward displacements of the anterior teeth and the maxilla were shown, and the downward displacements of the former were greater than those of the latter relatively. 5. The downward displacements of the anterior area to the anterior vertical elastic forces of the MEAW were greater than those of other archwires. In addition, the more tip-back bend was applied, the more displacement was seen. 6. As the Class II intermaxillary forces and the enough anterior vertical elastic forces were applied on the MEAW with tip-back bend, there was an intrusive effect of the posterior teeth.

Enlarged Lipogranuloma after Closed Reduction of Nasal Bone Fracture: A Case Report (비골골절 정복술 후 커진 지방육아종의 치험례)

  • Purpose: Lipogranuloma is the reaction of adipose tissue to various oils, paraffin, and other hydrocarbons injected into subcutaneous tissue for cosmetic or other reasons. The authors experienced a case of sclerosing lipogranuloma on the nasal dorsum. Methods: A 42-year-old female, without a history of the injection of any foreign materials, was admitted on our hospital for a painless, irregular, and firm mass located on her nasal dorsum with step-off deformity. It was considered that the mass had developed after augmentation rhinoplasty. The size of mass had been increased after closed reduction of nasal bone fracture. On April 2011, under general anesthesia, the mass was removed by open rhinoplasty technique. In addition, a pathologic examination was performed. After the mass extirpation, dermofat graft was performed for the correction of depression deformity. Results: The histopathological findings demonstrated a Swiss cheese pattern with variably-sized vacuoles, which corresponded to lipid removed with tissue processing, and variable foreign body giant cell reaction, fat necrosis, and hyalinized fibrous tissue. The pathologic diagnosis is lipogranuloma replacing nasalis muscle. It has been considered that sclerosing lipogranuloma is caused by nerve injury during augmentation rhinoplasty and the ointment used after the closed reduction of nasal bone fracture, which infiltrated through the injured mucosa. Conclusion: During the treatment of rhinoplasty or nasal bone fracture, the nerve injury or the ointment use can lead to lipogranuloma. Therefore, careful dissection for avoidance of the nerve injury and limited use of ointment seems to be helpful in decreasing incidence of lipogranuloma.

Periodontal tissue reaction to customized nano-hydroxyapatite block scaffold in one-wall intrabony defect: a histologic study in dogs

  • Purpose: This study evaluated histologically the tissue responses to and the effects of a customized nano-hydroxyapatite (n-HA) block bone graft on periodontal regeneration in a one-wall periodontal-defect model. Methods: A customized block bone for filling in the standardized periodontal defect was fabricated from prefabricated n-HA powders and a polymeric sponge. Bilateral $4{\times}{\times}4{\times}5$ mm (buccolingual width${\times}$mesiodistal width${\times}$depth), one-wall, critical-size intrabony periodontal defects were surgically created at the mandibular second and fourth premolars of five Beagle dogs. In each dog, one defect was filled with block-type HA and the other served as a sham-surgery control. The animals were sacrificed following an 8-week healing interval for clinical and histological evaluations. Results: Although the sites that received an n-HA block showed minimal bone formation, the n-HA block was maintained within the defect with its original hexahedral shape. In addition, only a limited inflammatory reaction was observed at sites that received an n-HA block, which might have been due to the high stability of the customized block bone. Conclusions: In the limitation of this study, customized n-HA block could provide a space for periodontal tissue engineering, with minimal inflammation.

A study on the preparation of phosphatic calcium compounds using the shell resources (패각을 이용한 인산칼슘계 화합물의 제조에 관한 연구)

  • The phosphatic calcium compounds such as calcium hydrogen phosphate, bone ash, hydroxyapatite and tricalcium phosphate were prepared using the high purity calcium hydroxide and calcium carbonate obtained from shell resources. Calcium hydrogen phosphate had been prepared using the high purity calcium hydroxide and phosphoric acid solution. Using the calcium hydrogen phosphate as a starting materials, bone ash have been prepared by solid state reaction method and hydroxyapatite could be obtained by hydrothermal treatment method, respectively. The tricalcium phosphate was prepared by the solid state reaction of a stoichiometic mixture of bone ash and high purity calcium carbonate. In this paper, the optimal preparation process and conditions of phosphatic calcium compounds were established.

Anti-Inflammatory Activity of Constituents Isolated from Ulmus davidiana var. japonica

  • Twenty six compounds (1-26) were isolated from the root barks of Ulmus davidiana var. japonica. The anti-inflammatory activity of the isolated compounds were evaluated agai nst the generation of inflammatory chemical mediators in bone marrow-derived mast cells. Among them, compounds 10, 11, 13, 15 and 19 inhibited not only cyclooxygenase-2 dependent prostaglandin $D_2$ generation but also 5-lipoxygenase dependent leukotrien $C_4$ generation in a concentration-dependent manner. In addition, compounds 11, 12, 13, 15 and 19 also inhibited $\beta$-hexosaminidase release, a marker of mast cell degranulation reaction, from bone marrow-derived mast cell. These results suggest that the anti-inflammatory activity of U. davidiana might in part occur by both the inhibition of eicosanoid generations and the degranulation reaction of mast cells.

Effect of Storage Conditions on the Setting Properties of Brushite Bone Cement Containing Granular β-Tricalcium Phosphate (과립형 β-삼인산칼슘을 사용한 Brushite계 골시멘트의 보관 조건에 따른 경화 특성)

  • In the bone cement composed of dense granules of $\beta-Ca_3(PO_4){_2}(\beta-TCP)$ and $Ca(H_2PO_4){_2}H_2O$, the compressive strength, setting time and temperature rise were measured to observe the degradation of cement with respect to the stored days before setting. Decreases of compressive strength and temperature rise were observed, while setting time increased with respect to the stored days. The similar trends were repeated with the increase of temperature of storage. Such a change virtually meant the fading of the character of cement and it took place only when the two starting materials were mixed during storage. The degradation could be mitigated taking advantage of granular $\beta$-TCP instead of powdery one. The formation of $CaHPO_4$, which resulted from reaction with ambient humidity, was attributed to the degradation observed during storage. Dependence of the degradation behavior on mixing and temperature during storage was discussed in terms of the driving force for reaction of cement.

ON THE BONE TISSUE REACTION TO IMPLANTS WITH DIFFERENT SURFACE TREATMENT METHODS (임플랜트 표면 처리 방법에 따른 골조직 반응에 대한 연구)

  • Statement of problem: Implant surface characteristics plays an important role in clinical success and many studies have been made for improvement of success by changing surface roughness. Purpose: Appropriate increase of surface roughness increases the activity of osteoblast and enhance contact and retention between bone and implant. Material and method- Machined, SLA and RBM surface implants, which are the most commonly used implants were implanted into the tibia of rabbits and after 1 week, 4 weeks, 8 weeks and 12 weeks there were histologic and histomorphometric analysis and study for bone gradient and change of Ca/P ratio using EDS(Energy Dispersive X-ray Spectroscope). Results: Comparison of bone-implant contact showed no significant difference among each implant. In comparison of bone area rates, SLA showed higher value with significant difference at 1 week and 4 weeks, and SLA and RBM at 8 weeks than Machined implant (p<0.05). In analysis of bone constituents with EDS, titanium was specifically detected in new bones and the rates were constant by surface treatment method or period. In case of Ca/P ratio, according to surface treatment method, each group showed significant difference. Lots of old bone fragments produced during implantation remained on the rough surface of RBM implant surface and each group showed histological finding with active synthesis of collagen fibers until 12 weeks. In transmission electronic microscopic examination of sample slice after elapse of twelve weeks, tens nm of borderline (lamina limitans like dense line)was seen to contact the bone, on the interface between bone and implant. Conclusion: SLA and RBM implant with rough surface shows better histomorphometrical result and the trend of prolonged bone formation and maturation in comparison with Machined implant. In addition, implant with rough surface seems to be helpful in early stage bone formation due to remaining of old bone fragments produced in implantation. From the results above, it is considered to be better to use implant with rough surface in implantation.

AN EXPERIMENTAL STUDY ON TISSUE RESPONSE FOLLOWING IMPLANTATION OF THE FREEZE - DRIED AND RADIATION - STERILIZED ALLOGENEIC BONE IN RECTUS ABDOMINICUS MUSCLE (백서 복근에 매식된 동결 건조 및 방사선 멸균처리된 동종골의 조직 반응에 관한 실험적 연구)

  • In this study, the healing changes of the implanted bone and its surrounding tissues were examined on the histopathologic basis following implantation of the freeze - dried and radiation - sterilized allogeneic bone in Rectus abdominicus of the rat. This study was performed to see the tissue recations after implantation of the freeze - dried and radiation - sterilized allogeneic bone and whether osteogenesis or osteo - induction or osteo - conduction is happened. And the results were as follows : 1. The shape of the implanted allogeneic bone of the 1, 2 - week group specimen was similar to that of normal bone in light - microscopic finding and the atrophy of cellular organells was found in trans - mission electron - microscopic finding. 2. The implanted allogeneic bone was surrounded with the dense fibroconnective tissues, and infiltration of the chronic inflammatory cells gradually became increased. 3. Hyaline degeneration was observed in the surrounding tissue at the 3, 4, 6 - week group specimen. 4. Light - microscopically the resorption of implanted bone became prominent after 4 - week group and the necrosis of allogeneic bone implant became severe with loss of cell components in lacuna. 5. Electron - microscopically, the osteoclast - like cells ere fond after, 2 - week group. It is summarized that the osteo - conduction potential of the bone is remained just after implanting the freeze - dried and radiation - sterilized allogeneic bone on Rectus abdominicus of the rat, but gradually it disappeared with the gradual increse of chronic inflammatory reaction and osteoclastic activity. So it is suggested that the antigenicity of the freeze - dried and radiation - sterilized bone is remained and it has little osteo - conductive activity when it is implanted in the muscle.

Lipopolysaccharide and Prostaglandin $E_2$ regulates the gene expression of IL-1$\beta$ in mouse osteoblast cells

  • Prostaglandin $E_2$(PG$E_2$) is an abundant eicosanoid in bone that has been implicated in a number of pathological states associated with bone loss, and is also known to stimulate matric metalloproteinase-1 systhesis and secretion in rat and human osteoblast cells, although the intracellular reaction remain unclear. Interleukin-1$\beta$ (IL-1$\beta$) is a cytokine that plays a critical role in bone remodelling and appears to act as a downstream effector of most bone-resorbing agents. However, it is still interesting to examine whether PG$E_2$ regulates IL-1$\beta$ expression by mouse osteoblasts or not. Here we demonstrate that PG$E_2$is a potent inducer of IL-1$\beta$ production by fetal osteoblasts.
