Sperm Component Inducing 2nd Polar Body Extrusion in Mouse Oocytes (생쥐 난자의 제2극체 방출을 유발하는 정자 성분)

  • This study was carried out to elucidate whether sperm contain a factor inducing second polar body extrusion and to search for an effective collection method of the sperm factor Thus, sperm extract, dialyzed sperm-extract or liquid chromatographic fractions of sperm extract was microinjected into ovulated oocytes. And the microinjected oocytes were incubated for 24 hours to investigate about the extrusion of second polar body. The results obtained were as follows; 1. Sperm extract significantly increased the second polar body extrusion. 2. Sperm extract showed five major fractions at retention volumes (RVs) 1.25, 1.37, 1.84, 2.10 and 2.67ml after separation with Superose 12 column. These sperm extract fractions did not significantly increase the second polar body extrusion. 3. Dialyzed sperm-extract significantly increased the second polar body extrusion 4. Dialyzed sperm-extract showed three maior fractions at RVs 1.88, 2.14 and 2.77ml after separation with Superose 12 column. Of these fractions, the fraction RV2.14 significantly increased the second polar body extrusion. In conclusion, sperm extract contained a factor inducing the second polar body extrusion and the factor was contained largely in fraction RV2.14 after dialysis and liquid chromatographic fractionation of sperm extract.

Effect of Yam Extract on Body Weight Levels and Serum Lipid Profiles in C57BL/6J Mice Fed a High Fat Diet (마 추출물이 고지방식이로 유도된 C57BL/6J 마우스의 체중 및 혈청지질 수준에 미치는 영향)

  • $Dioscorea$ $batatas$ Decne is a type of yam that eaten raw and used as a traditional oriental medicine in Asian countries. In this study, we evaluated the functional effects of yam water extracts on body weight levels and serum lipid concentrations in mice fed high fat diet. Mice were divided into four groups: normal diet control (ND), high fat diet control (HFD), HFD+yam extract 100 mg/kg (HFD-Y100), and HFD+yam extract 200 mg/kg (HFD-Y200). Yam extract was administrated orally to mice fed a high fat diet for 5 weeks. Treatment with yam extract significantly reduced body weight levels and energy efficiency in a dose-dependent manner in HFD-fed mice. Yam extract also attenuated serum levels of triglyceride, total cholesterol, and LDL-cholesterol, as well as organ weights of liver and abdominal adipose tissue in mice fed a high fat diet. Moreover, blood levels of glucose, insulin, and leptin significantly decreased in a dose-dependent manner upon administration of yam extract. Blood activities of GPT, GOT, and LDH were lower in the yam extract-treated groups compared to the HFD group. These results indicate that yam water extract may reduce elevated body weight and serum lipid concentrations in mice fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and hyperlipidemia.

Subacute Oral Toxicity of the Methanol Extract from Phellinus pini in Rats

  • The present investigation evaluated the safety of the methanol extract from the fruit body of Phellinus pini Ames (PPA) by determining its potential toxicity after a subacute administration in rats. The extract was orally administered in doses of 1 g/kg, 2 g/kg, and 4 g/kg daily for 14 days to rats. Body weight, biochemical, and hematological parameters were determined at the end of 14 days of daily administration. The no-observed adverse effect levels (NOAEL) of the extract were 4 g/kg, when given by gavage routes. Daily oral administration of PPA extract for up to 14 days did not result in the death of significant changes in the body weight, hematological, and mainly biological parameters. In biological analysis, some significant changes occurred, including triglyceride and blood urea nitrogen (BUN), indicating that the PPA extract has liver and kidney-modulating activity. The PPA extract was found to be low or non-toxic in rats.

Effects study of Aconiti Lateralis Radix Preparata extract on the regulation of heat and cold in PTU-induced hypothyroidism rats (포부자추출물의 갑상선기능저하증 흰쥐모델에서의 한열조절작용에 의한 개선효능 연구)

  • Objective : To suggest a scientific evidence of Aconitum carmichaeli Debx. (Aconiti Lateralis Radix Preparata: ALRP) as one of cooling and heating medicines on the regulation of body temperature, we investigated the effects of ALRP water extract on hypothyroidism. Methods : Hypothyroidism was induced by intradermal injection with PTU for 4 weeks in SD rats. ALRP extract or L-thyroxine as a control drug was orally administrated for 2 weeks with PTU injection in rats. The physiological and serological parameters were measured in rats. The histological change of thyroid tissues was observed by H&E staining, and also the expression of thermo-regulating proteins was determined by Western blot in dorsal root ganglia and brain tissues of rats. Results : The administration of ALRP extract in PTU-induced hypothyroidism rats was significantly increased body temperature, but did not changes on body weight, food and water intake. ALRP extract did not effect on the levels of TSH and T4 in the hypothyroidism rats. ALRP extract significantly decreased the levels of GPT, glucose, HDL-cholesterol, LDL-cholesterol, and total cholesterol in the hypothyroidism rats. In histological observation, the enlarged epithelium and atrophic follicles with higher concentration of follicular cells on hypothyroidism were improved by ALRP extract. In addition, ALRP extract increased the expression of TRPV1 and TRPM8 ion channel proteins in hypothyroidism rats. Conclusion : These results indicate that ALRP extract can improve PTU-induced hypothyroidism through regulation of body temperature and lipid accumulation. The action mechanism of ALRP extract is related with body temperature control by thermoregulation with TRP ion channels.

The Effect of Red Ginseng Ethanol Extract on the Immunotoxicity of Diethylstilbestrol in ICR Mice (마우스에 있어서 Diethylstilbestrol의 면역독성에 미치는 홍삼 Ethanol 유출물의 영향)

  • The effect of red ginseng ethanol extract on the immunotoxicity of diethylstilbestrol (DES) was studied in ICR mice. ICR male mice were divided into S groups (10 mice/group), and red ginseng ethanol extract (50, 100 and 200 mg/kg body wt., respectively) and DES (1 mg/kg body wt.) were injected intraperitoneally (i.p.) to ICR mice once a day for 2 weeks. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune response were evaluated by humoral immunity, cell-mediated immunity, non-specific immunity, and circulating leukocyte counts. The results of this study were summarized as followings: 1. The DES-treated control group as compared with normal group showed the tendency to decrease body weight rate and relative liver weight, decreased both humoral and cellular immune responses, phagocyte activity, and circulating leukocyte counts, but increased the natural killer (NK) cell activity. 2. Compared with the DES-treated control group, DES plus red ginseng ethanol extract-treated groups significantly decreased the body weight rate (P<0.01). Relative liver weight was significantly decreased in DES plus red ginseng ethanol extract (50mg/kg)-treated group (P<0.01), but significantly increased in DES plus red ginseng ethanol extract (100mg/kg)-treated group (P<0.01). Relative spleen and thymus weights were significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (200 mg/kg)-treated group (P<0.01). 3. Both humoral and cellular immune responses were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. 4. Phagocyte activity and circulating leukocyte counts were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. NK cell activity was significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (50 and 200 mg/kg)-treated groups (P<0.01).

Physicochemical Characteristics and Antioxidant activity, Antimutagenicity, and Cytotoxicity of Hot-water Extract of Hericium erinaceus (노루궁뎅이 버섯 열수 추출물의 이화학적 특성 및 항산화성, 항돌연변이성, cytotoxicity 분석)

  • The physicochemical characteristics and biological activities, including antioxidant activity, antimutagenicity, and cytotoxicity of hot-water extract of fruiting body of Hericium erinaceus, were investigated in this study. Hot-water extract of fruiting body of Hericium erinaceus contained carbohydrate (7.86%), protein (10.91%), and ${\beta}$-glucan (3.62%). Water solubility of hot-water extract was 42.58%. Antioxidant activities of the extract were evaluated by ABTS assay and FRAP assay. The $IC_{50}$ value was 312.21 ${\mu}g/mL$ in ABTS assay. Antimutagenic activity of the extract was evaluated by Ames test. Antimutagenicity of hot-water extract (5 mg/mL) on Salmonella Typhimurium TA100 mutagenated by sodium azide (0.15 ${\mu}g/mg$) was 69.2%. Cytotoxicity of hot-water extract was also evaluated by MTT and SRB assay. The cytotoxicity was highest (83.95%) on Hep3B treated with 2,000 ${\mu}g/mL$ of hot-water extract in SRB assay. Therefore, it is suggested that hot-water extract of fruiting body of Hericium erinaceus has high antioxidant activity, antimutagenicity, and cytotoxicity.

Effects of full body meridian massage using Rubus coreanus Miquel and Hovenia dulcis thunb extract

  • The purpose of this study was to verify the effect of dietary treatment of Hovenia dulcis thunb extract and Rubus coreanus Miquel extract combined with full-body nerve massage on fatigue recovery. The results are as follows. Homogeneity between groups was secured due to the general characteristics of the study subjects, and the abdominal group showed a significant decrease in fatigue and pain changes after full nerve massage. Changes in fatigue substances in the blood showed a significant decrease in the Hovenia dulcis thunb extract group, and changes in blood levels before and after the stress hormone experiment showed a significant decrease in cordicol in the Hovenia dulcis thunb extract group. Through the results of this study, we were able to confirm that oxidative stress was reduced in adult women in their 20s to 50s through a full-body nerve massage after eating Rubus coreanus Miquel extract and Hovenia dulcis thunb extract, using an objective and systematic experimental method for changes in oxidative stress.

Effect on Promoting Gastrointestinal Function and Inhibiting of Decreasing Body Temperature of Ginger Extracts(Zingiber Officinale) (생강(生薑) 추출물의 위장관 기능개선 및 체온저하 억제효과에 대한 실험적 연구)

  • This study was performed to investigate the effect of promoting gastrointestinal function and inhibiting of decreasing body temperature of ginger extract(Zingiber officinale) in rats. In order to elucidate the gastrointestinal function and inhibiting effect of body temperature of native ginger and improved ginger, water extracts of ginger were orally administrated into rats. The results are as follows: The gastrointestinal transit time was significantly decreased in native ginger(7.66hrs) and improved ginger(7.72hrs) extract administrated groups compare to control group(8.44hrs). The mean red faecal weight was increased in native ginger(30.6%) and improved ginger(31.1%) extract administrated groups compare to control group(24.9%) for 24hrs. Inhibiting effect of decreasing body temperature induced by serotonin was increased in native ginger($1.116^{\circ}C$) and improved ginger($1.416^{\circ}C$) extract administrated groups compare to positive control group($0.384^{\circ}C$) during 40 minutes. Gastrin and CGRP immunoreactive density was more strongly expressed in native ginger and improved ginger extract administrated groups compare to control group. Serotonin immunoreactive density was more weakly expressed in native ginger and improved ginger extract administrated groups compare to control group. These results suggest that ginger extracts may enhance physiological activity such as gastrointestinal motility, protection of mucosa and gastric acid secretion in gastrointestinal tracts, and inhibits decreasing body temperature

Effects of Green Gram Extract on Skin Lesion and Histopathological Changes in Mice with Contact Dermatitis (녹두 추출물이 생쥐에 유발된 접촉피부염의 피부 증상과 조직병리학적 소견에 미치는 영향)

  • Objectives : Green gram (mung bean) has a cold nature and has been known to detoxify against various side effects that belong to hot in nature. In particular, since it has the effect of detoxifying fever and detoxification to treat swelling, it was also used externally to treat febrile dermatological diseases such as erysipelas and rubella. This study was designed to determine whether green gram exhibits anti-inflammatory effects on contact dermatitis in mice. Methods : We investigated the effects of green gram extract (70% ethanol extract) on skin lesion, skin thickness and weights, melanin and erythema index and spleen body weight ratio in mice with contact dermatitis induced by repeated application of 1-Fluoro-2,4-dinitrobenzene. Results : Topical application of green gram extract ameliorates skin lesions of contact dermatitis such as scale and roughness induces by 1-Fluoro-2,4-dinitrobenzene. green gram extract also suppressed enlargement of skin thicknesses and weights significantly. In addition, green gram extract treatment also lowered erythema index significantly compared to those in the control group. In the histopathological observation, green gram extract prevented epidermal hyperplasia and hyperkeratosis in inflamed tissues. Finally, green gram extract did not affect changes in body weights and the spleen body weight ratio, unlike dexamethasone, which significantly prevented body weight gain and lowered the spleen body weight ratio. Conclusions : These results imply that green gram, which is known to have a detoxifying effect in Korean medicine, can be used in the treatment of contact dermatitis.

Effect of Green Coffee Bean Extract Supplementation on Body Fat Reduction in Mildly Obese Women (경도비만 여성에서 생커피두 엑기스의 섭취에 의한 체지방 감소 효과)

  • In previous studies, we performed joint animal studies and clinical trials between Yonsei University and Oryza Oil & Fat Chemical Co. Ltd. We have shown that coffee bean extract has potent anti-obesity and hypotriglyceridemic activities as well as beneficial effects on body fat reduction.In this study, the effects of coffee bean extract (100 mg/capsule) on body fat reduction were evaluated in overweight/obese women (body mass index of 25~30 $kg/m^2$ or body fat > 30%) not diagnosed with any type of disease. Subjects were randomly assigned to a coffee bean extract group (n=10) or placebo group (n=10). We measured anthropometric parameters, abdominal fat distribution by computed tomography and blood components before and after the 8week intervention period. After supplementation, the coffee bean extract group showed body weight (p=0.08), body mass index (p=0.06), hip circumference (p<0.05), and upper waist circumference (p< 0.01). In addition, after 8 weeks, the coffee bean extract group showed a significant decrease in abdominal internal fat area compared to 0 weeks (0 weeks : $155.8cm^2$; 8 weeks : $145.9cm^2$, ${\Delta}$ change : $-9.9cm^2$, respectively). However, there were no significant differences in lipid profiles or serological measurements between the coffee bean extract group and placebo group. The results of our human study demonstrated that coffee bean extract supplementation for 8 weeks has beneficial effects on reducing abdominal internal fat area as well as hip and waist circumferences.
