  • The effectiveness of bronopol and bithionol in treating Ichthyobodo sp. infestation in cultured olive flounder was investigated, and compared them with formalin. Bath with formalin at 200 ppm or bithionol at 100 ppm for 1 h showed complete removal of Ichthyobodo sp. from all the experimental fish. Baths with lower concentrations of formalin (50 and 100 ppm) or bithionol (25 and 50 ppm) significantly reduced infection intensities of Ichthyobodo sp. On the other hand, baths with bronopol or intubation of bithionol at various concentrations were not effective against Ichthyobodo sp. infestation.

  • Hexachlorophene, Bithionol and Dithiazanine iodide were used to examine the vermicidal effect on Fasciola hepatica in vitro. A kymographic study was performed to investigate the motility of Fasciola hepatica under the influence of these drugs. Following conclusions were made: 1) With 10,000 : 1 solution; exhibited the vermicidal effect on Fasciola hepatica, Hexachlorophene in 5 minutes, Bithionol in 10 minutes, and Dithiazanine, iodide in 40 minutes, respectively. 2) With 100,000 : 1 solution; exhibited the vermicidal effect on Fasciola hepatica, Hexachlorophene in 20 minutes and Bithionol in 30minutes. In the case of Dithiazanine iodide the stimulation upon the fluke continued for 120 minutes. 3) With 1,000,000 : 1 solution; exhibited the vermicidal effect on Fasciola hepatica, Hexachlorophene in 50 minutes and Bithionol in 80 minutes. In the case of Dithiazanine iodide the stimulation upon the fluke continued for 120 minutes. 4) With 10,000,000 : 1 solution; the only Hexachlorophene showed the vermicidal effect on Fasciola hepatica. In the case of Bithionol and Dithiazanine iodide the slight stimulation upon the fluke continued for 120 minutes.

  • The piperazine salts with 24 kinds of phenols and 6 kinds of organic carboxylic acids were prepared. The test of anthelmintic effect against Ascaris suilla, in vitro, for Hexachlorophene-piperazine salt, Bithionol-piperazine salt, Kainic acid-piperazine salt were also carried out compared with piperazine, Hexachlorophene, Bithionol and Kainic acid.

  • Four drugs, bithionol, hexachlorophee, hexachloroethane and tetrachloroethylene, were tested on the in vitro effects of vermicidal and laying capacity inhibitory action to the Eurytrema Pancreaticum of Korean cattle, and the results obtained were as follows : 1) Of the four drugs, bithionol was most effective at both high and low concentrations. 2) The effects of hexachlorophene, hexachloroethane and tetrachloroethylene were less effective than that of bithionol, and the laying activity, at low concentration, was observed to increase at the early stage and decrease gradually thereafter. 3) Tetrachloroethylene was least effective of the four drugs.

  • This study was done to assess an alternative method as a replacement of in vivo phototoxicity test. The human fibroblasts were exposed to several phototoxic chemicals (promethazine, chlorpromazine chlortetracycline, 8-methoxypsoralen, neutral red, bithionol) and non-phototoxic materials (cinnamic aldehyde, p-aminobenzoic acid, sodium lauryl sulfate, L-cysteine). The cell viability was measured by neutral red uptake (NRU) assay. The results of the NRU phototoxicity (PT) assay showed a close agreement with in vivo test except bithionol. We also have tested the cosmetic ingredients including $Medimin^{(R)}$ A, $Medimin^{(R)}$ D, $LG^{(R)}$ 106W, $Phytoclear^{(R)}$ EL-1, Carex humilis L. extract, Canna indica L. extract, Salvia miltiorrhira Bunge extract, $Parsol^{(R)}$ MCX and $Parsol^{(R)}$ 1789. Most materials except Salvia miltiorrhira Bunge extract did not show any phototoxicity.

  • Aspidium ether extract was obtained from Dryopteris crassirhizoma(Aspidiaceae), which is widely distributed in Korea and its vermicidal effect on the rats infected with Hymenolepis diminuta was investigated. Effects of Aspidium ether extract purchased from the market, several fractions obtained from the seed of Torreya nucifora and bithionol were also compared. Vermicidal effect of Aspidium against Hymenolepis diminuta was found only in one-third of the rats used for investigation and the effect of the fresh Aspidium ether extract was similar to that of Aspidium ether extract purchased from market. However, it was observed that the whole seeds of Torreya nucifera (Torreyaceae) and its residue from which oil fraction was removed as well as bithionol were highly effective against Hymenolepis diminuta.

  • The crystal structure of Bithionol, $C_{12}H_6Cl_4O_2S$, has been determined from X-ray intensity data measured by Enraf-Nonius CAD-4 diffractometer using graphite-monochromatized $Mo-K\alpha$ radiation. The crystal data as follows; triclinic space group P{\bar{1}}$, a = 8.879(2), b = 10.782(1), c = 8.511(1)${\AA}$, ${\alpha}$ = 115.43(1), ${\beta}$ = 115.22(1), ${\gamma}\;=\;74.44^{\circ}(1)$. ${\mu}\;=\;9.51\;cm^{-1}$, F(000) = 356, Z = 2. Final R value is 0.036 for independent 2669 observed reflections. Each six-membered benzene rings are coplanar within experimesntal errors and the dihedral angle between these planes is $81.28^{\circ}$ (1). The S-(1) and S-C(7) distances are 1.787(2) and 1.791(3)${\AA}$, respectively.

Comparison of Sensitivity Between Balb/c 3T3 Cell and HaCaT Cell by NRU Assay to Predict Skin Phototoxicity Potential

  • In order to find out the appropriate in vitro method for high correlation with in vivo, we com-pared the sensitivities of phototoxicity (PT) in vitro method between in human keratinocytes, HaCaT cells and in 3T3 fibroblast cells derived from Balb/c mice. Both cells were exposed to six known phototoxic chemicals : promethazine, neutral red, chlortetracycline, amiodarone, bithionol, 8-methoxypsoralen, or non-phototoxic chemical, ALS (ammonium laureth sulfate) and then irradiated with 5 J/$cm^2$ of UVA. Cell viability ($IC_{50}$ ) was measured by neutral red uptake (NRU) assay. The ratio of $IC_{50}$ value of chemicals in the presence and absence of UVA was determined by the cut-off value. The phototoxic potential of test chemicals in NRU assay was determined by measuring the photoirriation factor (PIF) with a cut-off value of 5. In both 3T3 and HaCaT cells, all known phototoxic chemicals were positive (over 5 of PIF value), except that bithionol was found to be non-phototoxic to HaCaT cells, and ALS, non-phototoxic chemical was negative. These results suggest that Balb/c 3T3 cell was more sensitive than HaCaT cell to predict phototoxicity potential.

Studies on Eurytrema pancreaticum (V) Experiment for anthelmintic effect (췌질(膵蛭)에 관한 연구(硏究) (V) 구충제(驅蟲劑)에 대하여)

  • The anthelmentic effects of Fuadin (sodium antimony-III-bis-pyrocatechin-disulphonate), Bithionol 2-2-thiobis 4,6-dichlorophenol)), hexachlorethane and carbon tetrachloride were studied on Eurytrema pancreaticum infected in sheep and goats. The evaluation on the effects was based upon the egg reduction in the feces of the experimental animals administered the drugs. The number of eggs per 5gm. of the feces was respectively calculated before the treatments, and the follw-up for the egg reductions was carried out over a period of three or four weeks(calculated twice a week). Oral administration of Bithionol (once a day for 2 succeed days, at the rate of 75 mg per kg of bady weight) and of hexachlorethane (once a day, at the rate of 300ml per kg of body weight) did not reveal any egg reduction in sheep host. Intramuscular injection of carbon tetrachloride with olive oil (once a day, at the rate of 0.05ml per kg of body weight) did not show the egg reduction in goats. Intramuscular injections of Fuadin for 5 days (the dosage schedule was 0.5ml at 1st day, 1.0ml at 2nd and 3rd day, 1.5ml at 4th and 5th day, to amount of 5.5ml) were completely devoid the fluke eggs in the feces of goats on 4 weeks after each treatment. Throughout the studies, Fuadin was only proved its anthelmintic effect to the goats infected with E. pancreaticum. On the other hand, Bithional, hexachlorethane and carbon tetrachloride did not reveal any effect as Fuadin showed.

Five Cases of Diphyllobothrium latum Infection (광절열두조충감염 5예)

  • Five cases of Diphyllobothrium latum infection were proved by collection of worms after treatment with bithionol, niclosamide or praziquantel, during 1975~1983. The patients were 4 males and one female aged 10~64 years residing in Seoul or Wando-gun, Chollanam-do, Korea. All of them remembered the history of eating several kinds of raw marine fish and 2 persons said they ate the raw mesh of perch, Lateolabrax japcnicus. Three amcnE them experienced abdominal pain, 2 natural discharge of a chain of segments, but none revealed any sign of anemia. The average egg size in stool varied $59~67{\times}41~48{\mu\textrm{m}}$ according to cases. The eggs were characterised by ovoid to elliptical shape, terminal operculum, and distinct abopercular protuberance. One naturally discharged segment (30cm) and 4 complete strobilae (320~680cm) with scolices obtained after treatments were examined. The morphological characters of proglottids such as rosette-form uterus with 3~6 loops, vaginal pore included in the cirrus sac, separated seminal vesicle from cirrus sac, etc. were all compatible with D. latum. These are the 7th~11th cases of D. latum infection proved by worms in Korea.
