이소플라본이 난소절제 쥐에서 골밀도와 골무기질 함량에 미치는 영향 (Effects of Isoflavones on Bone Mineral Density and Bone Mineral Content in Ovariectomized Rats)

  • A recent study reported that a diet rich in isoflavones is beneficial for bone formation in growing rats. It therefore seemed desirable to find out whether the beneficial effect of isoflavones in ovariectomized rats could also be reproduced with same amount of isoflavones which used for growing rats. To study the effect of isoflavones, an equal amount of isoflavones which used for growing rats, on bone mineral density and bone mineral content in ovariectmized rats were performed. Forty female Sprague-Dawley rats (body weight $210{\pm}5g$) were divided into two groups, ovariectomy and sham groups, which were each randomly divided into two subgroups that were fed casein and casein supplemented with isoflavones diets for 9 weeks after operation. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin) in spine and femur. Serum alkaline phosphatase activity (ALP) and osteocalcin and urinary DPD crosslinks value were measured as markers of bone formation and resorption. The results of this study indicate that body weight gain and food intake were higher in ovariectomy groups than in sham groups regardless of diets. Serum Ca concentration was lower in ovariectomy groups than in SHAM groups. Serum ALP, osteocalcin, and crosslink value were increased in ovariectomy groups. Spine BMD/weight, femur BMD/weight, and femur BMC/weight of ovariectomy groups were significantly lower than SHAM groups after 9 weeks. However, isoflavones supplemented group in ovariectomy groups, serum ALP and osteocalcin concentrations, spine BMD/weight and spine BMC, femur BMD/weight and femur BMC/weight were significantly increased after 9 weeks. In conclusion, the beneficial effect of isoflavones on bone in ovareiectomized rats was shown on 9 weeks after feeding with an equal amount of isoflavones supplementation which used for growing rats.

Effects of taurine supplementation on bone mineral density in ovariectomized rats fed calcium deficient diet

  • Taurine supplementation has been shown to have a beneficial effect on femur bone mineral content in ovariectomized rats. It therefore seemed desirable to find out whether the beneficial effect of taurine on ovariectomized rats fed calcium deficient diet could also be reproduced. Forty female Sprague-Dawley rats were divided into two groups, One group was OVX and the other group received sham operation (SHAM), and received either control diet or a taurine supplemented diet for 6 weeks. All rats were fed on calcium deficient diet (AIN-93: 50% level of calcium) and deionized water. Bone mineral density (BMD) and bone mineral content (BMC) were measured in spine and femur. The serum and urine concentrations of calcium and phosphorus were determined, Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations, Bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Urinary calcium and phosphorus excretion, osteocalcin in blood and cross link value were not significantly different among the groups. Within the OVX group, the taurine supplemented group had not higher femur bone mineral content than the control group. This study established the need for a study on the taurine effect on bone with different calcium levels.

실내·외 운동 환경의 차이가 노년기 여성의 골밀도와 신체구성에 미치는 영향 (Effects of Indoor and Outdoor Exercise Environments on Bone Mineral Density and Body Composition in Old Women)

  • 본 연구는 실내 외 운동 환경의 차이가 노년기 여성의 골밀도와 신체구성에 미치는 영향을 규명하는데 그 목적이 있다. 연구대상은 65세 이상 노년기 여성으로 실내운동집단 13명, 실외운동집단 13명 두 그룹으로 나누었으며, 20주간 운동프로그램을 실시하였다. 운동강도는 주3회, 1회 60분 동안 HRmax 50~70%, RPE 11~15로 점진적으로 증가시켰다. 결과는 다음과 같다. 첫째, 골밀도의 결과 집단 내 변화에서는 실외운동집단에서 유의하게 증가하였고, 집단 간 변화에서는 실외운동집단이 실내운동집단보다 유의하게 증가하였다. 둘째, 신체구성의 결과 집단 내 변화에서는 실내운동집단에서 체지방량이 유의하게 감소하였고, 실외운동집단에서 체중, 체질량지수, 체지방량이 유의하게 감소하였다. 집단 간의 변화에서는 체질량지수에서 실외운동집단이 실내운동집단보다 유의하게 감소하였다. 이와 같은 연구 결과들을 살펴볼 때 20주간 실외운동환경에서의 운동프로그램은 골밀도와 신체구성에 긍정적인 영향을 미친 것으로 나타났다.

이소플라본의 투여가 골질량이 감소된 저체중과 정상체중 여대생의 골밀도 및 골대사 지표에 미치는 영향에 관한 연구 (A Study of Soy Isoflavone Supplementation Effect on Bone Mineral Density and Bone Metabolism Markers in Female College Students with Low Bone Mass)

  • To investigate the effects of isoflavone supplementation on both bone mineral density and hormone variation in premenopausal women who had decreased bone mass, the 24 subjects were divided into two groups: one was the underweight group, consisting of 13 subjects, and the other was the normal weight group consisting of 11 subjects. For each group, we investigated the effects of isoflavone supplementation of 90 mg/day on both bone mineral density and hormone variation during 3 menstrual cycles. Anthropometric measurements, dietary recall, and analyses of blood and urine were assessed from baseline to post-treatment. The results were as follows: The average age of the underweight group was 21.8 years old and that of the normal weight group was 23.2 years old. The comparative results for the two groups at baseline were as follows: Onset of menarche, menstrual cycle, and menstrual length were not significantly different between the groups. Serum protein, total, HDL-, LDL-cholesterol, triglyceride, Ca, P, Mg, Cu, and Zn level were not significantly different between the groups. Serum estradiol, SHBG, LH, and FSH level were also not significantly different between the groups. Lumbar spine BMD by T scores of the underweight group was significantly lower than that of the normal weight group. Serum osteocalcin, urinary DPD, and urinary pH were not significantly different between the groups. The comparative results for the two groups at post-treatment were as follows: From baseline to post-treatment, the intake of energy, nutrients and isoflavone in food did not significantly change in either group. Serum protein, total cholesterol, HDL-, LDL-cholesterol, and triglyceride levels did not significantly change in either group. Serum Ca, Cu, and Zn levels were significantly lower in both groups and serum Mg level significantly decreased only in the underweight group. Serum estradiol levels were significantly lower in both groups, but serum SHBG, LH, and FSH levels did not significantly change in either group. Lumbar spine BMD by T score of the underweight group significantly increased to 15%, but that of the normal weight group did not significantly change. Serum osteocalcin of the underweight group significantly increased to 28%, while that of the normal weight group significantly increased to 40%. Urinary DPD of the normal weight group significantly increased to 12%. The results show that the BMD of the underweight group was lower than that of the normal weight group. Therefore, the underweight group had a disadvantage in obtaining maximum bone mineral density. The results also show that isoflavone supplementation during 3 menstrual cycles was effective in increasing the bone mineral density of the lumbar spine and affected bone metabolism markers in premenopausal underweight women. Therefore, it can be concluded that sufficient intake of isoflavone could be helpful in preventing decreases in bone mass among premenopausal women, especially underweight women.

채식과 일반식 폐경 후 여성의 식물성 에스트로겐 섭취와 골밀도와의 관련성 (The Study of Pytoestrogen Intake and Bone Mineral Density of Vegetarian and Nonvegetarian Postmenopausal Women)

  • There is some evidence that phytostrogen plays an important role in bone metabolism in postmenopausal women. In this study, we investigated the phytoestrogen intake levels and the relation between dietary phytoestrogens intake and bone mineral density of vegetarian (n = 77) and omnivore postmenopausal women (n = 122 . Vegetarian women, all of them were seven day adventists, who had been on vegetarian diet (almost lacto-ovo vegetarians;a few vegans) oyer 20 yrs. The average age of vegetarians and omnivores were 62.3 yrs and 60.2 yrs, respectively and, there was no significant difference. However, body weight (p < 0.001), body mass index (p < 0.001) of vegetarians were significantly lower than those of omnivores. The mean daily energy intake of vegetarians and omnivores were 1386.1 kcal (76.3% of RDA) and 1424.5 kcal (76.9% of RDA), respectively. The mean calcium intake of vegetarians (456.7 mg, 66.3 % of RDA) was not significantly different from that of omnivores (453.5 mg, 65.2 % of RDA). The mean daily isoflavones (daidzein +genistein) intake of vegetarians and omnivore were 33.9 mg and 23.9 mg, respectively. The vegetarians consumed significantly greater quantities of isoflavones (p < 0.05) and lignans precursor (p < 0.05). In the vegetarians, intake of isoflavones was significantly positively correlated with BMD of femoral neck, after adjusted for age and BMI. Also lignan precursor intake of vegetarians was significantly positively correlated with BMD of spine, after adjusted for age and BMI. In conclusion, in omnivore post menopausal women, intake of phytoestrogen such as isoflavones and lignans was little low, and it is not clear that positive association with bone mineral density. But in vegetarian postmenopausal women, phytoestrogen intakes be important factors related to bone mineral density.

골밀도와 수냉증과의 상관성 연구 (A Study on the Correlation between of Bone Mineral Density and Cold Hypersensitivity of Hands)

  • Objectives The purpose of this study was to examine the relationship between cold hypersensitivity and bone mineral density. Methods 28 outpatients who visited OO University Oriental Hospital from July 11th, 2013 to August 28th, 2013 were analyzed. Patients were subjected to thermometer, and those with thermal difference greater than 0.3 ℃ between upper arm and palm and also with more than VAS 4 of cold hypersensitivity were diagnosed with cold sensitivity. Lumba spine (L2-4) and heel bone measurement were obtained on dual energy X-ray absorptiometry (DEXA) and Quantitative Computed Tomography (QCT). Results 1. There was statistically significant correlation between T-score of DEXA and body weight (0.522), BMI (0.643), OSTA (Osteoporosis elf-assesment Tool for Asians, 0.579), BMI, T-score and Z-score of QCT. OSTA and T-score of QCT (0.521) also had statistically significance. 2. There was no statistically significant correlation between body fat, abdominal obesity rate, degree of cold hypersensitivity, upper arm and palm's temperature difference, palm temperature, height and Z-score, T-score of QCT or T-sore of DEXA. Conclusions Bone mineral density was lower in women who had cold hypersensitivity of hand and feet. But there was no statistically significant correlation between bone mineral density and degree of cold hypersensitivity, upper arm and palm’s temperature difference.

  • This study was performed to determine the effect of isoflavone on bone mineral density and bone mineral content in growing male rats. Twenty male, Sprague-Dawley rats were assigned to groups, that underwent 9 weeks of experi-ental treatment. Animals were assigned to one of two diet groups (casein group or casein supplemented with isoflavones). During 9 week of the study, food consumption was determined every other day through the measurement of total food given subtracting the food uneaten from original amount given. Rats in two experimental groups had similar initial body weights. At the end of experiment, however, the casein group had significantly greater body weights compared to casein supplemented with isoflavones group. It was also observed that the casein group had greater food intake comared to casein supplemented with isoflavones group. The difference in the final body weights of the groups was therefore due to difference in amount of food ingested, but could be due to the effect of isoflavones. Total BMD, spine BMD, and spine BMC per weight and femoral BMD per weight were significantly greater in casein supplemented with isolaones group than casein group. ALP and osteocalcin were significantly greater in the casein-fed group. Crosslink value was significantly lower in the casein supplemented with isoflavones group, All other variables were statistically similar between two groups. Overall, it can be concluded that casein supplemented with isoflavones beneficial for acquisition of bone mineral density and content on growing male rats.

Effects of isoflavone supplementation on the bone mineral density of growing female rats

  • This study was focused on whether or not isoflavones affect the increase in bone mineral density of growing females. Female Sprague-Dawley rats ($60{\pm}5\;g$) were randomly assigned to two groups and provided control diets (control group) or isoflavone-supplemented diet (IF group, 57.8 mg isoflavones/100 g diet) for 9 weeks in growing female rats. Measurements of Bone Mineral Density (BMD) and Bone Mineral Content (BMC) on the experimental animals were executed in the $3^{rd}$, $6^{th}$, $9^{th}$ weeks. In result, there was no significant difference in spine BMD between the isoflavones supplemented group and the control group. But, the IF group tended to have higher BMD than the control group in between 3 and 9 experimental weeks, and the striking difference could be shown in the $6^{th}$ week of feeding. In case of femur BMD, the effects of added isoflavones appeared in the $6^{th}$ week of feeding, and it became intensified in the $9^{th}$ week of feeding to the extent that the BMD in the IF group was significantly higher than that of the control group (p<0.05). In conclusion, isoflavone supplementation increased spine BMD per weight in the $6^{th}$ week of feeding, and affected the increase of femur BMD in the $9^{th}$ week. The result of the experiment implies that it affects positively the formation of spine and femur BMD of growing female rats. The study also suggests that the effects of isoflavone on the pattern of BMD formation might differ from the parts of bones.

난소절제 쥐에서 카페인 첨가식이가 골밀도 및 골함량에 미치는 영향 (Effects of Caffeine on Bone Mineral Density and Bone Mineral Content in Ovariectomized Rats)

  • 본 연구는 0.03%의 카페인 함유 식이를 폐경모델인 난소절제쥐에서 6주간 섭취시켜 골밀도와 골무기질함량에 미치는 영향을 아래와 같이 요약하였다. 1) 체중증가량은 난소절제군이 Sham군에 비해 유의적으로 높았으며 각 군내에서 카페인 섭취에 따른 차이는 없었다. 2) 혈 중 칼슘 농도는 난소절제군내에서 카페인군이 대조군보다 유의적으로 낮게 나타났다. 3) 혈 중 ALP는 Sham군과 난소절제군 모두에서 카페인 군이 대조군보다 높은 경향을 나타내었다. 혈중 Osteocalcin은 Sham 군과 난소절제군, 그리고 각 군내에서 카페인 섭취여부에 따라 유의적인 차이를 나타내지 않았다. 4) 요 중 칼슘 및 인의 농도, 요 중 Deoxypyridinoline(DPD)와 crosslinks value는 Sham 군과 난소절제군, 그리고 각 군내에서 카페인 섭취여부에 따라 유의적인 차이를 나타내지 않았다. 5) 척추골밀도는 Sham군에 비해 난소절제군이 유의적으로 낮았고, 난소절제군내에서 카페인 첨가군과 대조군간에 차이가 없었다. 6) 대퇴골밀도와 대퇴 골무기질 함량은 Sham군과 난소 절제군 간의 차이는 없었고, 각 군내에서 식이에 따른 차이도 없었다. 따라서 카페인 0.03% caffeine 섭취는 난소절제쥐에서 6주간 섭취 시킨 경우 척추와 대퇴골밀도에 부정적 영향을 미치지 않는 것으로 나타났다.

Effects of Rubus coreanus-Cheonggukjang on Bone Mineral Density and Bone Mineral Content in Growing Rats

  • The purpose of the present study was to investigate the bone-conserving effects of Rubus coreanus-Cheonggukjang (RC-CGJ) supplemented with more intensified phytochemicals compared to general Cheonggukjang (CGJ) in growing rats. Eighteen rats were divided into 3 treatment groups (Control, CGJ, and RC-CGJ) and were given experimental diets for 9 weeks. All of the rats in this study were fed a AIN-93G-based diet. Both CGJ groups were fed with 33.1% CGJ and RC-CGJ powder, respectively. The results of this study indicate that weight gain, mean food intake, and food efficiency ratio were not significantly different by the experimental diets among all groups. Spine bone mineral density (BMD) and femur BMD were not significantly different by the experimental diets. Spine bone mineral content (BMC) was significantly higher in the RC-CGJ and CGJ groups than in the control group, regardless of CGJ type. The femur BMC of the CGJ supplemented group was significantly higher compared with the control group and the RC-CGJ group. Compared with the control group, spine BMD and femur BMD per weight were markedly increased in the RC-CGJ and CGJ group regardless of CGJ type. Also, spine BMC per weight was significantly higher in the RC-CGJ group than in the CGJ group. However, femur BMC per weight was significantly higher in the CGJ group than in the RC-CGJ group. It can be concluded that RC-CGJ and CGJ supplemented diets have more beneficial effects on spine and femur peak bone mass in growing rats.
