Effects of Chunghyuljihwang-tang on Phenylhydrazine-induced Hemolytic Anemia in Rats (청혈지황탕(淸血地黃湯)이 Phenylhydrazine으로 유발한 용혈성 빈혈 흰쥐에 미치는 영향)

  • Objectives: This study was performed in order to investigate the anti-hemolytic effect of Chunghyuljihwang-tang(CHJHT, Qingxuedihuang-tang) on phenylhydrazine-induced hemolytic anemia in Rats. Materials and Methods: Hemolytic anemia model of rats were induced by phenylhydrazine(PHZ, 40mg/kg/day, i.p.) for 3days. Male Sprague-Dawley rats were divided into three groups for their treatment. Normal group was injected normal saline for 3 days with distilled water(d.w.) orally, control group was injected PHZ for 3 days with d.w. and CHJHT group was injected PHZ for 3 days with administration of CHJHT for 10 days. RBC(Red blood cell), Hb(hemoglobin), mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, reticulocyte were determined on day 0, 4, 7, 10. Body weight was investigated on day 0, 2, 4, 6, 8, 10 and the weight of spleen was measured on day 10. Results: For CHJHT group, the levels of RBC and Hb were significantly higher than control group on day 4, and the concentration of hemoglobin on day 7 was increased more than control group. Moreover, reticulocyte was significantly decreased on day 10 more than control group. These results could be concluded that CHJHT suppressed the hemolytic anemia by PHZ and improved hematological values of anemia earlier.

Improvement of Chronic Bleeding in the Patient with Unresectable Advanced Gastric Cancer Using the Decoction of Notoginseng Radix - a Case Report

  • Objectives: A 74-year-old male patient with unresectable advanced gastric cancer (clinical initial stage T3N+, Borrmann type III) admitted due to gastric bleeding at tumor site. On first admission day, hemoglobin level was 5.7g/dl and performance status was grade 3 according to Eastern Cooperative Oncology Group Performance Status(ECOG-PS). After performing red blood cell transfusion as an emergency treatment, hemoglobin level was increased up to 9.5g/dl. However, bleeding of oozing site was continued. For hemostasis, decoction of notoginseng radix (30g/day) was administered since day 7 after admission. The dose was elevated to 40g/day after hemoglobin level was decreased to 6.5g/dl on day 11. Since then, melena was stopped and hemoglobin level was maintained over 9.1g/dl. This case shows the hemostasis effect of decoction of notoginseng radix on gastric bleeding in unresectable advanced gastric cancer.

Depression Trends by Gender in Diabetic Patients of the Community (지역사회 당뇨병환자 성별 우울성향)

  • Purpose: The purpose of this study was to investigate the depression trends by gender in type2 diabetic patients. Method: One hundred Participants were recruited from the endocrinology outpatient department of a tertiary care hospital in an urban city. Depression was measured using the Beck Depression Inventory. Glycosylated hemoglobin was determined by a high-performance liquid chromatography technique. Fasting plasma glucose and 2 hour post-prandial blood glucose were analyzed by the glucose oxidase method. Results: The Depression score was significantly higher in diabetic women (l0.4) than in male diabetic patients (7.8). In male diabetic patients, depression was positively correlated with glycosylated hemoglobin (r=0.35, p=0.006). However, there was no correlation between depression and glycosylated hemoglobin in female diabetic patients. Conclusion: These findings indicate that depression score was significantly higher in female diabetic patients and the depression was positively correlated with glycosylated hemoglobin in male diabetic patients.

  • The purpose of this study was to enhance gas exchange by producing hemosome, a hemoglobin microencapsulated with phospholipid of egg, and perfluorocarbone(PFC) emulsion solution. In the experiment, stable emulsion solution with 437 nm of mean particle size could be produced by Flusol-DA sonication, and shortening of emulsion time could be attained with higher stability as well. $0.8{\mu}m$ sized hemosome could be produced by microencapsulation of hemoglobin with phospholipid extracted from egg yolk. The pattern of oxygen saturation curve of hemosome was S shape, which is similar to that found in normal blood, and $P_{50}$ was measured to be 24 mmHg. The oxygen saturation in the mixed solution of hemosome and blood in 1:4(V/V%) ratio was similar to that of normal blood, and the same result was found in the mixed solution of PFC emulsion and blood in 1:4(V/V%) ratio.

Hematologic and biochemical reference intervals for natural monument Korean goral (Naemorhedus caudatus) (천연기념물 한국 산양의 혈액 및 혈액 화학적 정상표준범위)

  • Korean goral (Neamorhedus caudatus) is registered as a natural monument number 217 by South Korea Cultural Heritage Administration. It is also recognized as the endangered species I by Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). In this study, blood samples of Korean goral were collected to make reference intervals. Blood sampling was conducted on 19 numbers of Korean gorals (ten females, nine males), which were reared in Korean Goral Restoration Center. For total samples, the reference intervals of hematological parameters were: white blood cell $7.69{\sim}10.87K/{\mu}{\Lambda}$; hematocrit 36.73~46.18%; red blood cell $10.72{\sim}12.86K/{\mu}{\Lambda}$; hemoglobin 12.79~15.14 g/dL; mean corpuscular volume 33.15~36.75 fL; mean corpuscular hemoglobin 11.53~12.23 pg; mean corpuscular hemoglobin concentration 32.64-35.91 g/dL and red blood cell distribution width 39.2~43.46%. For total samples of each parameters, the following results were obtained for serum biochemistry: glucose 111.81~153.77 mg/dL; blood urea nitrogen 22.35~28.91 mg/dL; creatine 1.22~1.84 mg/dL; phosphate 4.57~6 mg/dL; calcium 8.7~9.1 mg/dL; total protein 6.53~6.92 g/dL; albumin 3.1~3.48 g/dL; globulin 3.26~3.62 g/dL; alanine aminotransferase 56.7~158.56 U/L; aspartate aminotransferase 230.35~473.06 U/L; alkaline phosphatase 178.06~332.47 U/L; gamma-glutamyl transpeptidase 131.6-~181.24 U/L; total bilirubin 1.47~2.12 mg/dL; cholesterol 46.48~71.52 mg/dL; amylase 16.3~26.03 U/L; sodium 150.43~153.88 mmol/L; potassium 3.98~4.6 mmol/L and chlorine 109.48~113.26 mmol/L. The ranges of values were similar campared to previous studies except in the case of RDW value, which showed higher range than the RDW value of a previous study. The reference intervals from this study will be useful data for treatment and management of gorals.

Effects of Yeonlyeonggobon-dan on the Blood of Arsenic-poisoned Rats

  • Sodium arsenate and Yeonlyeonggobon-dan (nianlinggubendan) extract (YGD), a herbal restorative were treated p.o. 20 mg/kg and 500 mg/kg, respectively, and concurrently to rats, and examined the biochemical parameters in blood. The values of white blood cell (WBC), red blood cell (RBC), hemoglobin (Hgb) and hematocrit (Hct) in each group did not show significant variance. The value of aspartate aminotrasferase (AST) of arsenic-treated group was increased for 2 weeks significantly while that of the group of concurrent administration with YGD became low significantly compared with arsenic-treated group and the value of alkaline phosphatase (ALP) of arsenic-treated group was decreased while that of the group of concurrent administration with YGD was increased significantly compared with arsenic-treated group. In arsenic-treated groups, the value of glucose (Glu), and those of lactic dehydrogenase (LDH), blood urea nitrogen (BUN) and triglyceride (TG) were decreased at first but increased later while the groups of concurrent administration with YGD showed significant recovery from the toxicity of arsenic.

  • The study was conducted to investigate the phenotypes of blood proteins, hemoglobin and albumin, of 104 German shepherds in Dag-gu, Yae-chum Kim-hae and Kwang-ju area by the starch gel electrophoresis. The results obtained were summarized as follows: 1. The hemoglobin phenotypes were observed three(HbBB, HbBb and Hbbb). Fast migrating bands were same and slow migrating bands were divided by heavy(HbBB), light(HbBb) and non-stained(Hbbb). The frequencies of appearance in HbBB, HbBb and Hbbb were 17.31%, 51.92% and 30.77%, respectively. 2. The hemoglobin phenotypes were controlled by two allelic genes, $Hb^b$ and $Hb^B$. The gene frequencies were calculated at 0.567 in $Hb^B$ and 0.433 in $Hb^A$. 3. The albumin phenotypes were observed three(AlbFF, AlbSS and and AlbFS), which were divided by fast migrating band(AlbFF), slow migrating band(AlbSS) and mixed migrating band(AlbFS). The frequencies of appearance in AlbFF, AlbFS and AlbSS were 5.77%, 28.85% and 65.38%, respectively. 4. The albumin phenotypes were controlled by two alleic genes, $Alb^F$ and $Alb^S$. The gene frequencies were calculated at 0.202 in $Alb^F$ and 0.798 in $Alb^S$.

A Study on Formation of Hemoglobin Adduct in Blood of Mice Inhaled with Ethylene Oxide (에틸렌옥사이드에 폭로된 흰쥐의 혈액에 형성된 헤모글로빈 부가체에 대한 연구)

  • Ethylene oxide is a genotoxic carcinogen with widespread uses as industrial chemical intermediate and gaseous sterilant. 2-hydroxyethylated N-terminal valine in Hb is a good biomarker for biological monitoring of ethylene oxide exposure, because of its stability. For measuring the hemoglobin adduct formed by exposure of ethylene oxide, we studied the determination of (N-2-hydroxy-ethyl)valine(HEV) in hemoglobin adduct by using GC/MS. Firstly we synthesized HEV with 2-amino-ethanol and bromoisovaleric acid(BIVA) and confirmed it with GC/MS-FID. Its fragmentations were m/z 116(base ion, M+-45) and m/z 130(M+-31). For measuring HEV with higher sensitivity, we use derivatives which were PFPITH(pentafluorophenylisothiocianate) and TBDMS (tributyldimethylsilylation) by using Edman procedure. Its fragmentation were m/z 425(M+-57), m/z 383(M+-99) and m/z 172(M+-310) by using GC/MS. We did biological monitoring for mice inhalation exposure with 400 ppm ethylene oxide. The concentrations of hemoglobin adduct were $168{\pm}3.8\;and\;512{\pm}04$(nmol g-1 globin) at 0.5 hr/day 400 ppm ethylene oxide inhalation exposure group, and $631{\pm}17\;and\;2265{\pm}9.4$(nmol g-1 globin) at 1.0 hr/day 400 ppm ethylene oxide inhalation exposure for 1 and 4 weeks, respectively. We confirmed that (N-2-hydroxy-ethyl)valine(HEV) of hemoglobin was a good biomarker for biomonitoring of ethylene oxide exposure, and can measured with derivatives such as PFPITH(pentafluorophenylisothiocianate) and TBDMS(tributyldimethylsilylation) by using GC/MS.

Effects of Inonotus Obliqua Extract on Blood Glucose Levels in Genetically Diabetic Mice (차가버섯 추출물이 당뇨생쥐 $(C57BL/KsJ^-m^{+/+}Lepr^{db})$의 혈당수준에 미치는 영향)

  • This study investigated the therapeutic effects of Inonotus obliqua extract on blood glucose, insulin, and other biochemical parameters in genetically diabetic mice $(C57BL/KsJ^-m^{+/+}Lepr^{db})$. The mice were divided into four groups-control, Chaga 1 (dose of 0.09 mg/kg of body weight), Chaga 5 (5 times of Chaga 1), and Chaga 10 (10 times of Chaga 1) - according to supplemented dose. Inonotus obliqua extract was orally administered to the animals for 6 weeks. The body and organ (liver and kidney) weights were not different among groups. Fasting blood glucose level was significantly lower in the Chaga 5 group compared with the control (p < 0.05). Hemoglobin A1c content was significantly lower in the Chaga 5 group compared with either the control and Chaga 1 group (p < 0.05). There was no significant difference in serum insulin level among groups. The glucose-6-phosphatase activity in liver was significantly the lowest in Chaga 10 group and was significantly lower in Chaga 5 group as compared with those of control and Chaga 1 groups. Therefore, the results of this study demonstrate that Inonotus obliqua extract alleviates many of the symptoms of diabetes in genetically obese mice and may offer a possibility as a therapeutic supplement for the normalization of blood glucose levels in human with hyperglycemia and have beneficial effects in patients with non-insulin-dependent diabetes mellitus.

Effect of Oriental Postpartum Management on the Hematology and Blood Chemistry in Puerperium Maternity (한방산후조리가 산욕기 산모의 혈액 및 생화학검사에 미치는 영향)

  • Objectives : As participation of women in public affairs increases, they are more concerned about postpartum management. However, objectivity of its effect tends to be insufficient. This study was done to investigate the clinical applications of postpartum management in Oriental Medicine. Subjects and methods : This clinical study was done on 74 patients who were treated by postpartum management in the Oriental Medical Hospital of Woosuk University from November 1999 to August 2000. We analyzed the changes of hematology and blood chemistry. Then, we examined the differences between two groups : one group had undergone vaginal duct delivery and the other cesarean section in mode of delivery. Results : I) The ratio of vaginal duct delivery to cesarean section was 39:35. Two major age groups were 25-29 and 30-34, respectively 43% and 35%. 2) According to the hematology, hemoglobin, hematocrit, REC and platelet count significantly increased, while WBC and ESR level significantly decreased. 3) To the hematology by mode of delivery, WBC level decreased in both groups. WBC level of the vaginal duct delivery group was significantly lower than that of cesarean section. RBC level increased in both groups, the cesarean section group were statistically significant. Hemoglobin, hematocrit and platelet count increased but they were not significant in either group. 4) To the blood chemistry, Triglyceride (TG), protein, albumin and sodium levels were significantly increased but cholesterol, ALT, BUN, creatinine, potassium and chloride levels were not statistically significant. 5) According to the blood chemistry by mode of delivery, TG level of the vaginal duct delivery group was significantly reduced but the others were not significant. Conclusious : The effective results were shown that postpartum management by Oriental Medicine assisted postpartum health care as well as postpartum anemia.
