Effect of curcumin in the prevention of experimentally induced nephrolithiasis in rats by ethylene glycol and Vitamin D3

  • Curcumin (CMN) is known to have beneficial role in anorexia, coryza, cough, diabetic wounds, and hepatic disorders apart from its inherent antioxidant effects. Therefore, the present study was aimed to evaluate antioxidant effect of CMN in prevention of nephrolithiasis in rats-induced by ethylene glycol (EG) and Vitamin D3 (Vit. D3). Male Wistar rats (175 - 200 g) were randomized in groups like control, EG + Vit. D3 induced nephrolithiatiatic rats, CMN treated rats, CMN + EG + Vit. D3 treated rats, Vit. E + EG + Vit. D3 treated rats. Urine was collected weekly throughout the experimental protocol and estimated for calcium oxalate (CaO) count. After completion of experimental protocol serum was estimated for blood urea nitrogen and creatinine. Both the kidneys were excised and used to evaluate levels of biomarkers of oxidative stress and calcium oxalate crystal deposition by histopathological studies. Administration of EG and Vit. D3 to rats resulted in increased oxidative stress, hyperoxaluria and renal deposition of CaO crystals. Supplementation with CMN improves kidney function, reduces elevated oxidative stress, urinary oxalate level and renal deposition of CaO which shows its protective action in nephrolithiasis. The increased deposition of stone in the kidney and stone forming constituents of nephrolithiatic rats were effectively lowered by treatment of CMN.

  • Sesame (Sesamum indicum L.) is probably the most ancient oilseed crop known in the world. Sesame seed is known for its high nutritional value and for having oil (51%) and protein (20%) content. The fatty acid composition of sesame oil is palmitic acid (7.8%), stearic acid (3.6%), oleic acid (45.3%), and linoleic acid (37.7%). Sesame oil is characterized by a very high oxidative stability compared with other vegetable oils. Two lignan-type compounds, sesamin and sesamolin, are the major constituents of sesame oil unsaponifiables. Sesamol (a sesamolin derivative) can be present in sesame seeds and oils in very small amount. Other lignans and sesamol are also present in sesame seeds and oils in very small amount as aglycones. Lipid oxidation activity was significantly lower in the sesamolin-fed rats, which suggests that sesamolin and its metabolites contribute to the antioxidative properties of sesame seeds and oil and support that sesame lignans reduce susceptibility to oxidative stress. Sesaminols strongly inhibit lipid peroxidation related to their ability to scavenge free radical. The sesame seed lignan act synergistically with vitamin I in rats fed a low $\alpha$-tocopherol diet and cause a marked increase in a u-tocopherol concentration in the blood and tissue of rats fed an $\alpha$-tocopherol containing diet with sesame seed or its lignan. The authors are reviewed and discussed for present status and prospects of quality evaluation and researched in sesame seeds to provide and refers the condensed informations on their quality.

  • Ginseng has been used in China for at least two millennia and is now popular in over 35 countries. It is one of the world's popular herbs for complementary and alternative medicine and has been shown to have helpful effects on cognition and blood circulation, as well as anti-aging, anti-cancer, and anti-diabetic effects, among many others. The pharmacological activities of ginseng are dependent mainly on ginsenosides. Ginsenosides have a cholesterol-like four trans-ring steroid skeleton with a variety of sugar moieties. Nuclear receptors are one of the most important molecular targets of ginseng, and reports have shown that members of the nuclear receptor superfamily are regulated by a variety of ginsenosides. Here, we review the published literature on the effects of ginseng and its constituents on two main sex steroid hormone receptors: estrogen and androgen receptors. Furthermore, we discuss applications for sex steroid hormone receptor modulation and their therapeutic efficacy.

  • Cardiovascular diseases are a rapidly growing epidemic with high morbidity and mortality. There is an urgent need to develop nutraceutical-based therapy with minimum side effects to reduce cardiovascular risk. Panax ginseng occupies a prominent status in herbal medicine for its various therapeutic effects against inflammation, allergy, diabetes, cardiovascular diseases, and even cancer, with positive, beneficial, and restorative effects. The active components found in most P. ginseng varieties are known to include ginsenosides, polysaccharides, peptides, alkaloids, polyacetylene, and phenolic compounds, which are considered to be the main pharmacologically active constituents in ginseng. P. ginseng is an adaptogen. That is, it supports living organisms to maintain optimal homeostasis by exerting effects that counteract physiological changes caused by physical, chemical, or biological stressors. P. ginseng possesses immunomodulatory (including both immunostimulatory and immunosuppressive), neuromodulatory, and cardioprotective effects; suppresses anxiety; and balances vascular tone. P. ginseng has an antihypertensive effect that has been explained by its vasorelaxant action, and paradoxically, it is also known to increase blood pressure by vasoconstriction and help maintain cardiovascular health. Here, we discuss the potential adaptogenic effects of P. ginseng on the cardiovascular system and outline a future research perspective in this area.

  • Studies on the pharmaco-constituents from the leaves of Cercts chinensis which have been used for the treatment of inflammation, contusion, dilated blood, pain of heart and stomach, edema, etc. in Korean folk remedies were carried out. Dried leaves of the plant were extracted with MeOH. The MeOH extract was suspended in distilled water and subsequently fractionated with $Et_{2}O$ and n-BuOH. From the $Et_{2}O$ and n-BuOH fractions, six phenolic compounds were isolated and identified as myricitrin($C_{21}H_{20}O_{12}, {\;}m.p.{\;}199~200^{\circ}$. $4myricetin-3-O-{\alpha}-L-rhamnopyranoside$), kaempferol($C_{15}H_{10}O_{6}, {\;}m.p. 276^{\circ}$), quercetin($C_{15}Ha_{10}O_{7}, {\;}m.p.{\;}313~314^{\circ}$), quercitrin ($C_{21}H_{20}O_{12}, {\;}m.p.{\;}176~178^{\circ}, {\;}quercetin-3-O-{\alpha}-L-rhamnopyranoside$), gallicin ($C_{8}H_{8}O_{5}, {\;}m.p.{\;}202~203^{\circ}$. methyl gallate), gallic acid ($C_{7}H_{6}O_{5}, {\;}m.p.{\;}260~265^{\circ}) through their physico-chemical data and UV, IR, EI-MS, $^{13}C-NMR$, and $^{1}H-NMR$ analysis with authentics.

소리쟁이 뿌리 성분이 조골세포 분화에 미치는 영향 (The Stimulatory Effects on the Osteoblast Cells of the Root Constituents from Rumex crispus)

  • Rumex crispus (Curled Dock, Polygonaceae) is a perennial wild plant used in traditional medicine as a laxative, astringent, and to treat blood and skin disease. The ethanol extract of R. crispus was obtained and its carbohydrate contents were analyzed using high-performance anion-exchange chromatography with pulsed amperometric detection. The anabolic effects of R. crispus in human osteoblastic MG-63 cells were investigated using the WST-8 assay, alkalinephosphatase (ALP) assay, and mineralization assay. The ethanol extract increased the proliferation of MG-63 cells and stimulated ALP activity in a dose-dependent manner over a 72-hrs period. Additionally, the ethanol extract dose-dependently stimulated the formation of bone nodules in MG-63 cells treated for 12 days. The ethyl acetate fraction from the ethanol extract did not affect osteoblast viability but induced an increase in ALP activity. In conclusion, the ethanol extract of R. crispus increases the proliferation and bone-forming activity of osteoblasts, and hence it could be used in the development of bone-forming stimulatory nutraceuticals and osteoporosis-related medicines.

  • The application of antique medical instructions, practices, skills and knowledge has been considered as the most affordable treatment in many developing countries. The use of these preparations and prescriptions over generations has made a useful and valuable guide for drug discovery in modern medicine. Medical herbs have been of a high importance for this purpose. The genus Dorema, of Apiaceae family (Umbelliferae) has a wide use in ethnobotany and traditional medicine around the world. It has been used as a treatment for CNS disease, convulsion, upper respiratory tract problems, gastrointestinal disorder and high blood sugar. Furthermore, phytochemical investigations have reported Dorema species to contain a wide range of constituents including terpenes, coumarins and phenolic compounds. The current review summarizes comprehensive information regarding botany, phytochemistry and pharmacological aspects of Dorema spp.

Vasodilatation Effect of Complex Saponin Separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla Mixture Extract

  • The purpose of this study is to investigate vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery. In this study, to determine vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery, arterial rings with intact or damaged endothelium were used for experiment using organ bath, and were contracted by endothelin. complex saponin, major active constituents of Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract, showed a moderate vasodilatation effect on the basilar arteries of rabbits. Therefore, treatment with complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract may selectively accelerate cerebral blood flow through dilatation of the basilar artery. Theseis result suggest a potential role of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract as source of vasodilatation agent.

HCB와 PCBs에 노출된 나일틸라피아, Oreochromis niloticus의 혈액성분의 변화 (Changes of Haematological Constituents in the Nile tilapia, Oreochromis niloticus Exposed to HCB and PCBs)

  • 유용 양식어류인 틸라피아를 대상으로 어체중당 1일 0.05, 0.25 및 0.50mg/kg의 HCB와 PCBs의 경 구투여에 의해 30일 동안 혈액성상 및 혈액화학적 변동을 조사하였다. HCB의 투여에 의한 틸라피아의 RBC 수는 20일 이후 0.50mg/kg, 30일째는 0.25mg/kg의 농도, hemoglobin 농도와 hematocrit 값은 20일 이후 0.50mg/kg 농도에서 유의한 감소를 나타냈다. PCBs의 투여에 의한 틸라피아의 RBC 수, hemoglobin 농도 및 hematocrit 값은 20일 이후 0.25mg/kg 이상의 농도에서 유의한 감소를 나타냈다. 혈청 무기성분인 calcium, magnesium 및 inorganic phosphorus 농도는 HCB를 투여한 모든 실험에서 뚜렷한 차이가 인정되지 않았다. 또한, PCBs를 투여한 모든 실험에서도 magnesium과 inorganic phosphorus 농도의 변동은 관찰되지 않았으나, calcium 농도는 20일 이후 0.50mg/kg 농도, 30일째는 0.25mg/kg 이상의 농도에서 유의한 감소를 나타냈다 HCB의 경구투여에 의한 혈청 total protein 농도는 30일째 0.50mg/kg 농도에서, PCBs의 경구투여에 의해서는 20일 이후 0.25mgfg 이상의 농도에서 유의하게 감소하였다. HCB 및 PCBs의 경구투여에 의한 glucose 농도는 20일 이후 0.25mg/kg 이상의 농도에서 유의한 증가를 나타냈다. HCB 및 PCBs의 경구 투여에 의한 GOT와 GPT 활성은 20일 이후의 0.50mg/kg에서 유의한 증가를 나타냈다. 결론적으로 HCB 및 PCBs의 경구투여에 의한 틸라피아의 혈액화학적 변동은 0.25 mg/kg 농도 이상에서 적어도 20일 후부터 영향을 받을 것으로 생각된다.

고에너지 사양이 육종가 배치별 거세한우의 성장, 도체, 및 혈액성상에 미치는 영향 (Effects of high energy diet on growth performance, carcass characteristics, and blood constituents of Hanwoo steers distributed by estimated breeding value for meat quality)

  • 본 연구는 육종가를 이용하여 배치한 거세한우에 고에너지 사료를 급여하여 성장, 도체성적 및 혈액성분의 변화에 대하여 분석을 해보았다. 육종가의 적용은 소고기 고급육이 가능한 개체를 선발하는 방법으로 사용되어져 왔다. 본 연구는 육종가에 의해 고, 저로 나누어진 한우그룹에 고에너지 사료를 급여효과를 알아보기 위해 수행되었다. 고육종가 그룹에서 고에너지 사료를 급여할 경우 도체성적이 높아진다는 가정을 하였다. 연구를 수행하기 위해 26마리의 거세한우를 이용하여 $2{\times}2$ 완전임의배치를 하였다(고에너지, 일반사양 과 고육종가, 저육종가로 배치). 체중, 일당증체량, 건물섭취량, 사료요구율의 결과로 고 육종가 그룹에서 수치적으로 높게 나타났다. 혈청성분분석 결과로 비육후기(20 - 27개월령)의 혈청성분중 포도당과 중성지방 농도가 고육종가 그룹에서 유의적으로 높게 나타났고 이는 도체성적에도 긍정적인 효과를 가져오는 것으로 나타났다. 하지만 선행연구에서 나타났던 혈중 알부민과 인의 성분은 도체등급에 영향을 미치지 않는 것으로 보인다. 도체중 분석에서는 근내지방도가 고육종가 그룹이 수치적으로 높게 나타났으나 유의적인 차이는 없었다. 본 연구의 결과로 특정 혈중대사 성분들 중 포도당과 중성지방과 같은 지방대사에 관여하는 성분들은 비육후기 육종가가 높을수록 높은 농도로 나타났고 이는 혈중 대사성분이 유전적인 영향에 의해서 변화가 된다는 것을 알 수 있었다.
