Immunomodulatory Potential of Weissella cibaria in Aged C57BL/6J Mice

  • Aging is associated with distinct changes in immune cells and a decline in immune function, leading to increased susceptibility to infection and reduced responses to vaccination. Certain strains of lactic acid bacteria exert beneficial effects on the immune system. Previously, we reported that Weissella cibaria JW15 isolated from kimchi possesses immune stimulatory activity in vitro. In the present study, we further investigated whether oral administration of JW15 improves immune function in aged mice. Eighteen-month-old female mice were administered JW15 daily at low (JW15-L; $1{\times}10^8CFU/mouse$) or high dosage (JW15-H; $1{\times}10^9CFU/mouse$), or with Lactobacillus rhamnosus GG (LGG) using oral gavage. Two-month-old female mice were included as healthy young mice. After 4 weeks, the mice were euthanized and immune profiles were analyzed using whole blood and spleen. In complete blood count analysis, the numbers of white and red blood cells were significantly increased in the JW15-L group compared with those in the old mouse (OM) control group. In addition, administration of either JW15 of LGG resulted in higher numbers of splenocytes in comparison with the OM group. Furthermore, proliferative potentials were higher in all probiotic groups than OM. Cytokines such as IFN-${\gamma}$ and IL-6 were secreted at higher levels in splenocytes isolated from JW15-fed mice than in OM control mice. Similarly, mRNA expression of various cytokines was altered in the JW15 groups. Collectively, these results suggest that JW15 supplementation induces immunomodulatory effects in aged mice and indicate JW15 as a potential probiotic strain to improve immune function in aged animals.

Stress response of olive flounder, Paralichthys olivaceus and Japanese croaker, Nibea japonica on changes of water temperature

  • In this study, we investigate the physiological response of olive flounder, Paralichthys olivaceus and Japanese croaker, Nibea japonica, on a series of rising water temperatures (WT). At 1, 9, and 17 days, WT increased from 23℃ (control) to 26, 29 and 32℃ within 6 hours, respectively, and then maintained for 96 hours. At 5, 13, and 21 days, WT decreased from each WT to 23℃ within 6 hours and was maintained at 23℃ for 96 hours. Blood were collected at 0, 1, 5, 9, 13, 17, 21, and 60 days. In both species, hematocrit, red blood cell and hemoglobin of stressed groups were lower than non-stressed group at 13 days. The osmolality of stressed group was lower than non-stressed group at 21 and 60 days. In olive flounder, hemoglobin of stressed groups was lower than non-stressed group at 21 and 60 days, respectively, but osmolality of stressed group was higher than non-stressed group at 9 days. In Japanese croaker, hematocrit, red blood cell and hemoglobin of stressed groups were higher than non-stressed group at 21 and 60 days, respectively, and osmolality of stressed group was higher than that of non-stressed group at 1 day. At 1, 17, and 21 days, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) of stressed group were higher than non-stressed group in olive flounder. In Japanese croaker, AST and ALT of stressed group was lower than non-stressed group at 17, 21, and 60 days. The cortisol of olive flounder increased with increasing WT, but those of Japanese croaker increased with decreasing WT. In both species, the glucose increased with increasing WT. After changing WT, lactic acid of olive flounder and Japanese croaker were not significantly different. The physiological response of olive flounder increased directly to the increase in WT, and Japanese croaker contrasted with olive flounder.

Feasibility of sodium long chain polyphosphate as a potential growth promoter in broilers

  • The objective of this study was to evaluate in vitro antimicrobial and anti-biofilm activity of sodium long chain polyphosphate (SLCPP) and effect of dietary supplementation of SLCPP on growth performance, organ characteristics, blood metabolites, and intestinal microflora of broilers. Antimicrobial activities of SLCPP were observed against Escherichia coli O157:H7, Listeria monocytogenes, Salmonella enterica ser. Pullorum, Shigella sonnei, Klebsiella pneumonia, Pseudomonas aeruginosa in agar well diffusion assay. In addition, SLCPP demonstrated good anti-biofilm activity against K. pneumonia and P. aeruginosa. Furthermore, to investigate the dietary effect of SLCPP, a total of 480 1-day-old male Ross 308 broiler chicks were randomly allotted to three dietary treatment groups (4 replicates per group, 40 birds in each replicate): an antibiotic-free corn-soybean meal basal diet (NC); basal diet + enramycin 0.01% (PC); and basal diet + 0.1% SLCPP (SPP). The experiment lasted for 35 days. Results showed that birds fed with SLCPP had higher body weight (BW) and average daily gain (ADG), and lower feed conversion ratio (FCR) during the grower phase (days 7 to 21) (p < 0.05). Except for blood urea nitrogen, all other blood biochemical parameters remained unaffected by the dietary supplementation of SLCPP. Compared to the control group, lengths of the duodenum and ileum in the SPP group were significantly shorter (p < 0.05). Moreover, counts of lactic acid bacteria (LAB), total aerobes, and Streptococcus spp. in jejunum as well as LAB in cecum were increased in the SPP group than in the PC group (p < 0.05). These results suggest that dietary supplementation of SLCPP might promote the growth of broilers in their early growth phase.

고령자용 피트니스 압박웨어 착용시의 객관적 인체생리 평가 및 주관적 만족도 평가 (Objective Physiology Evaluation and Subjective Satisfaction Evaluation when Wearing Fitness Compression Wear for the Elderly)

  • Muscle reduction owing to aging causes changes in physical function among the elderly. Fitness compression wear reduces fatigue by compressing the main muscles, expanding blood vessels, and rapidly discharging lactic acid. The effect of clothing pressure when wearing fitness compression wear for the elderly was objectively and subjectively evaluated for six women aged 55-64 years. The evaluation clothes were three types of tops and bottoms, one type of design (A) preferred among the existing compression wear, and two types of design (B and C). The objective evaluation items included clothing pressure, blood flow, surface temperature, and subjective satisfaction. It was found that clothing A had the highest clothing pressure in the straight posture and five fitness movements. Blood flow increased the most when wearing the evaluation clothing. The surface temperature was found to be the highest for clothing C, and subjective satisfaction was found to be the best when wearing evaluation clothing B. If the tensile strength was low and the tensile elongation was high, as in evaluation clothing B, it was recognized as appropriate clothing pressure, and the subject was subjectively satisfied. When manufacturing fitness compression wear in the future, it will be possible to increase users' objective and subjective satisfaction with wearing it, if the incision of the garment were placed at the location of the line of non-extension (LoNE), and appropriate material characteristics are applied.

혈당 강하 요구르트 개발에 관한 연구 (Studies on the Development of the Yogurt Decreasing Blood Glucose)

  • 혈당 강하 요구르트의 개발을 위해 Streptococcus salivarius subsp. thermophilus와 Lactobacillus acidophilus145, bifido-bacterium infantis 혼합균주에 사균인 FK-23 유산구균, 피니톨, 식이섬유, 백강낭콩 추출 분말, 에리스리톨, 바나바 추출물 등의 기능성 성분을 포함한 요구르트를 제조하여 정상인과 혈당치가 높은 당뇨환자를 대상으로 간이 임상시험을 실시하였다. 1. 정상인의 식후 혈당치는 일반 요구르트를 섭취했을 때보다 혈당 강하 요구르트 음용 시 더 낮게 측정되었으여 임상대상자 대부분에서 동일한 현상이 나타났다. 2. 혈당치가 높은 당뇨환자 10명을 대상으로 식후 혈당치변화 및 음용기간 중 혈당치 변화를 확인한 결과 혈당 강하요구르트를 음용하기 전보다 음용 후 혈당치가 식후 30분, 식후 60분, 식후 120분 각각 19 mg/dL, 30 mg/dL, 31 mg/dL 감소하였다. 또한, 혈당 증가 억제율도 임상대상자의 80%가 감소하는 효과를 나타냈다. 3. 30일간 혈당 강하 요구르트를 음용한 후 식후 혈당치의 변화를 확인한 결과 음용전보다 음용 후 120분 경과 때, 혈당치가 59 mg/dL 감소하였으며 음용기간이 증가할수록 지속적으로 감소하는 경향을 나타내었다.

Effects of dietary Antrodia cinnamomea fermented product supplementation on antioxidation, anti-inflammation, and lipid metabolism in broiler chickens

  • Objective: This study was investigated the effects of dietary supplementation of Antrodia cinnamomea fermented product on modulation of antioxidation, anti-inflammation, and lipid metabolism in broilers. Methods: Functional compounds and in vitro antioxidant capacity were detected in wheat bran (WB) solid-state fermented by Antrodia cinnamomea for 16 days (FAC). In animal experiment, 400 d-old broiler chickens were allotted into 5 groups fed control diet, and control diet replaced with 5% WB, 10% WB, 5% FAC, and 10% FAC respectively. Growth performance, intestinal microflora, serum antioxidant enzymes and fatty acid profiles in pectoral superficial muscle were measured. Results: Pretreatment with hot water extracted fermented product significantly reduced chicken peripheral blood mononuclear cells death induced by lipopolysaccharide and 2,2'-Azobis(2-amidinopropane) dihydrochloride. Birds received 5% and 10% FAC had higher weight gain than WB groups. Cecal coliform and lactic acid bacteria were diminished and increased respectively while diet replaced with FAC. For FAC supplemented groups, superoxide dismutase (SOD) activity increased at 35 days only, with catalase elevated at 21 and 35 day. Regarding serum lipid parameters, 10% FAC replacement significantly reduced triglyceride and low-density lipoprotein level in chickens. For fatty acid composition in pectoral superficial muscle of 35-d-old chickens, 5% and 10% FAC inclusion had birds with significantly lower saturated fatty acids as compared with 10% WB group. Birds on the 5% FAC diet had a higher degree of unsaturation, followed by 10% FAC, control, 5% WB, and 10% WB. Conclusion: In conclusion, desirable intestinal microflora in chickens obtaining FAC may be attributed to the functional metabolites detected in final fermented product. Moreover, antioxidant effects observed in FAC were plausibly exerted in terms of improved antioxidant enzymes activities, increased unsaturated degree of fatty acids in chicken muscle and better weight gain in FAC inclusion groups, indicating that FAC possesses promising favorable mechanisms worthy to be developed.

Different Immune Regulatory Potential of Lactobacillus plantarum and Lactobacillus sakei Isolated from Kimchi

  • It is known that lactic acid bacteria (LAB) have many beneficial health effects, including anti-oxidative activity and immune regulation. In this study, the immune regulatory effects of Lactobacillus sakei and Lactobacillus plantarum, which are found in different types of kimchi, were evaluated. L. sakei and its lipoteichoic acid (LTA) have greater immune stimulating potential in IL-12, IFN-${\gamma}$, and TNF-${\alpha}$ production as compared with L. plantarum in an in vitro condition. On the other hand, L. plantarum is assumed to repress the Th1 immune response in murine experiments. After being injected with LPS, L. plantarum-fed mice maintained a healthier state, and the level of TNF-${\alpha}$ in their blood was lower than in other bacterial strainfed mice and in the LPS-only control mice. Additionally, IL-12 production was significantly decreased and the production of IL-4 was greatly increased in the splenocytes from L. plantarum-fed mice. Further experiments revealed that the pre-injection of purified LTA from L. plantarum (pLTA), L. sakei (sLTA), and S. aureus (aLTA) decreased TNF-${\alpha}$ and IL-4 production in LPS-injected mice. Mouse IL-12, however, was significantly increased by aLTA pre-injection. In conclusion, the L. sakei and L. plantarum strains have immune regulation effects, but the effects differ in cytokine production and the regulatory effects of the Th1/Th2 immune response.

Nutritional Quality of Napier Grass (Pennisetum purpureum Schum.) Silage Supplemented with Molasses and Rice Bran by Goats

  • In order to improve silage quality and utilization of napier grass (Pennisetum purpureum Schum.) by goats, the grass was ensiled with molasses (MOL) and/or defatted rice bran (DRB). Napier grass was harvested at the growing stage in July and cut into 3 cm length. The grass was mixed with 4% MOL and/or 15% DRB, ensiled 15 kg each into plastic bags and stored for 9 months. Dry matter content of the silage ensiled with MOL (MOL-silage) was 13.4%, but increased to 20% with DRB addition. The addition of MOL decreased pH value and ammonia nitrogen content, but increased lactic acid content. MOL-silage contained about 6% spoilage, but addition of DRB decreased spoilage to less than 1%. Goats were fed the silage at a level of 2.25% (DM basis) of their body weight. Goats fed DRB- or MOL/DRB-silages maintained nitrogen retention, but goats fed MOL-silage did not. The rumen fluid of goats fed DRB-silage tended to be higher in acetic acid and lower in propionic acid than those fed the other silages. Ammonia in the rumen fluids, urea nitrogen in the blood and the urinary nitrogen excretion were the lowest in goats fed MOL/DRB-silage. As the result, the ratio of retained nitrogen to nitrogen intake was the highest in goats fed MOL/DRB-silage. In conclusion, addition of DRB to napier grass increased DM of silage and decreased the volume of spoilage. The combination of MOL and DRB can improve the fermentation quality and thus enhance the utilization of the silage by goats, more than the MOL or DRB being as a single treatment.

단위가축을 위한 생균제의 급여 효과 (Effect of Dietary Probiotics Supplementation to Feed for Monogastric Animals)

  • 생균제는 항생제와 반대의 어원적 의미를 갖는 것으로 장내 미생물 균형을 기선함으로써 숙주 동물에게 유익한 작용을 유도할 수 있는 살아있는 미생물 첨가제로서 항생제를 대체할 수 있는 물질 중 하나로서 인식되어 왔다. Lactobacillus와 같은 젖산균은 유산을 생성하여 장의 산도를 pH를 저하시켜 잡균의 오염을 방지하며, 각종 비타민, 아미나산, 항균물질 등을 생산하여 가축 장내 미생물균총의 안정, 사료효율 증가, 내병성 증대 등의 효과가 있다. 또한, saccharomyces와 같은 효모균은 각종 소화효소를 생산하여 영양소 분해를 촉진하며 알코올과 글루타민산과 같은 사료의 기호성을 증진시키는 물질을 분비한다. 단위가축에 있어 생균제의 급여효과는 돼지에 있어 증체량과 사료효율을 증가시키고 설사발생빈도를 낮춘다. 또한, 가금에 있어서는 산란계의 산란율을 개선하고, 장내 유익한 미생물의 수를 증가시키며, 육계의 증체량을 증가시키며, 혈중 콜레스테롤 수치를 낮춘다. 그러나 다른 연구에서는 생균제의 급여가 가축의 성적에 별다른 영향을 미치지 못하였다고 보고하여 이를 규명할 수 있는 연구의 진행이 이루어져야 할 것으로 보인다. 또한, 생균제의 효율적인 이용, 균주의 선발과 광합성미생물과 같은 친환경적인 생균제의 개발등도 해결되어야 할 과제일 것이다.

Probiotic Functional Dairy Foods and Health Claims: an Overview

  • The commercial interests in functional foods containing probiotics are gaining significance in view of increasing studies on their role in digestive tract. Probiotic dairy foods containing health-promoting bacteria are important segment of functional food market. Various health benefits have been attributed to specific strains of lactic acid bacteria or food containing these probiotic cultures. Probiotic-containing foods are considered to improve general gut health and natural defenses of body, and lower blood cholesterol level. Specific probiotic microbes can alleviate or prevent diverse intestinal diarrhea-inducing disorders, cause prophylaxis of intestinal and urogenital infections, inhibit mutagenicity of intestinal contents, and reduce incidence of intestinal tumors. Recent increasing evidences on health effects of probiotics have triggered consumer interest in this category of functional foods. Rational approach needs to be applied in selection of strains for probiotic preparation to achieve required functionality. Present article focuses on some prominent probiotic candidates and criteria for their inclusion in functional food sector. Various health claims of probiotics on gastrointestinal disorders, anticarcinogenic effects, and anti-cholestrimic effects, and possible mechanistic explanations for their functionality are highlighted.
