Bioconversion of Citrus unshiu peel extracts with cytolase suppresses adipogenic activity in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.

  • This paper describes the development of RF front.end equipment of a wide band high precision satellite navigation receiver to be able to receive the currently available GPS navigation signal and the GALILEO navigation signal to be developed in Europe in the near future. The wide band satellite navigation receiver with high precision performance is composed of L - band antenna, RF/IF converters for multi - band navigation signals, and high performance baseband processor. The L - band satellite navigation antenna is able to be received the signals in the range from 1.1 GHz to 1.6 GHz and from the navigation satellite positioned near the horizon. The navigation signal of GALILEO navigation satellite consists of L1, E5, and E6 band with signal bandwidth more than 20 MHz which is wider than GPS signal. Due to the wide band navigation signal, the IF frequency and signal processing speed should be increased. The RF/IF converter has been designed with the single stage downconversion structure, and the IF frequency of 140 MHz has been derived from considering the maximum signal bandwidth and the sampling frequency of 112 MHz to be used in ADC circuit. The final output of RF/IF converter is a digital IF signal which is generated from signal processing of the AD converter from the IF signal. The developed RF front - end has the C/N0 performance over 40dB - Hz for the - 130dBm input signal power and includes the automatic gain control circuits to provide the dynamic range over 40dB.

Optimization of Ethanol Extraction Conditions for Effective Components from Gastrodia elata Blume (천마 유효성분의 에탄올 추출조건 최적화)

  • Gastrodia elata Blume (GEB) is a traditional herbal plant that has been used in Asian countries for centuries as an anticonvulsant analgesic, and a sedative for treating general paralysis, epilepsy, vertigo, and tetanus. This study was designed to optimize conditions for ethanol extracts or GEB by analyzing and monitoring the extraction characteristics with response surface methodology. The extract was used for analysis of the effective components of GEB. The estimated optimal conditions were 63.62% in ethanol of 5.06 mL/g in solvent per sample, and 6.25 hr in extract time. The optimal extraction conditions for $ \gamma-aminobutyric$ acid, were 45.52%, 5.67 mL/g, and 6.04 hr, while those for $\rho-hydroxybenzyl$ alcohol were 62.73%, 5.02 mL/g, and 5.95 hr. Regression equation was generated for each variables and then superimposed them, such as soluble solid, $ \gamma-aminobutyric$ acid and $\rho-hydroxybenzyl$ alcohol content thereby resulting in superimposed values of extinction conditions like $45\sim65%,\;5\sim7mL/g$ and $5\sim7$ hr, respectively.

Effect of Lighting Regimes on Performance and Meat Quality of Broilers (점등체계가 육계의 생산성과 육질특성에 미치는 영향)

  • The study was carried out to investigate the effect of lighting regimes on performance and meat quality of broiler. A total of 912 chicks was divided into 2 groups with male (3 replications/group, 34 heads/replication) and female (3 replications/group, 42 heads/replication). Treatments were control (C, natural lighting), T1 treatment (24L), and T2 treatment (1~2d, 24L; 2~4d, 23L+1D; 4~16d, 16L+8D; 16~21d, 18L+6D; 21~42d, 23L+1D) by lighting regimes. There was no significantly different on body weight, body weight gain, feed intake, and feed conversion ratio of male and female of broilers among treatments. There was no significantly different on cooking loss and water hold capacity of all broiler's meats among treatments. Share force of lighting regimes was higher compared to control (P<0.05). There was no significantly different on common ingredients (moisture, crude protein, crude fat, and ash) of broiler's meat. Finally, lighting regimes may decrease the stress as well as improve the growth performance and meat quality of broilers

  • BACKGROUND: Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. This investigated to determine the optimal culture conditions in ceramic balls for maximal sporulation of entomopathogenic fungi Beauveria bassiana M130 by use rice bran extract.METHODS AND RESULTS: METHODS AND RESULTS: A culture of entomopathogenic fungi for 12day on rice bran extract(1:8, w/v) incubated in ceramic matrix at 28℃. Natural zeolite ceramic ball was high production of 4.2×108 conidial/mL. The culture condition optimized initial pH, temperature, rice bran extract concentration, adhesives substance and concentration of NaCl, respectively. The high production of spore optimal conditions were temperature 28℃, initial pH 3, rice bran extract 3 mL, starch 33 g, 5 % NaCl and sopre suspension 7 mL, respectively.CONCLUSION: This study was carried out for the mass production of entomopathogenic fungi conidia recover rate 65% in matrix of natural zeolite ceramic ball, and to develop ingredient-used formulation of Beauveria bassiana M130 conidia for biological control agents.

Investigation of Microbial Contamination the Level in Fish Farms of Jeju East Coast. (제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사)

  • Level of microbial contamination was examined in four fish farms of Jeju east coast for sanitary indication of bacterial contamination such as heterotrophic bacteria, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus and total coliforms. The samples were collected and investigated from June to October in 2007. Bacterial species of S. aureus, B. cereus and Salmonella spp. were frequently detected from the seawater of above fish farms. Salmonella spp., S. aureus, B. cereus were detected in the range of $0{\sim}4.3{\times}l0^3,\;0{\sim}2.7{\times}l0^4,\;0{\sim}7.6{\times}l0^3$ CFU/ml, respectively, from the fish feed used in all four selected farms. Additionally, Salmonella spp., S. aureus, B. cereus were detected in oliver flounder (Paralichthys olivaceus) of these farms, however total coliforms and E. coli O157:H7 were not detected during our experimental period. For the production of microbiologically safe and healthy oliver flounder, proper quality control of feed, sanitation programs, and continuous monitoring of microorganism are essential practices, which required to include in the farm management system.

Effect of lighting regimes on amino acid and fatty acid contents of broiler chicken meat (점등체계가 육계 닭고기의 아미노산과 지방산 조성에 미치는 영향)

Lysine content of male chicken meats at T2 treatment was lower compared to that of other treatments (p<0.05). There was no significant difference in all nonessential amino acid (AA) among treatments. Methionine content of female chicken meats at T2 treatment was lower compared to that of other treatments (p<0.05). The rate of C16:0, C16:1n7, and C18:1n9 of male chicken meat at T2 treatment was higher than that of other treatments (p<0.05), while C18:2n6 was lower at T2 treatment than other treatment (p<0.05). Mono-saturated fatty acid was the highest at T2 treatment, however, polysaturated fatty acid was the lowest at that treatment (p<0.05). There was no considerable difference in fatty acid composition of female chicken meat among treatments. Finally, lighting regime didn' affect on amino acid and fatty acid contents, and used possibly on energy reduction.

Effect of Salicornia bigelovii Extract on the Activities of Whitening and Anti-wrinkle (추출용매에 따른 함초 추출물의 피부미용 효과)

  • This study investigates the effect of Salicornia bigelovii extract on the activities of antioxidant, anti-wrinkle and whitening. Among amino acids, the glutamic acid concentration was the highest with an order of proline > alanine > $\gamma$-amino-n-butyric acid > arginine. The total polyphenol concentration and antioxidant activity were an order of ethyl acetate extract > n-butanol extract > hexane extract > hot water extract > ethanol extract. Especially, when ethylacetate was used as extractant, the total polyphenol concentration and antioxidant activity were 31.6 mg/mL and 82.6%, respectively. The inhibition activity of tyrosinase was increased with the increase concentration of ethylacetate extract. Especially, when the extract concentration was increased from 20 to 100 mg/mL, it was raised from 15.2 to 98.2%. In the case of inhibition activities of elastase, and collagenase, they were also increased with the increase concentration of ethylacetate extract. When the extract concentrations were increased from 20 to 100 ug/mL, they were enhanced from 2.9 to 32.5% and from 3.8 to 29.7%, respectively. The results show that the ethyl acetate extract of Salicornia bigelovii can be applied as sources for functional cosmetics and pharmaceuticals.

Effect of Monounsaturated Fatty Acid-enriched Peanut Consumption on Serum Lipid in High Fat Diet-induced Mice (고지방 식이 유도 비만 마우스에서 고올레산 땅콩 섭취의 혈장 지질 개선 효과)

  • This study was conducted to evaluate the effect of high-oleate and normal-oleate peanut consumption on adipose mass and serum lipids in obese-induced C57BL/6J mice. After four weeks of the high-fat diet, mice were randomly divided into six groups: normal control (NC) diet, high-fat control (HFC) diet, high-oleate peanut-seed (HOPS) diet, normal-oleate peanut-seed (NOPS) diet, high-oleate peanut-oil (HOPO) diet, and olive-oil (OO) diet. After four weeks, all four experimental diet groups showed significantly lower body weight and epididymal fat weight than HFC group. In four experimental diet groups, serum triglycerides (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were significantly lower, and high-density lipoprotein cholesterol (HDL-C) was significantly higher than HFC group. TG was significantly decreased in HOPS group (92.1±1.2 mg/dL) than NOPS group (101.7±5.3 mg/dL, p<0.05). Similarly, LDL-C was significantly lower in HOPS group (66.1±2.8 mg/dL) than NOPS (76.9±1.5 mg/dL, p<0.05), on the other hand, HDL-C indicated a significant elevation in HOPS (50.5±2.1 mg/dL) than NOPS group (45.2±1.6 mg/dL, p<0.05). This result suggests that the consumption of high-oleate peanut has a favorable effect on the plasma lipid profile.

The Effect of Irrigation Control and the Application of Soil Ameliorators on Cadmium Uptake in Paddy Rice (수도의 카드뮴 흡수이행에 대한 물관리 및 개량제 시용효과)

  • In order to identify the effect of cadmium uptake by soil ameliorators in rice plants, compost, lime, silicate, and phosphate were treated. Plants were grown with irrigation water containing $0.01mg\;l^{-1}$ cadmium and treated with two irrigation systems, intermittent irrigation and continuous submersion. Soil pH was higher at the panicle formation stage compared to other growing stages. However, Eh value was decreased as growing stage was proceeded. Eh values measured at the continuous submersion plots were lower than those at the intermittent irrigation plots. Cadmium contents in the both leaves and roots of rice plants grown at the continuous submersion plots were lower than intermittent irrigation plots in the middle of growing season. Among the continuous submersion plots, cadmium uptake was highly reduced at the both silicate and lime treated plot compared to other treatments. In the harvest season, cadmium contents in the both leaves and brown rice were higher at the intermittent irrigation plots than the continuous submersion plots. Cadmium uptake was highly reduced at the compost and lime mixture plot compared to other treatments among the continuous submersion plots. Soil pH was negatively correlated with the cadmium contents of the both shoot and brown rice while Eh was positively correlated with those. The cadmium content of shoot was positively correlated with that of brown rice in the harvest season.
