  • This study described the spermatozoa of the discoglossidae Bombina orientalis using light, scanning and transmission electron microscopes. Sperm head possess a crescent or leaf shape, with a moderately flexible head, and with a sharp anterior and posterior tips. The nucleus are a thick cone shaped in the widest middle part of nucleus, and a slender anterior and posterior of nuclear tips. The chromatin is not completely compact, but irregularly imbricated such as roof. Some nuclear lacunae, irregular in shape, are scattered within the nucleus. No neck and middle piece were developed. The flagellum is composed of 9+2 axoneme, axial rod and undulating membrane. The mitochondria were distributed only in cytoplasmic membrane around the nucleus. In particular, the nuclear rod contains bundles of fibers, the rod penetrating from anterior portion to the middle of the nucleus, is extended roughly two-thirds of the nucleus such as eyelashes shaped.

  • Two more major pigments, lutein and 3,3'-dihydroxy-3,4,3',4'-bisdehydro-${\beta}$-carotene, were isolated from the abdominal skin of Bombina Orientalis and their structures were established by physical and chemical properties, visible and IR spectral characteristics and chromatographic behaviors.

  • Molinate is thiocarbamate herbicide used primarily in rice production. Chondrogenesis is a multistep process that is essential for endocondral bone formation. The transcription factor Sox9 has an essential role during the sequential steps of chondrocyte differentiation. Bombina orientalis is one of the most common amphibians in the world and comprises a large proportion of their total number. We examined the embryotoxic and survival effects of molinate at various concentration in B. orientalis embryos. The survival rates of embryos at 312h post fertilization treated with molinate was decreased with concentration dependent manner. Also, developmental malformations appeared by molinate in B. orientalis embryos. the expression levels of Sox9 mRNA was examined by RT-PCR. In our result showed that Sox9 expression was found to be increased in malformed tadpole compared to normal tadpole. These results suggested that molinate was detrimental for survival and development of B. orientalis embryo.

The Effects of Lead(II) Nitrate on the Embryo Development in Native Amphibians (질산납이 한국산 무미양서류의 배아발달에 미치는 영향)

  • An investigation of the effects of Pb for domestic anuran embryos were evaluated with the Frog Embryo Teratogenesis Assay; Xenopus (FETAX). Depending on the species, the difference between the embryo size and the embryonic development time was determined. As a result, mortality and malformation rates were increased, malformation patterns were changed and larval body length were decreased in a dose dependent manner of the Pb. The half maximal lethal concentration ($LC_{50}$) of the Bufo gargarizans, Hyla japonica, Rana nigromaculata and Bombina orientalis were 0.58, 0.49, 0.52, $0.54mg\;L^{-1}$, respectively. The half maximal effective concentration ($EC_{50}$) of the Bufo gargarizans, Hyla japonica, Rana nigromaculata and Bombina orientalis were 0.35, 0.74, 0.30, $0.29mg\;L^{-1}$, respectively. The teratogenic index (TI) were 1.66 in the Bufo gargarizans, 1.81 in the Hyla japonica, 1.73 in the Rana nigromaculata and 1.86 in the Bombina orientalis, respectively. Therefore, the Pb seems likely to have a teratogenic effect in all four species' embryonic development. The Bombina orientalis was the most sensitive to the Pb. This means that the difference between the different species, even if they have all been exposed to the same concentration of pollutants depending on the species. The result above show that the Pb acts as a teratogenic agent in the development of the four domestic frog species.

  • The cutaneous erythrophore differentiation from larvae to adult in bombina orientalis-the Korean fire bellied toad-is discussed electron microscopically to reveal the structural changes of carotenoid vesicles and pterinosomes in the process of erythrophore differentiation. Typical structure of adult erythrophores distributed beneath the basement membrane is composed much carotenoid vesicles and a small quantity of pterinosomes. Marginal electron density of these carotenoid vesicles is higher than those of internal regions and pterinosomes have many concentric lamellae. The very first differentiated erythrophores are apeared before and after the foreleg developing stage of metamorphosis and red ventral color is just appeared about $1\\sim2$ weeks after metamorphosis. About 1 year after metamorphosis, external body color of the young toad is similar to adult but the morphology of pigment granules observed in these erythrophores differs from that of adult in structure and electron density. At this stage, the differentiated level of pterinosomes is not remarkable and the accumulation state of carotenoid pigments in erythrophores is also relatively low.

  • Remodeling of epithelial cells during wound healing in the skin of the Korean fire-bellied toad, Bombina orientalis, were examined using the scanning and transmission electron microscopical analyses. Artificial wounds were induced on the dorsal surface of the skin by excision, and reared in special cages with normal diets for up to 31 days after injury. From 4 days after wounding, regenerated epithelial cells are more rapidly migrated to wounding area, and remodeling of tissue components are proceeded gradually. Especially, formation of basal lamina between regenerated epithelium and dermis, and reconstruction of cellular junctions such as desmosomes (among the regenerated epithelial cells) and hemidesmosomes (between basal epithelial cells and basal lamina) are detected through fine structural analysis from 10 days after injury. Parakeratosis of regenerated epithelial cells observed during 16 to 19 days after wounding.

Seminiferous Epithelium Cycle in Bombina orientalis

  • The purpose of the present study was to examine the seminiferous epithelium cycle of Bombina orientalis using a light microscope. The cycle was divided into a total of 10 stages, according to the morphological characteristics of the cells. The spermatogenetic cells included primary spermatogonia, secondary spermatogonia, primary spermatocytes, secondary spermatocytes, spermatid and sperm. At stage I, the primary spermatogonia was located closer to basal lamina of the seminiferous tubule without spermatocyst formations. Especially at the stage II, the secondary spermatogonia were located in the spermatocyst. The primary and secondary spermatocytes were found from stages III to VI. The secondary spermatocytes were smaller in size than the primary spermatocytes, but they had thicker nucleoplasm and smaller nuclei. The round-shaped, early sperm cells were formed in stage VII, and further divided at stage VIII to have more concentrated nucleoplasm before division to matured sperm cells. At stage X, the matured sperm cells emerged from the spermatocyst. Considering the above results, this study presented the special characteristics in the generation and type of sperm formation. The germ cell formation occurred in various stages, like the perspectives of Franca et al (1999), ultimately, providing taxonomically useful information.

Relationship of Maximal Take-off Speed to Power and Shortening llelocitv of Hindlimb Muscle in Anuran Amphibians (무미양서류의 도약속도와 다리근육의 동력 및 수축속도와의 관계)

  • To learn how maximal locomotory speed of animals is defined in terms of hindlimb structure and muscle contractile function, take-off speed, hindlimb length, thigh muscle mass, shortening velocity and power of the sastrocnemius muscle were measured with one fast species, Rono nigromaculota and one relatively slowresponding species, Bombina orientalis. Take-off speed (m.sec-1) was greater in R. nigromoculata $(2.4\pm0.2SD, $ n: 14) than in the Bombino $(1.6\pm0.1SD, $ n=8). Stvle of the take-off response was a long-iump type in the Rano and a short-ranged hopping in the Bombing. Faster take-off capacity of the ranid frogs was supported by the longer hindlimb length (relative to body length) and the more massive thigh muscles (relative to body mass), compared to the Bombina. Further, the ranids exhibited faster maximal shortening velocity and Breater maximal power generateion than the Bombina [Vmax $(ML.sec-1)=11.79\pm1.69SD$ for the Runa and $9.74\pm1.27SD$ for the Bombina; Pmax $nW.kg-1)=222.42\pm42.42SD$ for the Rono and $169.03\pm34.52SD$ for the Bombinal. With more massive thigh muscles and greater mechanical power, the ranids would generate greater total power and thus higher energy release per unit time to muscle tissues for the burst take-off. As a consequence, biomechanical properties seen in the ranids seem to be more effective for frost take-off than in the Bombina.
