The diagnosis of cause of death (COD) or estimation of postmortem interval (PMI) is hard to perform using postmortem blood or other bodily fluids due to various biochemical changes that occur during the agonal phase or after death. To solve these problems, new paradigms and new analytical methods are needed. In this study, postmortem blood was fractionated with specific gravity 1.021, 1.029, 1.038, and 1.045, and the absorbance patterns of each sample of the 131 total cases (12 kinds of COD) were analyzed using a spectrometer. The absorbance was grouped into 9 patterns (ABS pattern 1 to 9) according to the wave length and the signal intensity. These patterns of postmortem blood were found to be distinctly different from the absorbance pattern of fresh blood. The analysis of ABS patterns is useful for the diagnosis of deaths due to acute or rapid bleeding, fire death, drowning and, in some cases, poisoning, but is not useful for the estimation of PMI.
Possible correlations between nutrient intake add health status-as assessed by anthropometric measurements, physical performance and biochemical measurements-were investigated, using 514 healthy young men aged 20 years old who had no apparent health problems. The intakes of nutrients were estimated using a three-day dietary recall method. Height and body weight were measured, and body mass index (BMI) was calculated. Physical performance was tested using sit-ups, push-ups, a loom sprint and a 1,500m run. When compared with the Korean recommended dietary allowances (Korean RDA), the subjects nutrient intakes were adequate except for calcium. The intake of calcium was 516.66$\pm$293.43mg/day, which is 73.80 % of the Korean RDA. The subjects averaged 174.51$\pm$7.07cm in height, 68.17$\pm$9.25 kg in body weight and 22.23$\pm$2.16 in BMI. The associations between nutrient intakes and anthropometric measurements, and between nutrient intakes and physical performance, were weak. The intake of vegetable fat was positively correlated to body weight, whereas the intake of carbohydrate was negatively correlated to BMI. The intake of carbohydrate was positively correlated to the level of performance in the loom sprint, and the intake of vegetable Int was positively correlated to the level of performance of sit-ups. No correlation was found between nutrient intakes and the following biochemical measurements of the blood: the levels of glucose, total protein, total cholesterol, triglyceride, hemoglobin and hematocrit. These results suggest that anthropometric measurements and level of physical performance can be associated with energy nutrient intakes, even in moderately active, well-nourished, young men. No correlation was found between nutrient intake and biochemical measurements, probably because all subjects had a reasonably well-balanced diet.
The measurement of many biochemical changes that could be appeared as aging will be resulted in the very useful and basal marker to maintain healthy life and to predict the health condition and the retardation of the aging. We classified the geriatrics into the normal and same aged dementia on the based of clinical diagnostic as well as MMSE evaluation. And then we were collected blood samples from both groups and examined 13 biochemical assays to screen any valuable biochemical index for dementia. Among 13 assays, several assay data showed the statistically significant differences between two groups. The values of serum albumin, APase, and GOT were significantly decreased in demented group compared to control. On the other hand, the values of glucose and LDH were increased in dementia compared to control. Additionally, the value of Pi was increased but the values of calcium and GPT were decreased, without significant difference (p>0.05). The values of other assays such as bilirubin, BUN, cholesterol, creatinine and total protein did not show any statistically significant differences. Conclusively it may be suggested that dementia might be resulted in the variable biochemical changes which could be affected by many environmental factors as well as dementia itself.
Inflammaging in male reproductive organs covers a wide variety of problems, including sexual dysfunction and infertility. In this study, the beneficial effects of cordycepin (COR), isolated from potential medicinal fungi Cordyceps militaris, in aging-associated testicular inflammation and serum biochemical changes in naturally aged rats were investigated. Male Sprague Dawley rats were divided into young control (YC), aged control (AC), and COR (5, 10, and 20 mg/kg) treated aged rat groups. Aging-associated serum biochemical changes and inflammatory parameters were analyzed by biochemical assay kits, Western blotting, and real-time RT-PCR. Results showed a significant (p < 0.05) alteration in the total blood cell count, lipid metabolism, and liver functional parameters in AC group when compared with YC group. However, COR-treated aged rats ameliorated the altered biochemical parameters significantly (p < 0.05 and p < 0.01 at 5, 10, and 20 mg/kg, respectively). Furthermore, the increase in the expression of inflammatory mediators (COX-2, interleukin (IL)-6, IL-1β, and tissue necrosis factor-alpha) in aged rat testis was significant (p < 0.05) when compared with YC group. Treatment with COR at 20 mg/kg to aged rats attenuated the increased expression of inflammatory mediators significantly (p < 0.05). Mechanistic studies revealed that the potential attenuating effects exhibited by COR in aged rats was mediated by regulation of NF-κB activation and MAPKs (c-Jun N-terminal kinase, extracellular signal-regulated kinase 1/2, and p38) signaling. In conclusion, COR restored the altered serum biochemical parameters in aged rats and ameliorated the aging-associated testicular inflammation proving the therapeutic benefits of COR targeting inflammaging-associated male sexual dysfunctions.

This study aimed to evaluate the effects of Limosilactobacillus fermentum and Lactiplantibacillus plantarum isolated from human feces coordinating with inulin on the composition of gut microbiota and metabolic profiles in db/db mice. These supplements were administered to db/db mice for 12 weeks. The results showed that the Lactobacillaceae coordinating with inulin group (LI) exhibited lower fasting blood glucose levels than the model control group (MC). Additionally, LI was found to enhance colon tissue and increase the levels of short-chain fatty acids. 16S rRNA sequencing revealed that the abundance of Corynebacterium and Proteus, which were significantly increased in the MC group compared with NC group, were significantly decreased by the treatment of LI that also restored the key genera of the Lachnospiraceae_NK4A136_group, Lachnoclostridium, Ruminococcus_gnavus_group, Desulfovibrio, and Lachnospiraceae_UCG-006. Untargeted metabolomics analysis showed that lotaustralin, 5-hydroxyindoleacetic acid, and 13(S)-HpODE were increased while L-phenylalanine and L-tryptophan were decreased in the MC group compared with the NC group. However, the intervention of LI reversed the levels of these metabolites in the intestine. Correlation analysis revealed that Lachnoclostridium and Ruminococcus_gnavus_group were negatively correlated with 5-hydroxyindoleacetic acid and 13(S)-HpODE, but positively correlated with L-tryptophan. 13(S)-HpODE was involved in the "linoleic acid metabolism". L-tryptophan and 5-hydroxyindoleacetic acid were involved in "tryptophan metabolism" and "serotonergic synapse". These findings suggest that LI may alleviate type 2 diabetes symptoms by modulating the abundance of Ruminococcus_gnavus_group and Lachnoclostridium to regulate the pathways of "linoleic acid metabolism", "serotonergic synapse", and" tryptophan metabolism". Our results provide new insights into prevention and treatment of type 2 diabetes.
BACKGROUD/OBEJECTIVES: The mechanism of how black garlic effects lipid metabolism remains unsolved. Therefore, the objectives of this study were to determine the effects of black garlic on lipid profiles and the expression of related genes in rats fed a high fat diet. MATERIALS/METHODS: Thirty-two male Sqrague-Dawley rats aged 4 weeks were randomly divided into four groups (n=8) and fed the following diets for 5 weeks: normal food diet, (NF); a high-fat diet (HF); and a high-fat diet + 0.5% or 1.5% black garlic extract (HFBG0.5 or HFBG1.5). Body weights and blood biochemical parameters, including lipid profiles, and expressions of genes related to lipid metabolism were determined. RESULTS: Significant differences were observed in the final weights between the HFBG1.5 and HF groups. All blood biochemical parameters measured in the HFBG1.5 group showed significantly lower values than those in the HF group. Significant improvements of the plasama lipid profiles as well as fecal excretions of total lipids and triglyceride (TG) were also observed in the HFBG1.5 group, when compared to the HF diet group. There were significant differences in the levels of mRNA of sterol regulatory element binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and glucose-6-phosphate dehydrogenase (G6PDH) in the HFBG1.5 group compared to the HF group. In addition, the hepatic expression of (HMG-CoA) reductase and Acyl-CoA cholesterol acyltransferase (ACAT) mRNA was also significantly lower than the HF group. CONCLUSIONS: Consumption of black garlic extract lowers SREBP-1C mRNA expression, which causes downregulation of lipid and cholestrol metahbolism. As a result, the blood levels of total lipids, TG, and cholesterol were decreased.
Paeoniflorin (PAE) is the most abundant compound in Xuebijing injection widely used to treat sepsis. We aimed to investigate effect of PAE on expression of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) in a rat model of sepsis. Wistar rats were divided into Normal, Model, and PAE groups (n=20 each). Endotoxin was administrated at 5 mg/ml/kg in Model and PAE rats to establish rat sepsis model. 1 h after endotoxin administration, PAE was administrated at 4 ml/kg in PAE group once per day for 3 days. Routine blood tests and biochemical indexes were assessed, including aspartate aminotransferase (AST) and creatine kinase-MB (CK-MB). The plasma sTREM-1 level was measured using quantitative ELISA. At the end of experiment, the small intestine, liver, kidney and lung were subjected to pathological examinations. A rat model of sepsis-induced multiple organ dysfunction syndrome (MODS) was established successfully with endotoxin administration (5 mg/ml/kg), evidenced by histo-pathological examinations, routine blood tests and biochemical indexes: platelet count decreased and white blood cell count increased (p<0.05), CK-MB and AST increased (p<0.05). PAE treatment significantly reduced the plasma levels of AST, CK-MB, and sTREM-1, compared to Model group (p<0.05). Meanwhile, sepsis-induced damages in the liver, lung, stomach and intestinal mucosa were also markedly ameliorated by PAE treatment. PAE demonstrated a significantly protective effect in a rat model of sepsis by decreasing plasma sTREM-1 level, reducing inflammation, preventing MODS and protecting organ functions.
Purpose: The purpose of this study was to investigate the comparison of blood biochemical parameters, physical activity, stress, and sleep management between obese and non-obese subjects according to the body mass index (BMI). Methods: Data were collected from February 1 to May 31 in 2012. This research involved 403 subjects who visited an obesity clinic in Korea. They were divided into 4 groups: normal weight 100 ($18.5{\leq}BMI$ < $23.0kg/m^2$), overweight 100 ($23.0{\leq}BMI$ < $25.0kg/m^2$), obese 100 ($25.0{\leq}BMI$ < $30.0kg/m^2$), and severely obese 103 ($BMI{\geq}30.0kg/m^2$). Data were analyzed using SPSS version 19.0. Results: There were significant differences on the SGPT (F=22.98, p<.001), SGOT (F=12.91, p<.001), creatinine (F=4.82, p=.003), triglyceride (F=20.17, p<.001), and total cholesterol (F=6.55, p<.001) among the subjects according to the BMI. There was no significant association between frequency of physical activity according to the BMI. There was significant association regarding the management of sleep (F=5.13, p=.002), but no significant association regarding the management of stress. Overall, there were significant association regarding the management of stress and sleep (F=3.79, p=.011) among the groups. Conclusion: The result of this study suggests that it is possible to use the information as basic data for educational and nursing intervention programs based on the BMI.
Objective: The study was conducted to assess biochemical profiles in premenopausal and postmenopausal women having breast cancer. Materials and Methods: A hospital based case control study was carried out at Manipal Teaching Hospital (MTH), Pokhara, Nepal. The analysed variables were age, metabolic profile including total cholesterol, triglycerides, HDL-C, LDL-C, blood sugar, insulin concentration, C-peptide, HbA1c and selenium. Descriptive statistics and testing of hypothesis were used for the analysis using EPI INFO and SPSS 16 software. Results: In premenopausal women, significant differences were noted for total cholesterol (P value <0.001), triglycerides (P value 0.002), HbA1c level (P value <0.001), insulin concentration (P value 0.030), C-peptide concentration (P value 0.001), and selenium (P value <0.001) between cases and controls. Insignificant results were found for HDL-C (P value 0.749), LDL-C (P value 0.933), blood sugar (P value 0.59) and BMI (P value 0.746). Similarly, significant difference in total cholesterol (P value <0.001), triglycerides (P value 0.001), LDL-C (P value <0.001), HDL-C (P value 0.025), blood sugar (P value <0.001), insulin concentration (P value <0.001), c-peptide concentration (P value <0.001), HbA1c level (P value <0.001) and selenium (P value <0.001) were observed for postmenopausal patients and controls. Conclusions: Assessing metabolic changes and their management may be important for control of breast cancer and increased survival.
This study evaluated the effects of carnosine supplementation on carnosine concentration in muscles and blood biochemical indices of rats. Thirty-two eight-week-old Sprague-Dawley male rats were randomly divided into a control group (CON) as well as three carnosine-treated groups. The carnosine-treated groups included groups fed diets composed of 0.01% carnosine (LC), 0.1% carnosine (MC), and 1.0% carnosine (HC). Body weight gain, food intake, feed efficacy rate, protein efficacy rate, and organ weights were not significantly different among the groups. In all groups, the mean carnosine levels in gastrocnemius muscles were higher than the mean carnosine levels in soleus muscles. Carnosine concentrations in soleus muscles and gastrocnemius muscles were significantly higher in the HC group compared to all other groups (p<0.05). Serum triglyceride and LDL-cholesterol concentrations in all of the carnosine-supplemented groups were significantly lower than those of the control group (p<0.05), while HDL-cholesterol levels were significantly higher than those of the control group (p<0.05). Aspartate aminotransferase levels in rats supplemented with carnosine were significantly higher than those of the control group. In conclusion, diets supplemented with high levels of carnosine can increase carnosine concentrations in skeletal muscles, which might contribute to increased exercise capacity. Furthermore, these findings suggest that high levels of dietary carnosine improve the lipid profile of rats by lowering blood LDL-cholesterol and increasing HDL-cholesterol levels.
