Study on the Subacute Toxicity of Complex of Pine Needle Oil and Korean Medicinal Herbs against Rats

  • Pine needle oil and Korean medicinal herbs (KMH) are known as effective therapeutic agents on various blood vessel disease. We have already reported the ameliorative effect of complex of pine needle oil and Korean medicinal herbs against hyperlipidemia. But safety and non - toxicity of pine needle oil and Korean medicinal herbs to normal animal cells have not been studied clearly. In this study, we investigated whether pine needle oil and Korean medicinal herbs show side effects on rat or not. These materials were administered to rats, and subacute toxicity was examined by measuring the hematological values, CBC differentiation, biochemical levels of blood (TP, total protein; albumin; ALP, alkaline phosphotase; AST, aspatate aminotrans- ferase; ALT, alanine aminotransferase; T-Chol., total cholesterol; T-Bil., total bilirubin) and urine analysis, suggesting that the sample have no side effects and cytotoxicity. These results indicate that the complex of pine needle oil and Korean medicinal herbs may effective non- toxic, safety therapeutic agents on hepatocytes and hyperlipidemia.

Inhibitory Effect of Acetylmannan of Dioscorea bataras on Toxicity of Paraquat (마로부터 분리한 Acetylmannan의 Paraquat 독성 억제 효과)

  • Paraquat is a useful nonselective herbicide widely used throught the world. However accidental or intentional ingestion of the herbicide cause fatal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of acetylmannan from Dioscorea batalas on paraquat toxicity. 100mg/kg acetylmannan from wild or cultured Dioscorea bataras was administered orally to male SD rats for 3 days and the administration time interval was 24hours. After one hour of final administration, 50mg/kg paraquat was administered intraperitonially. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups, sGPT, BUN, creatinine, ALP levels were increased by 2 to 4 times of normal values. However in acetylmannan from wild Dioscorea batatas treated groups, sGPT, BUN, creatinine, ALP levels in blood and lung tissue were significantly decreased to normal levels. In acetylmannan from cultured Dioscorea batatas treated groups, BUN, creatinine were significantly decreased to normal values, but not in sGPT, ALP levels. Therefore, we concluded that acetylmannan from wild Dioscorea batatas can be used as an. antidote of paraquat toxicity.

  • The purpose of this study was to compare Black-pigmented Bacteroides isolated from necrotic pulp with the presence or absence of eight symptoms associated with pulpal necrosis and to identify the proportion of each Black-pigmented Bacteroides species. The canal contents of fourteen traumatically and cariously necrotized teeth were sampled with a special technique and cultured aerobically and anaerobically for growth in blood agar plate and for Black-pigmented Bacteroides on selective blood agar plate. Each Black-pigmented Bacteroides species were identified by Gram's stain, hemolysis reaction, colony color and morphology and biochemical tests. The results were as follows; 1. 60.9 percent of the bacteria isolated were anaerobic and 39.1 percent were aerobic. 2. Four Black-pigmented Bacteroides species were isolated; B. loescheii (74.1%), B. denticola (11.1%), B. intermedius (7.4%) and B. gingivalis (7.4%). 3. Black-pigmented Bacteroides was found to be significantly related to sinus tract formation and exudate.

  • In order to elucidate the mechanism of toxic action of a pisonous mushroom, Amanita pantherina, biochemical effects of the mushroom extracts on mice were studied. A hotwater extract of Amanita pantherina injected intraperitoneally into male ICR mice evoked signs similar to those observed clinically upon acute poisoning by the mushroom and also changed the levels of component enzyme activities in blood, liver and urine. The serum cholinesterase activity was decreased significantly during 1-3 h after injection.

  • A retrospective study was designed to determine whether there were any differences in prepartal blood biochemical parameters between cows developed subclinical ketosis (SCK) and non-SCK cows. Data on blood biochemistry from 33 Holstein parturient cows (15 SCK and 18 non-SCK cows) in one farm for one year, were compared. Plasma beta-hydroxybutyrate, non-esterified fatty acid and glucose concentrations were utilized in the diagnosis of SCK and showed significant differences between SCK and non-SCK cows after calving. Before calving, however, only plasma total cholesterol concentration in SCK cows was significantly lower than that in non-SCK cows. Consequently, it seems that lower plasma total cholesterol concentration before calving is related to the occurrence of SCK immediately after calving.

  • This study was performed to estimate the clinical signs and biochemical para meters of indicator on acute hepatic injury induced by the administration of CCl$_{4}$ in rats. Minor behavioral changer brittleness of skin hair and decreased volume of water and food intake were observed in rats after 2 hours of $CCl_{4}$ administration compared to control group. Concentration of serum albumin has shown lower than that of control group. However concentration of total bilirubin has shown higher than that of control group. As times go onto serum LDH activity was significantly increased compared to control Broup. Serum CPK activity hasn't shown change compared to control group. Passive hemagglutination that of afetoprotein was shown negative reaction in all the treatment groups and control group.

  • 1) Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. single treatment) alone partly showed blood pressure decreasing effect in SHRs and increasing effect of urinary volume in normal rats. 2) Cadmium nitrate (10mg/kg, s.c. single treatment) induced toxicity such as body weight decreasing effect, antidiuretic effect and muscle relaxant effect such as pull-up test, traction test and rota rod test in rats. However, Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. single treatment) showed antidotal effects on the above and also in acute toxicity test when coadministered with both of them. 3) Cadmium nitrate (1mg/kg, s.c. 7 days consecutive treatment) did not showed toxicity in body weight change, blood pressure, change, serum biochemical parameters in rats. Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. 7 days consecutive treatment) did not also showed any antidotal effects when coadministered with both of them for 7 days.

  • Objective : The present study was carried out to investigate the preventive effect of Sophorae Flos on streptozotocin - induced Diabetes mellitus. Method : Sophorae Flos was given to rats with oral administration. The experimental animals were divided into 3groups : normal group of rats, control group of Streptozotocin-induced diabetic rats, sample group with Sophorae Flos . The effect of Sophorae Flos on Streptozotocin-induced diabetes was observed by measuring the survival rate of rats, weight of rats, FER, blood glucose, each organ weight of rat, total cholesterol, HDL, GOT, GPT & creatinine. Result : Streptozotocin caused hyperglycemia and hypoinsulinemia by a selectively destroying pancreatic ${\beta}-cell$. Sophorae Flos treatment don't protected them from hyperglycemia and hypoinsulinemia. Organ weight liver, kidney, heart & spleen shows no significant changes. Sophorae Flos significantly don't recoverd the increase of several biochemical parameters such as blood glucose, total cholesterol, HDL, GOT, GPT, antioxidant & creatinine is vice versa. Conclusion : Sophorae Flos extract group did not show significant decrease than Streptozotocin control group.

Effects of Glycerol on the Oxygen Free Radical Reactions and Renal Functions in the Renal Cortex of Rats (Glycerol이 흰쥐 신피질에서의 산소유리기반응과 신기능에 미치는 영향)

  • In an attempt to define the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase in the renal cortex of rats, and the concentrations of blood urea nitrogen(BUH) and serum creatinine of rats at 24hr after the injection of a 50% solution of glycerol. Sprague-Dawley albino rats weighing 240 to 260 mg were injected intramuscularly with a 50% solution of glycerol(2 mι/kg, 4 mι/kg and 8 mι/kg). The group treated with glycerol showed significantlv higher MDA level and catalase activity, lower SOD activity and higher BUN and serum creatinine concentrations at 24 hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity.
