Synnemata Production by Isaria tenuipes using Colored Cocoon Silkworm, Golden Silk

  • This study was conducted to test the use of colored cocoon silkworm, Golden Silk, as a host for the fruiting body production of Cordyceps mushroom. Golden Silk variety had a shorter larval period than Yangwonjam at $25.3^{\circ}C$ and 78.5% RH: The average larval period of the Golden Silk was 24 days and 7 hr, and that of the Yangwonjam was 24 days and 15 hr. In the infection test of I. tenuipes on silkworm, the larval period of normal silkworms was longer than the silkworms inoculated with I. tenuipes in both Golden Silk and Yangwonjam. The pupae survival percentage of Golden Silk was 94.4%, whereas it was 91.9% in Yangwonjam. Golden Silk had a higher pupation rate than Yangwonjam. The pupation rate of normal silkworms was about 5% higher than that of silkworms inoculated with I. tenuipes. Infection rate of I. tenuipes into larvae of 5th instar newly exuviated silkworm was 93.8% in Golden Silk, and 96.9% in Yangwonjam. Synnemata production of I. tenuipes was better in Yangwonjam with an incidence rate of 99.8% than Golden Silk with 98.6%. The average synnemata weight of Golden Silk was 1.12 g and that of Yangwonjam 1.29 g. Golden Silk had a lower synnemata weight than Yangwonjam. But no significant difference was observed in synnemata weight varieties. The synnemata of I. tenuipes produced on pupae were white or milky-white in color, and similar in shape and color to wild synnemata collected in Korea.

Variation of RNase activities and nucleic acid content of non-irradiated and irradiated eggs of Bombyx mori during early development of embryo (잠란(蠶卵)의 초기발육과정(初期發育過程)에 따르는 RNase활성(活性) 및 핵산량(核酸量)의 변동(變動) 및 그 X선조사(線照射)에 의한 영향(影響)에 관(關)한 연구(硏究))

  • Previously identified female pupae were X-irradiated with a dose of 1000r one day prior to moth transformation. Female mothes from irradiated and non-irradiated pupae were copulated with normal male ones and allowed to lay eggs. Fertilized eggs were collected at 6 intervals such as 5, 15, 45, 90 minutes, 12 and 40 hours after laying, and deep-freezed immediately after each collection until measurements. RNase activity and nucleic acid content were determined with each sample and following results were obtained. 1) It was proved to exist two RNases in silk worm eggs as in mammalian tissues, one active maximally at pH 5.8 and the other at pH 8.0, and the acid RNase activity was much higher than that of alkaline RNase. 2) The activity of acid and alkaline RNases increased remarkably during early development of the embryo of silk worm eggs, reaching the maximum activity at 45 minutes from laying time in non-irradiated group. There was no appreciable difference in two RNase activities for 45 minutes after laying in both control and irradiated groups, but the activity of acid and alkaline RNases in latter group was three times as much as that in former group, at 90 minutes from laying time and it was also found the acid RNase activity was 1.8 times higher than alkaline one in irradiated group. 3) The RNA-P content of control group increased considerably for initial 45 minutes, followed by a decline 45 minutes later with sight but steady increase thereafter. The RNA-P content of irradiated group, however, increased at initial 5 minutes, followed by a marked fall 90 minutes after laying, with no change thereafter. The DNA-P of control group showed a sharp increase for initial 45 minutes, followed by a decline 45 minutes later with no appreciable change thereafter, whereas that of irradiated group showed an increase at initial 15 minutes, followed by a sharp decline for following 45 minutes with a gradual increase thereafter. It was thus proved that the synthesis of nucleic acid in silk worm eggs was much suppressed by X-irradiation during early development of embryo. 4) The RNase activity varied in parallel with the RNA-P content in control group, but the RNA-P content in irradiated group was shown to be minimum value in concidence with the maximum activity of both RNases.

A Study of Entomopathogenic Nematode at Heavy Metal Contents in the Polluted Soil of Kyungsangbuk-do Area (경상북도지역의 오염된 토양에서의 중금속 함량에 관한 곤충병원성 선충에 관한 연구)

  • The variety and density of entomopathogenic nematodes from the polluted soils of heavy metals were examined. In order to investigate the pollution of heavy metals in soil, 300 sites in kyungsangbuk-do were collected from March to October in 2001. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, Cr^{6+}$, CN) and than cheesed soil of 25 sites with high concentration of heavy metals. The seven strains of nematodes were isolated from seven samples by silkworm host (Bombyx mori mori) and white trap. Isolated nematodes are composed of two families, one order. The members of Rhabditida were isolated in the soil with mean Cd content of 0.870 ppm. And they were isolated in the soil samples with As content less than 0.745 ppm. However they were isolated regardless of concentration of Cu and Pb. The members of Cylindrocorpidae were isolated in the soil samples with Cr^{6+}$ content less than 0.05 ppm. Any entomopathogenic nematode was not detected in the CN polluted soil. Isolated nematodes successfully cultured on the silkworm host and were confirmed the pathogenicity, multiplicity, and tolerance against various condition of preservation. Which proved its potential usefulness as biological agent.

Bioactive and Chemical Properties by Silkworm (Bombyx mori L.) Powder Degradation with Kiwifruit, Papaya, Pineapple and Pear Juice (키위, 파파야, 파인애플 및 배 과즙 처리에 의한 누에분말의 이화학적 특성과 생리활성)

  • Bioactive and chemical properties of silkworm powder (SP) degradation by fruit extract containing the proteolytic enzymes of kiwifruit, papaya, pineapple and pear were investigated. Silkworm powder was incubated with extracts from each fruit at $60^{\circ}C$ for 24 hr. Protein content was slightly higher in the SP treated with fruit extract than that in the control SP. Major minerals were K, Ca, Mg, and Zn. Major fatty acids were linolenic acid, oleic acid, and palmitic acid. When total protein patterns were analyzed by SDS-polyacrylamide gel electrophoresis (PAGE), silkworm protein was strongly degraded by the treatment of fruit extract from pineapple, papaya, and pear, but little silkworm degradation was observed in kiwifruit extract treatment. Fibriolytic activity was only detected in the SP by the fruit extract treatments from papaya and pear. DPPH radical scavenging activity was slightly stronger in the SP treated with fruit extract than that in silkworm powder. However, all these samples exhibiteda relatively low activity compared with the butylated hydroxytoluene (BHT). These results may provide the basic data for understanding the biological activities and chemical characteristics of SP treated with fruit extract for development of functional foods.

One-step Separation of 30K Protein from the Silkworm Hemolymph by Anion-exchange Chromatography and Its Effect on the Proliferation of Human Cells (음이온교환 크로마토그래피를 이용한 누에체액 유래 30K 단백질의 정제와 정제된 단백질이 인간세포 배양 증식에 미치는 영향)

  • In order to investigate the feasibility of 30K protein from silkworm (Bombyx mori) hemolymph (SH) on the proliferation of human cells, a simple separating procedure by anion-exchange chromatography system with Q-Sepharose fast flow gel was established. The 30K protein was eluted with an optimized condition of 0.16 M sodium chloride in 20 mM tris buffer (pH 9.0). The separated 30K protein at three concentrations of 0.04, 0.12, and 0.4 mg/ml was added to the culture medium with various human cells, such as chondrocytes, periosteum-derived cells, and MRC-5 cells, and their growth rates were measured. The cell growth rate at protein concentration of 0.4 mg/ml was always higher than that without 30K protein in all human cells tested, suggesting that the 30K protein has positive effect on the increase of the life span of human cells.

${\alpha}$-Glucosidase Inhibitory Substances Exploration Isolated from the Herb Extract (생약재 추출물로부터 분리한 ${\alpha}$-Glucosidase 저해활성물질 탐색)

  • This is a part of the study on the food materials that are effective for diabetes treatment and for use in the development of functional bread products. In this study, various commercially available Oriental medicines with the intestinal absorption enzyme called ${\alpha}$-glucosidase, which is known to be effective for diabetes treatment, were explored. According to the research results on the water and methanol in 200 kinds of Oriental medicines, which were separated by layer to investigate the inhibitory activity of ${\alpha}$-glucosidase, Astragalus membranaceus (70.9%) in the water layer and Pericaeta communissima (72.9%) in the MeOH layer showed a strong inhibitory effect of over 70%. Myristica fragrans (69%), Morus alba (66.9%), Schisandra chinensis (65%), Panax notoginsens (63.9%), Anthriscus sylvestris (62.9%), Asparagus cochinchinensis (62.1%), Erycibe obtusifolia (60.9%), Polygonum cuspidatum (60.7%), Atractylodes lancea (60.2%), and Perilla frutescens (60.2%) in the water layer, and Codonopsis pilosula (67.8%), Prunus persica batsch (67.6%), Sinomenium acutum (63.5%), and Malvae semen (61.6%) in the MeOH layer, showed a more than 60% inhibitory effect. Thirty one species, including Polygonatum sibiricum (59.8%), Medicata fementata (59.7%), Alisma canaliculatum (59.5%), Coix lacryma-jobi (59.2%), Asiasarum sieboldi (59.0%), and Bupleurum falcatum (53.0%), in the water layer, and 10 species [Quisqualis indica (58.8%), Lycium chinense (58.3%), Trichosanthes kirilowii (58.0%), Thuja orientalis (55.9%), Bombyx mori (55.6%), Gallus domesticus (55.4%), Aralia continentalis (55.3%), Cibotium barometz (52.7%), Euphorbia pekinensis (52.7%), and Dolichos lablab (52.5%)] in the MeOHlayer, showed a more than 50% inhibitory effect. Therefore, such materials are expected to be the basic materials that will be used for the development of functional materials for diabetes treatment.

1-Deoxynojirimycin Content and Blood Glucose-Lowering Effect of Silkworm (Bombyx mori) Extract Powder

  • We investigated the 1-deoxynojirimycin (1-DNJ) content of extracts from silkworm larvae at each developmental stage within three silkworm varieties. We also compared the content of the following polyhydroxylated alkaloids in the silkworm extracts: 1-DNJ, fagomine, and 1,4-dideoxy-1,4-imino-d-arabinitol (DAB). In addition, we evaluated the glucose-lowering effects of silkworm extract powder in db/db mice. The 1-DNJ content was the highest in Yeonnokjam $5^{th}$ instar $3^{rd}$ d larvae and Hansaengjam $5^{th}$ instar $3^{rd}$ d larvae, which contained 18.4 mg/100 g dry weight and 18.3 mg/100 g dry weight, respectively. The $5^{th}$ instar $3^{rd}$ d larvae exhibited a higher content of 1-DNJ than that of $5^{th}$ instar $5^{th}$ d larvae among all varieties. The glucose-lowering effects of silkworm extracts and Yeonnokjam powder were tested on db/db mice, and the blood glucose levels were found to decrease significantly in the YR70 group. Silkworm extracts (180 mg/kg, 90 mg/kg, 45 mg/kg, and 22.5 mg/kg) and acarbose (50 mg/kg) were administered orally for 4 wk. Changes in water intake were not statistically significant between control and silkworm extract-treated groups. Compared to the control group, blood glucose levels in the silkworm extract powder-treated group decreased in the 22.5 mg/kg/d group after being administered for 4 wk. This decrease was statistically significant. Furthermore, biochemical changes in the AST(Aspartate aminotransferase), ALT(Alanine aminotransferase), TCHO(Total Cholesterol), TG(Triglyceride), LDL(Low density lipoprotein), and HDL(High density lipoprotein) levels in blood were not observed. However, statistically significant decreases in blood GLU in the 22.5 mg/kg/d group compared to that of the control group occurred. In addition, the epididymal fat weight of the silkworm extract powder-treated group decreased significantly in both the 22.5 mg/kg/d group and 180 mg/kg/d group compared to that of the control group, but there were no statistically significant changes in perirenal fat weight. These results demonstrate that silkworm extracts inhibit changes in blood glucose levels in model diabetic mice.

Biological Activity and Biochemical Properties of Silkworm (Bombyx mori L.) Powder Fermented with Bacillus subtilis and Aspergillus kawachii (유용식용 균주에 의한 발효 누에분말의 이화학적 특성과 생리활성)

  • Biological activities (${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) free radical scavenging activity, fibrinolytic activity and reducing power) and biochemical properties (protein content and electrophoretical protein patterns) were examined in solid state fermentation with Bacillus subtilis and Aspergillus kawachii using silkworm powder (SP) as substrate. The highest protein contents and free radical scavenging activities were seen in the SP fermented for 12 days with B. subtilis and A. kawachii, and these were in a time-dependent manner. The highest reducing power was seen in the SP fermented for 6 days with B. subtilis and for 12 days with A. kawachii, respectively. The highest fibrinolytic activities were seen in silkworm fermented for 6 days with B. subtilis and A. kawachii, but this activity was higher in the A. kawachii fermented SP than that of B. subtilis. When total protein patterns were analyzed by SDS-polyacrylamide gel electrophoresis (PAGE), the proteins of the SP fermented with B. subtilis for 3 days were completely degraded, while the protein degradation in the SP fermented with A. kawachii occurred after 12 days and this degradation increased proportionally to culture time. As a result, the SP fermented with both B. subtilis and A. kawachii showed higher fibrinolytic activities after 6 days of fermentation and antioxidative activity after 12 days, indicating that physiological activities of the fermented SP using these strains were highly improved compared to the unfermented SP, and that this compound could be a candidate material as a dietary supplement of healthy functional foods.

Construction of Transgenic Silkworms Expressing Human Stem Cell Factor (hSCF) (인간 유래 Stem Cell Factor (hSCF) 재조합단백질이 발현되는 누에형질전환체 제작)

  • Human Stem Cell Factor (hSCF) is a cytokine that binds to the c-Kit receptor and plays an important role in hematopoiesis, spermatogenesis, and melanogenesis. To produce the human Stem Cell Factor (hSCF) recombinant protein, we constructed a germline transgenic silkworm using the piggyback vector. The expression of the hSCF gene was driven by the Drosophila heat shock protein 70 (dHsp70) promoter. 3XP3 promotor-driven EGFP was used as a marker which allowed us to rapidly distinguish the transgenic silkworm. A mixture of the donor and helper vector was micro-injected into 1,020 eggs of bivoltin silkworms, Keomokjam. We obtained approximately 22 G1 broods that were EGFP-positive. The expression of the hSCF gene in the transgenic silkworm was analyzed by SDS-PAGE and immunoblotting. Also, analysis of insertion sites into the silkworm genome using inverse PCR showed that exogenous DNA was inserted into the transgenic silkworm genome. These results show that successfully constructed transgenic silkworm expresses the hSCF recombinant protein.

Studies on the Residual Contents of Sulfur Dioxide in Herbal Medicines Distributed at Domestic (국내 유통 한약재에서 이산화황 잔류량에 대한 연구)

  • We investigated the residual contents of sulfur dioxide ($SO_2$) on the 280 kinds of herbal medicines distributed at 8 cities including in Seoul, Busan, Daegu, Gwangju, Jecheon, Yeongju, Geumsan and Jeonju in Korea. The residual contents of $SO_2$ were determinated by modified Monier-Williams method. The residual contents of $SO_2$ were not detected at 206 products in total 280 products. However, it was detected below 100 ppm in 39 products, between 101 and 1000 ppm in 30 products and exceeded 1000 ppm in 5 products. $SO_2$ contents ranged 11 ${\sim}$ 2339 mg/kg (mean 293 mg/kg) at domestic samples distributed. Regardless of region, $SO_2$ contents were not found at Notopterygii Rhizoma, Ligustici Scinensis Rhizoma et Radix, Bombyx Batryticatus, Coicis Semen, Cnidii Rhizoma, Anemarrhenae Rhizoma, Gardeniae Fructus, Alismatis Rhizoma. But it's found at Batatatis Rhizoma, Paeoniae Radix and Codonopsitis Radix of every region collected the samples. $SO_2$ contents were not detected at 58 products which collected cultural fields of dometic. After treated with water wash and hot water extraction, the reduction rates of sulfur dioxide were appeared with 14.3% ${\sim}$ 40.4% and 55.2% ${\sim}$ 100.0%, respectively. These data will be used to establish a criteron of residual sulfur dioxide in herbal medicines.
