Solubilization of IH-901, a Novel Intestinal Metabolite of Ginseng Saponin, in Aqueous Solution (인삼사포닌의 소장내 최종대사물인 IH-901의 수용액중 가용화)

  • The purpose of the present study was to formulate the aqueous solution of $20-O-{\beta}-D-glucopyranosyl-20(S)-protopanaxadiol\;(IH-901)$, an intestinal bacterial metabolic derivative from Ginseng protopanaxadiol saponin. For this purpose, the effects of various solubilization agents such as cosolvents [ethanol, propylene glycol (PG), polyethylene glycol 300 (PEG 300), polyethylene glycol 400 (PEG 400), glycerin], surfactants $(Tween\;80,\;Cremophor^{\circledR}\;RH40,\;Cremophor^{\circledR}\;EL,\;Poloxamer\;407,\;Poloxamer\;188)$ and a complexation agent $[hydroxypropyl-{\beta}-cyclodextrin\;(HPBCD)]$, on the solubility of IH-90l in aqueous solution were evaluated. The solubility of IH-901 in water was under $1\;{\mu}g/ml\;at\;20^{\circ}C$. Cosolvents such as ethanol, PG, PEG 300, PEG 400 and glycerin did not enhance the solubility of IH-901 at the 0 - 40% concentration range. The solubility of IH-901 was significantly elevated by the addition of cosolvents over the 80% concentration range. On the other hand, tween 80, $Cremophor^{\circledR}\;EL,\;Cremophor^{\circledR}\;RH40$ and HPBCD showed enhanced effects on the solubility of IH-901. The enhanced effects of Poloxamer 407 or Poloxamer 188 on the IH-901 solubility were less pronounced compared with $Cremophor^{\circledR}\;EL\;or\;Cremophor^{\circledR}\;RH40$. As a results, $Cremophor^{\circledR}$ aqueous solution was selected as an optimum solvent system. The aqueous solutions containing 10% $Cremophor^{\circledR}\;EL$ and 7% $Cremophor^{\circledR}\;RH40$ were formulated as dosing solutions containing 5.0 mg/ml of IH-901 for its intravenous and oral administration, respectively. The formular showed physical stability after stored for 7 days at $4^{\circ}C$.

Potentiation of endothelium-dependent vasorelaxation of mesenteric arteries from spontaneously hypertensive rats by gemigliptin, a dipeptidyl peptidase-4 inhibitor class of antidiabetic drug

  • Dipeptidyl peptidase4 (DPP4) inhibitors such as gemigliptin are antidiabetic drugs elevating plasma concentration of incretins such as GLP-1. In addition to the DPP4 inhibition, gemigliptin might directly improve the functions of vessels under pathological conditions. To test this hypothesis, we investigated whether the acetylcholine-induced endothelium dependent relaxation (ACh-EDR) of mesenteric arteries (MA) are altered by gemigliptin pretreatment in Spontaneous Hypertensive Rats (SHR) and in Wistar-Kyoto rats (WKY) under hyperglycemia-like conditions (HG; 2 hr incubation with 50 mM glucose). ACh-EDR of WKY was reduced by the HG condition, which was significantly recovered by $1{\mu}M$ gemigliptin while not by saxagliptin and sitagliptin up to $10{\mu}M$. The ACh-EDR of SHR MA was also improved by $1{\mu}M$ gemigliptin while similar recovery was observed with higher concentration ($10{\mu}M$) of saxagliptin and sitagliptin. The facilitation of ACh-EDR by gemigliptin in SHR was not observed under pretreatment with NOS inhibitor, L-NAME. In the endothelium-denuded MA of SHR, sodium nitroprusside induced dose-dependent relaxation was not affected by gemigliptin. The ACh-EDR in WKY was decreased by treatment with $30{\mu}M$ pyrogallol, a superoxide generator, which was not prevented by gemigliptin. Exendin-4, a GLP-1 analogue, could not enhance the ACh-EDR in SHR MA. The present results of ex vivo study suggest that gemigliptin enhances the NOS-mediated EDR of the HG-treated MA as well as the MA from SHR via GLP-1 receptor independent mechanism.

Substrate Utilization Patterns During BTEX Biodegradation by an o-Xylene-Degrading Bacterium Ralstonia sp. PHS1

  • The biodegradation of BTEX components (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene) individually and in mixtures was investigated using the o-xylene-degrading thermo-tolerant bacterium Ralsronia sp. strain PHS1 , which utilizes benzene, toluene, ethylbenzene, or o-xylene as its sole carbon source. The results showed that as a single substrate for growth, benzene was superior to both toluene and ethylbenzene. While growth inhibition was severe at higher o-xylene concentrations, no inhibition was observed (up to 100 mg $l^-1$) with ethylbenzene. In mixtures of BTEX compounds, the PHS1 culture was shown to degrade all six BTEX components and the degradation rates were in the order of benzene, toluene, o-xylene, ethylbenzene, and m- and p-xylene. m-Xylene and p-xylene were found to be co-metabolized by this microorganism in the presence of the growth-supporting BTEX compounds. In binary mixtures containing the growth substrates (benzene, toluene, ethylbenzene. and o-xylene), PHS1 degraded each BTEX compound faster when it was alone than when it was a component of a BTEX mixture, although the degree of inhibition varied according to the substrates in the mixtures. p-Xylene was shown to be the most potent inhibitor of BTEX biodegradation in binary mixtures. On the other hand, the degradation rates of the non-growth substrates (m-xylene and p-xylene) were significantly enhanced by the addition of growth substrates. The substrate utilization patterns between PHS1 and other microorganisms were also examined.

Effects of Walking Exercise on Isokinetic Muscular Function (걷기운동이 등속성 근력에 미치는 효과)

  • The aim of this study was to examine the effect of a brisk walking intervention for 8weeks on isokinetic muscular function factor(strength, power, endurance). The following results were found from the current study: First, Isokinetic muscle function(strength, power, endurance) on $60^{\circ}/sec$ in extension and flexion was significant exercise group and interaction in left knee flexion and right knee extension(P<.00l). Secondly, Isokinetic muscle function(strength, power, endurance) on $120^{\circ}/sec$ in extension and flexion was significant exercise group and interaction in left knee flexion and right knee extension(P<.001). Thirdly, Isokinetic muscle function(strength, power, endurance) on $180^{\circ}/sec$ in extension and flexion was significant exercise group and interaction in left knee flexion and right knee extension(P<.001). Considering the above result of the study the brisk walking had more improvement knee flexor and extensor muscular power. Therefore the brisk walking in the study can be proposed as effective plan to prevention hurt from a sarcopenia and to raise quality in life.

Changes in the Quality Characteristics of Semi-dried Small Squid Idiosepius paradoxus during Refrigerated Storage (반 건조 새끼 오징어(Idiosepius paradoxus)의 냉장저장 중 품질특성의 변화)

  • This study evaluated the quality characteristics of semi-dried small squid Idiosepius paradoxus treated with grapefruit Citrus paradisi seed extract (GSE) during refrigerated storage for 6 months. The moisture content and water activity decreased significantly in both control and GSE-treated groups with increasing storage time, but there was no significant difference between groups. During storage, the degree of water activity reduction was lower in the GSE-treated group than in the control. The pH change was similar in both groups during storage. The thiobarbituric acid reactive substances (TBARS) value of the GSE-treated group was lower than that of the control until 4 months of storage, and was subsequently higher than in the control group. The Hunter color L value did not differ significantly during storage between groups, whereas the a and b values decreased significantly (P<0.05) in both groups with increasing storage time. The hardness value of the semi-dried small squid increased with storage time, but there was no statistical difference between GSE treatment and control. There were more viable cells in the control group than the GSE-treated group after 5 months of storage. According to the sensory evaluation, neither the taste, color, odor, nor texture differed statistically during storage between groups, while overall acceptability was significantly lower in the GSE-treated group than the control after 6 months of storage.

Analysis of Quality Change during Preservation by a Prototype Drink Containing a Helianthus annuus Seed Extract (해바라기씨 추출물 함유 음료의 저장기간 중 품질 특성 분석)

  • To explore the preservation parameters of a prototype anti-inflammatory drink containing a Helianthus annuus seed extract, we first examined sweetness, pH and color changes, and temperature effects during preservation. Over 25 days, pH rose slightly from 3.50 to 3.65. Small color changes in L, a, and b values occurred on prolonged storage. Temperature most affected storage properties, which differed considerably at $10^{\circ}C$ and $40^{\circ}C$. Microbiological safety tests showed that the prototype drink was safe; neither general bacterial contamination nor Escherichia coli growth occurred. We concluded that the prototype drink was biologically and microbiologically safe on preservation.

Fabrication and Characteristics of Lateral Type Field Emitter Arrays

  • We have proposed and fabricated two lateral type field emission diodes, poly-Si emitter by utilizing the local oxidation of silicon (LOCOS) and GaN emitter using metal organic chemical vapor deposition (MOCVD) process. The fabricated poly-Si diode exhibited excellent electrical characteristics such as a very low turn-on voltage of 2 V and a high emission current of $300{\;}\bu\textrm{A}/tip$ at the anode-to-cathode voltage of 25 V. These superior field emission characteristics was speculated as a result of strong surface modification inducing a quasi-negative electron affinity and the increase of emitting sites due to local sharp protrusions by an appropriate activation treatment. In respect, two kinds of procedures were proposed for the fabrication of the lateral type GaN emitter: a selective etching method with electron cyclotron resonance-reactive ion etching (ECR-RIE) or a simple selective growth by utilizing $Si_3N_4$ film as a masking layer. The fabricated device using the ECR-RIE exhibited electrical characteristics such as a turn-on voltage of 35 V for $7\bu\textrm{m}$ gap and an emission current of~580 nA/l0tips at anode-to-cathode voltage of 100 V. These new field emission characteristics of GaN tips are believed to be due to a low electron affinity as well as the shorter inter-electrode distance. Compared to lateral type GaN field emission diode using ECR-RIE, re-grown GaN emitters shows sharper shape tips and shorter inter-electrode distance.

A Study on Utilization of Mixed Wild Flowers as a Silage Materials (Silage 재료로서 혼합 야생화의 이용에 관한 연구)

  • This experiment was conducted to investigate the quality of wild flowers and mixed wild flower silages. Silage materials employed ochardgrass and mixed wild flower (orchardgrass, white clover, Achillea sibirica Ledeb., Coreopsistinctoria Nutt., Rudbeckiabicolor Nutt., Centaureacyanus L.). Six kilograms of silage materials cut from each treatment were divided into 3 containers, packed by vacuum, and fermented for 40 days. The orchardgrass silage showed significantly higher contents of crude protein than that of mixed wild flower silage. But the neutral detergent fiber and acid detergent fiber contents of orchardgrass silage were significantly lower than those of mixed wild flower silage. pH and butyric acid of orchardgrass silage were significantly lower than those of mixed wild flower silage, and significantly higher in lactic acid contents. It appears that water soluble carbohydrates needs to be supplemented, being grounded on the results above, into wild flower materials both with high fiber materials and with low substances which need lactic acid fermentation.

Effects of Low Level Laser on the Proliferation and Gene Expression of Fibroblasts and Immune Cells (저출력레이저조사가 섬유아세포와 면역세포의 증식 및 유전자발현에 미치는 영향)

  • The growth and synthetic activities of fibroblasts are regulated by cytokines and growth factors derived from activated inflammatory cells. Stimulatory effect of low level laser (LLL) radiation on wound healing seems to be in part due to direct stimulatory action on cell proliferation and synthetic activities of fibroblasts. Also indirect stimulatory effect on the fibroblast function through inflammatory or immune cells is another possible mechanism of biostimulatory action of LLL. This study was performed to determine the growth rate of human gingival fibroblasts obtained biopsy and culture, fibroblast cell line, and immune cell line by using $[^3H]-$ thymidine incorporation test. And gene expression pattern was also analyzed by using the DNA probe such as Hsp70, IL-1$\beta$, MIP-1$\alpha$ and actin cDNA. Proliferation rate of gingival fibroblast was increased by LLL irradiation, but no more effect was added by LPS or IL-1$\beta$ pretreatment Enhanced Hsp70 gene expression was found from gingival fibroblasts and fibroblast cell line COS by LLL irradiation., which was not more increased by LPS or IL-1$\beta$ pretreatment. LLL-irradiated promyelcytic cell line HL-60 and macrophage cell line RAW264.7 showed significant stimulatory effect of proliferation rate when compared with respective control. However there were no changes in growth rate of other immune cell tested in this study, such as B cell line WR19n.l and 230, helper T cell line Jurkat and Hut78, cytolytic T cell line CTLL-r8. By LLL-irradiation Hsp70 gene expression was increased in RAW246.7 and HL-60, not in CTLL-R8. And IL-1$\beta$ and MIP-1$\alpha$ gene expression were induced only from LLL-irradiated RAW264.7. These results led us to presume that LLL radiation may affect to the immune cells, especially to macrophage, through which it might promote wound healing process.

  • The purpose of this study was to examine industrial workers ; Knowledge, Attitude and Practice of occupational health management and service. The data was collected from Dec 4. 1993 to Jan 21, 1993. The data was collected from 352 industrial workers from 37 companies located in Kyng Ki Province and In cheon city. The measurement tool used in this study was a structured questionnaire developed by the community Nursing academy. The major findings attained from this study are summarized as follows : 1. The level of Knowledge, Attitude and Practice of industrial workers of occupational health management and service, were as follows : The total mean score was 72 out of 100. The total mean score of Knowledge was 22.5, out of 30. The mean score of Attitude was 39.2, out of 55. The mean score of Practice was 10. 6, out of 15. 2. The level of Knowledge, Attitude, and Practice of industrial workers of health management and service according to workers' general characteristics. The significant perceptions of health management as a whole that impact on the Knowledge score were occupational factors such as, age, income, number of workers, educational status, sex, and marital status. The significant factor of Practice which impact on the score was age, number of workers, sex, and marital status.
