  • The elution of di(2-ethylhexyl) phthalate (DEHP) from flexible bags into human blood or transfusion was studied. The conditions of determination of DEHP using high performance liquid chromatography (HPLC) was established as follows: Condition I-column, ${\mu}-Bondapak^{TM}\;C_{18}$; mobile phase, methanol: water=91 : 9 ; flow rate, 1.2ml/min; wavelength, 254nm; injection volume, $10{\mu}l$. Condition II-column, Lichorsorb RP-18$(10{\mu}m)$; mobile phase, methanol: water=94 : 6 ; flow rate, 1.1ml/min; wavelength, 254nm; injection volume, $10{\mu}l$. DEHP was found to be migrating from PVC blood and total parentral nutrient bags into methanol, but not into anti, coagulant drug solution.

과량의 부추 생즙 투여가 소형견의 혈액상에 미치는 영향 (Hematological Effect of Administration Excessive Amount of the Korean Native Chinese Chive Extracts in Small Dogs)

  • This study was conducted to observe the hematological effect of the small dogs when administrated excessive amount of the Korean native chinese chive(Allium tuberosum Rottler) extracts for a short period. In this experiment, clinically healthy dogs(n = 10, average weight = 4.17 kg) were used. They were inserted stomach tube and administrated the extracts(5ml/kg) for 7 days. The followings are the result of this experiment. The red blood cell counts, hemoglobin concentration and mean corpuscular hemoglobin concentration were significantly decreased. (P<0.05) The packed cell volume was slightly decreased. The mean corpuscular volume and methemoglobin concentration were significantly increased.(P<0.05) The white blood cell counts and reduced glutathione were slightly increased. Consequently, We determined that administrated excessive amount of the Korean native chinese chive extracts induced hemolytic anemia.

음곡에 시술한 비해약침이 Lipopolysaccharide로 유도된 흰쥐의 신장염에 미치는 영향 (The Effect of Tokoro Rhizoma Pharmacopuncture at $KI_{10}$ in Lipopolysaccharide Induced Acute Nephritis in Rats)

  • Objectives : This study was designed to evaluate the effects of Tokoro Rhizoma pharmat $KI_{10}$ in nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were divided into 4 groups and 3 groups(LPS, saline, Tokoro Rhizoma pharmacopuncture(TR-P) group) were injected LPS to induce nephritis. TR-P group was treated with TR at $KI_{10}$ three times for a week, saline group with normal saline. To evaluate the effects of TR at $KI_{10}$ on nephritis in rats, white blood cell(WBC), neutrophil in blood, creatinine, cytokine-induced neutrophil chemoattractant-1(CINC-1) in serum and urinary volume, creatinine in urine, renal myeloperoxidase(MPO) were measured and renal tissue was analyzed. Results : TR-P group significantly reduced WBC, neutrophil in blood, creatinine, CINC-1 in serum, creatinine in urine and renal MPO than LPS group. TR-P group increased urinary volume but, not significant. Conclusion : TR at $KI_{10}$ has a therapeutic effect on nephritis in LPS stimulated rat. Therefore, it is suggested that TR at $KI_{10}$ may be an useful therapeutics for nephritis in clinical field after further researches.

인공폐(산화기) 제작과 실험 (Design & Animal Experiment of Artificial Oxygenator)

  • We have designed a new type of bubble oxygenator (KOREA-KIM VENOTHERM OXYGENATOR) made of PVC sheet and deforming mesh incorporated in the heat exchanger, and evaluated in experimental animal for the analysis of it`s efficiency. The Oxygenator has low priming volume with high flow rate up to 6 L/rain, and efficiency of heat exchanger was excellent as 1-$1.5^{\circ}C.$ using total cardiopulmonary bypass method under moderate to deep hypothermia. Average priming volume of 1317 ml with 30% hemodilution method was perfused with an average of 1.1-3.0 L/min.$M^2$of arterial blood and pure oxygen at a rate of 2-3.4 L/min for 49.6 minutes continuously in average. During total cardiopulmonary bypass, average $PaO_2$ was $159.8{\pm}60$mmHg, $PaCO_2$ $41.0{\pm}3$mmHg respectively under $SaO_2$ over 96% with systolic arterial pressure of 70 mmHg and CVP of 5-10 cm$H_2O$. Plasma free Hemoglobin was $7.0{\pm}4$ mg/dl with 25% drop of hemoglobin and hematocrit at the end of cardiopulmonary bypass. This KKV Oxygenator was observed to have excellent capabillty of oxygen and carbon dioxide gas transfer with small amount of blood trauma, and the efficiency of heat exchanger was satisfactory during cooling and rewarming of the bubbled blood. Disadvantages have included the somewhat poor deforming effect due to loose PVC fiber mesh, the extracompact character of Teflon filters, and the rough inner surface of the heat exchanger copper pipes.

Experimental Study on Cerebral Hemodynamics during Observation of Plants

  • Psychological and physiological effects of plants were studied by investigating human responses while observing plants. Eighteen healthy adult male(aged between $19{\sim}25$ years) participated in this study. Semantic differential method(SD method) and multi-channel near-infrared spectroscopy(NIRS) were used to survey verbal and non-verbal response, respectively. Cerebral hemodynamics as a new evaluation index of brain activity was recorded for right brain hemisphere where visual information is mainly delivered. Thirty seconds of cerebral blood flow in forty seven channels were calculated when watching five types of picture images with different rates of hedge against gray block wall; 0:10, 3:7, 5:5, 7:3, 10:0. In the SD results, similar evaluations were found in all subjects. However, the change of cerebral hemodynamics as a non-verbal response varied among subjects. Largely two patterns of hemodynamics change were found with increasing plants rate in picture images; group A showed significant decreases of blood flow volume in many cortical regions, Group B had significant increase of blood flow volume in the occipital region for the scenes seen comparatively more plant. Our findings on the cerebral hemodynamics may indicate that there are two patterns of brain activity during observation of plants; group A in which brain areas associated with visual information and thinking work simultaneously to the visual stimuli; group B in which brain areas associated only with visual information work.

Daily Amperometric Monitoring of Immunoglobulin E in a Mouse Whole Blood: Model of Ovalbumin Induced Asthma

  • There is an increasing interest in monitoring of specific biomarker for determining progression of a disease or efficacy of a treatment. Conventional method for quantification of specific biomarkers as enzyme linked immunosorbent assay (ELISA) has high material costs, long incubation periods, requires large volume of samples and involves special instruments, which necessitates clinical samples to be sent to a lab. This paper reports on the development of an electrochemical biosensor to measure total immunoglobulin E (IgE), a marker of asthma disease that varies with age, gender, and disease in concentrations from 0.3-1000 ng/mL with consuming 20 µL volume of whole blood sample. The sensor provides rapid, accurate, easy, point-of-care measurement of IgE, also, sequential monitoring of total IgE with ovalbumin (OVA) induced mice is another application of sensor. Taken together, these results provide an alternative way for detection of biomarkers in whole blood with low volumes and long-term ex-vivo assessments for understanding the progression of a disease.

한국재래(韓國在來) 염소의 체액량(體液量) (Body Fluid Volumes of Korean Native Goats)

  • In order to approximate plasma volume, extracellular fluid volume and total body water volume of Korean native goats, measurements were made of the volumes of distribution of Evans blue, potasium thiocyanate and antipyrine. The results obtained in this work were summarized as follows: 1. Plasma volume showed a range of 50 to 72ml/kg with a mean of $62{\pm}6.2ml/kg$ (SD). 2. Blood volume showed a range of 69 to 98ml/kg with a mean of $85{\pm}7.9ml/kg$. 3. Extracellular fluid volume showed a range of 265 to 310ml/kg with a mean of $297{\pm}18.3ml/kg$. 4. Interstitial fluid volume showed a range of 204 to 261ml/kg with a mean of $236{\pm}16.8ml/kg$. 5. Intracellular fluid volume showed a range of 380 to 436ml/kg with a mean of $420{\pm}12.6ml/kg$. 6. The volume of total body water showed a range of 680 to 735ml/kg with a mean of $714{\pm}17.7ml/kg$.

초음파상 비전형적 소견을 보이는 간혈관종의 적혈구 표지 스캔소견 ($^{99m}Tc-Red$ Blood Cell Scintigraphy of Sonographically Atypical Hemangioma)

  • We evaluated the scintigraphic findings of sonographically atypical hemangioma, that is, a hypoechoic or heterogeneously hypoechoic nodule which contains hypoechoic areas more than 40% of the volume, and compared it with those of sonographically typical hemangioma in 26 patients with 31 nodules. Sonographically atypical hemangioma were 13 in 11 patients and sonographically typical hemangioma were 18 the 15 patients. Dynamic blood flow, planar and SPECT blood pool imaging was performed. In atypical hemangioma, increased blood pool activity was seen in 11 of 13 nodules on SPECT and in 10 of 13 nodules on planar image and increased blood flow was seen in 4 of 13 nodules on dynamic blood flow study. No significant difference in detecting increased blood pool activity between sonographically atypical and typical hemangioma. In conclusion, $^{99m}Tc-red$ blood cell SPECT is as useful in diagnosis of sonographically atypical hemangioma as in typical hemangioma and can be used as a confirmatory or complementary study.

운동선수들의 혈액분석을 통한 Etrythropoietin 간접도핑검사 (Blood Analysis for Indirect Doping Control of Erythropoietin in Sports)

  • The use of recombinant human erythropoietin (rhEPO), a stimulator of erythropoiesis, banned in sports because of the medical risk associated with thrombosis. Due to analytical difficulties to differentiate between natural human EPO (hEPO) and rhEPO, blood parameters of erythropoiesis such as contents of hemoglobin (cut-off value <17.5 g/d l for man, and < 16.0 g/dl for women), hematocrit and reticulocytes (cut-off value <2.0%) were measured to focus the misuse of rhEPO. We conducted anti-doping test for 122 blood samples of the World Cup athletes. The mean values of key parameters are as follows; 14.5$\pm$1.0 g/dl for hemoglobin, 41.7$\pm$2.8% for hematocrit, and 1.3$\pm$0.4% for reticulocyte. Blood sample was found to be stable up to 8 hours for the reticulocyte measurement. In addition, the soluble transferrin receptor and ferritin levels were measured by immunoassay methods using plasma samples (n=28) in which the mean value was 0.8$\pm$0.5 $\mu\textrm{g}$/$m\ell$ and 54.6$\pm$33.7 ng/$m\ell$, respectively. The results indicate that all samples tested were negative for the blood parameters of indirect anti-doping test for hEPO misuse. The statistical evaluation suggest that several other parameters such as red blood cell, mean corpuscular hemoglobin concentration, mean corpuscular volume, mean corpuscular hemoglobin and white blood cell could be considered as factors influencing hEPO function in addition to five parameters mentioned.

Effect of Glucose-Sweetened Drinks on Blood Glucose, Energy, and Water Intake at a Meal 3h Later in Healthy Males

  • The aims of this study were to describe the effects of glucose-sweetened drinks on blood glucose, energy, and water intake at a meal 3 hours later. The effect of blood glucose on prandial energy intake and the relationship between water and energy intake during a meal were also determined. Twenty healthy normal-weight men were fed pizza test meals 3h after consuming four drinks of 0, 50g, 65g, and 75g glucose in random order, within-subjects design. Blood samples were measured at baseline and every 30 min after ingestion of drinks and 30min after the end of the test meal and the appetite was also assessed by visual analog test at the same interval. The results of this study showed that various glucose drinks altered blood glucose responses compared with that of water control(p<0.0001). Blood glucose areas under the curve(AUC) for glucose-sweetened drinks were significantly(p<0.05) higher than that for the control over 3 hours after a drink and 30 min after the test meal. Consumption of the glucose-sweetened drinks significantly increased(p<0.05) energy and water intake at a test meal compared with the water control, except the drink containing 75g glucose. For all drinks combined, the energy intake was negatively correlated with the blood glucose and positively correlated with the volume of water consumed at a test meal at 3 hours later.
