흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과 (Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat)

  • 본 연구에서는 숙취해소에 좋은 것으로 알려진 식품 소재와 황칠나무 추출물을 복합하여 in vitro 및 in vivo에서 알코올 유도된 간 손상에 대한 보호효과를 검토하였다. HepG2세포에서 300 mM 알코올과 SBJ 혼합물을 처리하였을 때 농도 의존적으로 LDH 수치가 감소하였으나 황칠나무 추출물을 제외한 복합물인 SBJ-복합물에서는 그 효과가 유의적으로 낮아지는 것을 관찰하였다. In vivo에서 SBJ 혼합물의 보호효과를 확인하고자 흰쥐에 알코올과 SBJ 혼합물을 투여한 결과, 알코올 투여 후 1시간까지 급격하게 혈중 알코올 수치가 EtOH군에서 증가하는 것이 관찰되었으며, SBJ 혼합물 투여군은 유의적인 차이로 감소되었음이 관찰되었으며 또한 농도의존적인 경향을 확인하였다. ADH 및 ALDH 활성의 증가는 SBJ 혼합물의 알코올 분해 및 대사산물의 제거 활성에 기여할 것으로 예상된다. 뿐만 아니라, 알코올에 의하여 증가한 LDH의 농도가 대조군과 유사한 수준으로 유지하는 것을 확인하였으며, GST, SOD, GPx 및 reduced glutathione와 같은 항산화 인자 및 효소의 활성은 대조군보다 EtOH군이 유의적으로 감소했으며, SBJ 혼합물에 의해 개선되는 경향을 관찰하였다. 이를 통해 SBJ 혼합물은 ADH, ALDH 활성 증가 및 항산화 방어계를 향상시킴으로써 산화적 스트레스 감소를 통한 간보호 효과가 있는 것으로 생각되며, 이러한 간보호에 미치는 주요한 추출물은 황칠나무 추출물임을 알 수 있었다. 이러한 결과를 토대로 향후 숙취해소 작용을 갖는 신규 식품소재로 활용될 수 있을 것으로 기대된다.

가미방기황기탕(加味防己黃芪湯) 주정 추출물이 류마티스 관절염 동물병태모델에 미치는 영향 (Effects of GamiBangkeehwangkee-tang (fángjǐhuángqí-tāng) ethanol extract on Collagen-induced Rheumatoid Arthritis model of DBA/1 mice)

  • Objectives : The purpose of this study is to prove the effect of GamiBangkeehwangkee-tang (f$\acute{a}$ngj$\check{i}$hu$\acute{a}$ngq$\acute{i}$-t$\bar{a}$ng, BHT) ethanol extract on the immunity and rheumatoid arthritis related inflammatory cytokines. Methods : We checked viability in RAW 264.7 cell after treat by BHT. Then we measured inflammatory and immunity factors of DBA/1 mice with rheumatoid arthritis induced by collagen after BHT oral administration. Also, we checked micro-CT image, bone volume and bone inflammation ratio in micro-CT and structural parameter test. Results : BHT showed cell viability of 95% or higher in all concentration in RAW 264.7 cells. BHT treated group decreased level in serum of IgM and IgG test by 36% and 25% respectively. And BHT treated group showed significant decrease in WBC, neutrophil, lymphocyte and monocytes immune cell ratio in blood by 47%, 22%, 56% and 85% respectively. Also, BHT treated group decreased level in serum of $IL-1{\beta}$, IL-6, IL-17, $TNF-{\alpha}$ and hs-CRP tests by 29%, 33%, 32%, 24% and 56% respectively. Finally, BHT treated group showed increase ratio of bone volume that decrease ratio of bone inflammation. Conclusions : In this study, the results were observed rheumatoid arthritis factors cytokine decrease in serum. And BHT showed immunoglobulin and immune cells ratio decrease in serum and blood. Also, BHT depending on effects of inflammatory and immunity in hs-CRP test, micro-CT, structural parameter test. Thus, these results can used as a effective drug of BHT for inflammation and immunity.

해당화 열매 추출 분말의 항산화 활성과 고지방 식이로 유도된 Mice의 혈청지질 수준에 미치는 영향 (Antioxidant Activity of Rosa rugosa Thunberg and Effect on Serum Lipid Level in High Fat Diet-induced Mice)

  • Since Korean ancient times, powder of Rosa rugosa Thunberg has been used as a folk remedy. This study was conducted to verify the effects of Rosa rugosa Thunberg powder on in vitro antioxidant properties and serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. In the case of Rosa rugosa Thunberg powders, measurement of TPC, ABTS radical scavenging activity of trolox, DPPH radical scavenging activity, and measured value of FRAP were higher in ethanol extract than water extracts. For LDL-cholesterol concentration, mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a high numerical value compared with other groups (p<0.05). When testing for triglyceride concentrations in blood, mice fed 10% and 20% powder of Rosa rugosa Thunberg with high-fat diets showed the lowest numerical values (p<0.05). When testing for blood insulin concentrations, the high-fat diet group showed higher levels than compared to the control group (p<0.05). When testing for blood leptin concentrations, the high-fat diet group was $5.88{\pm}3.53ng/dL$, whereas mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a blood leptin level of $10.36{\pm}5.96ng/dL$ (p<0.05). Therefore, results prove that powder of Rosa rugosa Thunberg reduces triglyceride concentrations in the blood, and could be used as an excellent natural antioxidant in the future.

랫드에서 한방 혼합 추출물이 Ethanol의 약물동태학적 거동에 미치는 영향 (Effect of Mixed Extract for Elimination of Hangover on Ethanol Pharmacokinetics in Rats)

  • To investigate an efficacy of mixed extract with Ginseng radix, Puerariae lobata, Puerariae radix, Rubi pructus, Gomi pructus, Hoelen, Dried orange peel and Parvum comus cervi etc., on the hangover elimination, 12 hr-fasted male Sprague-Dawley rats weighing 150-200 g were given mixed extract (5 mL/kg, p.o.) and administered ethanol at a dose of 3 g/kg bw (25% in distilled water) orally 30 min postdosing. Blood was collected from caudal artery at 0.25, 0.5, 1, 2, 4, 6, 8, 10, and 12hr and then the animals were sacrificed at 24hr after the ethanol treatment. In these experiments, liver function indices, such as alanine aminotransferase and alkaline phosphatase activities, showed unaltered results in all treated groups compared with the normal group. The pharmacokinetics of ethanol after oral administration of mixed extract were also evaluated. From 0 min to 12hr, the administration of mixed extract showed 14% reduction of the area under the serum concentrations-versus-time curves (AUC) compared with the control group. The activities of alcohol dehydrogenase and aldehyde dehydrogenase measured at 24hr postdosing were also not altered by the administration of mixed extract compared with the control group. These studies demonstrate that oral administration of mixed extract, prepared by traditional prescription, decreases the ethanol concentration in serum and reduces AUC, suggesting that the mixed extract is effective for elimination of ethanol-induced hangover.

톳 열수추출물로부터 분리한 혈액 항응고성 다당류에 관한 연구 (Studies on the Blood Anticoagulant Polysaccharide Isolated from Hot Water Extracts of Hizikia fusiforme)

  • This study was focused on the purification, characterization and promotion mode of an anticoagulant polysaccharide from Hizikia fusiforme. The anticoagulant crude polysaccharide(HF 0) was obtained by using hot water extraction at 100oC for 3 hrs after homogenizing desalted Hizikia fusiforme. The anticoagulant polysaccharide(HF 2 3 1a) was purified from the crude extract(HF 0) through stepwise gradient ethanol precipitation(HF 2), DEAE Toyopearl 650C(HF 2 3), Sephadex G 75(HF 2 3 1), Sepharose CL 6B(HF 2 3 1a) chromatography and HPLC to homogeneity. HF 2 3 1a was estimated at 5.3$\times$105 Da molecular weight and composed of fucose(51.92%), galactose(19.34%), mannose(13.92%), xylose (7.14%), arabinose(3.95%) and rhamnose(3.78%), and comprimised 29.7 % sulfate residue. The sulfated anticoagulant polysaccharide from HF 2 3 1a was proposed to inhibit via the intrinsic pathway and common pathway in the blood coagulation. The HF 2 3 1a exhibited the anticoagulant activity by activating an antithrombin III and the activity depended on the concentration of HF 2 3 1a. Acute toxicity of HF 2 in mice was not detected. Only 14 of 33 control mice(11.4%) that had taken saline survived for 30 min after injecting thrombin(100 NIH unit/ml).

Improving Effects of Brassica oleracea L. var. italica Sprout Extract on Alcoholic Liver Dysfunction

  • Alcoholic fatty liver disorder has become a frequent health concern worldwide. To investigate the effects of Brassica oleracea (B. oleracea) sprout extract (BOE), the present study was designed with alcoholic fatty liver in the rat. Initially, the effects of BOE on liver parameters were examined. Male rats were divided into five groups. The normal control group was fed the normal diet, and the BOE group was fed the high fat diet and ethanol with/without BOE for 4 weeks. After 4 weeks feeding period, rats were sacrificed and their livers and blood were used for fatty liver-related biomarkers analyses. As a result, BOE ameliorated fatty liver-related enzymes profiles in liver tissues and also reduced blood alcohol concentration in rat model. We demonstrated that BOE protected the high fat diet and alcohol-induced fatty liver in rat model. Furthermore, BOE increased detoxificative abilities against alcohol.

귀뚜라미의 물 및 메탄올 추출물이 알코올 대사에 미치는 효과 (Effects of Water and Methanol Extracts of Cricket (Gryllus bimaculatus) on Alcohol Metabolism)

  • The cricket has been used in East Asia as crude drugs for treating fever and hypertension, and is presently reared as a pharmaceutical insect in China and a food for animals. For the purpose of evaluating protective extracts against alcohol-induced toxicity, the extracts of the cricket (Gryllus bimaculatus) were examined in animal models acutely administered alcohol by the cricket in ICR-mice. Water and methanol extracts from the cricket, were found to cause a significant decrease (37%) in the blood ethanol concentration as well as enhancement of liver mitochondrial alcohol dehydogenase (ADH) and acetaldehyde dehydogenase (ALDH) activitieson on a single intraperitoneal administration in mice. Futhermore methanol extract was demonstrated to exhibit more potent enhancing activity on ethanol metabolism than water extract. These results suggest that water/alcohol extract of G. bimaculatus may be used as a food for reducing the toxicity of alcohol.

청국장을 급여한 식이가 알코올 섭취쥐의 지질대사 및 간 기능개선에 미치는 영향 (Effects of Chongkukjang Intake on Lipid Metabolism and Liver Function in Ethanol Consumed Rats)

  • This study was performed to investigate the effect of chongkukjang intake on lipid metabolism and liver function in ethanol consumed rats. Twenty one Sprague-Dawley male rats aging 4 weeks old were used as experimental animals, which were divided into three dietary groups; casein diet(CA), soybean diet(SB) and chongkukjang diet(CJ). Alcohol was consumed with water as 25%(v/v) ethanol solution. After 4 weeks of experimental period, rats were sacrificed to get blood and liver samples for analysis of lipids, lipid peroxides, antioxidative enzymes and biochemical indices of liver function. The mean body weight, food intake and liver index were not significantly different among three groups. Serum level of total lipid, triglyceride and total cholesterol of chongkukjang diet group was the lowest among three groups although the difference was not significant. HDL-cholesterol level was significantly(p<0.05) higher in chongkukjang diet group than that of casein diet group. LDL-cholesterol level of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. Liver TBARS of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. The superoxide dismutase activity of chongkukjang diet group was significantly(p<0.05) higher than that of casein diet group. Catalase activity was not significantly different among three groups. As indices of liver function, glutamic oxaloacetic transminase(GOT), glutamic pyruvic transminase(GPT), $\gamma$-glutamyl transpeptidase($\gamma$-GTP) and lactate dehydrogenase(LDH) were not significantly different among three groups. Serum alcohol concentration and activities of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were not significantly different among three groups. The chongkukjang diet seems to give a beneficial effect for improving lipid metabolism by increasing HDL-cholesterol level and SOD activity while reducing liver TBARS level. However, effect on liver function has to be investigated further.

쇠비름 물, 에탄올 추출물의 항산화 및 항염증 활성 (Antioxidant and anti-inflammatory activities of water extracts and ethanol extracts from Portulaca oleracea L.)

  • 본 연구에서는 쇠비름의 활용가치를 높이고 기능성 소재로서의 이용 가능성을 타진하기 위해 쇠비름 물 및 에탄올 추출물의 항산화와 항염증 활성을 탐색하였다. DPPH 라디칼 소거능과 FRAP을 이용한 총 항산화 효능 평가 결과 에탄올 추출물이 물 추출물보다 항산화 활성이 우수한 것을 확인하였고, ABTS 라디칼 소거능은 $1,000{\mu}g/mL$의 농도에서만 쇠비름 물 추출물의 활성이 더 높았다. 한편, 세포독성을 살펴보기 위하여 쇠비름 물 및 에탄올 추출물을 이용해 대식세포에 MTT assay를 수행한 결과, $4,000{\mu}g/mL$의 이하 농도까지는 세포의 생존율에 영향을 미치지 않는 것을 확인하였다. $1.0{\mu}g/mL$ LPS로 염증반응을 유도시킨 세포에 대해 쇠비름 물 추출물에 비해 에탄올 추출물이 더 효과적으로 농도의존적인 NO와 ROS 생성 억제 효과가 있었다. 추가적으로 세포의 항염증 효능의 지표 단백질로 알려진 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 발현을 확인하였을 때 LPS 단독 처리군 대비 쇠비름 에탄올 추출물에서 3가지 단백질 모두의 발현양이 감소하는 것을 확인하였다. 이상의 결과를 종합하여 볼 때 쇠비름 에탄올 추출물은 물 추출물에 비해 높은 항산화와 항염증 활성을 가지는 것으로 확인되었으며, 추후 쇠비름의 유효성분 추출을 통한 항염증 물질의 연구와 염증 억제 성분의 분리 및 그 작용기전 연구에 기초 자료가 될 것으로 사료된다.

옥수수 껍질 분말의 In Vitro 에서의 항산화 특성과 고지방식이로 유도된 Mice의 혈청지질 수준에 미치는 영향 (Effect of In Vitro Antioxidant Properties and Extract of Corn Husk on Serum Lipids in Mice)

  • This study was performed to verify the effects of corn husk powder on serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. Powder of corn husks has been used as a folk remedy from ancient times in Korea. In the case of corn husk measurement of TPC (p<0.05), ABTS+ radical scavenging activity of trolox (p<0.05), DPPH radical scavenging activity (p<0.05), and measured value of FRAP (p<0.0001) were higher in ethanol extract than water extract. Weight of livers was significantly higher in the control group and high-fat diet group compared to groups fed 10% and 20% corn husk powders with high-fat diet (p<0.0001). For blood HDL-cholesterol concentration, mice fed 10% corn husk powder with high-fat diet ($160.00{\pm}16.17mg/dL$) showed significantly higher levels compared with the other three groups (p<0.05). Mice fed 10% and 20% corn husk powders with high-fat diet showed high numerical values of $13.50{\pm}2.51$ and $11.25{\pm}2.87mg/dL$, respectively. For triglyceride concentration in blood, mice fed 10% ($83.50{\pm}19.30mg/dL$) and 20% ($88.50{\pm}22.25$) corn husk powder with high-fat diet showed very low levels compared to the control group and high-fat diet group. In the case of leptin, the high-fat diet group showed a significantly high level of $5.88{\pm}3.53ng/dL$. Groups fed 10% and 20% corn husks with high-fat diet showed low numerical values of $2.72{\pm}2.34ng/dL$ and $1.10{\pm}0.62ng/dL$, respectively (p<0.05). Corn husk powder increased high HDL-cholesterol concentration in blood while lowering triglyceride concentration and leptin secretion. Therefore, powder of corn husks has utility in the food industry and food processing.
