Use of Stable Isotope Probing in Selectively Isolating Target Microbial Community Genomes from Environmental Samples for Enhancing Resolution in Ecotoxicological Assessment

  • In this study we attempted to develop a novel genomic method to selectively isolate target functional microbial genomes from environmental samples. For this purpose, stable isotope probing (SIP) was applied in selectively isolating organic pollutant-assimilating populations. When soil microbes were fed with $^{13}C-labeled $ biphenyl, biphenyl-utilizing cells were incorporated with the heavy carbon isotope. The heavy DNA portion was successfully separated by CsCl equilibrium density gradient. And the diversity in the heavy DNA was sufficiently reduced, being suitable for the current DNA microarray techniques to detect biphenyl-utilizing populations in the soil. In addition, we proposed a new way to get more genetic information by combining this SIP method with selective metagenomic approach. The increased selective power of these new DNA isolation methods will be expected to provide a good quality of new genetic information, which, in turn, will result in development of a variety of biomarkers that may be used in assessing ecotoxicology issues including the impacts of organic hazards, and antibiotic-resistant pathogens on human and ecological systems.

Effect of Biphenyl Dimethyl Dicarboxylate on the Humoral Immunosuppression by Ketoconazole in Mice

  • The present study was undertaken to investigate the effect of biphenyl dimethyl idcarboxylate (PMC) on the humoral immunosuppression by ketoconazole (KCZ) in ICR mice. PMC at a dose of 6 mg/kg was administered orally to mice daily for 14 consecutive days. KCZ was suspended in RPMI 1640 medium and orally administered at 160 mg/kg/day 2 hrs after the administration of PMC. Mice were immunized an challenged with challenged red blood cells (SRBC). The results of the present study are summarized as follows; a gain of body weight and relative weights of spleen and liver were significantly increased by combination of PMC and KCZ, as compared with those in mice treated with KCZ alone. Splenic plaque forming cells (PFC) and hemagglutination (HA) titers to SRBC were greatly enhanced by the combination of PMC and KCZ, compared with treatment of KCZ alone. The elevation of serum glutamicpyruvic transminase (S-GPT) and total protein levels caused by KCZ were reduced to normal level by the combination of PMC and KCZ. In addition, lower serum albumin and A/G ratio were also increased to normal level. These findings indicate that PMC has a protective effect against KCZ-induced humoral immunosuppression.

고분자전해질 연료전지용 부분 불소계 설폰화 Poly(biphenylene-co-sulfone)ether 막의 제조와 특성 (Preparation and Characteristics of Partially Fluorinated-Sulfonated Poly(biphenylene-co-sulfone)ether Membranes for Polymer Electrolyte Membrane Fuel Cell)

  • 불소 관능기인 perfluorocyclobutane(PFCB)기를 함유하는 설폰화된 부분 불소계 poly(biphenyleneco-sulfone)ether 공중합체(SPBS-X)를 제조하였다. 제조과정은 trifluorovinylether그룹을 양 말단에 포함하는 단량체의 합성, 고리화부가 반응형태의 열중합 그리고 후설폰화(post-sulfonation)의 3단계 합성과정을 통하였다. Trifluorovinylether그룹을 양 말단에 포함하는 biphenyl계와 sulfonyl계 단량체를 합성하였고, biphenyl계의 함량을 20 mol%에서 60 mol%까지 조절하면서 고리화부가 반응형태의 열중합을 통해 랜덤 공중합체가 얻어졌다(PBS-X). 후설폰화는 chlorosulfonic acid(CSA)를 사용하여 수행되었으며, 이를 통해 다양한 설폰화도를 가지는 고분자를 합성하였다. $^1H$-NMR, $^{19}F$-NMR 및 FT-IR 분석을 통하여, 제조된 화합물, 단량체 및 중합체들이 성공적으로 합성되었음을 확인하였다. 상온에서 CSA에 대해 설폰화 활성을 가지는 biphenyl기의 함량이 증가함에 따라 이온교환능력(IEC), 함수율 및 이온전도도가 증가하는 경향을 보였고, SPBS-60의 경우 넓은 온도범위(25~80 $^{\circ}C$)에서 Nafion$^{(R)}$-115보다 높은 이온전도도(80 $^{\circ}C$: 0.08 S/cm at RH 100%)를 나타내었다.

  • 본 연구에서는 자기장 하에서 증착 후 열처리된 NPD (4,4'-bis-[N-(1-napthyl)-N-phenyl-amino]biphenyl)박막의 토폴로지와 분자배열을 관찰하였다. NPD는 진공에서 열 증발법을 통하여 증착되었다. 분자 배열이 잘 되어진 유기/금속필름은 2전류밀도와 발광효율 같은 소자의 특성을 향상시키는 것이 특히 중요하다. 원자탐침현미경(AFM) 및 X선 회절 분석기(XRD)의 분석결과는 토폴로지와 NPD필름의 구조적 배열을 특성화하는데 사용되었다. 멀티소스미터는 ITO/NPD/Al 소자의 전류-전압 특성을 측정하는데 사용되었다. XRD 결과에 따르면 자기장 하에서 증착된 NPD 박막은 분자배열이 관찰되지 않았으나, $130^{\circ}C$에서 후(後)열처리한 NPD 박막에서는 고른 분자배열을 확인할 수 있었다. AFM 이미지에 따르면, 자기장 하에서 증착된 NPD 박막은 자기장 없이 증착된 박막보다 더 매끄러운 표면을 가졌다. NPD의 전류-전압 특성은 고른 분자 배열을 가진 NPD 필름의 더 높아진 전자이동도로 인해 향상되었다.
