A fourteen-month-old Holstein bull from a private cattle fattening farm at Ansung county in Kyunggi Province Korea was submitted on August 2nd 1997, for examination at the Pathology Division of the National Veterinary Research Institute. The bull died within 24 hours after developing clinical signs of abrupt inertia, complete anorexia high fever(40.3℃) diffuse severe emphysematous swelling of upper part of the right hind leg lateral recumbancy and paralysis. At necropsy diffuse severe subcutaneous redness and influx of serosanguinous exudate containing gas bubbles had accumulated under the thorax right hip and upper region of right hind leg. Muscles in upper right hind leg were blackish to dark red and yellowish brown in color. Muscle bundles were dry and separated by gas bubbles and serosanguinous fluids and muscles sections from affected regions floated in water. Histopathologically muscle fibers were partially or entirely degenerated fragmented and separated by exudate and gaseous substance mixed with polymorphonuclear cells. Blood vascular walls in affected regions showed severe acute fibrinoid necrosis. Typical large rod-shaped bacteria with or without oval central to subterminal spores were frequently observed in tissue sections stained with H-E and Gram stain. The large Gram-positive anaerobic endospore-producing rods were isolated from the suspension of muscle lesions. Isolated bacteria were identified as Clostridium(CL) chauvoei and CL. sordellii by biochemical tests. This case was diagnosed as blackleg based on the typical clinical signs gross finding histopathological observation and bacteriological results. This is the first case report on blackleg associated with Cl. chauvoei and Cl. sordellii in Holstein cattle in Korea.
An 11-year-old, 3.3 kg, male Maltese dog was referred to Veterinary Teaching Hospital of Konkuk University because of diarrhea and severe anemia. Abnormal physical examination findings included left submandibular lymph node enlargement, pale mucous membrane, cataract, and bloody diarrhea. Results of hematologic examination revealed a marked lymphocytosis resulting in leukocytosis and the markedly increased numbers of small, well-differentiated lymphocytes in the peripheral blood. Serum biochemical abnormalities consisted of elevated AST and ALP, hyperphosphatemia, hypoglycemia, and hypoalbuminemia. Radiographic examination showed cardiomegaly and hepatosplenomegaly. Results of urinalysis included bilirubinuria and proteinuria. Based on results of examination described above, chronic lymphocytic leukemia was diagnosed. Chemotherapy was initiated with cyclophosphamide ($300mg/m^2$, IV once every 2 weeks), vincristine ($0.75mg/m^2$, IV once every 2 weeks, alternating weeks with the cyclophosphamide), and plus prednisolone ($50mg/m^2$, PO, SID for a week, then $20mg/m^2$, PO every other day). The response to chemotherapy was partially present. This study first demonstrates clinicopathological findings and chemotherapeutic response of chronic lymphocytic leukemia in Korea.
Lectin was isolated from shiitake mushroom (Lentinula edodes) with 0.15 M NaCl solution, and purified by the following procedures : precipitation by ammonium sulfate, anion exchange column chromatography on DEAE Sephadex A-50 and hydroxyapatite column chromatography. The fresh pileus part of the mushroom contained more than two times of lectin compared to the stipe part, and lectins and its activity were reduced by heating. The extraction yield of crude lectin was 46.03%, 28% yield after purification on on DEAE Sephadex A-50 column chromatography. Some amino acids, aspartic acid, serine, alanine and histidine, were increased by purification process. Relatively low molecular weight parts of lectin had the agglutinating activity for rabbit blood, and its molecular weight was about 23 kDa The molecular weights of purified lectins, LA-a and LB-b, by the hydroxyapatite column chromatography were 24 kDa and 23 kDa, respectively.
To investigate the anti-complementary and cytotoxic effects of oriental prescription, Kuseonwangdogo, on the proliferation of preadipocyte 3T3- L1 cells, we examined biological effects of Kuseonwangdogo. The results obtained were as follows. 1. After 14 days, the body weight of rats treated with Kuseonwangdogo decreased more than that in the control group (p<0.05). However, the weights of liver, spleen and kidney were unchanged. In serum biochemical test, we examined the level of glucose (GLU) and glutamic pyruvic transaminase (GPT). The levels of GOT and CHOL in serum were decreased remarkably by the administration of Kuseonwangdogo (p<0.05). The haematological examination of the tested group showed significant increment of white blood cells (WBC), hemoglobin concentration (HGB), mean corpuscular hemoglobin (MCH) and monocyte (MO). 2. The effect of Kuseonwangdogo on the proliferation of 3T3-L1 cells was tested by the sulforhodamin B(SRB) assay. The high concentration ($100{\mu}l\;and\;200{\mu}l$) of extracts inhibited the proliferation of 3T3- L1 cells. The p-value was <0.01, respectively. 3. The extract of Kuseonwangdogo showed a potent anti -complementary activity. It was suggested that the active principle may be a kind of polysaccharide molecule. 4. The cytotoxic effects of Kuseonwang dogo and its composing herbs in human liver cells (WRL68) and monkey kidney cells (Vero) were examined by the SRB and 3- (4,5- Dimethylthiazol-2-yl) -2,5 diphenyl-2H- tetrazolium bromide (MTT) assay. Cytotoxic effects were not observed.
Objective : This research was performed to investigate protective effects of Sophora subprostrata, against ischemic brain damage after a middle cerebral artery(MCA) occlusion. The effect was estimated using histological test, neurobehavioural test, and biochemical test. Methods : Rats(Sprague-Dawley) were divided into four groups: Sham operated group, MCA occluded group, Sophora subprostrata administrated group after MCA occlusion, and Normal group. The MCA was occluded by intraluminal method. Sophora subprostrata was administrated orally twice(l and 4 hours) after middle cerebral artery occlusion. The neurobeavioural test was performed at 3 hours, 6 hours, 9 hours and 24 hours after the surgery by posture reflex test and swimming behavioural test. All groups were sacrificed at 24 hours after the surgery. The brain tissue was stained with 2% triphenyl tetrazolium chioride(TTC) or 1 % cresyl violet solution, to examine effect of Sophora subprostrata on ischemic brain tissue. The blood samples were obtained from the heart of rats. Tumor necrosis factor-a level was measured from sera using Enzyme-Linked Immunoabsorbent Assay(ELISA). Results : The results showed that (1) Sophora subprostrata reduced infarct size and total infarct volume by 54.8% compared to the control group, (2) that neuronal death, which was shown by decrease in cell number and size, was attenuated significantly in the boundary area of the infarction, (3) that serum $TNF-{\alpha}$ㆍlevel was reduced significantly, and finally, there was significant recovery of motor deficit at 3 hours after MCA occluded by Swimming behavioural test. Conclusions :In conclusion, Sophora subprostrata has protective effects against ischemic brain damage at the early stage of ischemia.
Isolation of the toxic substance which was produced by Streptentyces sp. and it's biorhemical characterestics and toxicity to fishes were reported in the previous paper. The present report includes antibiotic activity of the substance, inhibitory activity of the substance on the succinic dehydrogenase of fishes, and its effect on the blood corpuscles of a rabbit. An evident antibiotic activity of this substance was observed on Candida yeasts, but n$ot^1$on molds or bacteria. The substance inhibited the growth of Candida japonica and C. utilis to the 50% at the concentrations of 7.5 and 10.2ug per ml, respectively. The activity of succinic dehydrogenase obtained from various organs of Cyprinous carpio L. was also found to be inhibited by this substance. Original activities of the enzymes from tht brain, kidney, and liver, were inhibited by 75.4%, 38.2%, and 26.2%, respectively, but not the enzymes from the heart and spleen. Neither leukopenia nor leukocytosis was detected after the intravenous administration of the substince to the rabbit at the level of 6 mg per 2 kg body weight.

As utilization of radiation in medicine, industry and biochemical research increases, the protection against radiation damage has become an important issue. Natural products such as herbal medicines are beginning to receive attention as modifiers on the radiation response. In the present study, the protective effect of a herb, Menthae Herba, against radiation-induced DNA damage was evaluated using alkaline single-cell gel electrophoresis (SCGE; comet assay) in the mouse peripheral blood Iymphocytes and the micronucleus formation test in the Chinese hamster ovary (CHO) cells. The tail moment, which was a marker of DNA damage in the SCGE, and the frequency of micronuclei was decreased in groups treated with Mentae Herba extract before exposure to 200 cGy of gamma-ray. We also confirmed its activities to scavenge DPPH and hydroxyl radicals. These experiments demonstrated that Menthae Herba was effective at reducing the radiation-induced damage of DNA and scavenging free radicals. It is plausible that scavenging of free radicals by Menthae Herba may have played an important role in providing the protection against the radiation-induced damage to the DNA. These results indicated that Menthae Herba might be a useful radioprotector and that radical scavenging appears to be one of the mechanisms of radiation protection.

The effect of each azalea(Rhodoendron mucronulatum) pollen extract on phase I enzyme(aniline hydroxylase)was studied in this experiment. Serum aminotransferases were not changed in mice injected each azalea pollen extract, respectively compared to control group. The hepatic micros mal aniline hydroxylase activities in the presence of each azalea pollen extract were not affected in vitro. After treatment with azalea pollen water extract, hepatic microsomal aniline hydroxylase activity was increased with dose-dependent manner as compared to control group. The increment of hepatic microsmal aniline hydroxylase activity was more powerful by the treatment of water extract. As mice received aniline after pollen butanol and water extract-pretreatment once a day for 5 days, the blood and liver levels of aniline were decreased significantly.

The enhancement of the human body fat reduction of conjugated linoleic acid (CLA) with the supplementation of $\gamma$-oryzanol (OZ) was investigated on overweight Korean women (n=51, BMI> 23). Subjects were divided into 4 groups of control, CLA, glyceride form of CLA (GCLA), and CLA plus OZ (CLA-OZ). The soft-gel capsule (500 mg) was used to deliver control (500 mg olive oil), CLA (500 mg CLA), GCLA (500 mg GCLA) and CLA-OZ (500 mg CLA plus 50 mg OZ). Three capsules were taken twice a day for 12 weeks. The CLA-OZ supplementation reduced 1.35% body fat that was 0.34% enhancement against CLA supplementation. As considered subject variations, CLA-OZ reduced body fat ranged from 7.9% to -2.7%, equivalent to 5.6 kg loss to 0.7 kg gain in body fat mass, against CLA. The CLA-OZ reduced body weight and body mass index (BMI), relative to control, but the reductions by CLA-OZ were not different from those by CLA and GCLA. All biochemical markers analyzed for safty were not significantly different within or between groups and were within the normal range. The CLA-OZ supplementation significantly reduced blood pressure, as compared to the supplementation of CLA, GCLA and control. These results suggest that OZ could be a useful ingredient to mix with CLA for the reduction of human body fat.

The purpose of this study was to investigate the pharmacological effects of low-esterified pectin on carbon tetrachloride $(CCL_4)-induced$ hepatotoxicity in rats. The study included two experiments. In the first experiment the animals were given daily $CCL_4$ through gavage for 7 days and then 10, 50, or 250 mg/kg b.w. of pectin for 21 days. At the end of experiment rats were killed within 24 hours. The increased bilirubin level, enhanced alanine aminotransferase and aspartate aminotransferase activity in plasma induced by $CCL_4$ were partly normalized by pectin administration in a dose-dependent manner. The pectin treatment also resulted in significant recovery of $CCL_4-induced$ decrease of the liver glycogen content. In addition, pectin significantly improved $CCL_4-induced$ alterations of pro-oxidant and antioxidant biochemical parameters in liver and plasma compared to those of rats administered $CCL_4$. In the second experiment the animals were given daily 10, 50 or 250 mg/ kg b.w. of pectin for 21 days before a 7-day administration of $CCL_4$. Rats were killed 24 hours after the end of experiment. Pretreatment with pectin before $CCL_4$ administration resulted in significantly inhibited increase of the blood enzymatic activities of alanine and aspartate aminotransferases and bilirubin level in a dose-dependent manner. Also, preliminary administration of pectin prevented elevation of malondialdehyde and conjugated diene levels in liver and plasma as well as a reduction of glutathione content in liver of rats given $CCL_4$. These results suggest that low-esterified pectin exert healing and preventive effects on $CCL_4-induced$ hepatotoxicity in rats.
